Ultraviolet (UV) light is one of the major factors implicated in the pathogenesis of pterygium. The mechanism by which UV light induces this disease remains elusive. The aim of this study was to evaluate the effects of UVB irradiation on the expression of growth factors in cultured pterygium epithelial cells and to demonstrate their distribution within pterygium. We cultured pterygial epithelial cells from pterygium explants and these cells were exposed to 20 mJ/cm(2) of UVB. Total RNA was extracted at 0, 6, and 12 hours after irradiation. (32)P-labeled cDNA was synthesized and analyzed using

Chimeric Antigen Receptor (CAR) T-cell therapy has fundamentally altered the treatment paradigm for relapsed or refractory (R/R) B-cell malignancies. However, translating this success to T-cell lymphomas has been impeded by a critical biological barrier: fratricide. This process describes the self-destruction of therapeutic T-cells caused by their shared expression of target antigens, such as Tumor Necrosis Factor Receptor Superfamily Member 8 (CD30), following activation. Anaplastic Large Cell Lymphoma (ALCL), characterized by its uniform and high-level expression of CD30, represents both an

This paper reviews the history of aerosol therapy; discusses patient, drug, and device factors that can influence the success of aerosol therapy; and identifies trends that will drive the science of aerosol therapy in the future. Aerosol medication is generally less expensive, works more rapidly, and produces fewer side effects than the same drug given systemically. Aerosol therapy has been used for thousands of years by steaming and burning plant material. In the 50 years since the invention of the pressurized metered-dose inhaler, advances in drugs and devices have made aerosols the most com

Genomic studies in age-related macular degeneration (AMD) have identified genetic variants that account for the majority of AMD risk. An important next step is to understand the functional consequences and downstream effects of the identified AMD-associated genetic variants. Instrumental for this next step are 'omics' technologies, which enable high-throughput characterization and quantification of biological molecules, and subsequent integration of genomics with these omics datasets, a field referred to as systems genomics. Single cell sequencing studies of the retina and choroid demonstrated

BACKGROUND: In Huntington's disease (HD), a CAG repeat expansion mutation in the Huntingtin (HTT) gene drives a gain-of-function toxicity that disrupts mRNA processing. Although dysregulation of gene splicing has been shown in human HD post-mortem brain tissue, post-mortem analyses are likely confounded by cell type composition changes in late-stage HD, limiting the ability to identify dysregulation related to early pathogenesis.
METHODS: To investigate gene splicing changes in early HD, we performed alternative splicing analyses coupled with a proteogenomics approach to identify early CAG le

Glioblastoma (GB) is a highly aggressive tumor that demonstrates significant treatment resistance due to the blood-brain barrier (BBB) and resistance mechanisms of the tumor. Nanoparticle- based drug delivery systems (NDDS) can enhance drug bioavailability, enable targeted delivery, and provide controlled release. These nanoparticles can be engineered to target GB-specific biomarkers, such as EGFRvIII, which is expressed on glioma stem cells, to enhance treatment precision. A comprehensive literature review was conducted using the keywords "Glioblastoma," "Nanoparticles," "Pathogenesis," and "

Hereditary Multiple Osteochondromas (HMO) is a rare autosomal dominant skeletal disorder caused by heterozygous loss-of-function mutations in EXT1 or EXT2, which encode glycosyltransferases essential for heparan sulfate (HS) biosynthesis. Whether haploinsufficiency alone suffices or biallelic inactivation is required for osteochondroma formation remains a central unresolved question. In this study, we employed CRISPR/Cas9 combined with PiggyBac transposon technology to introduce a second pathogenic mutation (c.1883+1G>T) into patient-derived induced pluripotent stem cells (iPSCs) carrying a he

Fleshy fruit ripening involves broadly convergent physiological changes across phylogenetically distinct species, yet the extent to which shared chromatin-level mechanisms underlie this convergence remains poorly understood, partly due to technical challenges posed by metabolite-rich fruit tissues. Here, we develop iCUT&Tag, an improved chromatin profiling method requiring as few as 500 nuclei, and apply it to generate histone modification profiles across 12 fruit species spanning 9 botanical families, demonstrating its broad application in metabolite-rich fruit tissues. Comparative profiling

In severe childhood-onset neuromuscular disease (NMD), ventilatory muscle weakness and ineffective airway clearance drive recurrent infections and chronic colonization that is often culture-negative, polymicrobial, or both. The lung microbiome framework offers a unifying model: altered microbial immigration, elimination, and growth can produce dysbiosis with pathobiont expansion and antimicrobial resistance (AMR). We propose that pediatric NMD may follow a distinct developmental trajectory in which early-life secretion stasis, viral insults, and frequent antibiotics perturb immune-microbiome c

