A new phased assembly of the Antarctic spiny plunderfish provides novel insights into the evolution of the notothenioid radiation.

BACKGROUND: Notothenioids are a well characterised species flock endemic to the Antarctic and an important model group for studying genome adaptation to extreme cold. A new reference assembly and clade-wide comparative genomic analysis allow for investigation of cryonotothenioid evolution and the appearance of novel functionalities linked to cold adaptation. RESULTS: A new phased assembly of a model notothenioid, Harpagifer antarcticus, demonstrated low levels of haplotypic variability across the genome. Nevertheless, numerous insertions from multiple LINE-L2 clades were found, suggesting ong…

Comparative transcriptome analysis of false codling moth, Thaumatotibia leucotreta in response to high and low-temperature treatments.

The false codling moth (FCM), Thaumatotibia leucotreta, is a major quarantine pest native to Africa. Physical postharvest phytosanitary measures such as cold and heat treatments are championed to control its spread to new regions. However, the molecular changes that T. leucotreta undergoes as it attempts to adjust to its surroundings during the treatments and withstand the extreme temperatures remain largely unknown. The current study employs RNA-seq using the next-generation Illumina HiSeq platform to produce transcriptome profiles for differential gene expression analysis of T. leucotreta la

Pollution constrains thermal adaptation but not thermoregulation in dung beetles.

Insect heat and cold tolerance can be constrained by multiple factors, either natural, such as elevation, or anthropogenic, as pesticide exposure. Under a scenario of global change, alterations to physiological processes for tolerating thermal extremes and controlling self-temperature could make insect populations vulnerable. Therefore, this study sought to evaluate whether exposure to ivermectin, a pesticide used in livestock pastures around the world, affects thermal tolerance, thermoregulation and energetic status of several populations of the dung beetle Euoniticellus intermedius (Reiche)

Gene expression changes controlling distinct adaptations in the heart and skeletal muscle of a hibernating mammal.

Throughout the hibernation season, the thirteen-lined ground squirrel (Ictidomys tridecemlineatus) experiences extreme fluctuations in heart rate, metabolism, oxygen consumption, and body temperature, along with prolonged fasting and immobility. These conditions necessitate different functional requirements for the heart, which maintains contractile function throughout hibernation, and the skeletal muscle, which remains largely inactive. The adaptations used to maintain these contractile organs under such variable conditions serves as a natural model to study a variety of medically relevant co

Assessment of proline function in higher plants under extreme temperatures.

Climate change and abiotic stress factors are key players in crop losses worldwide. Among which, extreme temperatures (heat and cold) disturb plant growth and development, reduce productivity and, in severe cases, lead to plant death. Plants have developed numerous strategies to mitigate the detrimental impact of temperature stress. Exposure to stress leads to the accumulation of various metabolites, e.g. sugars, sugar alcohols, organic acids and amino acids. Plants accumulate the amino acid 'proline' in response to several abiotic stresses, including temperature stress. Proline abundance may

Mount Everest and Makalu cold injury amputation: 40 years on.

Freezing cold injuries (frostbite) of the extremities are a common injury among alpinists participating in high altitude expeditions, particularly during inclement weather conditions. Anecdotally, a digit that has suffered frostbite may be at greater risk to future cold injuries. In this case study, we profile a 62-year-old elite alpinist who suffered multiple digit amputations on both his hands and foot after historic summit attempts on Makalu (8481 m) and Mt. Everest (8848 m) in 1974-1979. We describe the clinical treatment he received at that time, and follow up his case 40 years after the

Vasodilative effect of isosorbide dinitrate ointment in complex regional pain syndrome type 1.

BACKGROUND: In complex regional pain syndrome type 1 (CRPS1) vascular changes occur from the initial, inflammatory event onto the trophic signs during chronicity of the disease, resulting in blood flow disturbances and marked temperature changes. Pharmacotherapeutic treatment is generally inadequate. AIM: To determine whether local application of the nitric oxide donor isosorbide dinitrate (ISDN) could cause vasodilation and thereby improve tissue blood distribution in the affected extremity. METHODS: In a pilot study, 5 female patients with CRPS1 in one hand were treated with ISDN ointment

Idiopathic chilblain: a case series from Injibara General Hospital, Ethiopia.

