Arbuscular mycorrhizal fungal community abundance, functions, and symbiotic interactions revealed by root metatranscriptomes.

Arbuscular mycorrhizal (AM) fungal communities are often profiled by metabarcoding, but this approach can bias abundance estimates and provide limited insight into community function and interactions with the host. Here, using weekly, field-based multi-omics, we find metatranscriptomes and metagenomes increase estimates of AM fungal relative abundance 6-15-fold over rDNA metabarcoding and that root AM fungal relative abundance correlates strongly with sorghum forage yield (R > 0.9) and moderately with grain yield/quality. AM fungal relative abundance is impacted directly by sorghum development

Bacterial and Fungal Community Shifts and Environmental Drivers in Talahao Coal Mine Water.

Underground coal extraction alters aquatic ecosystems. Quantitative evaluation of microbial responses serves as an indicator for ecological security. High-throughput amplicon sequencing and structural equation modeling were applied to analyze aquatic bacterial and fungal communities before and after operations at the Talahao coal mine. Mining activities correlated with altered physicochemical water properties and reduced microbial community evenness. Environmental variables, including ion accumulation and suspended solid concentrations, mediated the indirect effects of mining on microbial rest

Effects of Bacillus-based inoculum on odor emissions co-regulation, nutrient element transformations and microbial community tropological structures during chicken manure and sawdust composting.

This study aims to evaluate whether different doses of Bacillus-based inoculum inoculated in chicken manure and sawdust composting will provide distinct effects on the co-regulation of ammonia (NH3) and hydrogen sulfide (H2S), nutrient conversions and microbial topological structures. Results indicate that the Bacillus-based inoculum inhibits NH3 emissions mainly by regulating bacterial communities, while promotes H2S emissions by regulating both bacterial and fungal communities. The inoculum only has a little effect on total organic carbon (TOC) and inhibits total sulfur (TS) and total phosph

Phomapyrrolidones A-C, antitubercular alkaloids from the endophytic fungus Phoma sp. NRRL 46751.

Three new alkaloids, phomapyrrolidones A-C (1-3), bearing a cyclopenta[b]fluorene ring system were isolated from the mycelium extract of the endophytic fungal strain Phoma sp. NRRL 46751, inhabiting Saurauia scaberrinae. Methylation of 1 afforded its N-methyl derivative 4. The planar structures and relative configurations of 1-4 were elucidated by extensive spectroscopic analysis. Phomapyrrolidones B (2) and C (3) exhibited weak antitubercular activity at subcytotoxic concentrations.

A Modified Slide-Embedded Scanning Electron Microscopy Preparation Method to Visualize Antagonistic Interactions Between Trichoderma viride and Fusarium sp.

Mycoparasitism is an important mechanism of fungal antagonism in which one fungus parasitizes another. This type of interaction plays a major role in the biocontrol activity of Trichoderma spp. against phytopathogenic fungi. Detailed visualization of these interactions is essential for understanding the structural mechanisms involved in fungal antagonism, including hyphal attachment, coiling, penetration, and cellular distortion. Scanning electron microscopy (SEM) is widely used for structural examination of fungal interactions; however, conventional preparation methods such as filter paper sy

Plasma-based strategy for inhibiting Candida albicans growth and CaMCA1 gene expression in vitro and reducing fungal pathogenicity in a murine model of vulvovaginal candidiasis.

UNLABELLED: Vulvovaginal candidiasis (VVC) is a common mucosal infection, mainly caused by Candida albicans. The use of common antifungal drugs in treatment of VVC is limited due to emergence of resistant fungal strains and severe side effects. Cold atmospheric plasma (CAP) as a novel therapeutic approach is proven to display strong antifungal activity against C. albicans. In the present study, the effects of CAP treatment on virulence and pathogenicity of C. albicans in a murine model was investigated. Candida albicans was treated with CAP at different time exposures. Fungal cell morphology a

Lectin-Fc(IgG2a) fusion proteins binding to the cell wall of C. albicans cause structural, metabolic, oxidative, and other pleiotropic effects.

Candida albicans, an opportunistic fungal pathogen and the leading cause of hospital-acquired infections, exhibits diverse virulence factors that promote its survival within the human body. The rising incidence of multidrug resistance (MDR) in C. albicans requires urgent development of novel antifungal strategies. Through genetic engineering, our group developed Lectin-Fc(IgG2a) proteins [WGA-Fc(IgG2a) and Dectin-1-Fc(IgG2a)], which exhibit potent antifungal activity against multiple fungal pathogens and hold promise as broad-spectrum therapeutic agents. In this study, we investigated the dire

Gut mycobiome dysbiosis in digestive system cancers: immune remodeling and translational implications.

Fungal dysbiosis has been reported in colorectal cancer (CRC), gastric cancer (GC), and pancreatic ductal adenocarcinoma (PDAC), but evidence strength differs substantially by tumor type and model system. Human CRC studies provide the broadest ecological evidence, including altered fungal community structure and fungal-bacterial networks. Human GC evidence is mainly cross-sectional and identifies Candida albicans as a candidate tissue-associated signal rather than a proven driver. In PDAC, human tissue studies report fungal signals, whereas causal support for Malassezia-mannose-binding lectin

Antifungal activity of Anacardium occidentale against clinical strains of Sporothrix brasiliensis.

