Microvasculature of the human optic nerve.

PURPOSE: Methyl methacrylate vascular corrosion casting techniques were used to examine the normal anterior optic nerve microvasculature in 18 human eye bank eyes. METHODS: Selective cannulation of the central retinal artery, the short posterior ciliary arteries, or both, allowed the methyl methacrylate to be injected into the anterior optic nerve circulation. Preflushing with tissue plasminogen activator greatly enhanced the filling of the fine microvasculature by dissolving the intraluminal clots. RESULTS: The superficial nerve fiber layer of the optic nerve received its primary blood supp

Association Between Endothelial Cell Density and Transplant Suitability of Corneal Tissue With Type 1 and Type 2 Diabetes.

IMPORTANCE: The rate of types 1 and 2 diabetes in the United States is increasing. The effect of diabetes on corneal donor tissue is unknown … duced endothelial damage, reduced transplant suitability, or reductions in endothelial cell counts. Eye banks may need to collect medical history in a more robust manner. Additional studies may be valuable to determine the effect on long-term transplant outcomes of diabetes in cornea donors.

Effect of Optisol Supplementation With 0.255 μg/mL Amphotericin B on Elimination of Yeast at 5°C.

PURPOSE: Fungal infections in lamellar keratoplasty are a growing concern. Optisol-GS does not contain an antifungal agent and supplementation with 0.255 μg/mL Amphotericin B (AmpB) has been considered. This study tested the ability of 0.255 μg/mL AmpB in Optisol-GS to eliminate yeast contamination of corneal tissue. METHODS: Three isolates of Candida albicans, 1 of Candida parapsilosis, and 1 of Candida glabrata were tested in Optisol with and without AmpB. Corneoscleral rims stored at -80°C were thawed and placed in 10 multiwell plates (4 per plate). The rims were inoculated with 4 respecti

[Influence of the donor age on graft survival : Is the demographic change also important for corneal tissue donation?].

The success of corneal transplantation highly depends on the quality of the used graft. Various factors play a crucial role such as a perfec … failure. Extensive quality assurance procedures in tissue preparation and cornea processing in the eye banks in Germany (Europe) ensures that only corneas with tested high quality are allocated for transplantation regardless of the donor age. Against the background of an aging population, the use of grafts from older donors should not be waived.

Specular microscopy ancillary study methods for donor endothelial cell density determination of Cornea Donor Study images.

PURPOSE: To describe reliable methods for determining central corneal endothelial cell density (ECD) in a multicenter eye bank study. METHODS: The Specular Microscopy Reading Center utilized a dual-grading procedure and adjudication process to classify image quality and determine ECD for a subset of donor endothelial images obtained in the Specular Microscopy Ancillary Study, which is part of the Cornea Donor Study. Two certified readers classified images as analyzable (excellent, good, fair) or unanalyzable and determined the ECD using a variable frame technique. An adjudicator also evaluate

[Corneoscleral transplant excision in the cadaver. Experiences of the North Rhine Westphalia Lions Cornea Bank 1995 and 1996].

BACKGROUND: Donor corneas are normally obtained by whole globe enucleation-a procedure often refused by the bereaved. To increase the acceptance of cornea donation, we have exclusively obtained donor corneas by in situ excision since the end of 1994. There have been reports of increased endothelial damage and higher contamination rates. We report our experience in 1995 and 1996. METHODS: The in situ excision was performed by staff trained in microsurgical techniques. Only donor corneas with negative end-storage cultures after at least 10 days and an endothelial cell count of more than 2500 ce

Comparative Evaluation of Temperature, Bottle Type, and Amphotericin B on Fungal Detection in Corneal Preservation Media Using the BACT/ALERT System®.

Fungal contamination of donor corneas, although rare, constitutes a serious infection risk to recipients. Therefore, microbiological quality control with methods optimized for fungal detection is crucial in eye banks to detect and discard contaminated corneas. This study aimed to compare the performance of different BACT/ALERT® bottles (iFA Plus and iLYM) incubated at different temperatures (32 °C and 37 °C) when corneal preservation media, with or without amphotericin B, were cultured. A total of 6 culture conditions were compared by inoculation of fewer than 100 colony-forming units of 18 fu…

Factors Affecting the Density of Corneal Endothelial Cells Cultured from Donor Corneas.

We investigated which specific correlation exists between the endothelial cell density (ECD) of corneal endothelial cell (CEC) cultures and … ng task when corneas from elderly donors are used (i.e., the overall majority of those collected by eye banks) and particularly when corneas from elderly donors with ECD < 2000 cells/mm2 are used. Pooling more of two corneas to obtain suitable CECs could technically overcome the problem. The above issues should be tackled appropriately before moving into cl …

Evaluation of an animal product-free variant of MegaCell MEM as a storage medium for corneas destined for transplantation.

PURPOSE: The traditional medium for storing corneas in European Eye Banks is Gibco's MEM (Eagle's) with Earle's salts and Hepes containing 2% fetal calf serum, glutamine and antimycotics. Although serum-free MegaCell MEM has been reported to be more suitable for this purpose, it contains components of animal origin that potentially pose health risks to corneal recipients. The possibility of removing or replacing these components has therefore been investigated. METHODS: A MegaCell basal medium (DME) and a formulation of this (MegaCell DCS), which contains no components of animal origin, have …

Opportunities and challenges in leveraging multiomics and biobanks for vision science.

PURPOSE OF REVIEW: Emerging biobank resources allow large-scale integration of eye-specific phenotypes with clinical, genomic, and multiomic data. This convergence enables unprecedented opportunities to systematically dissect the genetic architecture, epidemiology, and mechanistic pathways of both rare monogenic and common polygenic diseases. The review aims to critically examine how contemporary data extraction, multiomics, and analytic methodologies are reshaping disease classification, genetic discovery, and translational research in ophthalmology, while highlighting the associated challeng

An improved spinning lens test to determine the stiffness of the human lens.

It is widely accepted that age-related changes in lens stiffness are significant for the development of presbyopia. However, precise details on the relative importance of age-related changes in the stiffness of the lens, in comparison with other potential mechanisms for the development of presbyopia, have not yet been established. One contributing factor to this uncertainty is the paucity and variability of experimental data on lens stiffness. The available published data generally indicate that stiffness varies spatially within the lens and that stiffness parameters tend to increase with age.

The swelling of cornea in bank cultivation. The role of barriers and peripheral edge.

The swelling of a rabbit cornea in organ culture is caused by influx of fluid via the surface. Data regarding the weight increase rates during the first 4 h of corneal buttons with different diameters, have been used to calculate the net influx per mm2 per h via the anterior + posterior surfaces on one hand, and via the peripheral edge on the other hand. In fresh corneas the contribution to the corneal swelling, of the net fluid uptake via the anterior + the posterior surfaces, is negligible. The influx via the peripheral edge is about the same in fresh and freeze treated buttons. For freeze t
