Comparative analysis of the effects of Origanum vulgare essential oil and carvacrol nanoemulsified and non-nanoemulsified forms against Salmonella Typhimurium strains.

This study investigates the potential of Origanum vulgare L. (OVEO), carvacrol (CAR), and their nanoemulsions (nOVEO and nCAR) in subinhibitory concentrations as antimicrobial agents against S. typhimurium strains in vitro and in situ. The minimum inhibitory concentrations (MIC) ranged from 0.59 to 0.60 mg/mL for the nanoemulsions and 0.68 to 1.5 mg/mL for their non-nanoemulsified counterparts. Growth kinetics analysis showed that all treatments at 1/4 MIC, 1/2 MIC, and MIC reduced the growth rate and prolonged the lag phase. Flow cytometry analysis revealed that these treatments caused permea

New Benzothiazole-Monoterpenoid Hybrids as Multifunctional Molecules with Potential Applications in Cosmetics.

The Thymus vulgaris and Origanum vulgare essential oils (contained thymol and carvacrol in a range of 35-80%) are used in various products in the fields of medicine, cosmetics, and foods. Molecular hybridization between benzothiazole (BT) and phenolic monoterpenoids is a promising method for the development of biologically active compounds. New benzothiazole-monoterpenoid hybrids were synthesized through a regioselective α-amidoalkylation reaction of thymol and carvacrol with high yields (70-96%). This approach is both simple and cost-effective, employing easily accessible and inexpensive reag

Tyramine-modified Ag nanoparticle loaded with carvacrol for antibacterial, anti-inflammatory, and anticoagulant treatment of sepsis.

Carvacrol (CA) exhibits excellent antibacterial and anti-inflammatory activities, but its clinical application is limited by low bioavailability and targeting capability. To overcome these limitations, tyramine-modified Ag nanoparticle loaded with CA (Ag-CA@Tyr) was prepared. It exhibited a regular spherical shape and demonstrated good drug release characteristics, stability, and biosafety. It significantly reduced the levels of pro-inflammatory cytokines and inhibited the growth of bacteria. In animal experiments, Ag-CA@Tyr alleviated liver and lung damage in sepsis rats, while also exhibitin

Carvacrol Inhibits Expression of Transient Receptor Potential Melastatin 7 Channels and Alleviates Zinc Neurotoxicity Induced by Traumatic Brain Injury.

Carvacrol is a monoterpenoid phenol produced by aromatic plants such as oregano. Although the exact mechanism by which carvacrol acts has not yet been established, it appears to inhibit transient receptor potential melastatin 7 (TRPM7), which modulates the homeostasis of metal ions such as zinc and calcium. Several studies have demonstrated that carvacrol has protective effects against zinc neurotoxicity after ischemia and epilepsy. However, to date, no studies have investigated the effect of carvacrol on traumatic brain injury (TBI)-induced zinc neurotoxicity. In the present study, we investi

Carvacrol acts as a potent selective antagonist of different types of nicotinic acetylcholine receptors and enhances the effect of monepantel in the parasitic nematode Ascaris suum.

The neuromuscular system of parasitic nematodes has proven to be an efficient pharmacological target for antihelmintics. Some of the most frequently used antiparasitic drugs are agonists or antagonists of nicotinic acetylcholine receptors (nAChRs). The antinematodal mechanism of action of carvacrol involves the inhibition of parasite muscle contraction. We have examined the interaction of carvacrol with antinematodal drugs that are agonists of different subtypes of nAChRs and monepantel, which is a non-competitive antagonist of this receptor in A. suum. Additionally, we investigated the effect

Carvacrol and human health: A comprehensive review.

Carvacrol (CV) is a phenolic monoterpenoid found in essential oils of oregano (Origanum vulgare), thyme (Thymus vulgaris), pepperwort (Lepidium flavum), wild bergamot (Citrus aurantium bergamia), and other plants. Carvacrol possesses a wide range of bioactivities putatively useful for clinical applications such antimicrobial, antioxidant, and anticancer activities. Carvacrol antimicrobial activity is higher than that of other volatile compounds present in essential oils due to the presence of the free hydroxyl group, hydrophobicity, and the phenol moiety. The present review illustrates the sta

Larvicidal potential of carvacrol and terpinen-4-ol from the essential oil of Origanum vulgare (Lamiaceae) against Anopheles stephensi, Anopheles subpictus, Culex quinquefasciatus and Culex tritaeniorhynchus (Diptera: Culicidae).

Mosquito-borne diseases represent a deadly threat for millions of people worldwide. However, the use of synthetic insecticides to control Culicidae may lead to resistance, high operational costs and adverse non-target effects. Nowadays, plant-borne mosquitocides may serve as suitable alternative in the fight against mosquito vectors. In this study, the mosquito larvicidal activity of Origanum vulgare (Lamiaceae) leaf essential oil (EO) and its major chemical constituents was evaluated against the malaria vectors Anopheles stephensi and An. subpictus, the filariasis vector Culex quinquefasciatu

Use of rotary fluidized-bed technology for development of sustained-release plant extracts pellets: potential application for feed additive delivery.