AIM: To investigate the antifungal activity of Anacardium occidentale leaf extract can fight Sporothrix brasiliensis, a fungus that cause sporotrichosis. MATERIAL AND METHODS: We tested the extract against 20 fungal samples obtained from humans. We also evaluated whether the extract could reduce the growth of fungal communities called biofilms and examined changes in fungal cells after treatment. RESULTS AND CONCLUSION: The extract, rich in gallic acid, rutin, and ellagic acid, was able to stop fungal growth at low concentrations and reduced mature biofilms by up to 68%. Microscopy showed da

Sphingolipids as targets for treatment of fungal infections.

Invasive fungal infections have significantly increased in the last few decades. Three classes of drugs are commonly used to treat these infections: polyenes, azoles and echinocandins. Unfortunately each of these drugs has drawbacks; polyenes are toxic, resistance against azoles is emerging and echinocandins have narrow spectrum of activity. Thus, the development of new antifungals is urgently needed. In this context, fungal sphingolipids have emerged as a potential target for new antifungals, because their biosynthesis in fungi is structurally different than in mammals. Besides, some fungal s

Neutron Scattering Analysis of Cryptococcus neoformans Polysaccharide Reveals Solution Rigidity and Repeating Fractal-like Structural Patterns.

Cryptococcus neoformans is a fungal pathogen that can cause life-threatening brain infections in immunocompromised individuals. Unlike other fungal pathogens, it possesses a protective polysaccharide capsule that is crucial for its virulence. During infections, Cryptococcus cells release copious amounts of extracellular polysaccharides (exo-PS) that interfere with host immune responses. Both exo-PS and capsular-PS play pivotal roles in Cryptococcus infections and serve as essential targets for disease diagnosis and vaccine development strategies. However, understanding their structure is compl

Two closely related β-1,2-xylosyltransferases differentially impact cryptococcal glycan synthesis.

Cryptococcus neoformans is an opportunistic fungal pathogen that causes pulmonary infection in immunocompromised patients, which in severe cases leads to fatal meningoencephalitis. Cryptococcus exhibits unique glycobiology that plays important roles in pathogenesis. Unlike model yeast and other common fungal pathogens, Cryptococcus incorporates xylose, a five-carbon monosaccharide, into its glycans. One trimer motif, which consists of xylose in β-1,2 linkage to the reducing mannose of an α-1,3-mannose dimer, occurs in key cryptococcal glycoconjugates that include protein N- and O-linked glycan

Regional species turnover and local host and forest stand characteristics jointly structure early foliar mycobiomes of Scots pine seedlings.

The foliar mycobiome plays an important role in tree health and stress resilience, yet factors shaping foliar fungal communities in boreal conifer seedlings remain poorly understood. We investigated how geographic region, seedling origin (planted vs. naturally regenerated), needle nitrogen status, and local stand structure influence needle-associated fungal communities of Scots pine (Pinus sylvestris) seedlings. Needle samples were collected from seedlings without visible disease symptoms across 23 forest stands in southern Finland. Fungal communities were characterized using Illumina ITS2 amp

Cryptococcus neoformans Evades Pulmonary Immunity by Modulating Xylose Precursor Transport.

Cryptococcus neoformans is a fungal pathogen that kills almost 200,000 people each year and is distinguished by abundant and unique surface glycan structures that are rich in xylose. A mutant strain of C. neoformans that cannot transport xylose precursors into the secretory compartment is severely attenuated in virulence in mice yet surprisingly is not cleared. We found that this strain failed to induce the nonprotective T helper cell type 2 (Th2) responses characteristic of wild-type infection, instead promoting sustained interleukin 12p40 (IL-12p40) induction and increased IL-17A (IL-17) pro

T218 and K376 Control Phenamacril Sensitivity in Fusarium solani Myosin I and Enable Structure-Based Antifungal Discovery.

Phenamacril (PHA) is a Fusarium-specific fungicide targeting myosin I, but its sensitivity differs among Fusarium species. The molecular basis of this differential sensitivity remains unclear. Here, we investigated how site-specific variations in Fusarium solani myosin I (FsMyoI) affect PHA sensitivity and evaluated FsMyoI as a target for inhibitor discovery. Molecular dynamics simulations, site-directed mutagenesis, binding assays, and ATPase activity measurements revealed that FsMyoIK376M and FsMyoIT218S mutations reshaped the dynamics of key functional regions, influencing PHA binding and A

Microbiome network remodeling is associated with soil lipid metabolism in safflower under different cropping system-location combinations: an omics-based dissection.

This study investigated differences in safflower rhizosphere soil microbiota and metabolic functions among three planting pattern-location combinations (PLMEs): soybean-safflower rotation at Chenghai (CH), tobacco-safflower rotation at Longpan (ZYHY), and apple orchard intercropping at Lijiang (LJ). Each PLME comprised 6 plots. Bacterial α-diversity was significantly higher in the CH system, whereas fungal α-diversity peaked under the ZYHY system. Correlation analysis showed that bacterial α-diversity was negatively correlated with available potassium, while fungal α-diversity was negatively c

Advances on anticancer fungal metabolites: sources, chemical and biological activities in the last decade (2012-2023).

Among microorganisms, fungi are the ones that have the most imagination in producing secondary metabolites with the most varied structural differences, which are produced through different biosynthetic pathways. Therefore, they synthesize secondary metabolites classifiable into numerous families of natural compounds such as amino acids, alkaloids, anthraquinones, aromatic compounds, cyclohexene epoxides, furanones, macrolides, naphthoquinones, polyketides, pyrones, terpenes, etc. They also produced metabolites with very complex structures that can not be classified in the known families of nat
