The adherent mucous gel layer lining the colonic epithelium is the first line of host defense against invasive pathogens, such as Entamoeba histolytica. Poliovirus (PV) can persist in vivo in the intestine of immunocompromised hosts for years. This paper briefly reviews commonly used procedures for separation of mononuclear cells (MNC) and granulocytes from human blood with X-ray contrast media as gradient material, and also presents new and modified procedures for leucocyte preparation. Standard techniques for human blood do not always yield satisfactory results with blood from other species. Unilateral ischemia in the right cerebral hemisphere of the rat was induced by ligation of the right common carotid artery coupled with controlled hemorrhage to produce hypotension (25 +/- 8 mm/Hg). A comparison between a microsomal and a cytosolic source of receptor-like androgen-binding activity was made in the rat ventral prostate. Microsomal binders remain in solution at 90% saturation with (NH4)2SO4 and display equal affinity for 5 alpha-dihydrotestosterone (DHT) and mibolerone. The nature of the complexes formed between histone H1 and linear double-stranded DNA is dependent on ionic strength and on the H1 : DNA ratio. At an input ratio of less than about 60% (w/w) H1 : DNA, there is a sharp transition from non-co-operative to co-operative binding at a critical salt concentration that depends on the DNA size and is in the range 20 to 50 mM-NaCl. In sucrose density gradient analysis, the cytosol of human ovarian capsules was found to contain two (3H)dihydrotestosterone (DHT)-binding macromolecules with sedimentation constants of 9S and 4.6S, respectively. The PGs produced in the growth medium by skin fibroblast cultures from two O.I. affected patients were investigated. After density gradient centrifugation, in the most dense fraction two main families of molecules appeared. The patient with the more severe clinical picture showed a lower content of the PGs with the highest molecular weight. The GAG composition of PGs was different in the two patients. The more severely affected one showed an increase of HS and a decrease of ChS content, in agreement with the lower value of galactosamine to glucosamine ratio in urinary GAGs.

Purification of epizootic hemorrhagic disease virus (EHDV) from sonicated cell culture supernatant was effected using polyethylene glycol (PEG) precipitation and two CsCl density gradient centrifugations. During purification procedures the pH was maintained at values close to pH 8.0 as pH stability for EHDV was maximal between pH 7.2 and pH 9.0.
