COL9A1 encodes the alpha-1 chain of type IX collagen heterotrimer, which is a vital component of collagen fibrils in hyaline cartilage. There are preliminary lines of evidence suggesting that COL9A1 mutations may be associated with autosomal dominant multiple epiphyseal dysplasia (MED), a disorder affecting the epiphysis of long bones. With only 2 reported cases (both from the same family) of MED in autosomal dominant COL9A1-related disorders (MIM 614135) in the clinical scientific literature hitherto, the phenotype is poorly understood at present. Here, we report the clinical and imaging find

A 17-year-old boy presented with a history of longstanding hematuria and non-nephrotic proteinuria without renal insufficiency, for which renal biopsy was performed. The findings by routine light microscopy and direct immunofluorescence study were mild and nonspecific. Electron microscopy, however, demonstrated the unexpected finding of distinct collagen fibrils within capillary wall basement membranes, typical of the nail-patella syndrome. Repeat physical examination following the biopsy confirmed the presence of normal nails and patellae, and radiographs of the knees were also normal. The bo

Prolongation of the PR interval is a nonspecific condition. Etiologies include normal physiologic variance, idiopathic bundle branch fibrosis, calcific valvular heart disease, ischemic heart disease, cardiomyopathies, infections resulting in myocarditis, drugs (especially type I antidysrhythmic medications), collagen vascular diseases, tumors, trauma, electrolyte imbalances, and many other miscellaneous causes. Isolated first degrees atrioventricular block is generally not associated with an increased risk of sudden death, syncope, or progression to higher degrees of atrioventricular block. Ex

Giant cell tumors of bone (GCTB) are rare but aggressive, locally destructive tumors. They typically affect young people, significantly reducing their quality of life and increasing mortality rates. Giant cell tumors of bone are composed of osteoclast-like giant cells that respond to increased secretion of RANKL by stromal cells, triggering osteolysis. For over a decade, denosumab, a monoclonal antibody targeting this receptor activator, has been approved as a neo-adjuvant to facilitate surgical resection or in the setting of inoperable tumors. Denosumab treatment has shown rapid pain improvem

By the Shields classification, articulated over 30 years ago, inherited dentin defects are divided into 5 types: 3 types of dentinogenesis imperfecta (DGI), and 2 types of dentin dysplasia (DD). DGI type I is osteogenesis imperfecta (OI) with DGI. OI with DGI is caused, in most cases, by mutations in the 2 genes encoding type I collagen. Many genes are required to generate the enzymes that catalyze collagen's diverse post-translational modifications and its assembly into fibers, fibrils, bundles, and networks. Rare inherited diseases of bone are caused by defects in these genes, and some are o

Mucous membrane pemphigoid (MMP) is an autoimmune blistering disease characterized by autoantibodies against the basal membrane zone of skin and surface-close epithelia and predominant mucosal lesions. The oral cavity and conjunctivae are most frequently affected, albeit clinical manifestations can also occur on the skin. MMP-associated lesions outside the oral cavity typically lead to scarring. Mechanisms underlying scarring are largely unknown in MMP and effective treatment options are limited. Herein, we assessed the collagen architecture in tissue samples of an antibody-transfer mouse mode

The hemidesmosome is a membrane-associated supramolecular dermal epidermal complex linking the cytoskeleton of the basal keratinocyte to structures within the papillary dermis. Different components of this complex have been identified as autoantigens in autoimmune bullous skin diseases. Some of the autoantigens have been characterized at the molecular level. Little is known, however, about the factors that initiate the production of autoantibodies. By histopathology, acquired skin diseases of hemidesmosomes show subepidermal blisters and by direct immunofluorescence, linear deposits of IgG, C3

BACKGROUND: Despite gluteus medius (GMED) tendinosis being relatively common, its presence in association with hip osteoarthritis (OA) or total hip arthroplasty (THA) is not well studied. It was hypothesized that more tendon degeneration would be found in patients with OA of the hip and in those that had undergone THA than that in a control group.
METHODS: One hundred patients were included between 2016 and 2019 and were included into 4 groups; the patients were undergoing revision surgery in two groups and primary THA in the other two groups; 22 patients had previously undergone primary THA

