Relationship between autotomy behaviour and spinal cord monoaminergic levels in rats.

In the rat, unilateral neurectomy of the sciatic and saphenous nerves causes autotomy, a self-mutilation behaviour, against the denervated limb that is variable in both its onset and severity. To study some of the possible neurochemical sources of this variability, spinal cord levels of norepinephrine (NE), dopamine (DA), serotonin (5-HT0 and 5-hydroxyindoleacetic acid (5-HIAA) were analysed ipsi- and contralateral to the lesioned side by high performance liquid chromatography at C5-T1 and L1-S1. According to the early or late onset and to the slight or intense autotomy behaviour, the animals

[Behavioral teratogenesis and placental, milk, and blood-brain barrier transfer following prenatal and postnatal exposures to chlorpromazine in rats].

In order to determine behavioral teratogenesis of fetal and neonatal exposures to chlorpromazine (CPZ) in rats, conditioned avoidance learning by shuttle box in pups was examined. CPZ was administered sc to pregnant and nursing rats during fetal organogenesis period (on days 11-15 of gestation, at doses of 9, 21 mg/kg/day), perinatal period (on days 16-20 of gestation, at 9, 18 mg/kg/day), and nursing period (on days 1-10 postpartum, at 9, 18 mg/kg/day). The results showed that high doses of CPZ during all periods caused learning impairment in pups, although learning impairment by the exposure

Distribution of sympathetic preganglionic neurons and monoaminergic nerve terminals in the spinal cord of the rat.

A study was made of the distribution of sympathetic preganglionic neurons identified by retrograde labeling with horseradish peroxidase from various peripheral nerve trunks and of the distributions of monoaminergic terminals in the spinal cord of the rat. Nerve terminals were stained immunohistochemically by using antisera raised against tyrosine hydroxylase, phenylethanolamine-N-methyl-transferase, neuropeptide Y, and 5-hydroxytryptamine and by using formaldehyde-induced fluorescence. The three-dimensional distribution of sympathetic preganglionic neurons was described by using computer recon

Diazepam, a positive allosteric modulator of the GABAA receptor, induces strain-specific responses in object recognition, ultrasonic vocalizations and monoaminergic profiles in the Wistar Kyoto rat model of endogenous depression.

Major depressive disorder (MDD) involves cognitive impairment, altered affective states and dysregulation of neurotransmitter homeostasis. The Wistar Kyoto (WKY) rat is characterized by inherent depressive-like behaviour, anxiety, cognitive deficits and dysregulated monoaminergic/GABAergic neurotransmission. This study investigated strain-specific differences in learning and memory, affective states and neurochemical responses to Diazepam (DZP), a positive allosteric modulator of the GABAA receptor, in Wistar and WKY rats. Learning and memory were assessed using the novel object recognition (N

Brain monoamine changes modulate the corticotropin-releasing hormone receptor 1-mediated behavioural response to acute thermal stress in zebrafish larvae.

While central monoamines play a role in regulating stress-related locomotory activity, the modulation of monoamines by the corticosteroid stress axis in shaping acute behavioural responses are unclear. We investigated whether the corticotropin-releasing hormone receptor 1 (Crhr1) modulation of stress-related behavioral response involves monoamine regulation by subjecting Crhr1 knockout (crhr1-/-) zebrafish (Danio rerio) to an acute thermal stressor (TS: +5 °C above ambient for 60 min). The TS-induced cortisol response and hyper locomotory activity in the WT larvae was abolished in fish lacking

BEHAVIORAL AND NEUROCHEMICAL CHANGES DURING INTRANASAL ADMINISTRATION OF ALPHA-GLUTAMYL-TRYPTOPHAN AND CHELATE COMPLEX OF ZINC ARGINYL-GLYCINATE ON MONOAMINE SYSTEMS DYSFUNCTIONS KNOCK-OUT MODELS.

UNLABELLED: Monoamine neurotransmitter system dysfunctions lead to behavioral disorders, cognitive metabolic, and other pathological conditions. In this case, different amino acids are precursors of monoamines, while the parenteral path of monoamine administration has pharmacological restrictions. Therefore, intranasal administration one of the most promising methods of delivering an active substance is. The purpose of the work is to study the effect of intranasal administration of a chelate complex of zinc arginyl-glycinate and alpha-glutamyl-tryptophan dipeptide on behavioral and neurochemic

Mu opioid receptor knockout mouse: Phenotypes with implications on restless legs syndrome.

