Tuning of resonance energy transfer from 4',6-diamidino-2-phenylindole to an ultrasmall silver nanocluster across the lipid bilayer.

Herein we have developed a simple liposome-based donor-acceptor system across the lipid bilayer using 4',6-diamidino-2-phenylindole (DAPI) as the donor and an ultrasmall ligand-capped silver nanocluster (Ag NC) as the acceptor. The process of Förster resonance energy transfer (FRET) between DAPI and the Ag NC across the liposome bilayer has been demonstrated using steady-state and time-resolved fluorescence spectroscopy. The synthesized Ag NCs with a majority of Ag4 and Ag5 cores have been characterized using FTIR, mass spectrometry, HRTEM, UV-Vis and PL spectroscopy. Scanning electron microsc

Membrane topology of human presenilin-1 in SK-N-SH cells determined by fluorescence correlation spectroscopy and fluorescent energy transfer.

Presenilin-1 (PS1) protein acts as passive ER Ca(2+) leak channels that facilitate passive Ca(2+) leak across ER membrane. Mutations in the gene encoding PS1 protein cause neurodegeneration in the brains of patients with familial Alzheimer's disease (FAD). FADPS1 mutations abrogate the function of ER Ca(2+) leak channel activity in human neuroblastoma SK-N-SH cells in vitro (Das et al., J Neurochem 122(3):487-500, 2012) and in mouse embryonic fibroblasts. Consequently, genetic deletion or mutations of the PS1 gene cause calcium (Ca(2+)) signaling abnormalities leading to neurodegeneration in F

Synthesis and characterization of fluorescence resonance energy transfer-based nanoprobes by coating CdTe QDs with rhodamine B in gelatin nanoparticles.

A FRET-based nanoprobe consisting of CdTe quantum dot as donor and rhodamine B as acceptor embedded in gelatin nanoparticles was constructed and the photophysics of the system was characterized. The FRET-based sensors were led to at fixed donor/acceptor distance by synthesizing via a two-step desolvation way. This approach allowed for short acceptor-donor separation and thus for high energy transfer efficiencies. The morphology of the nanoprobes was presented spherically and relatively uniform with a diameter of about 100 nm, which was characterized by transmission electron microscope. The lum

Dissociative excitation energy transfer in the reactions of protonated cysteine and tryptophan with electronically excited singlet molecular oxygen (a1Δ(g)).

We report a study on the reactions of protonated cysteine (CysH(+)) and tryptophan (TrpH(+)) with the lowest electronically excited state of molecular oxygen (O(2), a(1)Δ(g)), including the measurement of the effects of collision energy (E(col)) on reaction cross sections over the center-of-mass E(col) range of 0.05 to 1.0 eV. Electronic structure calculations were used to examine properties of complexes, transition states and products that might be important along the reaction coordinate. For CysH(+) + (1)O(2), the product channel corresponds to C(α)-C(β) bond rupture of a hydroperoxide inter

Fluorescence resonance energy transfer analysis of cell surface receptor interactions and signaling using spectral variants of the green fluorescent protein.

BACKGROUND: Fluorescence resonance energy transfer (FRET) is a powerful technique for measuring molecular interactions at Angstrom distances. We present a new method for FRET that utilizes the unique spectral properties of variants of the green fluorescent protein (GFP) for large-scale analysis by flow cytometry. METHODS: The proteins of interest are fused in frame separately to the cyan fluorescent protein (CFP) or the yellow fluorescent protein (YFP). FRET between these differentially tagged fusion proteins is analyzed using a dual-laser FACSVantage cytometer. RESULTS: We show that homotyp

Modelling dose and dose-averaged linear energy transfer to predict high-grade temporal lobe necrosis following skull-base proton therapy.

