Discriminative pentobarbital stimulus in rats immediately after intravenous administration.

Rats with chronically implanted venous cannulas were trained to make differential active or passive shock-avoidance responses in a 2-compartment box at 15 sec after i.v. adminstration of pentobarbital (5 mg/kg) or saline. The discriminative pentobarbital stimulus persisted for several min after adminstration. The pentobarbital response also was elicited in tests at 15 sec after i.v. injection of chlordiazepoxide (2.5 and 5 mg/kg) or alcohol (500 mg/kg). Subsequent training at 10 min after i.p. injection of pentobarbital (10 mg/kg), followed by tests after i.p. administration of other drugs, in…

A new method for the simultaneous determination of 1,4-benzodiazepines and amfepramone as adulterants in phytotherapeutic formulations by voltammetry.

The use of synthetic pharmaceuticals in phytotherapeutics can be defined as an illegal practice, since these compounds are normally present as non-declared compounds in the phytotherapeutical formulations. This work aims to show the development of an analytical method based on adsorptive cathodic stripping voltammetry (AdCSV) for the simultaneous determination of 1,4-benzodiazepines and amfepramone. The developed method was used to measure seven benzodiazepines (clonazepam, flurazepam, alprazolam, midazolam, medazepam, chlordiazepoxide, and diazepam) and amfepramone in slimming formulations th…

Use and abuse of the benzodiazepines.

Since chlordiazepoxide was introduced in 1961, the benzodiazepines have had many important roles in the pharmacotherapy of various disorders. This drug class for the central nervous system has been considered one of the safest in use for 35 years, especially when the benzodiazepines are compared with the barbiturates they often replaced. The objective of this article is to provide an update on the availability and distribution of benzodiazepines around the world and to discuss their most common clinical applications. Adverse effects of benzodiazepines, observed after long-term therapeutic use …

Pharmacological analysis of hyperventilation in arthritic rats.

The study examined the validity of increased minute volume of ventilation as a measurement of chronic pain in arthritic rats. The opiates morphine and R 62 818 attenuated arthritic hyperventilation, but only at doses which also reduced the ventilatory response to CO2 in normal rats. The non-steroidal anti-inflammatory drugs (NSAIDs), indomethacin and suprofen, the corticosteroids, cortisone and dexamethasone, and the tranquillizers, haloperidol and chlordiazepoxide, were essentially ineffective except at doses that also produced anti-inflammatory and/or toxic effects. A combination of an in it…

Psychotropic drug use in an ambulatory elderly population.

Medication histories were obtained from 3,192 ambulatory elderly persons participating in a health screening program. A total of 2,009 (63%) women and 1,183 (37%) men were included in the drug use study. Flurazepam, pentobarbital and the combination of amobarbital and secobarbital were used by 4.5, 0.6 and 0.4% of participants, respectively, and these three agents accounted for 87.6% of all prescribed hypnotic drugs. Diazepam, meprobamate and chlordiazepoxide were the most commonly used minor tranquilizers. Barbiturates were contained in nearly one fourth of minor tranquilizers used by these e…

Examining behavioural test sensitivity and locomotor proxies of anxiety-like behaviour in zebrafish.

This study assessed the sensitivity of four anxiety-like behaviour paradigms in zebrafish: the novel tank dive test, shoaling test, light/dark test, and the less common shoal with novel object test. A second goal was to measure the extent to which the main effect measures are related to locomotor behaviours to determine whether swimming velocity and freezing (immobility) are indicative of anxiety-like behaviour. Using the well-established anxiolytic, chlordiazepoxide, we found the novel tank dive to be most sensitive followed by the shoaling test. The light/dark test and shoaling plus novel ob…

The stimulus properties of gamma-hydroxybutyrate.

Fourteen rats were trained to discriminate the effects of gamma-hydroxybutyrate (GHB) (sodium salt, 200 mg/kg) and saline in a two-lever choice task using a fixed ratio 10 schedule of water reinforcement. Intermediate responding, i.e., responding not fully appropriate for either training condition was observed in tests following morphine, lysergic acid diethylamide, chlordiazepoxide, and the presumed GABA-mimetics muscimol, gamma-butyrolactone, baclofen, and 3-aminopropane sulfonic acid. Naloxone blocked the intermediate results following morphine, but had no effect on GBH-induced stimulus con…

A study of the mutagenicity of anesthetics and their metabolites.

