Photoreceptive retinal ganglion cells control the information rate of the optic nerve.

Information transfer in the brain relies upon energetically expensive spiking activity of neurons. Rates of information flow should therefore be carefully optimized, but mechanisms to control this parameter are poorly understood. We address this deficit in the visual system, where ambient light (irradiance) is predictive of the amount of information reaching the eye and ask whether a neural measure of irradiance can therefore be used to proactively control information flow along the optic nerve. We first show that firing rates for the retina's output neurons [retinal ganglion cells (RGCs)] sca

An Inhibitory Septum to Lateral Hypothalamus Circuit That Suppresses Feeding.

UNLABELLED: Feeding behavior is orchestrated by neural circuits primarily residing in the hypothalamus and hindbrain. However, the relative influence of cognitive and emotional brain circuits to the feeding circuitry in the hypothalamus and hindbrain remains unclear. Here, using the cell-type selectivity of genetic methods, circuit mapping, and behavior assays, we sought to decipher neural circuits emanating from the septal nucleus to the lateral hypothalamus (LH) that contribute to neural regulation of food intake in mice. We found that chemogenetic and optogenetic activation of septal vesicu

Obesogenic diet impairs social memory through alterations of hippocampal CA2 excitability and oxytocin signaling.

While obesity induces cardio-metabolic disorders and cognitive deficits, the underlying neural mechanisms remain unexplored. In mice, exposure to an obesogenic high-fat and sugar diet (HFD) resulted in social recognition memory deficits in both males and females, a process that is dependent upon hippocampal area CA2 and oxytocin signaling. HFD-fed mice had stronger inputs onto CA2 pyramidal neurons that led to increased action potential firing, without altering intrinsic properties or inhibitory transmission. Chemogenetic CA2 pyramidal neuron inhibition in HFD-fed mice rescued social novelty d

A dopaminergic projection from the ventral tegmental area to the dorsal raphe nucleus critically regulates propofol anesthesia in mice.

General anesthesia induces a reversible loss of consciousness, yet the precise neural circuits mediating this state transition remain incompletely understood. The ventral tegmental area (VTA) dopaminergic (DA) neurons and the dorsal raphe nucleus (DRN) serotonergic (5-HT) neurons are key components of the ascending arousal system. This study investigated the existence and functional role of VTADA→DRN5-HT circuit in regulating propofol anesthesia. Using virus-mediated neural circuit tracing in DAT-Cre and TPH2-CreER transgenic mice, we demonstrated an anatomically connected circuit appositions

Entorhinal Cortex Hyperexcitability Drives Selective Hippocampal Injury and Cognitive Impairment After Global Cerebral Ischemia.

BACKGROUND: The hippocampus is highly susceptible to global cerebral ischemia, yet the contribution of extrahippocampal regions to this vulnerability remains poorly defined. In particular, afferent inputs to the hippocampus, such as those from the entorhinal cortex (EC), have received limited investigation in the context of ischemic brain injury. METHODS: Global cerebral ischemia was performed using a 2-vessel occlusion methods in mice. In vitro electrophysiological techniques were used to detect the excitability of EC during ischemia-reperfusion and after chemogenetic inhibition. Cell damage

Role of dopaminergic projections from ventral tegmental area to paraventricular thalamus in heroin-induced behavioural sensitization of rats.

The ventral tegmental area (VTA) is involved in the behavioral sensitization to heroin, but the possible neural projections and molecular mechanisms remain unclear. Rats first underwent 8 days of heroin sensitization induction, followed by a 10-day drug-free period. Subsequently, a M5 muscarinic acetylcholine receptor antisense oligonucleotide was microinjected into the VTA, and the expression of heroin sensitization was assessed. Secondly, retrograde viral tracing combined with immunofluorescence labeling was used to characterize VTA neuronal populations projecting to the paraventricular thal

A brain-lung circuit drives LPS-induced lung injury via sympathetic neutrophil axis.