BACKGROUND: In skull-base proton therapy (PT), severe toxicity outcomes such as temporal lobe necrosis (TLN) may be associated with inadequate management of the actual radiobiological effectiveness (RBE) of proton beams. Combining dose-averaged linear energy transfer (LETd) and dose may be crucial in treatment plan optimization and evaluation. PURPOSE: To gather evidence on the combined effect of LETd and dose on voxel- and structure-wise levels in determining a high-grade TLN (CTCAE v.5 grade ≥G2, G2-TLN) in skull-base tumors treated with PT; a dose-LETd-volume histogram (DLVH)-based model w

Time-Resolved Förster Resonance Energy Transfer Nanoassay Based on CdTe Quantum Dots for Sensitive Detection of Prostate Cancer Antigen 3.

Prostate Cancer Antigen 3 (PCA3) is a long noncoding RNA highly expressed in prostate cancer cells, making it a promising biomarker for noninvasive prostate cancer diagnosis. Simple and rapid detection using nanoprobes can potentially overcome the limitations of traditional diagnostic techniques. Here, we designed, characterized, and applied a DNA-strand displacement assay based on Förster Resonance Energy Transfer (FRET) between terbium (Tb) ions and semiconductor quantum dots (QDs) as a proof-of-concept for sensitive and specific mix-and-measure quantification of a synthetic DNA analogue of

Voxel-level linear energy transfer-driven dose conversion factors between LEM- and MKM-based treatment plans in carbon-ion radiotherapy.

BACKGROUND: Carbon-ion radiotherapy employs distinct relative biological effectiveness (RBE) models-the local effect model (LEM) and the microdosimetric kinetic model (MKM)-which differ in their dependencies on dose-averaged linear energy transfer (LETd) and absorbed dose. Conversion factors (CFs) between these models are essential for translating clinical outcomes across institutions; however, previous studies have neglected LETd-driven spatial heterogeneity. PURPOSE: This study aimed to quantify the impact of LETd on CFs by establishing a voxel-level framework for iso-effective dose convers

Biochemical clocks in fingerprints energy transfer between free tryptophan and NADH for potential estimation of postmortem interval, age, and sex.

NADH is an essential coenzyme involved in key metabolic processes. This study introduces a novel, non-invasive spectroscopic approach for analyzing age, sex, and time of death based on this coenzyme and other sweat-fatty substance components, while considering external factors such as drug exposure, acidic environments, and silver nanoparticles (AgNPs). The non-invasive mode involves the analysis of sweat-fatty substances collected from a fingerprint on a non-absorbent surface from both living and deceased individuals. The novel analysis considers excitation energy transfer (EET) in the form o

Efficient energy transfer from organic triplet states to Mn2+ dopants for dynamic tunable multicolor afterglow in 1D hybrid cadmium chloride.

Metal ion-doped organic-inorganic hybrid metal halides have emerged as promising room-temperature phosphorescence (RTP) materials owing to their tunable afterglow properties and significant potential in information security applications. However, optimizing RTP performance and achieving dynamic control over afterglow colors remain challenging in 1D hybrid systems, primarily because of the inefficient energy transfer from RTP-active organic components to external emissive sites. Herein, we report a novel 1D hybrid metal halide benchmark material, [(NBP)Cd2Cl5H2O] (NBP-Cd, NBP = N-benzylpiperido

Luminescence and Energy Transfer Properties of Color-Tunable Dy3+, Eu3+ co-Activated NaGdSiO4 Phosphor for WLED with Great Thermo-Stability.

A series of NaGd1-x-ySiO4: y Dy3+-x Eu3+ phosphors were synthesized by a high-temperature solid-phase method. The optimal doping ion concentration of Dy3+ ions for this phosphor was determined to be 1 % from the emission spectra. The energy transfer between Dy3+ and Eu3+ ions at 351 nm was investigated by photoluminescence spectra and fluorescence decay curves. At the excitation wavelengths of 275 nm, 351 nm, 366 nm, and 394 nm, a change from yellow to white to red light can be realized by adjusting the doping concentration of Eu/Dy ions. Particularly, by testing the temperature-dependent fluo

Synthesis of phenol formaldehyde resin (PFR) based fluorescence resonance energy transfer (FRET) composites for double channel detection of latent fingerprints.

