Antenatal and early life tobacco smoke exposure in an African birth cohort study.

BACKGROUND: Exposure to tobacco smoke in African infants has not been well studied, despite the high burden of childhood respiratory disease in these communities. OBJECTIVE: To investigate the prevalence of antenatal and early life tobacco smoke exposure and associations with infant birth outcomes in an African birth cohort, the Drakenstein Child Health Study. METHODS: Self-report questionnaires assessing maternal and household smoking were administered. Maternal and infant urine cotinine testing was conducted antenatally, at birth and at 6-10 weeks of life to measure tobacco smoke exposure.

Green tea extract supplement inhibition of HMGB1 release in rats exposed to cigarette smoke.

Tobacco-smoke exposure is linked to carcinogenic, oxidative and inflammatory cellular reactions. Green tea has been reported to have anti-release properties against various pro-inflammatory cytokines. To determine the effects of green tea extract (GTE) on serum high mobility group box-1 (HMGB1) levels in rats exposed to cigarette smoke (CS), we divided rats into 4 treatment groups: (1) CS only, (2) dietary supplement with GTE (3 mg/d) and CS (GCS1), (3) dietary supplement with GTE (4.5 mg/d) and CS (GCS2) and (4) a control group. HMGB1 and cotinine serum levels were analyzed by ELISA. The aver

Factors Predicting 6-Month Smoking Cessation in Korean Adults.

The purpose of this study was to identify factors predicting 6-month smoking cessation in Korean adult smokers. This descriptive correlation study assessed levels of urine cotinine, serum cotinine, serum serotonin, and 5-hydroxyindoleacetic acid; tobacco withdrawal symptoms; and resilience among 164 Korean adult smokers. Serum cotinine levels were negatively related to resilience at six months (r = -.42, p = .019), but were positively related to the amount of smoking (r = .32, p = .008) and with the Week 6 tobacco withdrawal symptoms score (r = .48, p = .001, n = 41). Higher resilience was ass

Effects of excessive alcohol drinking on nicotine biotransformation in rats.

Alcohol and nicotine (tobacco smoke) are often used together, and taking both addictive substances is associated with an increased risk of certain diseases. It is extremely important to understand the pharmacodynamic and pharmacokinetic mechanisms of the interaction between nicotine and ethanol, which are still not fully understood. The study aimed to evaluate the influence of chronic alcohol consumption on nicotine biotransformation in ethanol-preferring and non-preferring male and female rats. Rats were divided into four groups depending on their alcohol preferences and gender. Nicotine, nor

Urinary Cyanoethyl Mercapturic Acid, a Biomarker of the Smoke Toxicant Acrylonitrile, Clearly Distinguishes Smokers From Nonsmokers.

INTRODUCTION: Cyanoethyl mercapturic acid (CEMA) is a urinary metabolite of acrylonitrile, a toxicant found in substantial quantities in cigarette smoke, but not in non-combusted products such as e-cigarettes or smokeless tobacco and rarely in the diet or in the general human environment. Thus, we hypothesized that CEMA is an excellent biomarker of combusted tobacco product use. AIMS AND METHODS: We tested this hypothesis by analyzing CEMA in the urine of 1259 cigarette smokers (urinary cotinine ≥25 ng/mL) and 1191 nonsmokers. The analyses of CEMA and cotinine were performed by validated liqu

Design and optimization of a total vaporization technique coupled to solid-phase microextraction.

Solid-phase microextraction (SPME) is a popular sampling technique in which chemical compounds are collected with a sorbent-coated fiber and then desorbed into an analytical instrument such as a liquid or gas chromatograph. Typically, this technique is used to sample the headspace above a solid or liquid sample (headspace SPME), or to directly sample a liquid (immersion SPME). However, this work demonstrates an alternative approach where the sample is totally vaporized (total vaporization SPME or TV-SPME) so that analytes partition directly between the vapor phase and the SPME fiber. The imple

Biological monitoring of environmental exposure to mutagens among high school students in Jeddah.

Mutagenic and/or carcinogenic substances, which are present in the environment, are numerous. They include electrophilic compounds, polycyclic aromatic compounds such as benzo(a)pyrene which is a polyaromatic hydrocarbon. It is well known to be mutagenic and carcinogenic. Biological monitoring that depends on estimating a substance or one of its metabolites or one of its effective components in biological fluids such as urine and blood was carried out. Thioether concentration in urine indicates absorption of mutagenic and/or carcinogenic substances. Thiocyanates' concentration in urine was use

Formation of N-(2-hydroxyethyl)valine due to exposure to ethylene oxide via tobacco smoke: A risk factor for onset of cancer.

Human exposure to ethylene oxide (EtO) occurs mainly through inhalation of occupational polluted air and tobacco smoke. EtO is able to react with DNA and proteins producing some molecular adducts. One of these, resulting from reaction between EtO and valine in hemoglobin, is N-(2-hydroxyethyl) valine (HOEtVal). This adduct represents a biological effective dose marker, the level of which correlates linearly with the alkylating activity occurring in DNA. The aim of the present study was to measure HOEtVal in 146 urbanized adult and healthy subjects, nonoccupationally exposed to EtO, and to corr

Assessment and Counseling Gaps Among Former Smokers Eligible for Lung Cancer Screening in US Adults : A Cross-Sectional Analysis of National Health and Nutrition Examination Surveys (NHANES), 2013-2018.

