Chronic Deep Brain Stimulation of the Human Nucleus Accumbens Region Disrupts the Stability of Intertemporal Preferences.

When choosing between rewards that differ in temporal proximity (intertemporal choice), human preferences are typically stable, constituting a clinically relevant transdiagnostic trait. Here we show, in female and male human patients undergoing deep brain stimulation (DBS) of the anterior limb of the internal capsule/NAcc region for treatment-resistant obsessive-compulsive disorder, that long-term chronic (but not phasic) DBS disrupts intertemporal preferences. Hierarchical Bayesian modeling accounting for temporal discounting behavior across multiple time points allowed us to assess both shor

Infant expectations of instant or delayed gratification.

Choices between immediate gratification and long-term (but larger) gains are prevalent in human life, which is why the decision-making processes to delay gratification have been studied extensively throughout different developmental ages. Children's delay-of-gratification behaviors have been examined in the well-known "marshmallow test," in which 3- to 5-year-olds are given a marshmallow and told by an experimenter that they can eat it immediately or wait for an unspecified duration of time (which can be capped at 15 min) until the experimenter returns so that they can receive another marshmal

What weighs more-low compliance with self-deferral or minor medical procedures? Explaining the high rate of hepatitis C virus window-period donations in Poland.

BACKGROUND: Since the introduction of nucleic acid testing (NAT) for routine blood donor screening, hepatitis C virus (HCV) RNA-only detection rates reported from Poland have been higher than in most other European countries. STUDY DESIGN AND METHODS: To examine factors that likely contribute to these window-period donations, we conducted a case-control study among 47 recently HCV-infected blood donors (cases), who gave blood between July 2002 and June 2014, and 141 controls matched by age, sex, and donation dates. Firth-corrected, conditional logistic regression models were fitted to estimat

Unrelated donor bone marrow transplantation for chronic myelogenous leukemia: a decision analysis.

BACKGROUND: Chronic myelogenous leukemia (CML) is an indolent but ultimately fatal disease. Because the natural history of CML varies and quality of life with CML may be excellent until shortly before death, deciding whether and when to pursue unrelated donor bone marrow transplantation is often difficult. OBJECTIVE: To compare early transplantation, delayed transplantation, and no transplantation for patients with chronic-phase CML on the basis of discounted, quality-adjusted life expectancy. DESIGN: A markov model comparing different strategies was constructed. This model considers patient

Compensatory activation in fronto-parietal cortices among HIV-infected persons during a monetary decision-making task.

HIV infection can cause direct and indirect damage to the brain and is consistently associated with neurocognitive disorders, including impairments in decision-making capacities. The tendency to devalue rewards that are delayed (temporal discounting) is relevant to a range of health risk behaviors. Making choices about delayed rewards engages the executive control network of the brain, which has been found to be affected by HIV. In this case-control study of 18 HIV-positive and 17 HIV-negative adults, we examined the effects of HIV on brain activation during a temporal discounting task. Functi

Methylation Patterns of the HTR2A Associate With Relapse-Related Behaviors in Cocaine-Dependent Participants.

Relapse during abstinence in cocaine use disorder (CUD) is often hastened by high impulsivity (predisposition toward rapid unplanned reactio … thy controls. Impulsivity was assessed by self-report (Barratt Impulsiveness Scale; BIS-11) and the delay discounting task, while levels of cue reactivity were determined by performance in the cocaine-word Stroop task. Genomic DNA was extracted from lymphocytes and the bisulfite-treated DNA was subjected to pyrosequencing to determine degree of methylation at four …

The phenotype of recovery XI: associations of sleep quality and perceived stress with discounting and quality of life in substance use recovery.

PURPOSE: Sleep and stress show an interdependent relationship in physiology, and both are known risk factors for relapse in substance use disorder (SUD) recovery. However, sleep and stress are often investigated independently in addiction research. In this exploratory study, the associations of sleep quality and perceived stress with delay discounting (DD), effort discounting (ED), and quality of life (QOL) were examined concomitantly to determine their role in addiction recovery. DD has been proposed as a prognostic indicator of SUD treatment response, ED is hypothesized to be relevant to the…

Effects of tDCS on reward responsiveness and valuation in Parkinson's patients with impulse control disorders.

Parkinson's disease (PD) patients with impulse control disorders (ICD) frequently report hypersensitivity to rewards. However, a few studies have explored the effectiveness of modulation techniques on symptoms experienced by these patients. In this study, we assessed the effect of anodal tDCS over the DLPFC on reward responsiveness and valuation in PD patients with ICD. 43 participants (15 PD patients with ICD, 13 PD without ICD, and 15 healthy matched controls) were asked to perform a reward-craving test employing both explicit (self-ratings of liking and wanting) and implicit (heart rate and

Prior commitment: Its effect on suboptimal choice in a gambling-like task.

