The heritable diseases of connective tissue are caused by known or putative defects in the synthesis of collagens, proteoglycans, glycoproteins, or attachment proteins of the extracellular matrix. Abnormal synthesis of type I collagen has been reported in several clinical variants of osteogenesis imperfecta. Because clinical classification of these variants is limited by genetic heterogenity and variable expression, biochemical criteria should be used for precise definition of the variants. Newly recognized molecular defects in osteogenesis imperfecta include the diminished formation of type I

Multiple metatarsal osteotomy was performed in 50 children and found to be a reliable procedure for treatment of metatarsus adductus in a child. As a prerequisite to the operation, the epiphysis of the first ray should be ossified. During the operation, the epiphyseal plate must be identified and avoided. After the correction, displacement of the bony fragments may be prevented by wire fixation. The consequences of failure to observe these rules were noted in a patient who was observed for 13 years; despite premature arrest of the physis resulting in 2.5 cm shortening of the first ray, he is a

A case of brachyolmy in a 13 year-old girl is reported. Growth retardation was detected when the child was 5 year old and became severe when 13 years. It concerned only the trunk, limbs being normal. Kyphoscoliosis, akromikia of hands and feet, slight facial dysmorphy with flat nose and thick lips were present. Characteristic radiological features were observed: kyphoscoliosis, platyspondyly with transversally enlarged and irregular vertebrae. Morphological changes of the other epiphyses were minimal: short and squat colla femorii and reduced size of the aleae ilii. Mental development was norm

A 16-day-old male Japanese Black calf affected by skeletal dysplasia, a recessive genetic disorder characterized by marked enlargement of the limb joints due to epiphyseal enlargement. Histological examination revealed chondrocyte proliferation in both the humeral and femoral growth plates, with disorganized chondrocyte alignment and fewer bone trabeculae observed in the humerus. These findings suggest that the morphological changes of chondrocytes may be imbalanced during endochondral ossification at the growth plate. Joint enlargement was evident in the epiphyseal region at birth, despite th

Focal fibrocartilaginous dysplasia (FFCD) is a rare developmental disorder frequently leading to angular deformities in young children. It most commonly affects the proximal tibia followed by distal femur. There are totally 22 reported cases of FFCD of femur. We report a case of FFCD of distal femur as a cause of varus deformity that was managed nonoperatively. The deformity resolved spontaneously over a period of 8 years. This is the second reported case of spontaneous resolution of angular deformity secondary to FFCD of distal femur. We strongly recommend to observe the behavior of deformity

A patient with Engelmann's disease of bone (progressive diaphyseal dysplasia) also suffered from Raynaud's phenomenon, multiple nail fold infarcts, anaemia, leucopenia, hepatosplenomegaly, and a raised erythrocyte sedimentation rate. Her mother, who also had this inherited bone disease, was known to have Raynaud's phenomenon, necrotising vasculitis, and digital gangrene. Review of other published cases of Engelmann's disease indicates that some of these features are not uncommon. It is argued that Engelmann's disease is not primarily a metabolic bone disease but may be a systemic disorder whic

Valgus deformities were created in 6 pony foals by hemicircumferential transection of the periosteum and periosteal stripping (HCTP and PS) just proximally to the distal physis on the medial side of 1 radius (principal thoracic limb). The opposite thoracic limb served as a control. One month after this surgical procedure was done, the limbs were radiographed and the angle of deviation was determined. All horses developed a valgus deformity of the principal limb. In an effort to correct the acquired valgus deformity, the 2nd surgical procedure was performed--HCTP and PS on the lateral aspect of

The pathomorphological features of two economically important skeletal disorders in broilers are described. So-called 'runting and leg weakness' in two-week-old broilers presents the features of rickets, whereas so-called 'lesions of the femur' in six-week-old broilers is marked by resorptive osteoporosis. Both pathomorphological features are observed during the intervening period. Hyperplastic parathyroid glands are present in all birds. The possible aetiology and pathogenesis of the two conditions are discussed. In view of the epidemiological and histopathological features, it is suggested t

The sternocostoclavicular (SCC) region is subject to the same diseases that occur in joints, with congenital and developmental anomalies, inflammatory and infectious diseases, soft tissue and bone tumors as well as the seronegative spondyloarthropathies, including ankylosing spondylitis, reactive arthritis, enteropathic arthritis, psoriatic arthritis, pustulosis palmoplantaris and other syndromes. Most of these conditions present with swelling of the joint, which may be associated with pain and/or tenderness. These disorders are characterised by onset usually before the age of 40 years, absenc

Over the past 11 years, mounting evidence has suggested a significant association between ferroptosis and the development and progression of musculoskeletal (MSK) diseases, such as osteoporosis and osteoarthritis. However, a comprehensive bibliometric analysis summarizing the relationship between ferroptosis and MSK diseases is currently lacking. The present study collected articles and reviews on the topic of ferroptosis in MSK diseases. The data were collected from January 1st, 2012 to June 30th, 2023 by screening the Web of Science database. Various tools, including VOSviewer, CiteSpace, Pa

Mucopolysaccharidosis (MPS) VII is a lysosomal storage disorder characterized by deficient β-glucuronidase activity, which leads to the accumulation of incompletely degraded glycosaminoglycans (GAGs). MPS VII patients present with severe skeletal abnormalities, which are particularly prevalent in the spine. Incomplete cartilage-to-bone conversion in MPS VII vertebrae during postnatal development is associated with progressive spinal deformity and spinal cord compression. The objectives of this study were to determine the earliest postnatal developmental stage at which vertebral bone disease ma

The segmental structure of the vertebrate body plan is most evident in the axial skeleton. The regulated generation of somites, a process called somitogenesis, underlies the vertebrate body plan and is crucial for proper skeletal development. A genetic clock regulates this process, controlling the timing of somite development. Molecular evidence for the existence of the segmentation clock was first described in the expression of Notch signaling pathway members, several of which are expressed in a cyclic fashion in the presomitic mesoderm (PSM). The Wnt and fibroblast growth factor (FGF) pathwa

Nucleotide excision repair requires the action of multiple interacting proteins that locate damage in DNA, remove it as a short oligonucleotide and synthesize a replacement patch. Mutations in genes coding for these proteins give rise to a wide range of diseases involving skin carcinogenesis, neuronal decline and developmental disorders of bone and central nervous system. Complex clinical symptoms of more than one clinical disorder may occur because of mutations that influence protein-protein interactions. Significant differences in repair occur between individuals and species for which the mo

One to two percent of all children are born with a developmental disorder requiring pediatric hospital admissions. For many such syndromes, the molecular pathogenesis remains poorly characterized. Parallel developmental disorders in other species could provide complementary models for human rare diseases by uncovering new candidate genes, improving the understanding of the molecular mechanisms and opening possibilities for therapeutic trials. We performed various experiments, e.g. combined genome-wide association and next generation sequencing, to investigate the clinico-pathological features

OBJECTIVES: Hutchinson-Gilford progeria syndrome is a rare and uniformly fatal segmental "premature aging" disease that affects a variety of organ systems. We sought to more clearly define the bone and weight abnormalities in patients with progeria as potential outcome parameters for prospective clinical trials.
PATIENTS AND METHODS: We collected and analyzed longitudinal medical information, both retrospectively and prospectively, from a total of 41 children with Hutchinson-Gilford progeria syndrome spanning 14 countries, from the Progeria Research Foundation Medical and Research Database at
