Functionalized Coacervates for Cancer Therapy: Advances and Prospects in Phase-Separation-Based Intelligent Drug Delivery.

Functionalized coacervates, a type of nanomaterial inspired by biomolecular condensation, have emerged as an important research direction in biomedicine, particularly in cancer therapy. These membraneless structures formed by liquid-liquid phase separation (LLPS) self-assembly have high drug-loading capacity, favorable biocompatibility, and tunable responsiveness to tumor microenvironment (TME) cues. This programmability enables targeted delivery and controlled release. This review summarizes the latest progress in the design and therapeutic application of functionalized coacervates in cancer

Electrochemical Biosensing Strategies for Oral Health: From Engineered Interfaces to Advanced Transistor Architectures.

Oral diseases represent a major global health burden, underscoring the need for sensitive, accessible, and noninvasive diagnostic technologies. Electrochemical biosensing offers a powerful route for point-of-care oral health monitoring by translating biomolecular interactions in saliva, gingival crevicular fluid, and exhaled breath condensate into quantifiable electrical signals. This review systematically discusses electrochemical biosensing strategies for oral disease diagnosis, beginning with the structure and operating mechanisms of electrochemical sensors and then summarizing their applic

Micropipette aspiration reveals differential RNA-dependent viscoelasticity of nucleolar subcompartments.

The nucleolus is a multiphasic biomolecular condensate that facilitates ribosome biogenesis, a complex process involving hundreds of proteins and RNAs. The proper execution of ribosome biogenesis likely depends on the material properties of the nucleolus. However, these material properties remain poorly understood due to the challenges of in vivo measurements. Here, we use micropipette aspiration (MPA) to directly characterize the viscoelasticity and interfacial tensions of nucleoli within transcriptionally active Xenopus laevis oocytes. We examine the major nucleolar subphases, the outer gran

Distinguishing near- versus off-critical phase behaviors of intrinsically disordered proteins.

Intrinsically disordered prion-like low complexity domains (PLCDs) drive phase transitions that underlie the biogenesis of many biomolecular condensates. Here, we report results from large-scale Monte Carlo simulations on lattices aided by computations of Binder cumulants and rigorous finite-size scaling. These approaches enable accurate mapping of the critical regime and computations of the full binodal of an archetypal PLCD. This weakly associating polymer undergoes phase separation coupled to percolation. Between the lowest temperature and the critical point, the concentrations along the le…

Small GTPase RAN-driven PNET2 oligomerization and phase separation at the nuclear lamina promote nuclear envelope integrity in plants.

The nuclear envelope is a fundamental organizer of eukaryotic cells, yet how plants regulate its architecture and integrity remains poorly understood. In this study, we identified the plant inner nuclear membrane protein PLANT NUCLEAR ENVELOPE TRANSMEMBRANE 2 (PNET2) as a scaffold that maintains nuclear envelope integrity and genome stability. Loss of PNET2 function compromises nuclear membrane structure and sensitizes cells to DNA damage, whereas overexpression drives aberrant nuclear membrane expansion. Biochemically, PNET2 cooperates with the nuclear lamin protein KAKU4 and CROWDED NUCLEI 1

Combined ab initio/empirical approach for optimization of Lennard-Jones parameters for polar-neutral compounds.

The study of small functionalized organic molecules in aqueous solution is a useful step toward gaining a basic understanding of the behavior of biomolecular systems in their native aqueous environment. Interest in studying amines and fluorine-substituted compounds has risen from their intrinsic physicochemical properties and their prevalence in biological and pharmaceutical compounds. In the present study, a previously developed approach which optimizes Lennard-Jones (LJ) parameters via the use of rare gas atoms combined with the reproduction of experimental condensed phase properties was ext

Emergent 3D genome reorganization and graded gene control from the stepwise assembly of transcriptional condensates.

Transcriptional condensates are clusters of transcription factors, coactivators, and RNA Pol II associated with gene activation, yet how they assemble and function within the cell remains unclear. Here, we show that transcriptional condensates form in a stepwise manner to enable both graded and three-dimensional (3D) gene control in the yeast heat shock response (HSR). First, the transcription factor Hsf1 (heat shock factor 1) clusters upon partial dissociation from the chaperone Hsp70. Next, the coactivator Mediator partitions following further Hsp70 dissociation and Hsf1 phosphorylation. Fin

Molecular Motion of Viscous Solvents from Solid-State NMR.

Liquid-liquid phase separation (LLPS) is a ubiquitous phenomenon in biological systems and involves molecular motions across multiple time scales, especially in viscous, crowded environments. These dynamics include not only biomacromolecules but also small solutes, ions, and water. To establish an NMR framework for characterizing such heterogeneous dynamic regimes, we employ two homogeneous viscous model systems, glycerol and PEG400, which reproduce the slowed molecular motions characteristic of condensed biomolecular environments while avoiding the structural complexity of phase-separated sys

Biofunctionalized zinc nanoparticles from Alangium salvifolium seeds extract: a potent antibacterial and anti-Cancer therapeutic agent.

The rise of multidrug-resistant (MDR) pathogens and the limitations of traditional therapies have prompted the discovery of biocompatible nanomaterials. In this study, green, one-pot synthesis of zinc oxide nanoparticles (AS-ZnO-NPs) was performed using Alangium salvifolium seed extract (aqueous) as both reducing and stabilizing agent.Nanoparticles were characterized physicochemically, showing an absorption peak at 387 nm by UV-visible spectroscopy. Dynamic light scattering produced a hydrodynamic diameter of 53.28 nm, which is consistent with a biomolecular capping layer, while transmission e

Structurally characterized Ni(II) complex exhibiting potent anti-Candida activity: biomolecular interaction mechanism and in silico ADMET insights.

