Novel brominated flame retardant TBPH upregulates thyroid synthesis markers via S1PR3 and involves IL-10 with atypical JAK-STAT activation.

Bis(2-ethylhexyl) tetrabromophthalate (TBPH), an emerging brominated flame retardant, has raised environmental concern due to its endocrine-disrupting potential. While epidemiological evidence and our previous animal studies have linked TBPH to thyroid disruption, the molecular mechanisms remain unclear. This study aimed to identify potential molecular targets and downstream pathways involved in TBPH-induced effects on thyroid. We found that TBPH (100 nM) significantly upregulated the expression of key thyroid hormone synthesis markers PAX-8, TPO, and TG in human thyroid follicular cells (Nthy

Endocrine disruption by emergent brominated flame retardant Decabromodiphenyl ethane (DBDPE) promotes cancer-associated characteristics in human breast cells and zebrafish xenografts.

Reports show that at least 1000 manufactured chemicals released into the environment are associated with endocrine disruption. Among these, halogenated flame retardants are a source of concern due to their ubiquity, toxic properties, and adverse health effects. Replacing polybrominated diphenyl ethers (PBDEs), a well-studied family of compounds whose use is currently restricted, with compounds of similar chemical structure and fire resistance, such as Decabromodiphenyl ethane (DBDPE), leads to the presumptions of comparable health effects. Because of the reported toxicity of PBDEs, the presenc

Age, Gender, and BMI Modulate the Hepatotoxic Effects of Brominated Flame Retardant Exposure in US Adolescents and Adults: A Comprehensive Analysis of Liver Injury Biomarkers.

Brominated flame retardants (BFRs), commonly found in consumer products, have been identified as potential hazards to liver function. While the individual effects of specific BFRs are somewhat understood, there is limited evidence on how mixtures of these chemicals, especially when influenced by demographic factors, interact to affect liver function. This study utilized data from 10,828 participants aged 12 and above from the National Health and Nutrition Examination Survey (2005-2016) to investigate the associations between BFRs (both individually and in combinations) and biomarkers of liver

Development and validation of a multi-residue method for the simultaneous analysis of brominated and organophosphate flame retardants, organochlorine pesticides, and polycyclic aromatic compounds in household dust.

Household dust is a sink for multiple toxic chemicals with known or suspected potential health effects. However, most dust exposure studies focus on a few chemicals, which may limit overall understanding of human exposure characteristics because people spend most of their time indoors. This paper describes the development and evaluation of a multi-residue analysis of 20 organochlorine pesticides (OCPs), 15 polycyclic aromatic hydrocarbons (PAHs), 8 polybrominated diphenyl ether congeners (PBDEs), 3 hexabromocyclododecane (HBCDs), 8 synthetic musks (Musks), and 7 organophosphate esters (OPEs) i

Complex Mixtures of Brominated/Chlorinated Diphenyl Ethers and Dibenzofurans in Soils from the Agbogbloshie e-Waste Site (Ghana): Occurrence, Formation, and Exposure Implications.

The distribution and toxic equivalents (TEQs) of brominated and chlorinated dibenzo- p-dioxins/dibenzofurans (PBDD/Fs and PCDD/Fs) in soils from Agbogbloshie e-waste site (Ghana) were investigated. The composition of brominated/chlorinated dibenzofurans (PXDFs) and diphenyl ethers (PBDEs, PCDEs, and PXDEs) was examined using two-dimensional gas chromatography-time-of-flight mass spectrometry to elucidate possible formation pathways of dioxins from e-waste recycling. The highest concentrations of PCDD/Fs and PBDD/Fs were found, respectively, in the open burning (1.3-380 ng/g dry weight) and dis

Brominated flame retardants and organochlorine contaminants in winter flounder, harp and hooded seals, and North Atlantic right whales from the Northwest Atlantic Ocean.

Various brominated flame retardants (BFRs), including polybrominated diphenyl ethers (PBDEs) and current-use, non-PBDE BFRs, as well as organochlorine (OC) pesticides and polychlorinated biphenyls (PCBs), were measured in winter flounder, harp and hooded seals, and North Atlantic right whales from the Eastern United States and Canada. The concentrations of PBDEs in winter flounder and right whales were similar in magnitude to the levels of PCBs, which was unlike the pattern observed in seals. In these marine mammals, the levels of PBDEs were orders of magnitude lower than the levels of OCs and

Theoretical evaluation of inter-ion and intra-ion isotope effects in fragmentation: insights into chlorine and bromine isotope effects of halogenated organic compounds occurring in electron ionization mass spectrometry.

