Occurrence of Pineal Gland Tumors in Combined Chronic Toxicity/Carcinogenicity Studies in Wistar Rats.

Pineal gland tumors are very rare brain lesions in rats as well as in other species including humans. A total of 8 (out of 1,360 examined) Wistar rats from 3 different combined chronic toxicity/carcinogenicity or mere carcinogenicity studies revealed pineal gland tumors. The tumors were regarded to be spontaneous and unrelated to treatment. The morphology and immunohistochemical evaluation led to the diagnosis malignant pinealoma. The main characteristics that were variably developed within the tumors were the following: cellular atypia, high mitotic index, giant cells, necrosis, Homer Wright

Exposure limits for occupational and environmental chemical pollutants in various media.

Exposure limits for occupational and environmental chemical contaminants are based on animal experimentation and observation in humans. Usually, the final recommendations are "predictions" which can hardly guarantee the absolute safety for all. Late effects were discovered to lately in the past. The impact of the biotransformation of pollutants on their biological activity is demonstrated with trichloroethylene, styrene and N,N-dimethylformamide, which are suspected of having carcinogenic potential. In addition, trichloroethylene and N,N-dimethylformamide cause intolerance of alcohol with hype

Metal mutagens and carcinogens effectively displace acridine orange from DNA as measured by fluorescence polarization.

Several metals are known mutagens and carcinogens. These metals effectively displace acridine orange from DNA when measured by fluorescence polarization. Displacement of 50% of the acridine orange is obtained with less than 0.5 mM concentrations of lead, manganese, cobalt, zinc, cadmium, nickel, iron, copper, and cis-platinum. In contrast, greater than 80 mM concentrations of lithium, sodium, and potassium are required to displace an equivalent amount of acridine orange from calf thymus DNA. Although cis-platinum shows the best DNA reactivity in this assay, the interaction between this metal a

How to evaluate and select cervical screening and triage tests in vaccinated populations.

The substantial changes of human papillomavirus (HPV) type distribution and reductions of cervical precancer and cancer incidence in vaccinated populations, which are entering screening programs now, require reassessment of screening and triage approaches to maintain the balance of benefits and harms of cervical cancer screening. The reduction of precancer and cancer in vaccinated populations affects the risk indicated by most screening and triage tests. Among HPV-positive women, the type distribution changes in vaccinated women to less carcinogenic types that are less likely to progress to pr

Historical Control Data of Spontaneous Pathological Findings in C57BL/6J Mice Used in 18-Month Dietary Carcinogenicity Assays.

A retrospective analysis in C57BL6/J mice used in dietary carcinogenicity studies was performed to determine the survival rate, causes of death and incidences of spontaneous non-tumoral and tumoral findings. Data were collected from 1600 mice from control dose groups of sixteen 18-month carcinogenicity assays performed between 2003 and 2021 at the same test facility with similar environmental conditions and experimental procedures. The survival rate was high in both sexes (81%-85%) and the causes of humane euthanasia or death were mainly non-tumoral (chronic ulcerative dermatitis, atrial throm

Causes of Mortality and Profile of Spontaneous Tumors in Young CD-1 Mice.

A retrospective study was performed to establish the causes of mortality and incidence patterns of tumors in young (<50 weeks) control CD-1® mice from Charles River Laboratories. Tumor incidences (fatal and nonfatal) and nonneoplastic causes of death observed during the first 50 weeks of the study were collected from 48 thirteen-week toxicity studies conducted between 2009 and 2018 and from 43 carcinogenicity studies conducted between 2005 and 2018. Thirteen-week studies had a mortality rate of 8/620 (1.3%) in males and 4/620 (0.65%) in females. The major factors contributing to death were int

Toxicological evaluation of smokeless tobacco: 2-year chronic toxicity and carcinogenicity feeding study in Wistar Han rats.

UNLABELLED: A comprehensive 2-year oral chronic toxicity/carcinogenicity study was conducted with smokeless tobacco using modern toxicological test methods and well-accepted standards. The study included a 1-year interim subgroup to assess toxicity at that intermediate time point. Test groups consisted of a tobacco blend (B) used in snus, and an aqueous tobacco extract of that tobacco blend (E) administered at 0.2, 2, or 5 mg nicotine/kg body weight/day via dosed feed to male and female Wistar Han rats. The dosages were selected to simulate potential exposure in humans ingesting smokeless toba

The effects of diet, ad libitum overfeeding, and moderate dietary restriction on the rodent bioassay: the uncontrolled variable in safety assessment.

Ad libitum (AL) overfeeding is the most significant, uncontrolled variable affecting the outcome of the current rodent bioassay. There is a highly significant correlation between AL food consumption, the resultant obesity and body weight, and low 2-yr survival in rodents. AL feeding of diets with lowered protein, metabolizable energy (ME), and increased fiber does not improve survival. Only dietary restriction (DR) of all diets tested significantly improves survival and delays the onset of spontaneous degenerative disease (i.e., nephropathy and cardiomyopathy) and diet-related tumors. Moderate

Polymerase Chain Reaction Human Papillomavirus (HPV) Detection and HPV Genotyping in Invasive Cervical Cancers With Prior Negative HC2 Test Results.

OBJECTIVES: Recently, three large Chinese cohort studies showed that 7.5% to 15.5% of patients with cervical carcinoma had negative high-risk human papillomavirus (hrHPV) test results on prior cytology specimens. These studies raise the question as to whether these negative hrHPV results represent truly hrHPV-negative carcinomas or false-negative hrHPV test results due to limitations in the testing of cytology specimens. This is increasingly an important question with increasing push to use hrHPV testing alone to screen for cervical cancer. METHODS: We investigated the hrHPV status on the sur

From TD50 to benchmark dose in nitrosamine risk assessment: evidence from N-Nitrosotrimetazidine carcinogenicity and TGR mutation data.

