Antifungal activity and mechanism of tetramycin against Alternaria alternata, the soft rot causing fungi in kiwifruit.

Kiwifruit rot caused by the fungus Alternaria alternata occurs in many countries, leading to considerable losses during kiwifruit production. In this study, we evaluated the antifungal activity and mechanism of tetramycin against kiwifruit soft rot caused by Alternaria alternata. Tetramycin exerted antifungal effects through the suppression of mycelial growth, conidial germination, and the pathogenicity of A. alternata. Scanning electron microscopic observations revealed that tetramycin destroyed the mycelial structure, causing the mycelia to twist, shrink, and even break. Furthermore, transmi

Influence of sex and geographical location on culturable midgut bacterial diversity of pink bollworm, Pectinophora gossypiella larvae and their enzyme production potential.

Gut microbes play a dynamic role in many metabolic and physiological activities in insects. Recent studies have shown the difference in the gut microbiota associated with both sex and geographical locations. Deciphering the gut microbiota hosted with respect to their function in an economic pest like pink bollworm, Pectinophora gossypiella, is essential to understand the relationship between insect and its gut microbiome. In the present study, culturable midgut bacteria were isolated from Bt cotton resistant P. gossypiella larvae (male and female) collected from different cotton growing region

Therapeutic effects of pentoxifylline on invasive pulmonary aspergillosis in immunosuppressed mice.

BACKGROUND: The most common and severe infection of Aspergillus fumigatus is invasive pulmonary aspergillosis (IPA), which is usually seen in immunocompromised patients. Neutropenia is the primary risk factor implicated in IPA; however, IPA also occurs in patients without neutropenia, namely, those who are immunosuppressed owing to long-term corticosteroid use. With IPA-associated mortality as high as 51-79%, novel and effective treatment strategies are urgently needed. Pentoxifylline (PTX) has been shown to competitively inhibit the family 18 chitinases in fungi, which may be an new antifunga

The chitobiose transporter, chbC, is required for chitin utilization in Borrelia burgdorferi.

BACKGROUND: The bacterium Borrelia burgdorferi, the causative agent of Lyme disease, is a limited-genome organism that must obtain many of its biochemical building blocks, including N-acetylglucosamine (GlcNAc), from its tick or vertebrate host. GlcNAc can be imported into the cell as a monomer or dimer (chitobiose), and the annotation for several B. burgdorferi genes suggests that this organism may be able to degrade and utilize chitin, a polymer of GlcNAc. We investigated the ability of B. burgdorferi to utilize chitin in the absence of free GlcNAc, and we attempted to identify genes involve

Pseudomonas protegens as a biocontrol agent against phytopathogenic fungi - mini review.

Pseudomonas protegens, a member of the P. fluorescens complex, is an important biocontrol agent with high potential in sustainable agriculture. It produces diverse secondary metabolites, including 2,4-diacetylphloroglucinol (DAPG), pyoluteorin (PLT), orfamides, and protegenins, as well as hydrolytic enzymes such as chitinases, which contribute to antifungal activity, fungal cell wall degradation, and induction of systemic resistance (ISR) in plants. Despite these insights, gaps remain in understanding the regulation of metabolite biosynthesis, variability of ISR across crops, and the field eff

Onchocerca volvulus Molting Inhibitors Identified through Scaffold Hopping.

The anthelmintic closantel has shown promise in abrogating the L3 molting of Onchocerca volvulus, the causative agent of the infectious disease onchocerciasis. In our search for alternative scaffolds, we utilized a fragment replacement/modification approach to generate novel chemotypes with improved chitinase inhibitory properties. Further evaluation of the compounds unveiled the potential of urea-tropolones as potent inhibitors of O. volvulus L3 molting.

Endochitinase Overexpression in Transgenic Tomato Plants Improves Resistance to Phytopathogens.

Engineering broad-spectrum disease resistance remains a major challenge in crop improvement because most resistance genes are effective against a limited range of pathogens. Pathogenesis-related (PR) proteins, particularly endochitinases, are promising targets for enhancing plant immunity due to their antimicrobial activity and ability to activate defense responses. Here, we generated transgenic tomato (Solanum lycopersicum L.) plants overexpressing an endochitinase gene to assess their potential for enhanced resistance against bacterial and fungal pathogens. Three independent overexpression l

Downregulating CHI3L2 via the STAT3 Pathway: The Mechanism of Calcitriol in Suppressing Psoriatic Inflammation and Keratinocyte Hyperproliferation.

Psoriasis is a chronic inflammatory skin disorder driven by immune dysregulation and keratinocyte hyperproliferation. Calcitriol, the active form of vitamin D, shows therapeutic potential, but its molecular mechanisms remain incompletely understood. CHI3L2 (chitinase-3-like protein 2) is a member of the chitinase-like protein family implicated in inflammatory and tissue-remodelling processes, yet its role in psoriasis has not been explored. Here, we investigated whether CHI3L2 mediates the anti-psoriatic effects of calcitriol. Topical application of calcitriol alleviated imiquimod (IMQ)-induce

Pharmacokinetic profile of sarcin and thionin from Aspergillus giganteus and in vitro validation against human fungal pathogen.

