Infradian Rhythm of the Content of Secretory Granules in Pinealocyte Cytoplasm in Mice and Rats.

The numerical density of secretory granules dense-core vesicles (DCV) in the cytoplasm of pinealocytes of the pineal gland was estimated by transmission electron microscopy in male white mice and Wistar rats. The 3-day biorhythm and lunaphase changes in the DCV content in the perikaryon and the processes of pinealocytes, which are manifested significantly in different seasons of the year, are established. The three-day biorhythm in adult male mice in comparison with younger male rats is not expressed uniformly in different phases of the moon. The in-phase manifestation of infradian biorhythms

Drug-induced endovesiculation of erythrocytes is modulated by the dynamics in the cytoskeleton/membrane interaction.

Recent studies on erythrocyte membrane fluctuations revealed that the erythrocyte cytoskeleton actively modulates its membrane association thereby regulating crucial membrane properties. Cationic amphiphilic drugs like chlorpromazine are known to induce a cup-like cell shape and vesicle formation into the cell interior, effectors of this process, however, are largely unknown. Using flow cytometry, this study explored conditions that influence endovesiculation induced by chlorpromazine. We found that inhibitors of membrane fluctuations, like ATP depletion, vanadate or fluoride, also inhibited e

Comparative biochemistry of Giardia, Hexamita and Spironucleus: Enigmatic diplomonads.

The diplomonad genera are here represented by three highly diverse species, both free-living (Hexamita inflata), and parasitic (Spironucleus vortens and Giardia intestinalis). All three are moderately aerotolerant flagellates, inhabiting environments where O2 tensions are low and fluctuating. Many diplomonads are opportunistic pathogens of avian, terrestrial and aquatic animals. Hexamitids inhabit deep waters and sediments of lakes and marine basins, S. vortens commonly infects the intestinal tract of ornamental fish, particularly of cichlids and cyprinids, and G. intestinalis, the upper intes

Repairing a torn cell surface: make way, lysosomes to the rescue.

Biological membranes are often described as 'self-sealing' structures. If indeed membranes do have an inherent capacity for repair, does this explain how a cell can rapidly reseal a very large (1-1000 microm(2)) disruption in its plasma membrane? It is becoming increasingly clear that, in nucleated animal cells, the cytoplasm plays an active and essential role in resealing. A rapid and apparently chaotic membrane fusion response is initiated locally in the cytoplasm by the Ca(2+) that floods in through a disruption: cytoplasmic vesicles are thereby joined with one another (homotypically) and w…

Aggregation and beyond: alpha-synuclein-based biomarkers in synucleinopathies.

Parkinson's disease is clinically known for the loss of dopaminergic neurons in the substantia nigra pars compacta and accumulation of intraneuronal cytoplasmic inclusions rich in alpha-synuclein called 'Lewy bodies' and 'Lewy neurites'. Together with dementia with Lewy bodies and multiple system atrophy, Parkinson's disease is part of a group of disorders called synucleinopathies. Currently, diagnosis of synucleinopathies is based on the clinical assessment which often takes place in advanced disease stages. While the causal role of alpha-synuclein aggregates in these disorders is still debat

Pathological changes induced by BaH1, a hemorrhagic proteinase isolated from Bothrops asper (Terciopelo) snake venom, on mouse capillary blood vessels.

The pathological changes induced in capillaries by BaH1, a hemorrhagic metalloproteinase isolated from the venom of Bothrops asper, were studied after i.m. injection in mouse gastrocnemius. Hemorrhage was observed macroscopically, and corroborated histologically, within the first 5 min. At the ultrastructural level, the earliest changes in endothelial cells, observed 1 min after toxin administration, consisted of a decrease in the number of pinocytotic vesicles, the presence of blebs and cytoplasmic projections pinching off to the vascular lumen and the detachment of endothelial cells from the

Bicarbonate absorption in eel intestine: evidence for the presence of membrane-bound carbonic anhydrase on the brush border membranes of the enterocyte.

