Combined cancer immunotherapy with lipid nanoparticle delivery of oligo-based cGAS-agonistic adjuvant and peptide or mRNA vaccines.

Therapeutic vaccines are promising for cancer immunotherapy in combination with immune checkpoint blockade (ICB). Though lipid nanoparticles (LNPs) hold great potential to deliver cancer therapeutic vaccines, LNPs delivering peptide or mRNA vaccines often induce suboptimal T cell responses. Type I interferon (IFN-I) responses can enhance antigen presentation and potentiate T cell responses. Here, we report LNP codelivery of peptide or mRNA vaccines with a cyclic GMP-AMP synthase (cGAS) agonist that can specifically induce IFN-I responses to potentiate anticancer T cell responses for robust ICB

Design and Efficacy of Oncolytic Viruses and Antitumor Vaccines: A Dead End in the Immunotherapy of Pancreatic Cancer?

Pancreatic ductal adenocarcinoma (PDAC) remains one of the deadliest malignancies, marked by late diagnosis, limited responsiveness to conventional therapies, and an immunosuppressive tumor microenvironment. While immunotherapy has transformed treatment paradigms in several cancers, its efficacy in PDAC has been minimal. Oncolytic viruses and therapeutic cancer vaccines have emerged as promising immunotherapeutic strategies designed to stimulate robust, tumor-specific immune responses and reshape the immune landscape. However, despite encouraging preclinical data, clinical translation in PDAC …

Meeting report: Considerations for trial design and endpoints in licensing therapeutic HPV16/18 vaccines to prevent cervical cancer.

Cervical cancer is a major cause of morbidity and mortality globally with a disproportionate impact on women in low- and middle-income countries. In 2021, the World Health Organization (WHO) called for increased vaccination, screening, and treatment to eliminate cervical cancer. However, even with widespread rollout of human papillomavirus (HPV) prophylactic vaccines, millions of women who previously acquired HPV infections will remain at risk for progression to cancer for decades to come. The development and licensing of an affordable, accessible therapeutic HPV vaccine, designed to clear or

Restoring anti-oncodriver Th1 responses with dendritic cell vaccines in HER2/neu-positive breast cancer: progress and potential.

HER2/neu is expressed in the majority of in situ breast cancers, but maintained in 20-30% of invasive breast cancer (IBC). During breast tumorigenesis, there is a progressive loss of anti-HER2 CD4(pos) Th1 (anti-HER2Th1) from benign to ductal carcinoma in situ, with almost complete loss in IBC. This anti-HER2Th1 response can predict response to neoadjuvant therapy, risk of recurrence and disease-free survival. Vaccines consisting of HER2-pulsed type I polarized dendritic cells (DC1) administered during ductal carcinoma in situ and early IBC can efficiently correct anti-HER2Th1 response and hav

Translating nanoparticulate-personalized cancer vaccines into clinical applications: case study with RNA-lipoplexes for the treatment of melanoma.

The development of nucleic acid based vaccines against cancer has gained considerable momentum through the advancement of modern sequencing technologies and on novel RNA-based synthetic drug formats, which can be readily adapted following identification of every patient's tumor-specific mutations. Furthermore, affordable and individual 'on demand' production of molecularly optimized vaccines should allow their application in large groups of patients. This has resulted in the therapeutic concept of an active personalized cancer vaccine, which has been brought into clinical testing. Successful t

Preclinical development of a mutant KRAS targeting therapeutic cancer vaccine.

KRAS mutations are frequent oncogenic drivers in several indications, yet limited targeted therapy options are available. Here we engineered our clinically validated artARENA platform to develop an "off-the-shelf" shared neoantigen vaccine, capable of triggering potent CD8+ T-cell responses against the five most prevalent KRAS mutations (G12D, G12V, G12C, G12R, and G13D). Alternating two-vector therapy (sequential administration of artPICV-based vector followed by artLCMV-based vector, encoding the same optimized antigen construct) induced KRAS neoepitope-specific polyfunctional T-cell respons

Vaccine Therapy for the Management of Penile Cancer: Evidence, Opportunities and Challenges.

Penile squamous cell carcinoma (PSCC) is a rare malignancy with limited therapeutic options in advanced and recurrent diseases. Advanced PSCC is typically managed with multimodal therapy, including neoadjuvant chemotherapy or chemoradiation followed by surgery; however, durable responses remain uncommon, and outcomes after recurrence are poor. Cancer vaccines represent a promising immunotherapeutic strategy, as these treatments induce tumor-specific immunity and heightened immune surveillance against penile cancer cells. While therapeutic cancer vaccines have not yet demonstrated consistent cl…

Cancer Vaccine Targeting Mutated GNAQ-Expressing Uveal Melanoma.

