ERG3-Encoding Sterol C5,6-DESATURASE in Candida albicans Is Required for Virulence in an Enterically Infected Invasive Candidiasis Mouse Model.

Gastrointestinal colonization by Candida species is considered the main source of candidemia. The ERG3 gene in Candida albicans encodes a sterol C5,6-desaturase, which is essential for ergosterol biosynthesis. Although ERG3 inactivation shows reduced virulence in mouse models of disseminated candidiasis, the role of ERG3 in intestinal infections is unknown. Here, we infected mice with the C. albicans strains CAE3DU3 and CAF2-1, containing mutant and wild-type ERG3, respectively, and studied gut infection and colonization by these strains. We found that the CAE3DU3 strain showed reduced coloniz

Improved detection of deeply invasive candidiasis with DNA aptamers specific binding to (1→3)-β-D-glucans from Candida albicans.

Deeply invasive or disseminated candidiasis is a serious and often fatal complication that can occur frequently in immuno-compromised individuals. However, conventional diagnostic methods of Candida albicans display low sensitivity and lack of specificity; the development of rapid and accurate detection methods remains a high priority. Aptamers are single-strand DNA or RNA oligonucleotides that specifically bind to target molecules with high affinity. In this study, we sought to screen high-affinity DNA aptamers that specifically bound to (1→3)-β-D-glucans from cell wall of Candida albicans us

Fluconazole prophylaxis in extremely low birth weight neonates reduces invasive candidiasis mortality rates without emergence of fluconazole-resistant Candida species.

OBJECTIVE: We evaluated the impact of fluconazole prophylaxis for extremely low birth weight infants on invasive candidiasis incidence, invasive candidiasis-related mortality rates, and fluconazole susceptibility of Candida isolates. METHODS: Extremely low birth weight infants <5 days of age, except those with liver dysfunction, were eligible for fluconazole prophylaxis. NICU infants (all birth weights) with invasive candidiasis between April 2002 and March 2006 were compared with those with invasive candidiasis before fluconazole prophylaxis (2000-2001). RESULTS: Twenty-two infants had inva

Temporal Trends and Seasonality of Invasive Candidiasis During and After the COVID-19 Pandemic: An Interrupted Time Series Analysis in Colombia.

Invasive candidiasis is a severe opportunistic infection whose incidence may be influenced by major disruptive events. The COVID-19 pandemic substantially altered hospital dynamics in Colombia. This study aimed to evaluate temporal trends, seasonality, and potential changes in the incidence of invasive candidiasis between 2019 and 2024. We conducted an observational time-series study using confirmed cases of invasive candidiasis from medium- and high-complexity hospitals in three major Colombian cities. Cases were aggregated quarterly. An interrupted time-series (ITS) analysis was performed. A

Synthesis and characterization of two novel amphotericin B-chitosan nanoparticles cross-linked with glutaraldehyde and phthaldialdehyde for the treatment of invasive candidiasis.

Amphotericin B (AmB) is a potent broad-spectrum antifungal agent widely used to treat invasive candidiasis (IC); however, its clinical application is limited by severe toxicity, particularly nephrotoxicity. In this study, chitosan (CS)-AmB nanoparticles (NPs) cross-linked with glutaraldehyde (Glu) and phthaldialdehyde (Ph) were developed to enhance therapeutic efficacy and reduce AmB toxicity. CS-Glu-AmB and CS-Ph-AmB NPs were synthesized by cross-linking CS with Glu or Ph, followed by AmB incorporation. NP formation was confirmed by FT-IR, 1H NMR, and XRD analyses. The average particle sizes

Correlation of broad-spectrum antibiotic utilization patterns with invasive candidiasis in critically ill patients and development of an early prediction model.

Invasive candidiasis (IC) remains an infection with high incidence and mortality rates in the ICU setting, particularly among patients treated with broad-spectrum antibiotics. This study aims to investigate the association between detailed antibiotic usage profiles and the occurrence of IC, and to develop an IC-predictive model specialized for patients who received broad-spectrum antibiotics. We retrospectively collected detailed information on antibiotic categories, treatment duration, combination therapies and other clinical data of enrolled patients. Univariate and multivariate logistic reg

Creating a prediction model for invasive candidiasis in the intensive care unit using a case control design: a European multicentre approach.

PURPOSE: Invasive candidiasis (IC) has a high attributable morbidity and mortality in patients in the intensive care unit (ICU). Current diagnostic tools lack sensitivity, introduce delay or have not been validated for regular use. As early treatment has proven vital for survival, multiple prediction models have been proposed but have not been validated for multinational implementation. In this study we propose to find factors predisposing the ICU patient to develop IC. We hope to develop an alternative prediction model using a large international dataset. METHODS: Using ICU-acquired IC as pr

Iron Chelator Deferasirox Reduces Candida albicans Invasion of Oral Epithelial Cells and Infection Levels in Murine Oropharyngeal Candidiasis.

Candida albicans, the causative agent of mucosal infections, including oropharyngeal candidiasis (OPC), as well as bloodstream infections, is becoming increasingly resistant to existing treatment options. In the absence of novel drug candidates, drug repurposing aimed at using existing drugs to treat off-label diseases is a promising strategy. C. albicans requires environmental iron for survival and virulence, while host nutritional immunity deploys iron-binding proteins to sequester iron and reduce fungal growth. Here we evaluated the role of iron limitation using deferasirox (an FDA-approved

Synergistic antifungal effect of lactoferrin with azole antifungals against Candida albicans and a proposal for a new treatment method for invasive candidiasis.

The combination of lactoferrin with fluconazole has been reported to synergistically enhance the antifungal activity of fluconazole against Candida spp. and inhibit the hyphal formation in fluconazole-resistant strains of Candida albicans. In this study, we investigated the association between the therapeutic effects of this combination and the pharmacological characteristics of fluconazole and itraconazole and the variation in these effects with differences among the strains in terms of the susceptibility and resistance mechanisms. Lactoferrin enhanced the growth-inhibitory activity of flucon