Revelation of chlorine and bromine isotope effects in fragmentation is crucial for compound-specific isotope analysis of chlorine/bromine (CSIA-Cl/Br) using gas chromatography-electron ionization mass spectrometry (GC-EI-MS), but theoretical fundamentals of the isotope effects remain unclear. Herein, this study provides a theoretical basis for elucidating the details and implications of chlorine and bromine isotope effects occurring in dehalogenation reactions in EI-MS. Inter-ion and intra-ion isotope effects can occur in dehalogenation reactions in EI-MS, and affect chlorine/bromine isotope r…

Manipulating Heavy Halogenated Asymmetric Terminals Affords Regio-Regular Hetero-Fluorinated/Brominated Dual Asymmetric Acceptor With a Binary Photovoltaic Efficiency of 20.3.

Asymmetric terminal engineering of small molecular acceptors (SMAs) is one promising approach to efficient organic solar cells (OSCs), while synthesizing regio-regular dual asymmetric terminal-based SMAs for boosting binary OSCs remains a critical challenge, and heavier halogenated effects are still underexplored. Herein, three global asymmetric SMAs (AY2F-ClF, AY2F-BrF, and AY2F-IF) and symmetric SY-2(FBr), featuring one or two regio-regular, locally asymmetric hetero-dihalogenated terminals with successively heavier halogens, were successfully synthesized by applying the dual asymmetric term

Formation of volatile chlorinated and brominated products during low temperature thermal decomposition of the representative PFAS perfluorohexane sulfonate (PFHxS) in the presence of NaCl and NaBr.

Per- and polyfluoroalkyl substances (PFAS) are synthetic organofluorine compounds known for their chemical and physical stability as well as their wide range of uses. Some PFAS are widely distributed in the environment, leading to concerns related to both environmental and human health. High temperature thermal treatment (i.e., incineration) has been utilized for PFAS treatment, but this requires significant infrastructure and energy, prompting interest in lower temperature approaches that may still lead to efficient destruction. Lower treatment temperatures, however, increase the potential fo

New brominated flame retardant decabromodiphenyl ethane (DBDPE) in water sediments: A review of contamination characteristics, exposure pathways, ecotoxicological effects and health risks.

As an alternative to polybrominated diphenyl ethers (PBDEs), decabromodiphenyl ethane (DBDPE) has become one of the most important new brominated flame retardants (NBFRs). However, little is known about whether this emerging contaminant may has an environmental fate similar to PBDEs. Sediments are the main sink for DBDPE in the aqueous phase. Worldwide concentration data, since it was first found in sediments to date, have been collated, and the following conclusions have been drawn. (1) DBDPE concentrations in sediments have increased rapidly, often with a higher risk of contamination in sour

Evaluation of the toxic effects of brominated compounds (BDE-47, 99, 209, TBBPA) and bisphenol A (BPA) using a zebrafish liver cell line, ZFL.

The toxic effects of three polybrominated diphenyl ether (PBDE) congeners (BDE-47, -99, and -209), tetrabromobisphenol A (TBBPA) and bisphenol A (BPA), were evaluated by determining their 24h and 96 h median lethal concentrations using a zebrafish liver cell line, ZFL. It was found that BDE-47, BDE-99 and TBBPA showed comparative cytotoxicity within the range of 1.2-4.2 μM, and were more toxic than BPA (367.1 μM at 24 h and 357.6 μM at 96 h). However, BDE-209 induced only 15% lethality with exposures up to 25 μM. The molecular stresses of BDE-47, -99, TBBPA and BPA involved in thyroid hormone

Developmental toxicity of brominated flame retardants, tetrabromobisphenol A and 1,2,5,6,9,10-hexabromocyclododecane, in rat offspring after maternal exposure from mid-gestation through lactation.

To evaluate developmental exposure effects of two brominated flame retardants, tetrabromobisphenol A (TBBPA) and 1,2,5,6,9,10-hexabromocyclododecane (HBCD), pregnant Sprague-Dawley rats were administered either chemical at doses of 100, 1000 or 10,000 ppm in a soy-free diet from gestation day 10 until the day 20 after delivery. Offspring exposed to TBBPA showed dose-unrelated slight decreases of serum triiodothyronine (T(3)) concentration at postnatal day 20, and there was no evidence of hypothyroidism-related neuronal mismigration and impaired oligodendroglial development as judged by morphom

Effects of methyl mercury in combination with polychlorinated biphenyls and brominated flame retardants on the uptake of glutamate in rat brain synaptosomes: a mathematical approach for the study of mixtures.