BACKGROUND: Lung cancer screening (LCS) for former and current smokers requires that current smokers are counseled on tobacco treatment. In the USA, over 4 million former smokers are estimated to be eligible for LCS based on self-report for "not smoking now." Tobacco use and exposure can be measured with the biomarker cotinine, a nicotine metabolite reflecting recent exposure. OBJECTIVE: To examine predictors of tobacco use and exposure among self-reported former smokers eligible for LCS. DESIGN: Cross-sectional study using the 2013-2018 National Health and Nutrition Examination Survey. PAR

Prenatal exposure to tobacco and adverse birth outcomes: effect modification by folate intake during pregnancy.

BACKGROUND: Fetal exposure to tobacco increases the risk for many adverse birth outcomes, but whether diet mitigates these risks has yet to be explored. Here, we examined whether maternal folate intake (from foods and supplements) during pregnancy modified the association between prenatal exposure to tobacco and with preterm delivery, small-for-gestational age (SGA) births, or neonatal adiposity. METHODS: Mother-child pairs (n = 701) from Healthy Start were included in this analysis. Urinary cotinine was measured at ~ 27 weeks gestation. Diet was assessed using repeated 24-h dietary recalls.

Racial differences in exposure and glucuronidation of the tobacco-specific carcinogen 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK).

BACKGROUND: In the United States, Blacks who smoke cigarettes have a higher mean blood concentration of the nicotine metabolite cotinine than White smokers. It has not been determined whether there are racial differences in the exposure to the cigarette smoke carcinogen 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) and in the detoxification of NNK metabolites. METHODS: A community-based cross-sectional survey of 69 Black and 93 White smokers was conducted in lower Westchester County, New York. Cigarette yields of tar and nicotine and markers of exposure to tobacco smoke.

Although cigarette yields of tar and nicotine have been declining since the early 1970s, little information is available for the general population on the consequences of their use on exposure to tobacco combustion products. In a population-based sample of 298 smokers, the majority of whom were Hispanic, we examined the relationships between yields of cigarettes currently smoked and levels of salivary cotinine and end-expired carbon monoxide. Spearman correlation coefficients between the current number of cigarettes smoked and cotinine (r = 0.52) or carbon monoxide (r = 0.51) were higher than

Environmental tobacco smoke exposure during pregnancy has limited effect on infant birthweight and umbilical vein endothelial nitric oxide synthase.

INTRODUCTION: Women who smoke, deliver significantly smaller infants. These infants have reduced levels of the vasodilator endothelial nitric oxide synthase (eNOS) levels in the umbilical vessels, which may reduce fetal growth. Serum cotinine, the degradation product of nicotine, can be used to determine the level of tobacco exposure. Newborns of environmental smokers are suggested to be smaller and shorter in weight, length, and head circumference. eNOS levels have not yet been studied in these infants. We investigated the existence of a relation between maternal environmental tobacco smoke e

Elevated blood levels of carcinogens in passive smokers.

The hypothesis that involuntary exposure to tobacco smoke--passive smoking--results in greater risk of cancer was assessed by measuring the levels of two known carcinogens in the blood of 57 nonsmokers with varying degrees of involuntary exposure, including six heavily exposed bartenders. The concentrations of hemoglobin adducts of 4-aminobiphenyl, a bladder carcinogen, were significantly higher in subjects with confirmed involuntary exposure (plasma cotinine concentrations between 2 and 23 ng/ml) compared with subjects with undetectable levels of cotinine. Similarly, adducts of 3-aminobipheny

[Passive smoking and positive intracutaneous allergen tests in children with respiratory diseases].

In children with respiratory disease (bronchitis, pneumonia and bronchial asthma) skin hypersensitivity towards tobacco leaf crude extract has been tested and related to urinary cotinine level, the latter being most informative in assessing exposure to cigarette smoke. Besides routine intracutaneous tests a mixture of seven main brands of Bulgarian tobaccos (Burley, Virginia, Oriental etc.) has been applied as allergen. Urinary cotinine has been estimated by the method of BARLOW et al. [1]. Skin sensitization towards tobacco was positive in 2 (8%) of the children hospitalized for pneumonia and

Adductomics of Newborn Dried Blood Spots Detects Constituents of Maternal Smoking During Pregnancy and Associated Oxidative Stress Exposure.

Maternal Smoking During Pregnancy (MSDP) remains a major source of fetal toxicant exposure. We applied adductomics to profile reactive adducts at the human serum albumin cysteine-34 (HSA-Cys34) locus, which integrates longer-term exposures. HSA-Cys34 adducts formed by acrylonitrile and ethylene oxide, two tobacco-related toxicants previously linked to smoking in adults, were quantified and compared with cotinine and MSDP status. Their relationships with other reactive adducts were also examined. Neonatal dried blood spots (DBS) from 110 children were analyzed. Cotinine and 55 Cys34 adducts wer

Biochemical Validation of Self-Reported Smokeless Tobacco Abstinence among Smokeless Tobacco Users: Results from a Clinical Trial of Varenicline in India.

The validity of self-reported tobacco use is often questioned given the potential for underestimation of use. This study used data from a double-blind, placebo-controlled clinical trial of varenicline for smokeless tobacco dependence in India to evaluate the accuracy of self-reported smokeless tobacco cessation using biochemical validation procedures and to evaluate correlates of reporting inaccuracy. Smokeless tobacco users attending a dental clinic at AIIMS were randomized to placebo or varenicline; all participants received counseling. Detailed smokeless tobacco use was recorded and abstine
