BPDCN MYB fusions regulate cell cycle genes, impair differentiation, and induce myeloid-dendritic cell leukemia.

MYB fusions are recurrently found in select cancers, including blastic plasmacytoid DC neoplasm (BPDCN), an acute leukemia with poor prognosis. They are markedly enriched in BPDCN compared with other blood cancers and, in some patients, are the only obvious somatic mutation detected. This suggests that they may alone be sufficient to drive DC transformation. MYB fusions are hypothesized to alter the normal transcription factor activity of MYB, but, mechanistically, how they promote leukemogenesis is poorly understood. Using CUT&RUN chromatin profiling, we found that, in BPDCN leukemogenesis, M

Prevalence of the BCR/ABL fusion gene and T cell stimulation capacity of dendritic cells in chronic myelogenous leukemia.

Dendritic cell (DC) vaccines are promising for immunotherapy, and their production using CD34+ hematopoietic stem cells (HPSCs) from patients with chronic myelogenous leukemia (CML) and healthy donors is well established. However, the generation of CD1a+CD14- DCs and their functional properties in patients with CML remain elusive. Here, we aimed to study the biology of DCs generated from CD34-/low HPSCs and evaluate the status of their BCR/ABL translocation, ability to stimulate T cells, and capacity of endocytosis compared to DCs derived from CD34+ HPSCs from both patients with CML and health

Tr1 cell-mediated protection against autoimmune disease by intranasal administration of a fusion protein targeting cDC1 cells.

Curative therapies against autoimmune diseases are lacking. Indeed, most of the currently available treatments are only targeting symptoms. We have developed a novel strategy for a therapeutic vaccine against autoimmune diseases based on intranasal administration of a fusion protein tolerogen, which consists of a mutant, enzymatically inactive, cholera toxin A1 (CTA1)-subunit genetically fused to disease-relevant high-affinity peptides and a dimer of D-fragments from protein A (DD). The CTA1 R7K mutant - myelin oligodendrocyte glycoprotein (MOG), or proteolipid protein (PLP) - DD (CTA1R7K-MOG/

Incomplete cytokinesis and re-fusion of small mononucleated Hodgkin cells lead to giant multinucleated Reed-Sternberg cells.

Multinucleated Reed-Sternberg (RS) cells are pathognomonic for classical Hodgkin lymphoma (HL), and their presence is essential for diagnosis. How these giant tumor cells develop is controversial, however. It has been postulated that RS cells arise from mononucleated Hodgkin cells via endomitosis. Conversely, continuous single-cell tracking of HL cell lines by long-term time-lapse microscopy has identified cell fusion as the main route of RS cell formation. In contrast to growth-induced formation of giant Hodgkin cells, fusion of small mononuclear cells followed by a size increase gives rise t…

Target-cell specificity of fusogenic liposomes: membrane fusion-mediated macromolecule delivery into human blood mononuclear cells.

Fusogenic liposome, a unique vector prepared by fusing ultraviolet-inactivated Sendai virus and liposome, is known to efficiently deliver content into various animal cells through membrane fusion. In this study, we examined the target-cell specificity of fusogenic liposome (FL)-mediated macromolecule delivery into human blood cells using diphtheria toxin fragment A (DTA) as a probe. Among the peripheral blood mononuclear cells (PBMC), FL was able to deliver its encapsulates into CD14+ monocytes and CD4-/CD8- T-cells, but not into CD19+ B-lymphocytes, CD4+ T-cells or CD8+ T-cells. The susceptib

In vitro and in vivo study of pluripotency in intraspecific hybrid cells obtained by fusion of murine embryonic stem cells with splenocytes.

Hypoxanthine phosphoribosyltransferase-deficient (HPRT-) mouse embryonic stem (ES) cells, HM-1 cells (genotype XY), were fused with adult female DD/c mouse spleen cells. As a result, a set of HAT-resistant clones was isolated. Four hybrid clones most similar in morphology and growth characteristics to the HM-1 cells were studied in detail with respect to their pluripotency. Of these, three clones contained 41-43 chromosomes, and one clone was nearly tetraploid. All the clones had the XXY set of sex chromosomes and expressed the HPRT of the somatic partner only. The hybrid clones shared feature

Appearance of Epstein-Barr virus-determined nuclear antigen in human epithelial cells following fusion with lymphoid cells.

When human epithelial FL cells were fused with lymphoid cells harboring Epstein-Barr virus (EBV) genome, EBV-determined nuclear antigen (EBNA) immunofluorescence became evident in high frequency in FL nuclei of heterokaryons several days after fusion. The implications of the findings are discussed in relation to the presence of EBV genome in nasopharyngeal carcinoma cells.

Expanding the Spectrum of Adult NTRK3-Rearranged Spindle Cell Neoplasms: A Recurrent NTRK3-SQSTM1 Fusion Spindle Cell Tumor With Deceptively Bland Morphology.

