Nanostructure of mouse otoconia.

Mammalian otoconia of the inner ear vestibular apparatus are calcium carbonate-containing mineralized structures critical for maintaining balance and detecting linear acceleration. The mineral phase of otoconia is calcite, which coherently diffracts X-rays much like a single-crystal. Otoconia contain osteopontin (OPN), a mineral-binding protein influencing mineralization processes in bones, teeth and avian eggshells, for example, and in pathologic mineral deposits. Here we describe mineral nanostructure and the distribution of OPN in mouse otoconia. Scanning electron microscopy and atomic forc

Biomimetic oligopeptide formed enamel-like tissue and dentin tubule occlusion via mineralization for dentin hypersensitivity treatment.

OBJECTIVE: Dentin hypersensitivity (DH) is a common oral disease with approximately 41.9% prevalence. Reconstruction of dental hard tissues is the preferred treatment for relieving DH. Here, we applied biomineralization method using oligopeptide simulating cementum protein 1 (CEMP1) to regenerate hard tissues on demineralized dentin. METHODS: The self-assembly and biomineralization property of the oligopeptide were detected by scanning electron microscopy (SEM), circular dichroism spectroscopy, and transmission electron microscopy. Oligopeptide's binding capacity to demineralized dentin was e…

Identification of novel candidate genes involved in mineralization of dental enamel by genome-wide transcript profiling.

The gene repertoire regulating vertebrate biomineralization is poorly understood. Dental enamel, the most highly mineralized tissue in mammals, differs from other calcifying systems in that the formative cells (ameloblasts) lack remodeling activity and largely degrade and resorb the initial extracellular matrix. Enamel mineralization requires that ameloblasts undergo a profound functional switch from matrix-secreting to maturational (calcium transport, protein resorption) roles as mineralization progresses. During the maturation stage, extracellular pH decreases markedly, placing high demands …

Purification, characterization, and in vitro mineralization studies of a novel goose eggshell matrix protein, ansocalcin.

Biomineralization is an important process in which hard tissues are generated through mineral deposition, often assisted by biomacromolecules. Eggshells, because of their rapid formation via mineralization, are chosen as a model for understanding the fundamentals of biomineralization. This report discusses purification and characterization of various proteins and peptides from goose eggshell matrix. A novel 15-kDa protein (ansocalcin) was extracted from the eggshell matrix, purified, and identified and its role in mineralization evaluated using in vitro crystal growth experiments. The complete…

Radiolyo-chemiluminescence of bones and seafood shells - a new, promising method for the detection of food irradiation?

Biomineral substances are dissolved in an alkaline mixture of luminol solution and EDTA. Light emission is observed upon dissolution of irradiated calcium carbonate in mollusk and crustacean shells and calcium hydroxyapatite in bones, showing a significantly increased chemiluminescence compared to unirradiated samples. This enables the determination of exposure to ionizing radiation and in some cases even dosimetry. The method described is simple in sample preparation and fast for the detection of irradiated seafood and poultry.

Cave microorganisms: hidden players in global greenhouse gas cycling and climate regulation.

Caves are unique among ecosystem types because of their physical structures and biological functions. Embedded in rocks, the geological setting defines the boundaries of caves and dictates how energy and matter move through them. General features of caves, compared to surface ecosystems, include absence of light, relatively stable temperature and humidity, and oligotrophic conditions. Despite these conditions, caves are highly diverse ecosystems whose environmental properties are shaped by geological, hydrological, and climatic factors. Cave microbiomes metabolize atmospheric trace gases, such

Arbuscular mycorrhizal fungi influence the speciation and subcellular abundance of uranium in plant roots.

Uranium (U) is a natural radioactive metal and a persistent environmental pollutant. Characterising the influence of arbuscular mycorrhizal fungi (AMF) on U bioaccumulation and partitioning in plants is crucial to understand U soil-to-plant transfer mechanisms. High resolution elemental mapping, spectroscopy and microscopy techniques were conducted on uranyl nitrate dosed Plantago lanceolata roots colonised with Rhizophagus irregularis. U-rich particles accumulated within the root cells, with higher abundance in epidermal and outer cortex cells of mycorrhizal root samples than in non-mycorrhiz

Spectral Detection of Nanophase Iron Minerals Produced by Fe(III)-Reducing Hyperthermophilic Crenarchaea.