The aim of this study was to develop sustained release plant extracts as a potential alternative to antibiotic growth promoters for growing pigs. Pellets with a core based on microcrystalline cellulose and 3 active compounds (eugenol, carvacrol, and thymol) were prepared using rotary fluidized-bed technology. Two particle sizes were produced that had a mean size of approximately 250 and 500 mum. Results show the process was able to produce pellets with a spherical and homogenous form when 10% of the active compounds were incorporated into the core. When active compounds were increased to 20%,

Spinosin-carvacrol co-treatment attenuates deltamethrin-induced hepatotoxicity via modulation of oxidative stress, inflammation, and apoptosis in mice.

Exposure to certain pesticides remains a significant environmental and public health concern due to their capacity to induce oxidative damage in vital organs. Deltamethrin (DEL), a widely used synthetic pyrethroid, disrupts hepatic function primarily through oxidative stress-mediated mechanisms. This study investigated the protective effects of spinosin (SP) and carvacrol (CAR), administered individually or in combination, against DEL-induced liver toxicity and explored the underlying molecular pathways. Hepatic injury was induced in male Swiss albino mice by oral gavage of DEL (15 mg/kg bw).

Carvacrol Improves Vascular Function in Hypertensive Animals by Modulating Endothelial Progenitor Cells.

Carvacrol, a phenolic monoterpene, has diverse biological activities, highlighting its antioxidant and antihypertensive capacity. However, there is little evidence demonstrating its influence on vascular regeneration. Therefore, we evaluated the modulation of carvacrol on endothelial repair induced by endothelial progenitor cells (EPC) in hypertension. Twelve-week-old spontaneously hypertensive rats (SHR) were treated with a vehicle, carvacrol (50 or 100 mg/kg/day), or resveratrol (10 mg/kg/day) orally for four weeks. Wistar Kyoto (WKY) rats were used as the normotensive controls. Their systol

Carvacrol treatment opens Kir6.2 ATP-dependent potassium channels and prevents apoptosis on rat testis following ischemia-reperfusion injury model.

Testicular torsion is a urological problem that causes subfertility and testicular damage in males. Testis torsion and detorsion lead to ischemia-reperfusion (IR) injury in the testis. Testicular IR injury causes the increase of reactive oxygen species (ROS), oxidative stress (OS) and germ cell-specific apoptosis. In this study, we aimed to investigate whether Carvacrol has a protective effect on testicular IR injury and its effects on Kir6.2 channels, which is a member of adenosine triphosphate (ATP)-dependent potassium channels. In the study, 2-4 months old 36 albino Wistar rats were used. F

Phytochemical- and Cyclodextrin-Based Pickering Emulsions: Natural Potentiators of Antibacterial, Antifungal, and Antibiofilm Activity.

We present self-assembled Pickering emulsions containing biocidal phytochemical oils (carvacrol and terpinen-4-ol) and β-cyclodextrin able to potentiate the antimicrobial and antibiofilm activity of miconazoctylium bromide. The carvacrol-containing emulsion is 2-fold more sensitive against C. albicans and S. aureus and highly active against E. coli, compared to the commercial cream containing miconazole nitrate. Moreover, this emulsion shows a synergistic effect against fungi, additive responses against bacteria, and remarkable staphylococcal biofilm eradication. These results are associated w

Experimental and molecular docking analyses of antibacterial activity in moroccan Rosmarinus officinalis essential oil.

Antimicrobial resistance motivates antibacterial agents with multi-target mechanisms. We evaluated Moroccan rosemary essential oil (ROEO) against four pathogens (Escherichia coli, Citrobacter freundii, Staphylococcus aureus, Enterococcus faecalis) by disk diffusion and broth microdilution. ROEO inhibited all strains (zones 11.3-21.0 mm); activity was bactericidal for E. faecalis (MBC/MIC = 2.0) and bacteriostatic for others (MBC/MIC > 4). Bioassay-guided silica chromatography, using E. faecalis as the pre-specified indicator, localised activity to a polar fraction (F8; 18.5% of the oil; 14.0 ±

Chemical Composition of Eriodictyon californicum (California Yerba Santa) Cultivated in Ontario, Oregon, USA.

Leaves from California yerba santa (Eriodictyon californicum) have been used historically by indigenous peoples for medicinal purposes. Recent research has ascribed potential pharmaceutical effects to leaf polyphenols, without a consideration of other constituents. Based on prior analyses of polyphenols in leaves sampled in nature, five accessions known to be rich in sterubin and five accessions known to be rich in eriolic acid C were grown from seeds in Ontario, Oregon, and samples of their leaves were harvested and evaluated for their essential oil and polyphenol contents. The major essentia