OBJECTIVES: NTRK-rearranged spindle cell neoplasms (other than infantile fibrosarcoma) are an emerging entity of tumors that demonstrate wide variation in clinical and histopathologic features. We report a case of an NTRK-rearranged spindle cell neoplasm bearing a deceptively bland morphology. METHODS: We performed histopathologic, immunohistochemical, and molecular evaluation on resection tissue. We also conducted a literature review on adult NTRK3-rearranged spindle cell neoplasms. RESULTS: The tumor presented as a recurrent ankle mass in an elderly patient. Histologically, it was composed

DEK :: AFF2 Fusion Sinonasal and Skull Base Nonkeratinizing Squamous Cell Carcinoma : A Clinical Outcome Study Compared With Conventional Sinonasal Squamous Cell Carcinoma.

DEK :: AFF2 fusion nonkeratinizing squamous cell carcinoma (NKSCC) is an emerging entity in the sinonasal tract, temporal bone, and skull base. However, the clinical behavior of these tumors has not been well studied. Here, we report the largest cohort of DEK :: AFF2 carcinomas to determine if morphology, mitotic rate, and/or Ki-67 IHC are associated with patient outcomes, including a comparison with high-risk human papillomavirus (HPV)-associated and independent patients. We solicited cases of molecularly or AFF2 immunohistochemistry (IHC) proven DEK :: AFF2 SCC from surgical pathologists to

Prime-boost vaccines encoding an intracellular idiotype/GM-CSF fusion protein induce protective cell-mediated immunity in murine pre-B cell leukemia.

Two vaccines against an intracellularly expressed B cell idiotype were assessed for their ability to induce protective immunity in mice against challenge with a pre-B cell leukemia. One vaccine was based on a plasmid expression vector and the other was a recombinant vaccinia virus; both vaccines expressed a polypeptide derived from the complementarity-determining regions (CDR(2)-CDR(3)) of the leukemic clone-specific immunoglobulin heavy chain (IgH), as a fusion product with mouse granulocyte-macrophage colony-stimulating factor (mGM-CSF). Mice inoculated with either vaccine showed significant

Reveal the Effect of Mechanical Stretch on Virus-Cell Membrane Fusion Based on a Microfluidic Chip.

Lung-on-a-chip models are important for studying the onset and progression of respiratory diseases. However, existing lung chips often lack the ability to precisely regulate alveolar stretch deformation and rarely apply the 3D physiological microenvironment of alveoli to respiratory virus research. Here, we present a biomimetic 3D microfluidic chip microphysiological system for alveolar cells stretch visualization and investigate the effect of mechanical stretch on viral infection. We utilize a SARS-CoV-2 pseudovirus to infect alveolar epithelial cells cultured on the lung alveolus chip. Subse

Improving B-cell Linear Epitope Prediction via Multiple Feature Fusion and an Integrated Machine Learning Algorithm.

INTRODUCTION: The identification of linear B-Cell epitopes (BCEs) is significantly important for the discovery of drugs, such as antibody production, peptide-based vaccines, and other therapeutics. MATERIALS AND METHODS: Unlike traditional laboratory-based methods, computational techniques can save cost and time in predicting large-scale BCEs. For this purpose, numerous in-silico methods have been designed to enhance the overall efficacy of BCE prediction. However, research gaps exist for further improvement in the context of using novel feature representations and learning models for BCE pre

Improved cross-presentation enabled by fusion-dependent mini-proteins that efficiently reach the dendritic cell cytosol.

Cross-presentation is the process by which dendritic cells communicate to CD8 + T-cells the detection of exogenous foreign substances known as antigens. This process primes naïve CD8 + T-cells to eradicate antigen-expressing pathogens and tumors. It is well known that pathogens and tumors evade CD8 + T-cell immunity by reducing antigen uptake into the endocytic pathway and/or limiting endosomal escape of the antigen to the cytosol. Here we employ biophysical and cell biology tools to separately and quantitatively probe the efficiency of antigen uptake, internalization, display, and activity in

Clear Cell Tumor With MITF::CREM Fusion: A Rare Case Report of a Newly Described Entity.

The increasing application of molecular diagnostics in pathology has enabled the identification of novel tumor entities, including clear cell tumor with MITF::CREM translocation, as recognized in the 5th edition of the WHO Classification of Tumours. We describe a case involving a 2-year-old boy with a left scalp lesion, initially suspected clinically to represent a pyogenic granuloma. Histopathological assessment, corroborated by molecular analysis, established the diagnosis of clear cell tumor with MITF::CREM translocation. This case emphasizes the need to consider rare entities in the differ

A dual-pathway Wnt-IL-13 fusion protein enhances human intestinal regeneration through tuft cell activation.

Compromised intestinal regenerative responses drive severe inflammatory conditions, such as inflammatory bowel disease and graft-versus-host disease, affecting millions of people worldwide each year. Despite extensive research, effective therapies remain limited, and no curative treatments are currently available. We recently discovered that human intestinal tuft cells promote tissue repair following injury through Wnt and IL-4/IL-13 signaling pathways. Building on this discovery, here, we report the engineering and functional validation of a synthetic Wnt-IL-13 fusion protein that simultaneou