The commonly used volatile anesthetics, several of their metabolites, and drugs frequently employed by the anesthesiologist were screened for mutagenicity in the Salmonella/rat-liver microsomal assay system developed by Dr. B. Ames and his colleagues. Chloral hydrate, both a sedative and metabolite of trichloroethylene, was found to be weakly mutagenic. Other compounds testing including halothane, isoflurane, methoxyflurane, diazepam and chlordiazepoxide were not mutagenic. Non-volatile compounds were tested for their ability to inhibit growth of bacterial strains with decreased capacity to re…

Drug therapy in the treatment of minimal brain dysfunction.

The pharmacotherapy of minimal brain dysfunction (MBD) is reviewed. Studies using central nervous system (CNS) stimulants (amphetamines and methylphenidate, deanol, pemoline, caffeine), antidepressants (imipramine and desipramine), anticonvulsants (phenytoin and primidone), antianxiety agents (chlordiazepoxide, hydroxyzine, meprobamate), antipsychotic agents (phenothiazines, thioxanthenes, butyrophenones) and miscellaneous agents (benztropine, thyrotropin-releasing hormone, megavitamins) are discussed. When drugs are indicated, the CNS stimulants are the agents of choice in the treatment of MB…

Drug effects in squirrel monkeys trained on a multiple schedule with a punishment contingency.

The behavior of four monkeys trained on a multiple schedule was differentially sensitive to selected pharmacological agents. The three components of the multiple schedule were: (1) a variable-interval schedule in which responses were reinforced on the average of once per minute; (2) a concurrent schedule in which every tenth response was reinforced and every fifteenth response, on the average, was shocked; and, (3) a neutral stimulus in the presence of which responses were neither reinforced nor shocked. Pentobarbital, chlordiazepoxide, and meprobamate increased responding during each of the c…

Inverse agonist properties of the FG 7142 discriminative stimulus.

A two-lever, food-motivated discrimination was established between the benzodiazepine receptor partial inverse agonist FG 7142 (5.0 mg/kg) and its vehicle. The FG 7142 discriminative stimulus was pharmacologically characterized by testing trained rats with a variety of benzodiazepine receptor ligands. Administration of the inverse agonist DMCM (0.15-0.30 mg/kg) dose-dependently mimicked the FG 7142 stimulus. In contrast, the benzodiazepine receptor agonist chlordiazepoxide, partial agonist ZK 91 296, mixed agonist/antagonist CGS 9896 and antagonist RO 15-1788 blocked the FG 7142 cue. These res…

Effects of wisanine and dihydrowisanine on aggressive behaviour in chicks.

The effects of wisanine and dihydrowisanine were studied on stereotyped and aggressive behaviour induced by apomorphine (0.5 mg/kg s.c.) in chicks. Pretreatment of chicks with wisanine (0.3-0.6 g/kg i.p.) and dihydrowisanine (0.2-0.4 g/kg i.p.) dose dependently antagonised fighting episodes and pecking of the chick's own feet and those of other chicks. It has been suggested that the antagonism is due to dopaminergic receptor blockade and demonstrates the antipsychotic effect of these drugs.

Benzodiazepines: some aspects of their clinical pharmacology.

Benzodiazepine derivatives are the most commonly prescribed anti-anxiety agents in clinical practice. Six benzodiazepine anxiolytics are now available in the United States. Additional drugs are used in other parts of the world, and many others are in various stages of clinical testing. All these benzodiazepine derivatives have similar neuropharmacological properties--they reduce anxiety, produce sedation and sleep, have anticonvulsant effects, and can produce muscle relaxation. Faced with this bewildering array of drugs from the same class which are very similar in intrinsic effects upon the b

Self-control, biofeedback, and change in 'psychosomatic' approach.

The new psychosomatic treatment modalities of biofeedback and relaxation techniques have in common an emphasis on self-control or self-regulation. The surging popularity of the self-control techniques at present may reflect a change in the self-image of medicine, which emphasizes its role as a collaborator of the patient in his self-control of disease processes. The modern concept of psychosomatic medicine emphasizes a systems or field approach to evaluation and treatment rather than 'psychogenic' causes of physical illness. The self-control treatment modalities form an important part of the a

Comparison of the effects of the benzodiazepine midazolam and three serotonin antagonists on a consummatory conflict paradigm.

A consummatory conflict procedure that involves an abrupt reduction in magnitude of an expected reward (negative contrast) has been shown to be particularly sensitive to the effects of anxiolytic agents. As previously reported with chlordiazepoxide, another benzodiazepine (BDZ), midazolam released suppressed consummatory performance in a dose-dependent manner. This effect was not due to a general appetitie stimulatory effect of the drug. The effects of three 5-HT antagonists on negative contrast were examined to evaluate the role serotonin may play in the anxiolytic action of BDZ. Methysergide…