The current study reports the synthesis, structural interpretation, biomolecular interaction, and antifungal appraisal of a Ni(II) Schiff base complex demonstrating effective activity against Candida albicans. The complex, formulated as [Ni(L)2], features a NiN4O2 coordination environment and was synthesized using the ligand L, [4-bromo-2-((3-(methylamino)propylimino)methyl)phenol], derived from the condensation of 5-bromo-2-hydroxybenzaldehyde with N-methylpropane-1,3-diamine, followed by complexation with nickel(II) chloride in methanol. The complex was comprehensively characterized by FTIR,

Targeting autophagy in oral squamous cell carcinoma chemoresistance: molecular mechanisms, therapeutic strategies, and emerging nanotherapeutic approaches.

Autophagy is a lysosome-dependent recycling process that maintains cellular homeostasis and helps cells adapt to therapeutic stress. In oral squamous cell carcinoma (OSCC), dysregulated autophagy may promote chemoresistance by supporting metabolic adaptation, removing damaged cellular components, and limiting treatment-induced cell death. Its effects are nevertheless context dependent, as autophagy can also interact with apoptosis, ferroptosis, and other cytotoxic pathways. This review summarizes molecular mechanisms linking autophagy to OSCC chemoresistance, focusing on non-coding RNAs, p53/T

Stress-driven emergence of heritable non-genetic drug resistance.

Drug resistance is the chief cause of treatment failure for therapies targeting chronic and infectious diseases. Whether the emergence of resistance is accelerated by environmental exposure to low levels of therapeutics remains controversial. Here, we report a non-genetic mechanism of stress adaptation that promotes heritable resistance to the widely used antifungal drug fluconazole. In the human fungal pathogen Candida albicans, transient exposure to subtherapeutic fluconazole doses induces a protective response that we term para-resistance. Like conventional resistance mechanisms, para-resis

Synthesis of cyclic oligomeric squaramides for oral protein delivery to treat inflammatory bowel disease.

Cyclic squaramide oligomers are attractive supramolecular building blocks, but their use as biomolecular delivery materials remains underexplored. Here, we report the one-step preparation of a low-molecular-weight cyclic oligomeric squaramides (COS) mixture by equimolar condensation of dimethyl squarate and 2,2'-diamino-N-methyldiethylamine. Reaction-condition studies and comparative DFT-refined scan calculations indicate that linker architecture and competing hydrogen-bonding interactions influence cyclic oligomer formation. In aqueous buffer, COS forms dynamic supramolecular assemblies and a

From RNA to DNA: How Cargo Identity Reprograms Lipid Nanoparticle Architecture and Function.

Lipid nanoparticles (LNPs) have become the leading platform for delivering genetic material, gaining global recognition through the success of mRNA-based COVID-19 vaccines such as mRNA-1273 (SpikeVax, Moderna) and BNT162b2 (Comirnaty, BioNTech/Pfizer). Yet, while RNA-LNPs have reached clinical maturity, their DNA counterparts remain comparatively underexplored, despite holding great promise for gene replacement and genome-editing therapies. In this review, we turn the spotlight on DNA-loaded LNPs, examining how their structure, composition, and biological behavior differ from RNA-LNPs, their n

Nucleolar stress controls mutant Huntington toxicity and monitors Huntington's disease progression.

Transcriptional and cellular-stress surveillance deficits are hallmarks of Huntington's disease (HD), a fatal autosomal-dominant neurodegenerative disorder caused by a pathological expansion of CAG repeats in the Huntingtin (HTT) gene. The nucleolus, a dynamic nuclear biomolecular condensate and the site of ribosomal RNA (rRNA) transcription, is implicated in the cellular stress response and in protein quality control. While the exact pathomechanisms of HD are still unclear, the impact of nucleolar dysfunction on HD pathophysiology in vivo remains elusive. Here we identified aberrant maturatio

Leveraging diverse liquid-liquid phase separation patterns to predict the prognosis and immunotherapy of pediatric acute myeloid leukemia.

BACKGROUND: Despite improvements in 5-year survival rates for pediatric acute myeloid leukemia (P-AML) over recent decades, the relapse rate remains high. This challenge is compounded by the absence of reliable prognostic biomarkers, which limits the effectiveness of predictive, preventive, and personalized approaches in P-AML management. Emerging evidence has indicated that the aberrant liquid-liquid phase separation (LLPS) can alter the spatiotemporal coordination of biomolecular condensates, thereby contributing to tumorigenesis and progression. However, the role of LLPS in P-AML remains un…

LINE-1 Retrotransposon Protein ORF1p Forms Condensates That Drive cGAS-Induced Immune Evasion in Lung Squamous Cell Carcinoma.

UNLABELLED: Lung squamous cell carcinoma (LUSC) is a lethal malignancy with limited treatment options, driven by an immunosuppressive tumor microenvironment (TME) and a paucity of actionable targets. Pathogenic LINE-1 (pL1HS) retrotransposons promote LUSC progression by fostering immune evasion. Elucidation of the mechanistic link between retrotransposon activity and TME reprogramming could help identify strategies to stimulate antitumor immunity. In this study, we conducted integrated multiomics analyses, including bulk and single-cell RNA sequencing combined with proteomics, that revealed hy

Charge Transfer Interactions in Quantum Mechanics and in Classical Force Fields.

Charge transfer (CT) is the delocalization of electron density from a donor to an acceptor. It is a crucial component of noncovalent interactions that significantly influences hydrogen bonding, ion solvation, and the structure of aqueous and biomolecular systems. Because direct experimental measurement of CT is challenging, theoretical methods rooted in quantum mechanics (QM) are essential. However, defining CT uniquely within QM and separating it from polarization remains a major methodological hurdle. This review surveys the theoretical methodologies developed to quantify CT, focusing primar