The presence of N-nitrosamine drug substance-related impurities (NDSRIs) in pharmaceuticals represents a significant regulatory and safety challenge due to their classification as a "cohort of concern." This paper describes the toxicological evaluation of N-Nitrosotrimetazidine (NTMZ), performed to refine the initial default acceptable intake (AI) limits of 18-26.5 ng/day and the later published value of 400 ng/day, established using the Carcinogenic Potency Categorization Approach (CPCA). The evaluation followed a tiered approach: NTMZ was first confirmed as mutagenic in vitro in a standard A

Interaction of the cytochrome P4501A2, SULT1A1 and NAT gene polymorphisms with smoking and dietary mutagen intake in modification of the risk of pancreatic cancer.

Aromatic amines, N-nitroso compounds and heterocyclic amines are suspected human pancreatic carcinogens. Cytochrome P450 (CYP) 1A2, N-acetyltransferase (NAT) 1, NAT2 and sulfotransferase (SULT) are enzymes involved in the metabolism of these carcinogens. To test the hypothesis that genetic variations in carcinogen metabolism modify the risk of pancreatic cancer (PC), we investigated the effect of single-nucleotide polymorphisms (SNPs) of the CYP1A2, NAT1, NAT2 and SULT1A1 gene on modification of the risk of PC in a hospital-based study of 755 patients with pancreatic adenocarcinoma and 636 hea

Bone marrow spontaneous lesions in rodents from nonclinical 104-week carcinogenicity studies.

The authors performed a retrospective study to determine the incidences and range of spontaneous lesions in the bone marrow (sternum and femur) of control mice and rats. Data was collected from 2186 mice (Crl:CD-1(ICR)BR), and 2347 rats (Han Wistar and CD(SD) rats) from the control dose groups of 104-week carcinogenicity studies carried out between 2005 and 2014. The incidence of spontaneous lesions in the bone marrow was higher in mice than in rats, and in both species non-neoplastic lesions were more common than neoplastic lesions. In mice, the most common non-neoplastic lesions in the bone

Occurrence of thymic lymphoma in carcinogenesis bioassay specimens of the Japanese medaka (Oryzias latipes).

Multiple cases of thymic lymphoma were diagnosed in bioassay specimens of Japanese medaka (Oryzias latipes), a small fish species. Lymphoma in chemically exposed specimens was not clearly related to specific compounds, dose, or sex. Multiple cases of lymphoma occurred in three specimens held in one aquarium and in two specimens in two other aquaria, suggesting the possibility of horizontal transmission. Forty cases were observed in 9802 specimens (0.41%), with an incidence of 0.46% in exposed specimens (5690 total) and 0.34% in controls (4112 total). Lesions were dominated by small cells with

Somatic cell mutagenicity in Drosophila melanogaster in comparison with genetic damage in early germ-cell stages.

With the intention of assessing the general performance, sensitivity and the underlying mechanisms of somatic cell mutagenicity assays in Drosophila, a study was undertaken to compare the effectiveness of 5 procarcinogens and 4 direct-acting agents in the white/white-coral eye mosaic assay (SMART) with their activity in early (premeiotic) male and female germ-cell stages, after exposure of Drosophila larvae. The outcome indicated a lack of agreement in the results from recessive lethal assays (SLRL) in comparison with the somatic mutation and recombination test (SMART). The procarcinogens 2-na

Trapping of chemical carcinogens with magnetic polyethyleneimine microcapsules: II. Effect of membrane and reactant structures.

Effects of the membrane structure and reactant type on the trapping of carcinogens and other reactive species by semipermeable magnetic polyethyleneimine (PEI) microcapsules are investigated. A series of these microcapsules with poly(hexamethyleneterephthalamide) membranes was prepared by interfacial polymerization with an 8-fold variation of hexamethylenediamine concentration in the aqueous emulsion phase. Although little change was found in the encapsulation of PEI (within the microcapsule core) and magnetite, the microcapsule membrane showed a 6-fold alteration in regard to mass and associa

Chemical interactions with herpes simplex type 2 virus: enhancement of transformation by hydrazine and 1,2-dimethylhydrazine.

Quantitative assays for the morphological transformation of 3T3 Swiss mouse cells by herpes simplex type 2 virus (HSV-2) were employed to examine the effect on cell transformation of chemical carcinogens and suspected carcinogens. Exposure of the cells to the chemical compound, followed by virus infection, resulted in enhancement of transformation when compared to that observed with chemical or virus alone. Enhancement occurred in tests utilizing either UV light-inactivated HSV-2 (strain 333) or a temperature-sensitive (ts) mutant of HSV-2 (A8(293)]. A series of seven ts-mutants were tested an

Human carcinogenic risk evaluation, Part III: Assessing cancer hazard and risk in human drug development.

Assessing cancer risk for human pharmaceuticals is important because drugs are taken at pharmacologically active doses and often on a chronic basis. Epidemiologic studies on patient populations have limited value because of the long latency period for most cancers and because these studies lack sensitivity. The Center for Drug Evaluation and Research (CDER) of the U.S. Food and Drug Administration relies on short-term surrogate assays (genetic toxicology studies) to assess risk to patients involved in clinical trials and on rodent carcinogenicity studies to assess cancer risk for drug approval