Fungal infections are more predominant in agricultural and clinical fields. Aspergillosis caused by Aspergillus fumigatus leads to respiratory failure in patients along with various illnesses. Due to the limitation of antifungal therapy and antifungal drugs, there is an emergence to develop efficient antifungal compounds (AFCs) from natural sources to cure and prevent fungal infections. The present study deals with the investigation of the mechanism of the active compounds from Aspergillus giganteus against aspergillosis. Primarily, the bioavailability and toxicological properties of antifunga

A comparative study of molecular diagnostic methods designed to detect the crayfish plague pathogen, Aphanomyces astaci.

Crayfish plague is the most important disease of freshwater crayfish with a significant impact on European species. We compared the analytical test sensitivity and specificity of three published PCR assays for the detection of Aphanomyces astaci, the causative agent of crayfish plague: a conventional PCR assay targeting the ITS region and two TaqMan(®) real time assays, targeting either the ITS region or the chitinase gene. We also tested a variation of the conventional assay, by changing one of the primers. Test specificity was assessed using DNA from a range of A. astaci strains and an array

Apical surface expression of aspartic protease Plasmepsin 4, a potential transmission-blocking target of the plasmodium ookinete.

To invade its definitive host, the mosquito, the malaria parasite must cross the midgut peritrophic matrix that is composed of chitin cross-linked by chitin-binding proteins and then develop into an oocyst on the midgut basal lamina. Previous evidence indicates that Plasmodium ookinete-secreted chitinase is important in midgut invasion. The mechanistic role of other ookinete-secreted enzymes in midgut invasion has not been previously examined. De novo mass spectrometry sequencing of a protein obtained by benzamidine affinity column of Plasmodium gallinaceum ookinete axenic culture supernatant

A chitin-like component in Aedes aegypti eggshells, eggs and ovaries.

An insoluble white substance was prepared from extracts of eggshells of Aedes aegypti, the yellow fever mosquito and dengue vector. Its infrared and proton NMR spectra were similar to that of standard commercial chitin. This putative chitin-like material, also obtained from ovaries, newly laid and dark eggs, was hydrolyzed in acid and a major product was identified by HPLC to be glucosamine. The eggshell acid hydrolysate was also analyzed by ESI-MS and an ion identical to a glucosamine monoprotonated species was detected. The presence of chitin was also analyzed during different developmental

Production of 6-methylsalicylic acid by expression of a fungal polyketide synthase activates disease resistance in tobacco.

Salicylic acid (SA) has been shown to act as a signal molecule that is produced by many plants subsequent to the recognition of potentially pathogenic microbes. Increases in levels of SA often trigger the activation of plant defenses and can result in increased resistance to subsequent challenge by pathogens. We observed that the polyketide 6-methylsalicylic acid (6-MeSA), a compound that apparently is not endogenous to tobacco, can mimic SA. Tobacco leaves treated with 6-MeSA show enhanced accumulation of the pathogenesis-related (PR) proteins PR1, beta-1,3-glucanase, and chitinase and also d

Unraveling the mechanism of action of the PepGAT peptide against Candida albicans through proteomics approaches.

Candida albicans is a major opportunistic human pathogen, frequently implicated in infections with increasing antifungal resistance. As such, the search for new therapeutic alternatives is essential. Therefore, synthetic antimicrobial peptides (SAMPs) have emerged as promising alternatives because resistance development against these molecules appears to occur less frequently than against many conventional antimicrobial drugs. Here, the mechanisms of action of the chitinase-derived synthetic peptide PepGAT, which is bioinspired by the chitinase of Arabidopsis thaliana, were evaluated against C

Paracoccin Overexpression in Paracoccidioides brasiliensis Enhances Fungal Virulence by Remodeling Chitin Properties of the Cell Wall.

BACKGROUND: The thermodimorphic fungi Paracoccidioides spp. are the etiological agents of paracoccidioidomycosis. Although poorly studied, paracoccin (PCN) from Paracoccidioides brasiliensis has been shown to harbor lectinic, enzymatic, and immunomodulatory properties that affect disease development. METHODS: Mutants of P. brasiliensis overexpressing PCN (ov-PCN) were constructed by Agrobacterium tumefaciens-mediated transformation. ov-PCN strains were analyzed and inoculated intranasally or intravenously to mice. Fungal burden, lung pathology, and survival were monitored to evaluate virulenc

Chitinolytic activity in nasal polyps.

BACKGROUND: Chitin is a recognition element for tissue infiltration by innate cells implicated in allergy and immunity. This process can be negatively regulated by vertebrate chitinases. Both acidic mammalian chitinase (AMCase) and chitotriosidase (ChT) have chitinolytic activity. This study aimed to determine the activities of AMCase and ChT in nasal polyps (NPs), as well as their in situ localization in NP tissue. METHODS: AMCase and ChT activities in NPs were compared with those in inferior turbinate tissue samples. Tissue samples were measured for AMCase and ChT activities at a range of p