Regulatory limit values for toxicants are in general determined by the toxicology of the single compounds. However, little is known about their combined effects. Methyl mercury (MeHg), polychlorinated biphenyls (PCBs), and brominated flame retardants (BFRs) are dominant contaminants in the environment and food. MeHg is a well known neurotoxicant, especially affecting the developing brain. There is increasing evidence that PCB and BFRs also have neurotoxic effects. An enhanced effect of these toxicants, due to either synergistic or additive effects, would be considered as a risk for the fetal d

Synthesis, crystal structure, Hirshfeld surface analysis, density function theory calculations and photophysical properties of methyl 4'-[(4-bromobenzo-yl)-oxy]biphenyl-4-carboxyl-ate: a compound with bromine⋯oxygen contacts.

In the mol-ecular title compound, C21H15BrO4, the dihedral angles between the aromatic bromo-benzene ring and the immediate neighbors (first and second aromatic ring of the biphenyl moiety) are 56.57 (2) and 50.91 (4)°. The dihedral angle between the aromatic rings of the biphenyl fragment is 5.78 (4)°. The torsion angles across the ester groups associated with bromo-benzene and methyl moieties are 178.0 (1) and 176.86 (2)°, respectively, revealing an anti-periplanar conformation in both cases. In the crystal, the packing of the mol-ecules is stabilized by Br⋯O contacts running infinitely alon

An updated review of new brominated flame retardant decabromodiphenyl ethane (DBDPE): environmental fate, toxicokinetics, toxicity, and mechanisms based on adverse outcome pathway framework.

Decabromodiphenyl ethane (DBDPE), a novel brominated flame retardant (NBFR) widely adopted as a primary replacement for banned decabromodiphenyl ether (BDE-209), now permeates global environmental matrices. Emerging evidence indicates DBDPE elicits biota toxicity and poses human health risks. This review synthesizes current knowledge on DBDPE's environmental distribution, contamination characteristics, and toxicokinetic profiles-including absorption, distribution, metabolism, and excretion (ADME) in vivo. We critically evaluate its multi-system toxicity (neurological, thyroid, hepatic, intesti

Debromination of novel brominated flame retardants using Zn-based additives: A viable thermochemical approach in the mitigation of toxic effects during e-waste recycling.

Brominated flame retardants (BFRs) are bromine-bearing additives added to the polymeric fraction in various applications to impede fire ignition. The Stockholm Convention and various other legislations abolished legacy BFRs usage and hence, the so-called novel BFRs (NBFRs) were introduced into the market. Recent studies spotlighted their existence in household dust, aquifers and aquatic/aerial species. Co-pyrolysis of BFRs with metal oxides has emerged as a potent chemical recycling approach that produces a bromine-free stream of hydrocarbon. Herein, we investigate the debromination of two pro…

Bromination and conversion of tetrahydro-1H-indene to bisoxirane with a new approach: synthesis, structural characterization by spectroscopic and theoretical methods, and biological analysis supported by DFT and docking.

In this study, a new method for synthesizing diepoxides is proposed. Tetrahydroindene 1 was brominated with NBS in the presence of LiClO4 and acetic acid, resulting in the formation of dibromodiacetate derivatives 2 and 3. Treatment of compounds 2 and 3 with NaOH in methanol produced a mixture of diepoxides 4 and 5. Additionally, direct bromination of tetrahydro-1H-indene yielded tetrabromo octahydroindene isomers 6 and 7. The structures of the compounds were characterized using spectroscopic techniques such as 1H NMR, 13C NMR, APT, COSY, and XRD. The new method provides an easy and selective

Polybrominated diphenyl ethers in serum from residents living in a brominated flame retardant production area: Occurrence, influencing factors, and relationships with thyroid and liver function.

Polybrominated diphenyl ethers (PBDEs) have been used as flame retardants (FRs) in China for decades, even after they were identified as persistent organic pollutants. In this study, serum samples were collected from 172 adults without occupational exposure who were residents of a well-known FR production region (Laizhou Bay, north China), and PBDE congeners were measured to assess their occurrence, congener profile and influencing factors in serum. Moreover, the relationships between serum concentrations of PBDEs and thyroid/liver function indicators were analyzed to evaluate whether human ex