Animals choose suboptimally when provided with cues that signal whether reinforcement is coming or not. For example, pigeons do not prefer an alternative that always provides them with a signal for reinforcement over an alternative that provides them with a signal for reinforcement only half of the time and a signal for the absence of reinforcement the rest of the time. In the present research, we tested the hypothesis that if the results of the choice are delayed, pigeons will choose less suboptimally. We tested this hypothesis by forcing pigeons to wait following their choice, requiring them

Identifying individual differences of fluoxetine response in juvenile rhesus monkeys by metabolite profiling.

Fluoxetine is the only psychopharmacological agent approved for depression by the US Food and Drug Administration for children and is commonly used therapeutically in a variety of neurodevelopmental disorders. Therapeutic response shows high individual variability, and severe side effects have been observed. In the current study we set out to identify biomarkers of response to fluoxetine as well as biomarkers that correlate with impulsivity, a measure of reward delay behavior and potential side effect of the drug, in juvenile male rhesus monkeys. The study group was also genotyped for polymorp

Applying the reinforcer pathology model to alcohol use and suicide risk: An initial test.

Alcohol use and suicide risk have a well-established relation. In the United States, up to a third of suicide decedents have alcohol in their blood at the time of death and half of those have a blood alcohol level above the legal driving limit (i.e., 0.08% blood alcohol content). Despite the established relation, the mechanisms linking alcohol and suicide risk are unclear. The reinforcer pathology framework of addiction, pioneered by Warren Bickel, can be adapted to make predictions about suicide risk among those who use alcohol. To assess this, we conducted two crowdsourced studies (using Ama

The dorsomedial prefrontal cortex promotes self-control by inhibiting the egocentric perspective.

The dorsomedial prefrontal cortex (dmPFC) plays a crucial role in social cognitive functions, including perspective-taking. Although perspective-taking has been linked to self-control, the mechanism by which the dmPFC might facilitate self-control remains unclear. Using the multimodal neuroimaging dataset from the Human Connectome Project (Study 1, N =978 adults), we established a reliable association between the dmPFC and self-control, as measured by discounting rate-the tendency to prefer smaller, immediate rewards over larger, delayed ones. Experiments (Study 2, N = 36 adults) involving hig

Inducing forward head posture recalibrates the valuation of future rewards.

BACKGROUND: The ability to prioritize long-term goals over immediate rewards is a cornerstone of human self-regulation and life-course success. Emerging evidence suggests that axial postural orientation provides a continuous stream of proprioceptive feedback that modulates this temporal valuation process. Using the framework of grounded cognition, this study investigated whether inducing Forward Head Posture (FHP) influences the valuation processes underlying self-regulation and future-oriented choice. METHODS: Healthy young adults (N = 59) performed a Delay Discounting Task (DDT) and a contr…

First, we utilized a regularized partial correlation network to estimate relationships between target processes and subjective well-being. Subsequently, we applied a directed acyclic graph (DAG …

Age-Dependent Differences in Cerebellar CB1 Receptor Expression and Its Association With Impulsivity and Alcohol Intake in Rats.

The cannabinoid receptor 1 (CB1R) in the hippocampus has been involved in impulsivity and alcohol consumption in adolescent rats. However, the role of CB1R in the cerebellum, despite that structure's exceptionally high CB1R density, remains unexplored. We therefore tested the hypothesis that cerebellar CB1R contributes to regulating impulsive behaviour, alcohol consumption and seeking. Male Wistar rats, adolescent (PND25), adult (PND90) and aged (PND365), were assessed on the delay discounting task (DDT) and for voluntary alcohol intake-conditioned place preference (ACPP). CB1R levels were qua…

Behavioral mechanisms of oxycodone's effects in female and male rats: Reinforcement delay and impulsive choice.

μ-Opioid agonists (e.g., morphine) typically increase impulsive choice, which has been interpreted as an opioid-induced increase in sensitivity to reinforcement delay. Relatively little research has been done with opioids other than morphine (e.g., oxycodone), or on sex differences in opioid effects, on impulsive choice. The present study investigated the effects of acute (0.1-1.0 mg/kg) and chronic (1.0 mg/kg twice/day) administration of oxycodone on choice controlled by reinforcement delay, a primary mechanism implicated in impulsive choice, in female and male rats. Rats responded under a co

Impulsivity in spontaneously hypertensive rats: Within-subjects comparison of sensitivity to delay and to amount of reinforcement.

Previous research has shown that spontaneously hypertensive rats (SHR) display higher levels of impulsive choice behavior, which is accompanied by a higher sensitivity to the delay of reinforcement, and by a normal sensitivity to the amount of reinforcement. Because those results were based on three different samples of subjects, in the present report we evaluated these three processes in the same individuals. SHR and WIS rats were exposed to concurrent-chains schedules in which the terminal links were manipulated to assess impulsivity, sensitivity to delay, and sensitivity to amount. For expl