Intraepidermal and dermal-epidermal cohesion are of paramount importance for the integrity of the skin. Some constituent molecules of keratinocyte adhesion complexes and basement membrane-associated structures are the targets of antibody-mediated autoimmune reactions that give rise to various (muco-)cutaneous blistering diseases. The current state of our knowledge about these molecules--along with the main clinical, histological, and immunohistochemical features of the corresponding autoimmune diseases and their pathogenetic mechanisms--comprise the subjects surveyed in this review. Among the

Decellularization represents a robust strategy for generating biologically derived scaffolds that retain the native architecture and biochemical complexity of the extracellular matrix (ECM), thereby providing a conducive microenvironment for germ cell adhesion, proliferation, and differentiation-processes fundamental to the reconstitution of testicular function. While decellularized ECM (dECM) scaffolds have been extensively utilized in mammalian organoid systems for in vitro spermatogenesis and fertility-related research, the development of standardized protocols tailored to teleost models re

OBJECTIVE: Knee osteoarthritis (OA) is a chronic joint disease associated with pain and reduced function. The mechanisms underlying OA and how mechanical properties change with the disease are not fully understood, partly due to large variability during mechanical testing of cartilage. We characterized the mechanical properties of human femoral cartilage with varying levels of degeneration using a tissue constitution specific fibril-reinforced poroelastic (FRPE) material model.
METHOD: We created sample-specific finite element (FE) models based on synchrotron-based x-ray tomography to reduce

Fibroblasts play a crucial role in the development and progression of heart failure in many heart-related diseases. However, the contributions of fibroblast and its subtypes to the pathogenesis of ischemic cardiomyopathy (ICM) remain unclear. In this study, we analyzed single-nucleus RNA sequencing data from cardiac tissues of seven ICM patients with heart failure and eight control subjects. Our findings indicate that, compared to controls, fibroblasts from ICM patients exhibit dysregulation of genes associated with cell-substrate adhesion and matrix or collagen fibril organization. We further

Cutaneous asthenia is a hereditary connective tissue disease, primarily of dogs and cats, resembling Ehlers-Danlos syndrome in man. Collagen dysplasia results in skin hyperextensibility, skin and vessel fragility, and poor wound healing. The purpose of this study was to describe the histological findings in a dog with a collagenopathy consistent with cutaneous asthenia. An 8-month-old crossbreed female dog presented with lacerations and numerous atrophic and irregular scars. The skin was hyperextensible and easily torn by the slightest trauma. Ultrastructurally, the dermis was comprised of ela

Amyloidosis is a dysproteinemia characterized by extracellular deposition of amyloid fibrils. Its diagnosis depends on the demonstration of the characteristic apple-green birefringence in tissue stained with Congo red. Aspiration of subcutaneous fat as a means of procuring tissue has received attention in the internal medicine literature. However, this application of fine needle aspiration biopsy has not been investigated extensively by cytopathologists. We therefore report our experience. Over the past two years we performed 18 aspirations of subcutaneous fat on 17 patients in whom amyloidosi

Myopia alters the microstructural and biomechanical properties of the posterior sclera, which is characterized as a layered structure with potentially different inter-layer collagen fibril characteristics. Scanning acoustic microscopy (SAM) has been used to investigate how the micron-scale bulk mechanical properties of the posterior sclera are affected by myopia. Other investigators have employed second harmonic generation (SHG) imaging to characterize the collagen microstructure of tissues. In the present study, SAM and SHG imaging were used to investigate the existence of biomechanically-dis

BACKGROUND: It is commonly accepted that canine dystrophic mineralization of the hair follicle glassy membrane can be seen in hyperadrenocorticism and as a senile change in poodles. Pathology textbooks define this change as deposition of calcium salts in the form of basophilic, amorphous, granular material along collagen fibrils.
HYPOTHESIS/OBJECTIVES: The aim was to evaluate whether the incidence of the lesion is specific to poodles and if it is always associated with calcium deposition.
ANIMALS: One hundred and forty-seven dogs divided into three groups: (i) 91 normal poodles; (ii) 40 dogs

Valgus bracing is a common intervention aiming to reduce medial knee compartment loading in individuals with varus malaligned knees and medial knee osteoarthritis (KOA). However, its effects on cartilage mechanics relevant to tissue health and failure remain unknown. We investigated whether valgus bracing reduced cartilage surface stresses and strains, and thus potential risk for further cartilage degradation, in individuals with knee varus malalignment and medial KOA. Twenty-two participants (Kellgren-Lawrence grade ≥ 2) completed unbraced and braced walking trials. Participant-specific knee