Background/Objectives: Uveal melanoma (UM) is the most common intraocular malignancy in adults. Although brachytherapy of the primary tumor provides an approximate 80% five-year survival, with time, nearly half of patients experience predominant liver metastases. It was proposed that malignant cells migrate early and stay dormant as they adapt to the liver microenvironment. We propose that cancer vaccine-mediated activation of UM-targeted immunity in primary UM patients could prevent progression of metastatic disease from dormant cells or malignant seeds. Thus, this study explored DNA vaccinat

Closing the Gap: HPV Vaccination and Head and Neck Cancer Prevention.

PURPOSE OF REVIEW: The association between Human Papillomavirus (HPV) infection and Head and Neck Squamous Cell Carcinoma (HNSCC), particularly Oropharyngeal SCC (OPSCC), represents a growing public health concern due to the rising incidence and the lack of effective screening programs. This review synthesizes current clinical evidence supporting prophylactic HPV vaccination as a primary prevention strategy against HNSCC and highlights emerging innovative management strategies. RECENT FINDINGS: Multiple randomized controlled trials and meta-analyses confirm the high efficacy of available HPV v

Polysaccharide hydrogels as immunologically active vaccine depots for cancer immunotherapy.

Cancer vaccines aim to elicit durable, antigen-specific antitumour immunity, yet clinical translation remains constrained by inefficient antigen retention, weak innate immune activation, and poor spatiotemporal coordination of immune priming. Biomaterial-based vaccine depots, particularly hydrogels incorporating natural polysaccharides, can address these barriers by creating immune-instructive microenvironments that support localised and sustained antigen delivery. Polysaccharide-based hydrogels combine structural support, tuneable release kinetics, and intrinsic immunomodulatory properties th…

Artificial intelligence (AI)-enabled platforms for cancer vaccine engineering.

Cancer vaccines are emerging as a promising approach in immuno-oncology, with the potential to generate tumour-specific and long-lasting immune responses. However, shared-antigen strategies often show limited efficacy because of tumour heterogeneity, immune escape and challenges in identifying immunogenic targets, ultimately limiting progress towards effective immunotherapies. Recent breakthroughs in artificial intelligence (AI) are facilitating a shift towards systematic and data-driven vaccine design, effectively bridging conventional design gaps. AI-driven integration of immunopeptidomics, …

Artificial intelligence for translational personalized neoantigen cancer vaccine development.

Personalized neoantigen cancer vaccine is a promising strategy for precision immunotherapy by targeting patient-specific and mutation-derived tumor antigens. Early clinical studies have demonstrated the feasibility, safety, and immunogenicity of these vaccines across multiple solid tumors, with encouraging outcomes particularly when combined with immune checkpoint blockade. However, broader clinical translation remains limited by sequential bottlenecks across the vaccine development pipeline, including false-positive neoantigen selection,  imperfect modeling of antigen processing and HLA prese

mRNA cancer vaccine: A novel and potential immunotherapy for multiple myeloma.

Multiple myeloma (MM) is a malignant haematologic tumour originating from bone marrow plasma cells, characterised by the abnormal proliferation of monoclonal plasma cells, invasion of the bone marrow, and resulting end-organ damage, with continuous clinical progression. Although standardised treatment regimens and the application of novel cellular immunotherapies have helped control disease progression, the risks of drug resistance and relapse remain, and a complete cure is currently not achievable. mRNA cancer vaccines aim to target personalized neoantigens, inducing strong and specific anti-…

FcRn-guided antigen trafficking enhances cancer vaccine efficacy.

The development of tumor vaccines represents a significant focus within cancer therapeutics research. Nonetheless, the efficiency of antigen presentation in tumor vaccine remains suboptimal. We introduce an innovative mRNA-lipid nanoparticle platform designed to express tumor antigenic epitopes fused with the transmembrane domain and cytoplasmic tail of the neonatal Fc receptor (FcRn). This novel design exploits FcRn trafficking signals to direct the epitope-FcRn fusion toward endolysosomal degradation, thereby generating epitopes capable of eliciting targeted T cell responses and establishing

HPV vaccination and cervical cancer control in Georgia: evidence and projections.

INTRODUCTION: Human Papillomavirus (HPV) is the primary cause of cervical cancer. In Georgia, cervical cancer is the fifth leading cause of cancer-related deaths among women, with approximately 330 new cases and 200 deaths annually. Although HPV vaccines are included in the national immunization programme, coverage remains suboptimal. This study aimed to assess national trends in HPV vaccination, cervical cancer screening, and incidence in Georgia, and to project the potential impact of improved vaccination coverage. MATERIALS AND METHODS: National data from 2016–2022 were analysed using descr