A reversible two-channel fluorescent nanocomposite with fluorescence resonance energy transfer (FRET) effect was designed for the development, analysis, and characterization of latent fingerprints (LFPs). For the construction of the FRET probe, a core of mesoporous silicas (MSNs) were used to encapsulate the organic dye rhodamine 6G (RhD-6) as an acceptor, while green-emitting monodisperse phenolic resin nanoparticles (PFR NPs) were selected as a donor. The up-conversion material (UC) of NaYF4:Yb,Er was synthesized using a simple hydrothermal method, and the MSNs-RhD-6/PFR (PRM) was electrosta

Novel Förster Resonance Energy Transfer probe with quantum dot for a long-time imaging of active caspases inside individual cells.

With the goal to investigate biological phenomena at a single-cell level, we designed, synthesized and tested a molecular probe based on Förster resonance energy transfer (FRET) between a highly luminescent quantum dot (QD) as a donor and a fluorophore or fluorescence quencher as an acceptor linked by a specific peptide. In principle, QD luminescence, effectively dissipated in the probe, is switched on after the cleavage of the peptide by a protease and the release of the quencher. We proposed a novel synthesis strategy of a probe. A two-step synthesis consists of: (i) Conjugation of CdTe QDs

Spatial energy transfer with observation of bimolecular singlet oxygen emission using quantum dots as donors and zinc-phthalocyanine as acceptors.

This study reports the influence of CdSe-ZnS core-shell quantum dots (QDs) for formation of singlet oxygen using zinc-phthalocyanine (ZnPc) dyes in colloidal solutions. Using a microluminescence surface scan technique it was possible to measure accurately the photon diffusion length, or photon mean free path, inside the medium. Analyses were performed for a range of QD concentrations. Photon diffusion length was assigned to the bimolecular singlet oxygen emission at 707 nm. Related singlet oxygen emission was predicted by observing quenching of the photon diffusion length measured at the speci

Human Neutrophil Elastase Activated Fluorescent Probe for Pulmonary Diseases Based on Fluorescence Resonance Energy Transfer Using CdSe/ZnS Quantum Dots.

There is an increasing demand for effective noninvasive diagnosis against common pulmonary diseases, which are rising sharply due to the serious air pollution. Human neutrophil elastase (HNE), a typical protease highly involved in pulmonary inflammatory diseases and lung cancer, is a potential predictor for disease progression. Currently, few of the HNE-targeting probes are applicable in vivo due to the limitation in sensitivity and biocompatibility. Herein, we reported the achievement of in vitro detection and in vivo imaging of HNE by incorporating the HNE-specific peptide substrate, quantum

Mitochondria Targeting Fluorescent Probes Based on through Bond-Energy Transfer for Mutually Imaging Signaling Molecules H2 S and H2 O2.

Reactive signaling molecules participate in varieties of biochemical reactions, and methods to detect their mutual existence and crosstalk are in urgent demand. A benzothiadiazole-based handle was designed to fluorescently respond to the co-existence of H2 S and H2 O2 under pseudo-physiological conditions on a basis of a thiyl-radical-mediated mechanism that accounts for the rapid and efficient domino-like reaction processes. Then the handle motif was attached to a rhodamine moiety by means of an ethynylene linkage, and achieved a significant H2 S-H2 O2 mutual response in the mitochondria of l

Exploiting the fluorescence resonance energy transfer (FRET) between CdTe quantum dots and Au nanoparticles for the determination of bioactive thiols.

This work focused the implementation of FRET processes between CdTe quantum dots (QDs), acting as donors, and gold nanoparticles (AuNPs), behaving as acceptors, for the determination of several bioactive thiols such as captopril, glutathione, l-cysteine, thiomalic acid and coenzyme M. The surface chemistry of the QDs and AuNPs was adjusted with adequate capping ligands, i.e. mercaptopropionic acid and cysteamine, respectively, to guarantee the establishment of strong electrostatic interaction between them and promoting the formation of stable FRET assemblies. Under these circumstances the fluo