Mineral transformations by two hyperthermophilic Fe(III)-reducing crenarchaea, Pyrodictium delaneyi and Pyrobaculum islandicum, were examined using synthetic nanophase ferrihydrite, lepidocrocite, and akaganeite separately as terminal electron acceptors and compared with abiotic mineral transformations under similar conditions. Spectral analyses using visible-near-infrared, Fourier-transform infrared attenuated total reflectance (FTIR-ATR), Raman, and Mössbauer spectroscopies were complementary and revealed formation of various biomineral assemblages distinguishable from abiotic phases. Carbon and steel surfaces modified by Leptothrix discophora SP-6: characterization and implications.

Leptothrix discophora SP-6, a type of manganese(Mn)-oxidizing bacteria, has been known to accumulate Mn oxides from the aqueous environment … ells featuring a biocathode. In this preliminary study, factors affecting the biofilm formation and biomineralization processes were examined. The inflow of air into the culture medium was found essential to sustain the ennoblement of substrate electrodes. The OCP and FESEM/EDS data indicated that a smoother initial substrate surface generally led to better ennoble …

Essential magnetosome proteins MamI and MamL from magnetotactic bacteria interact in mammalian cells.

To detect cellular activities deep within the body using magnetic resonance platforms, magnetosomes are the ideal model of genetically-encoded nanoparticles. These membrane-bound iron biominerals produced by magnetotactic bacteria are highly regulated by approximately 30 genes; however, the number of magnetosome genes that are essential and/or constitute the root structure upon which biominerals form is largely undefined. To examine the possibility that key magnetosome genes may interact in a foreign environment, we expressed mamI and mamL as fluorescent fusion proteins in mammalian cells. Loc

Biomolecular Minerals and Volcanic Glass Bio-Mimics to Control Adult Sand Flies, the Vector of Human Leishmania Protozoan Parasites.

Sand flies (Diptera: Psychodidae) serve as vectors for transmitting protozoan parasites, Leishmania spp., that cause the disease called leishmaniasis. The main approach to controlling sand flies is the use of chemical insecticides. The discovery of alternative methods for their control is needed because of potential health risks of chemical insecticides and development of sand fly resistance to these pesticides. The biomineral produced by diatoms (diatomaceous earth, DE; Celite) and a volcanic glass bio-mimic (Imergard) have been shown by our group to be efficacious against mosquitoes, filth f

Graphite accelerate dissimilatory iron reduction and vivianite crystal enlargement.

Biomineralized vivianite induced by dissimilatory iron reduction bacteria (DIRB) has received increasing attention because it alleviates phosphorus crisis and phosphorus pollution simultaneously. However, the relatively small crystal size and low Fe(III) reduction rate restrict the separation and recovery of vivianite. In this study, graphite was selected as additive to enhance vivianite biomineralization with soluble ferric citrate and insoluble hematite as two representative electron acceptors. As soluble ferric citrate provided abundant accessible electron acceptors, relatively inconspicuou…

DMP1-derived peptides promote remineralization of human dentin.

Remineralization of dentin during dental caries is of considerable clinical interest. Dentin matrix protein 1 (DMP1) is a non-collagenous calcium-binding protein that plays a critical role in biomineralization. In the present study, we tested if peptides derived from DMP1 can be used for dentin remineralization. Peptide pA (pA, MW = 1.726 kDa) and peptide pB (pB, MW = 2.185), containing common collagen-binding domains and unique calcium-binding domains, were synthesized by solid-phase chemistry. An extreme caries lesion scenario was created by collagenase digestion, and the biomineral-nucleati…

Differential impacts of ocean acidification and alkalinization on shell microstructure and molecular responses in Mytilus edulis.

Anthropogenic CO2 emissions are intensifying ocean acidification (OA), disrupting carbonate chemistry and threatening marine calcifiers such … hanced shell integrity and stimulated growth-associated processes. Across treatments, a core set of biomineralization-related genes (e.g., VWA7, CA14, ALPL) exhibited expression shifts, suggesting central roles in carbonate homeostasis. In contrast, differential regulations of genes such as CA10 and VWDE revealed pH-specific responses. Notably, OAE induced minimal …

The Synthetic Collagen-Binding Peptide NIPEP-OSS Delays Mouse Myeloma Progression.

Multiple myeloma (MM) is the second most common hematological malignancy. It is a clonal B-cell disorder characterized by the proliferation of malignant plasma cells in the bone marrow, the presence of monoclonal serum immunoglobulin, and osteolytic lesions. An increasing amount of evidence shows that the interactions of MM cells and the bone microenvironment play a significant role, suggesting that these interactions may be good targets for therapy. The osteopontin-derived collagen-binding motif-bearing peptide NIPEP-OSS stimulates biomineralization and enhances bone remodeling dynamics. Due …
