Identification of Francisella species and discrimination of type A and type B strains of F. tularensis by 16S rRNA analysis.

Tularemia is a zoonotic disease, occurring throughout the Northern Hemisphere. The causative agent, the bacterium Francisella tularensis, is represented by two main types. Type A is found in North America, whereas type B is mainly found in Asia and Europe and to a minor extent in North America. No routine technique for rapid diagnosis of tularemia has been generally applied. We have partially sequenced 16S rRNAs of two F. tularensis strains, as well as the closely related Francisella novicida. Of 550 nucleotides analyzed, only one difference in 16S rRNA primary sequence was found. This 16S rRN

Pancreatic stone protein versus conventional biomarkers for prognostic discrimination in sepsis: a meta-analysis of paired AUC comparisons.

BACKGROUND: Pancreatic stone protein (PSP) has been proposed as a prognostic biomarker in sepsis, but its comparative performance versus C-reactive protein (CRP) and procalcitonin (PCT) remains uncertain. METHODS: We performed a meta-analysis of direct within-study paired AUC comparisons between PSP and CRP and/or PCT in critically ill adults with sepsis, septic shock, or acute infection-related conditions in whom sepsis development or clinically relevant adverse outcomes were prospectively evaluated. Differences in AUC (ΔAUC) were pooled using random-effects models. The principal conservativ

FEN1-Gated Rolling Circle Amplification Achieves Single-Base Mismatch Discrimination for Breast Cancer Biomarker Analysis.

Rolling circle amplification (RCA) is a powerful isothermal nucleic acid amplification technique, yet its specificity is often compromised because padlock probes can be circularized even with imperfectly matched targets, leading to false-positive signals. Herein, we report a strand displacement-assisted FEN1 cleavage strategy to substantially improve the specificity of RCA. A dumbbell-shaped padlock probe is designed to generate dual 5' flaps only upon perfect hybridization with the target. Flap endonuclease 1 (FEN1) specifically recognizes the three-base overlap structure and cleaves the 5' f

The role of ultrasound texture analysis in the discrimination of pleomorphic adenoma and Warthin tumor in subjects with well-defined tumor borders.

BACKGROUND: Pleomorphic adenoma (PA) and Warthin tumor (WT) are the two most common benign parotid tumors, and their distinct management strategies necessitate accurate preoperative discrimination. Therefore, this study aimed to investigate the value of ultrasound texture analysis in differentiating between them. METHODS: This retrospective study analyzed ultrasound images from patients with pathologically confirmed PA or WT. Following lesion segmentation, texture features were extracted from the regions of interest (ROI). Feature selection was subsequently performed to identify the most disc

Using AI-Based Gait Analysis to Establish a 5-Meter Walk Time Cutoff for Discriminating Alzheimer's Disease.

Introduction A decline in gait function has been reported to occur early in Alzheimer's disease (AD), a major cause of dementia, suggesting that gait analysis may be a useful tool for dementia screening. However, a simple and practical analysis method or clear cutoff values have yet to be established. This study aimed to analyze gait function using the AI-powered smartphone application "Toruto," identify gait indicators characteristic of elderly patients with AD, and propose effective cutoff values for dementia discrimination. Methods A total of 147 participants were included in the study: 86

Parenchymal Neuro-Behçet's disease or Comorbid Behçet's disease with multiple sclerosis: A discriminative analysis of a complex clinical entity.

BACKGROUND: Patients with Behçet's disease (BD) may rarely manifest with cerebral white matter lesions resembling multiple sclerosis (MS). This may result in misdiagnosis due to diagnostic difficulties between parenchymal neuro-BD (pNBD) and MS. This study aims to elucidate the distinguishing features of patients with comorbid BD and MS (BD+MS) in comparison to those with pNBD and MS alone by focusing on clinical and laboratory features. We also aimed to identify the distinctive characteristics of BD+MS patients by comparing them to patients with pNBD and MS. METHODS: The methodology of this

Machine learning encodes urine and serum metabolic patterns for autoimmune disease discrimination, classification and metabolic dysregulation analysis.

There is a wide variety of autoimmune diseases (ADs) with complex pathogenesis and their accurate diagnosis is difficult to achieve because of their vague symptoms. Metabolomics has been proven to be an efficient tool in the analysis of metabolic disorders to provide clues about the mechanism and diagnosis of diseases. Previous studies of the metabolomics analysis of ADs were not competent in their discrimination. Herein, a liquid chromatography tandem mass spectrometry (LC-MS) strategy combined with machine learning is proposed for the discrimination and classification of ADs. Urine and serum

A Combined Flow Cytometric Semen Analysis and miRNA Profiling as a Tool to Discriminate Between High- and Low-Fertility Bulls.

Predicting bull fertility is one of the main challenges for the dairy breeding industry and artificial insemination (AI) centers. Semen evaluation performed in the AI center is not fully reliable to determine the level of bull fertility. Spermatozoa are rich in active miRNA. Specific sperm-borne miRNAs can be linked to fertility. The aim of our study is to propose a combined flow cytometric analysis and miRNA profiling of semen bulls with different fertility to identify markers that can be potentially used for the prediction of field fertility. Sperm functions were analyzed in frozen-thawed se

Exhaled volatilome analysis as a useful tool to discriminate asthma with other coexisting atopic diseases in women of childbearing age.

The prevalence of asthma is considerably high among women of childbearing age. Most asthmatic women also often have other atopic disorders. Therefore, the differentiation between patients with atopic diseases without asthma and asthmatics with coexisting diseases is essential to avoid underdiagnosis of asthma and to design strategies to reduce symptom severity and improve quality of life of patients. Hence, we aimed for the first time to conduct an analysis of volatile organic compounds in exhaled breath of women of childbearing age as a new approach to discriminate between asthmatics with oth