Acute respiratory distress syndrome (ARDS) is a fatal complication of sepsis. However, the neural mechanisms underlying the exacerbation of pulmonary inflammation during systemic infection remain largely undefined. We employed an intraperitoneal lipopolysaccharide (LPS)-induced systemic inflammatory lung injury model and found that systemic inflammation strongly activates corticotropin-releasing hormone (CRH) neurons in the paraventricular nucleus (PVN), whereas bilateral subdiaphragmatic vagotomy abolished this response. Chemogenetic inhibition of CRHPVN neurons alleviated lung injury and the

Cortistatin neurons in the prelimbic cortex regulate seizure susceptibility in female mice via BDNF-TrkB signaling.

The risk of developing psychiatric disorders, particularly stress-related disorders such as major depressive disorder (MDD) and post-traumatic stress disorder (PTSD), is increased three fold in patients with epilepsy. While this increased risk may arise as a consequence of living with epilepsy, shared neurobiological mechanisms, particularly dysregulation of GABAergic signaling, may also contribute. To investigate this link, we investigated the function of GABAergic neurons co-expressing the neuropeptide cortistatin (CST), which has anticonvulsant effects and is implicated in both MDD and PTSD

The vagus nerve activated by inflammation co-transmits acute visceral pain via the NTS-PVN pathway.

BACKGROUND: Acute pancreatitis (AP) induces visceral pain through inflammation and mechanical stress, yet current treatments remain inadequate. This therapeutic limitation reflects our limited knowledge of how nociceptive signals are transmitted from peripheral visceral afferents to central neural pathways. METHODS: A neural circuit connecting the pancreas and the brain was mapped using various viral tracer techniques. The role of this circuit in acute pancreatitis-induced visceral pain was verified using optogenetics, chemogenetics, fiber-optic in vivo calcium signal recording, and electroph…

DRN-SNc serotonergic circuit drives stress-induced motor deficits and Parkinson's disease vulnerability.

Stress is a recognized risk factor for Parkinson's disease (PD), but the mechanisms by which stress exacerbates PD symptoms through the sero … ra compacta (SNc) dopaminergic (DA) neurons. Manipulating the DRN-SNc pathway with optogenetics and chemogenetics confirmed its critical role in stress-induced motor deficits. Activation of the SNc 5-HT2C receptor with an agonist replicated these deficits, while receptor inhibition prevented them, underscoring its importance. Chronic EP stress worsened MPTP-ind …

Minimally-invasive manipulation of spared and hypoactive interneurons reduces CA1 synchronization and nonspatial behavior alterations in epilepsy models.

Temporal lobe epilepsy (TLE) is associated with seizures and severe cognitive impairments, including memory deficits. The dysfunction of a large population of hippocampal interneurons, composed of various subtypes, is proposed as a key mechanism and a therapeutic target. However, the nature and extent of alterations in hippocampal inhibitory neurons remain unclear, as does their impact on TLE pathology. To address this issue, we combined immunolabeling, calcium imaging, electrophysiology, chemogenetic tools, and behavioral assays in mouse pilocarpine TLE models to investigate the survival and

Oxytocin Improves Autistic Behaviors by Positively Shifting GABA Reversal Potential via NKCC1 in Early-Postnatal-Stage.

Accumulating evidence has identified disrupted oxytocin signaling in both autistic patients and animal models of autism. Nevertheless, the specific timing of the impact of oxytocin on social behavior has remained unclear. Using mouse strains from oxytocin-Cre mice crossed with Cre-dependent chemogenetic mice, oxytocinergic neuronal activity is selectivity manipulated during the early or late postnatal stages and revealed, for the first time, that the suppression of oxytocinergic neurons in the early rather than late postnatal stage led to the emergence of autistic-like behaviors. Notably, sign

A projection from the medial prefrontal cortex to the lateral septum modulates coping behavior on the shock-probe test.