BACKGROUND: Idiopathic chilblains are inflammatory lesions on the skin that typically appear after being in a cold, wet environment. The condition manifests itself as burning, soreness, and pruritis in the extremities (fingers and toes), usually occurring 12 to 24 h following a triggering event. Conservative measures are the mainstay of treatment for idiopathic chilblain; however pharmaceutical therapy may be necessary in cases that are severe or persistent. This is the only case series in Ethiopia concerning idiopathic chilblain. CASE PRESENTATION: From June to August of 2024, 12 individuals

Brachial Artery Aneurysm as a Limb Threatening Condition: a Case Report.

Brachial artery aneurysms are rare but potentially limb threatening condition. The presented case here is a 52-year old male referred to the emergency department complaining a sudden onset and progressive pain with coldness of his right upper extremity during brushing. The right upper extremity was pulseless and three-dimensional computed tomography showed an aneurysm of the proximal right brachial artery associated with arterial occlusion in its distal branch. Embolectomy was done, the aneurysm resected, and the artery successfully re-vascularised by interposing a saphenous vein graft.

Early-life exposure to non-optimal temperature and emotional and behavioral problems in preschool children.

BACKGROUND: The effects of early-life exposure to extreme temperatures on adverse neurodevelopmental outcomes have been increasingly investigated. However, less attention has been given to emotional and behavioral problems (EBPs). This study aimed to examine the association between early-life exposure to non-optimal temperature and EBPs among preschool children aged 2 - 6 years. METHODS: We conducted a cross-sectional study among over 4000 preschool children in Guangzhou, China. EBPs were assessed using the parent-reported Strengths and Difficulties Questionnaire (SDQ). Residential daily ambi

Molecular evolution in Panagrolaimus nematodes: origins of parthenogenesis, hermaphroditism and the Antarctic species P. davidi.

BACKGROUND: As exemplified by the famously successful model organism Caenorhabditis elegans, nematodes offer outstanding animal systems for investigating diverse biological phenomena due to their small genome sizes, short generation times and ease of laboratory maintenance. Nematodes in the genus Panagrolaimus have served in comparative development and anhydrobiosis studies, and the Antarctic species P. davidi offers a powerful paradigm for understanding the biological mechanisms of extreme cold tolerance. Panagrolaimus nematodes are also unique in that examples of gonochoristic, hermaphroditi…

[Frostbites: cold-freezing injuries due to a biting cold].

Frostbites are cold-freezing injuries of the extremities due to the exposure to temperatures below 0°C. The risk, in the absence of treatment, is the amputation of frostbitten areas which become gangrenous. Victims of frostbite must be treated and evacuated urgently. Nurses play an active role in the secondary long-term treatment.

Rumen DNA virome plasticity and viral metabolic potential are associated with seasonal adaptation in grazing yak and cattle on the Qinghai-Tibet Plateau.

BACKGROUND: As a diverse and abundant component of the rumen ecosystem, viruses interact with other microorganisms and are thought to influence microbial metabolism and host productivity. However, how the rumen virome responds to seasonal fluctuations in extreme environments remains poorly understood. Here, metagenomic analyses were used to investigate temporal dynamics of viral diversity, functional potential, and virus-host associations in the rumen virome of yak and cattle on the Qinghai-Tibet Plateau across warm and cold seasons. RESULTS: Rumen viral communities exhibited pronounced seaso

Lumbar Sympathetic Block for the Treatment of Intractable Erythromelalgia: A Case Report.

BACKGROUND: Erythromelalgia is a rare pain disorder, which causes hyperperfused, red, and painful extremities. Symptoms can be so extreme that patients often resort to soaking their limbs in ice-cold water for hours on end and socially isolating themselves to avoid triggering an episode. Treatment is challenging, as many cases are refractory to medical management, which is first line. Invasive procedures are typically trialed next, some of which are still considered experimental. CASE REPORT: We describe a case of a 26-year-old woman with erythromelalgia, who remained symptomatic despite exte

Short-latency afferent inhibition is reduced with cold-water immersion of a limb and remains reduced after removal from the cold stimulus.

The experience of pain that is induced by extremely cold temperatures can exert a modulatory effect on motor cortex circuitry. Although it is known that immersion of a single limb in very cold water can increase corticomotor excitability it is unknown how afferent input to the cortex shapes excitatory and inhibitory processes. Therefore, the purpose of this study was to examine motor-evoked potentials (MEP), short-latency afferent inhibition (SAI) and long-latency afferent inhibition (LAI) in response to immersion of a single hand in cold water. Transcranial magnetic stimulation (TMS) was used