BACKGROUND: miRNAs are short RNA transcripts that modulate gene expression after transcription and have emerged as pivotal regulators of cancer biology. A subset, termed oncomiRNAs, functions as oncogenes or tumor suppressors, influencing key cellular events such as cell growth, programmed cell death, neovascularization, tissue invasion, and metastatic spread. Dysregulation of these miRNAs drives tumor initiation and progression, underscoring their role in cancer evolution. Traditionally, studies have relied on bulk tissue analyses, overlooking the profound spatiotemporal heterogeneity of onco

The potential link between viral infections and the onset of Alzheimer's disease (AD) has been debated for several years. The emergence of the SARS-CoV-2 pandemic has raised concerns regarding its potential role in predisposing individuals to AD or aggravating its progression. The widespread transmission of SARS-CoV-2 has introduced novel aspects to AD research, driving comprehensive investigations into the possible correlation between COVID-19 and neuropathological manifestations observed in patients with AD. This review explores the complex connection between COVID-19 and Alzheimer's disease

Alzheimer's disease (AD) is the most common cause of dementia in older adults. Neuropathological and imaging studies have demonstrated a progressive and stereotyped accumulation of protein aggregates, but the underlying molecular and cellular mechanisms driving AD progression and vulnerable cell populations affected by disease remain coarsely understood. The current study harnesses single cell and spatial genomics tools and knowledge from the BRAIN Initiative Cell Census Network to understand the impact of disease progression on middle temporal gyrus cell types. We used image-based quantitativ

The coding regions account for only a small part of the human genome, and the remaining vast majority of the regions generate large amounts of non-coding RNAs. Although non-coding RNAs do not code for any protein, they are suggested to work as either tumor suppressers or oncogenes through modulating the expression of genes and functions of proteins at transcriptional, posttranscriptional and post-translational levels. Acute Lymphoblastic Leukemia (ALL) originates from malignant transformed B/T-precursor-stage lymphoid progenitors in the bone marrow (BM). The pathogenesis of ALL is closely asso

The increasing dementia prevalence worldwide is driving the testing of novel therapeutic approaches, such as invasive brain technologies, despite limited clinical evidence and the risk of accelerating cognitive decline. Our manuscript (a) reviews the NIH Clinicaltrials.gov database for deep brain stimulation, stem cell implantation, and gene therapy trials on people with dementia; (b) discusses issues on beneficence, nonmaleficence, and autonomy associated with these trials; and (c) proposes nine recommendations that build on elements from the Declaration of Helsinki. We found 49 preregistered

Proteins are one of the essential macromolecules that constitute the foundation of life. Amino acids are the building blocks of proteins. Since the early production of monosodium glutamate (MSG), many amino acids have been widely produced for applications in pharmaceuticals, feed additives, and animal nutrition. In livestock and poultry industries, feed amino acids help reduce crude protein content and balance animal nutrition. Amid global food security concerns, demand is growing for amino acids to lessen the reliance on soybean meal in feed. Thanks to their higher quality and lower cost rela

Diabetic nephropathy (DN) has become the primary cause of end-stage renal disease globally, and its epidemiological burden intensifies alongside the surging prevalence of diabetes. The pathogenesis involves complex interactions among metabolic dysregulation, oxidative stress, inflammatory responses, and fibrotic signaling pathways. Hyperglycemia drives renal injury through activation of the RAAS and accumulation of advanced glycation end products (AGEs), while aberrant activation of key signaling pathways such as TGF-β/Smad3 and NF-κB further promotes renal fibrosis. Current clinical diagnosis

First-in-class (FIC) oncology drugs-defined by their novel mechanisms of action or targeting of previously unaddressed molecular targets-have emerged as a leading force in cancer therapeutic innovation. In this Review, we delineate the global landscape of FIC oncology drug approvals between 2009 and 2024 and analyze the evolving characteristics of both approved FIC therapies and potential FIC (PFIC) candidates in clinical development. To clarify the underlying dynamics of this evolution, we construct an analytic framework encompassing four distinct archetypes of FIC development: new targets, n

Pseudomonas species are major psychrotrophic spoilage agents of dairy products, yet the molecular basis of spoilage variability among closely related strains remains poorly understood. This study integrates whole-genome sequencing, phenomics, and computational structural biology to elucidate how nucleotide variations in the aprX gene drive differences in spoilage potential-a fundamental question in the microbiology of food borne isolates. Phylogenetic analysis of the aprX operon from representative Pseudomonas isolates classified them into five distinct types. Cold-storage simulation experimen