Bicarbonate absorptive fluxes through the isolated intestine of the European eel (Anguilla anguilla) were evaluated by the pH-stat method under short-circuited conditions. It was found that bicarbonate absorptive flux was dependent on the luminal Na+ and was inhibited by luminal 4-acetamido-4' stilbene-2-2' disulfonic acid (SITS; 2.5 x 10(-4) M) and luminal acetazolamide (10(-4) M), while luminal amiloride (1 mM) was without effect. Furthermore, by using brush border membrane vesicles (BBMV) isolated from eel intestine, the existence of two carbonic anhydrase (CA) isoforms, one tightly associa

Intramuscular streptomycin effect on dark cells of utricle in guinea pigs.

Streptomycin has been used in the treatment of Meniere's disease for almost half a century. Clinical studies showed that streptomycin may eliminate vertigo attacks and stabilize or improve hearing in the majority of patients. Animal experiments have demonstrated severe damage to the vestibular hair cells after streptomycin treatment. This study is to observe the effect of intramuscular streptomycin on dark cells of utricle. Guinea pigs that had received intramuscular injections of streptomycin sulfate 400 mg/kg per day for 1 and 2 weeks were studied. The dark cells of utricle were observed und

The Treatment of Hairy Cell Leukemia: A Review.

Hairy cell leukemia is a chronic lymphoproliferative malignancy first described by Bouroncle et al. over 30 years ago(1). Representing less than 2% of adult leukemias, hairy cell leukemia has become the focus of intense interest over the past few years as a result of therapeutic advances. The malignant cell has characteristic irregular cytoplasmic projections visible on the peripheral blood smear and cytoplasmic vesicles staining positive for tartrate-resistant acid phosphatase. The origin of the hairy cell appears to be a preplasma cell B lymphocyte based on immunoglobulin gene rearrangements…

First Characterization of the Neospora caninum Dense Granule Protein GRA9.

The obligate intracellular apicomplexan parasite Neospora caninum (N. caninum) is closely related to Toxoplasma gondii (T. gondii). The dense granules, which are present in all apicomplexan parasites, are important secretory organelles. Dense granule (GRA) proteins are released into the parasitophorous vacuole (PV) following host cell invasion and are known to play important roles in the maintenance of the host-parasite relationship and in the acquisition of nutrients. Here, we provide a detailed characterization of the N. caninum dense granule protein NcGRA9. The in silico genomic organizatio

Lansoprazole is an uncompetitive inhibitor of tissue-nonspecific alkaline phosphatase.

Lansoprazole, a known H(+)/K(+)-ATPase inhibitor, is currently used as a therapeutical option for the initial treatment of gastroesophageal reflux disease. Recently, lansoprazole has been found to be an inhibitor of cytosolic PHOSPHO1 (a phosphatase which hydrolyses phosphocholine and phosphoethanolamine), providing a possible therapeutical target to cure pathological mineralization. Since PHOSPHO1 is present inside matrix vesicles, we tested the effect of lansoprazole on matrix vesicles containing several key enzymes for the mineralization process including tissue-nonspecific alkaline phospha

Lipid imbalance in the neurological disorder, Niemann-Pick C disease.

Niemann-Pick C (NPC) disease is a progressive neurological disorder in which cholesterol, gangliosides and bis-monoacylglycerol phosphate accumulate in late endosomes/lysosomes. This disease is caused by mutations in either the NPC1 or NPC2 gene. NPC1 and NPC2 are involved in egress of lipids, particularly cholesterol, from late endosomes/lysosomes but the precise functions of these proteins are not clear. An important question regarding the function of NPC proteins is: why do mutations in these ubiquitously expressed proteins have such dire consequences in the brain? This review summarizes th

Modes of action of a small molecule antiviral compound targeting yellow fever virus NS4B protein.

Yellow fever virus (YFV) replicates its RNA genome in membranous vesicles derived from the invagination of endoplasmic reticulum membranes, designated as replication organelles (ROs). Nonstructural protein 4B (NS4B) of flaviviruses play essential roles in the biogenesis of ROs and evasion of innate immune responses. We report herein that the binding of an antiviral agent, acetic acid benzodiazepine (BDAA), to YFV NS4B not only rapidly inhibits YFV RNA synthesis, but also induces the activation of cytoplasmic double-stranded RNA (dsRNA)-sensing pathways to accelerate the apoptosis of infected c

Wiskott-Aldrich syndrome proteins in the nucleus: aWASH with possibilities.