Restless legs syndrome (RLS) is characterized by an irresistible need to move the legs while sitting or lying at night with insomnia as a frequent consequence. Human RLS has been associated with abnormalities in the endogenous opioid system, the dopaminergic system, the iron regulatory system, anemia, and inflammatory and auto-immune disorders. Our previous work indicates that mice lacking all three subtypes of opioid receptors have a phenotype similar to that of human RLS. To study the roles of each opioid receptor subtype in RLS, we first used mu opioid receptor knockout (MOR KO) mice based

Modulation of Monoaminergic Systems by Antidepressants in the Frontal Cortex of Rats After Chronic Mild Stress Exposure.

The standard pharmacological treatment of the major depressive disorder (MDD) is still grounded in a monoaminergic approach. Consequently, antidepressant treatments pursue to heighten serotonergic and noradrenergic neurotransmissions. Thus, the aim of this study was to assess the impact of exposure to a well-characterized animal model, the chronic mild stress (CMS) on serotonin (5-HT) and noradrenaline (NE) levels, and reuptake transporters and receptors in the frontal cortex (FC) of CMS-exposed rats. Moreover, considering the diverse pharmacological profiles of existing antidepressants and th

Alpha Adrenergic Induction of Transport of Lysosomal Enzyme across the Blood-Brain Barrier.

The impermeability of the adult blood-brain barrier (BBB) to lysosomal enzymes impedes the ability to treat the central nervous system manifestations of lysosomal storage diseases. Here, we found that simultaneous stimulation of the alpha1 and alpha2 adrenoreceptor restores in adult mice the high rate of transport for the lysosomal enzyme P-GUS that is seen in neonates but lost with development. Beta adrenergics, other monoamines, and acetylcholine did not restore this transport. A high dose (500 microg/mouse) of clonidine, a strong alpha2 and weak alpha1 agonist, was able to act as monotherap

The metabotropic glutamate receptor 7 allosteric modulator AMN082: a monoaminergic agent in disguise?

Metabotropic glutamate receptor 7 (mGluR7) remains the most elusive of the eight known mGluRs primarily because of the limited availability of tool compounds to interrogate its potential therapeutic utility. The discovery of N,N'-dibenzhydrylethane-1,2-diamine dihydrochloride (AMN082) as the first orally active, brain-penetrable, mGluR7-selective allosteric agonist by Mitsukawa and colleagues (Proc Natl Acad Sci USA 102:18712-18717, 2005) provides a means to investigate this receptor system directly. AMN082 demonstrates mGluR7 agonist activity in vitro and interestingly has a behavioral profil

Love and attachment: the psychobiology of social bonding.

Basic animal studies and human imaging studies have contributed to our understanding of the psychobiology of love and attachment. There are overlaps and distinctions in the neuronal circuitry of maternal love, romantic love, and long-term attachment. In these circuits, important molecules, which have been demonstrated to play a role in the psychobiology of social bonding include dopamine, serotonin, opioids, oxytocin, and vasopressin. Particular genetic and environmental variations contribute to social-bonding phenotypes, consistent with an evolutionary perspective on the value of these behavi

Changes in behavior and monoamine levels in microdialysate from dorsal striatum after 6-OHDA infusions into ventral striatum.

Long-Evans rats received bilateral 6-hydroxydopamine infusions into the nucleus accumbens and were tested immediately (1 and 2 days) or after a recovery period (14 and 15 days) for changes in extracellular levels of dorsal striatal monoamines using in vivo microdialysis. Compared to controls, the monoamine metabolites 3,4-dihydroxyphenylacetic acid, homovanillic acid and 5-hydroxyindoleacetic acid were generally enhanced when tested immediately after 6-hydroxydopamine treatment, including spontaneous levels and those following depolarizing infusions of potassium (60 mM, 20 min) through the mic

Olfactory dysfunction, central cholinergic integrity and cognitive impairment in Parkinson's disease.

Olfactory dysfunction is common in subjects with Parkinson's disease. The pathophysiology of such dysfunction, however, remains poorly understood. Neurodegeneration within central regions involved in odour perception may contribute to olfactory dysfunction in Parkinson's disease. Central cholinergic deficits occur in Parkinson's disease and cholinergic neurons innervate regions, such as the limbic archicortex, involved in odour perception. We investigated the relationship between performance on an odour identification task and forebrain cholinergic denervation in Parkinson's disease subjects w

[Narcolepsy].

First described as a separate entity by Gelineau in 1880 and later considered as a symptom, narcolepsy has eventually been recognized as a disease on clinical and polygraphic grounds. Its prevalence stays between 0.04 and 0.06 percent. Age at onset varies from 5 to 50 with a peak in the second decade. Clinical symptoms include excessive daytime somnolence, overwhelming daytime sleep episodes, attacks of cataplexy, hypnagogic hallucinations, sleep paralysis and disturbed nocturnal sleep; sleep onset REM episodes are the main polygraphic feature. Natural history varies with the different symptom
