Clinical Validation of a Machine Learning-Based Biomarker Signature to Predict Response to Cytotoxic Chemotherapy Alone or Combined with Targeted Therapy in Metastatic Colorectal Cancer Patients: A Study Protocol and Review.

Metastatic colorectal cancer (mCRC) is a severe condition with high rates of illness and death. Current treatments are limited and not always effective because the cancer responds differently to drugs in different patients. This research aims to use artificial intelligence (AI) to improve treatment by predicting which therapies will work best for individual patients. By analyzing large sets of patient data and using machine learning, we hope to create a model that can identify which patients will respond to chemotherapy, either alone or combined with other targeted treatments. The study will i

Tropical diseases in the ICU: A syndromic approach to diagnosis and treatment.

Tropical infections form 20-30% of ICU admissions in tropical countries. Diarrheal diseases, malaria, dengue, typhoid, rickettsial diseases and leptospirosis are common causes of critical illness. Overlapping clinical features makes initial diagnosis challenging. A systematic approach involving (1) history of specific continent or country of travel, (2) exposure to specific environments (forests or farms, water sports, consumption of exotic foods), (3) incubation period, and (4) pattern of organ involvement and subtle differences in manifestations help in differential diagnosis and choice of i

Radiomics feature stability and distinction power in organic low-contrast phantoms for novel CBCT imaging.

PURPOSE: Due to the prevalence of daily cone-beam computed tomography (CBCT) imaging in radiation therapy, radiomics analysis has great potential to detect early radiation induced tissue changes. Clinical applications of radiomics using CBCT imaging have been hindered by lack of stability in radiomics features and comparably poor image quality. Novel CBCT imaging devices promise improved quality comparable to those of fan-beam CTs. Using organic low-contrast phantoms and patient data, this study aims to assess the potential of novel CBCT imaging for radiomics analysis by testing the robustness

Machine Learning in Cardio-Oncology: New Insights from an Emerging Discipline.

A growing body of evidence on a wide spectrum of adverse cardiac events following oncologic therapies has led to the emergence of cardio-oncology as an increasingly relevant interdisciplinary specialty. This also calls for better risk-stratification for patients undergoing cancer treatment. Machine learning (ML), a popular branch discipline of artificial intelligence that tackles complex big data problems by identifying interaction patterns among variables, has seen increasing usage in cardio-oncology studies for risk stratification. The objective of this comprehensive review is to outline the

Ranking the Predictive Power of Clinical and Biological Features Associated With Disease Progression in Huntington's Disease.

Huntington's disease (HD) is characterised by a triad of cognitive, behavioural, and motor symptoms which lead to functional decline and loss of independence. With potential disease-modifying therapies in development, there is interest in accurately measuring HD progression and characterising prognostic variables to improve efficiency of clinical trials. Using the large, prospective Enroll-HD cohort, we investigated the relative contribution and ranking of potential prognostic variables in patients with manifest HD. A random forest regression model was trained to predict change of clinical out

Plasmodium knowlesi in a traveller returning to New Zealand.

The recent discovery that Plasmodium knowlesi causes malaria in human populations, established it as the fifth species of plasmodium that may do so. A case of P. knowlesi malaria is described in a helicopter pilot from New Zealand, who became ill after returning from recurring visits to Malaysian Borneo in June 2010. His P. knowlesi infection was not detected using microscopic examination and a rapid diagnostic test for malaria, but was confirmed by both PCR (polymerase chain reaction) and sequence analysis showing homology with the ribosomal RNA gene for P. knowlesi. He responded rapidly to t

Association between salivary microbiota and tacrolimus pharmacokinetic variability in kidney transplant.

Kidney transplantation (KT) serves as a highly effective treatment for end-stage renal disease (ESRD). Nonetheless, the administration of tacrolimus, a commonly used immunosuppressant in KT, faces challenges due to the lack of dependable biomarkers for its efficacy and the considerable variability in tacrolimus pharmacokinetics (TacIPV). In this study, 183 saliva samples from 48 KT recipients under tacrolimus therapy, alongside 9 healthy control samples, were subjected to 16S rRNA sequencing. The analysis revealed significant differences in the composition of salivary microbiota among KT recip

Optimizing lipid nanoparticles for fetal gene delivery in vitro, ex vivo, and aided with machine learning.

There is a clinical need to develop lipid nanoparticles (LNPs) to deliver congenital therapies to the fetus during pregnancy. The aim of these therapies is to restore normal fetal development and prevent irreversible conditions after birth. As a first step, LNPs need to be optimized for transplacental transport, safety on the placental barrier and fetal organs and transfection efficiency. We developed and characterized a library of LNPs of varying compositions and used machine learning (ML) models to delineate the determinants of LNP size and zeta potential. Utilizing different in vitro placen

In silico Comparison of Left Atrial Ablation Techniques That Target the Anatomical, Structural, and Electrical Substrates of Atrial Fibrillation.

Catheter ablation therapy for persistent atrial fibrillation (AF) typically includes pulmonary vein isolation (PVI) and may include additional ablation lesions that target patient-specific anatomical, electrical, or structural features. Clinical centers employ different ablation strategies, which use imaging data together with electroanatomic mapping data, depending on data availability. The aim of this study was to compare ablation techniques across a virtual cohort of AF patients. We constructed 20 paroxysmal and 30 persistent AF patient-specific left atrial (LA) bilayer models incorporating

MRI Based Radiomics as an Imaging Biomarker for Locally Advanced Carcinoma Rectum: Predicting Tumor Response Following Neoadjuvant Chemoradiotherapy as an Organ Preservation Strategy.

BACKGROUND: Patients with locally advanced rectal cancer show variable responses to neoadjuvant chemoradiotherapy (NACTRT), making early prediction of treatment response clinically important. Radiomics and machine learning may offer a potential non-invasive imaging biomarker to identify patients who are more likely to respond to treatment. AIM: We investigated a machine learning-based radiomic approach to predict tumor response (TR) to NACTRT in patients with adenocarcinoma of the rectum from baseline and post-NACTRT magnetic resonance imaging (MRI). METHODS: One hundred patients with locall

PerioTwin-DeltaCRED: a validation-aware physiological digital twin for longitudinal periodontal treatment response prediction.

BACKGROUND: Predicting individual periodontal treatment response remains clinically unresolved. Existing machine-learning models are predominantly cross-sectional, lack a longitudinal response-vector design, and are rarely evaluated within validation-aware digital-twin frameworks that capture multidimensional credibility rather than single-endpoint discrimination alone. This study aimed to develop and evaluate PerioTwin-DeltaCRED, a validation-aware periodontal digital twin that models treatment response as a longitudinal PPD and CAL response vector, and to benchmark classical and quantum mach