Actin and proteins that regulate its dynamics or interactions have well-established roles in the cytoplasm where they function as key components of the cytoskeleton to control diverse processes, including cellular infrastructure, cellular motility, cell signaling, and vesicle transport. Recent work has also uncovered roles for actin and its regulatory proteins in the nucleus, primarily in mechanisms governing gene expression. The Wiskott Aldrich Syndrome (WAS) family of proteins, comprising the WASP/N-WASP, SCAR/WAVE, WHAMM/JMY/WHAMY, and WASH subfamilies, function in the cytoplasm where they

Effects of colchicine on the gallbladder of the mouse.

The effects of colchicine on the mouse gallbladder followed a course depending on the dosage given (0.4-4 mg/100 g body weight). Following 0.5 mg/100 g, by 16 h there was a marked cholestasis with dilatation of the gallbladder and steatosis. There were progressive alterations in the Golgi apparatus and accumulation of vesicles. The apical mucous droplets decreased in number and became pleomorphic and dispersed throughout the cytoplasm. Lipid droplets appeared in numbers on the epithelial cytoplasm. By 48 h the tissues had reverted to normal appearances. When cholecystokinin, pilocarpine or cer

The sulcus medianus organ in the fourth cerebral ventricle of the rabbit: a previously undescribed circumventricular organ.

The sulcus medianus of the fourth ventricle in the rabbit contains a previously undescribed circumventricular organ. The cells in this region are predominantly nonciliated. The apical surface of each cell has characteristic microvillous projections with pinocytotic vesicles. The cell body is elongated, with the nucleus situated in the body or basal process; the cytoplasm of the cell is pale and contains microtubules and filaments. The general morphology of the cells is similar to tanycytes found in other circumventricular organs. Supraependymal cells were present on the non-ciliated ependyma i

Effects of Daunomycin and Daunomycin-protein conjugates on the ultrastructure of Trypanosoma rhodesiense.

The nucleus (but not the kinetoplast) of Trypanosoma rhodesiense is the principal target for trypanocidal attack by Daunomycin in vitro and Daunomycin-protein conjugates in vivo; free Daunomycin is inactive in vivo. Nucleolar segregation and abnormal distribution of heterochromatin occur after drug treatment as in tumour cells. Other abnormal changes include formation of cytoplasmic clefts and autophagic vacuoles, increased prominence of the tubular collection system, and exocytotic vesicle production. Large masses of ferritin accumulate in white cells after treatment with the drug-ferritin co

Structural details of anthrax spores during stages of transformation into vegetative cells.

Moberly, Betty J. (The University of Michigan, Ann Arbor), F. Shafa, and Philipp Gerhardt. Structural details of anthrax spores during stages of transformation into vegetative cells. J. Bacteriol. 92:220-228. 1966.-Anthrax spores in stages of dormancy, activation, germination, and outgrowth into vegetative cells were examined in an electron microscope. The fine structure proved to be much like that observed in related species of Bacillus, except for a visible alteration after heat activation and clusters of vesicle-like bodies in the cytoplasm of vegetative cells.

Key Regulators of Autophagosome Closure.

Autophagy is an evolutionarily conserved pathway, in which cytoplasmic components are sequestered within double-membrane vesicles called autophagosomes and then transported into lysosomes or vacuoles for degradation. Over 40 conserved autophagy-related (ATG) genes define the core machinery for the five processes of autophagy: initiation, nucleation, elongation, closure, and fusion. In this review, we focus on one of the least well-characterized events in autophagy, namely the closure of the isolation membrane/phagophore to form the sealed autophagosome. This process is tightly regulated by ESC

Production of CCHF virus-like particle by a baculovirus-insect cell expression system.

Crimean-Congo Haemorrhagic Fever Virus (CCHFV) is a tick-born virus of the Nairovirus genus within the Bunyaviridae family, which is widespread and causes high fatality. The nucleocapsid of CCHFV is comprised of N proteins that are encoded by the S segment. In this research, the N protein of CCHFV was expressed in insect cells using a recombinant baculovirus. Under an electron microscope, Virus-Like Particles (VLPs) with various size and morphology were observed in cytoplasmic vesicles in the infected cells. Sucrose-gradient purification of the cell lysate indicated that the VLPs were mainly l…