Effective coping plays an important role in preventing stress-induced neuropsychiatric conditions. The ventromedial prefrontal cortex (vmPFC) has been associated with active, adaptive coping in humans and rodents. Chronic or severe stress has been shown to induce a maladaptive shift from active to passive coping behavior; however, the neural circuits for effective coping strategies remain unclear. In the current study, we demonstrated that neurons in the infralimbic (IL) subregion of rat vmPFC that project to the lateral septum (LS) were recruited by exposure to the shock probe in the shock-pr

Regulating astrocytic activity in the dorsal striatum mitigates L-dopa-induced dyskinesia in Parkinson's disease.

In Parkinson's disease (PD), long-term 3,4-dihydroxy-L-phenylalanine (L-dopa) therapy leads to the development of motor complications, including L-dopa-induced dyskinesia (LID). Increased numbers of reactive astrocytes in the brains of patients with PD are a key feature of this disease. Astrocytes are involved in the development of LID; however, whether the regulation of astrocytic activity influences LID development remains unclear. Therefore, this study aimed to determine the effect of the direct modulation of glial fibrillary acidic protein (GFAP)-expressing glia on LID development during L

Tac1-expressing neurons in the central amygdala predominantly mediate histamine-induced itch by receiving inputs from parabrachial Tac1-expressing neurons.

Itch is a distinct and bothersome sensation closely associated with a strong urge to scratch. Both the parabrachial nucleus (PBN) and the central amygdala (CeA) are responsive to itch stimuli and contain neurons that express tachykinin 1 (Tac1), which are known for their significant involvement in itch-induced scratching at both spinal and supraspinal levels. Significantly, the PBN neurons project their axons to form close connections with the CeA neurons. However, the role of the PBNTac1-CeATac1 pathway in modulating itch remains to be determined. We utilized immunohistochemistry, fiber photo

Electroacupuncture reduces visceral pain and anxiety associated with inflammatory bowel disease by inhibition of vHPC-to-mPFC pathway.

EA effectively treats gastrointestinal diseases, pain symptoms, and emotional disorder. Furthermore, vHPC and mPFC are two of the crucial nuclei involved in controlling chronic pain and anxiety-like behaviors. In the present study, it is investigated whether EA may reduce visceral pain and anxiety associated with inflammatory bowel disease (IBD) by inhibition of vHPC-to-mPFC pathway. We found that EA alleviated visceral hyperalgesia and anxiety in TNBS-treated IBD mice. EA decreased the numbers of c-Fos and neurogranin (labeled glutamatergic neurons) co-labeled neurons in both vHPC and mPFC. E

Parvalbumin neurons in the nucleus accumbens shell modulate seizure in temporal lobe epilepsy.

A growing number of clinical and animal studies suggest that the nucleus accumbens (NAc), especially the shell, is involved in the pathogenesis of temporal lobe epilepsy (TLE). However, the role of parvalbumin (PV) GABAergic neurons in the NAc shell involved in TLE is still unclear. In this study, we induced a spontaneous TLE model by intrahippocampal administration of kainic acid (KA), which generally induce acute seizures in first 2 h (acute phase) and then lead to spontaneous recurrent seizures after two months (chronic phase). We found that chemogenetic activation of NAc shell PV neurons c

Inhibition of prefrontal cortex parvalbumin interneurons mitigates behavioral and physiological sequelae of chronic stress in male mice.

Chronic stress leads to hypofunction of the medial prefrontal cortex (mPFC), mechanisms of which remain to be determined. Enhanced activation of GABAergic of parvalbumin (PV) expressing interneurons (INs) is thought to play a role in stress-induced prefrontal inhibition. In this study, we tested whether chemogenetic inhibition of mPFC PV INs after chronic stress can rescue chronic stress-related behavioral and physiological phenotypes. Mice underwent 2 weeks of chronic variable stress (CVS) followed by a battery of behavioral tests known to be affected by chronic stress exposure, e.g. an open
