Regulation of RELM/FIZZ isoform expression by Cdx2 in response to innate and adaptive immune stimulation in the intestine.

Host immune responses to commensal flora and enteric pathogens are known to influence gene expression in the intestinal epithelium. Although the Cdx family of caudal-related transcription factors represents critical regulators of gene expression in the intestinal epithelium, the effect of intestinal immune responses on Cdx expression and function has not been determined. We have shown that bacterial colonization and Th2 immune stimulation by intestinal nematode infection induce expression of the intestinal goblet cell-specific gene RELM beta. In this study, we investigated the transcriptional

Enhancement of carrimycin production in Streptomyces spiramyceticus 54-IA by the reporter-guided selection method with traditional mutagenesis.

Carrimycin, a new drug approved in recent years, is produced by recombinant Streptomyces spiramyceticus harboring a heterogeneous 4″-O-isovaleryltransferase gene (ist). The regulatory gene bsm42 plays a crucial role in activation of carrimycin biosynthesis, its high expression can improve the carrimycin production. The xylE-neo reporter cassette under control of the the two different lengths of bsm42 promoters, F1R and F2R, can be used to monitor the expression of bsm42 to screen carrimycin high-yield mutants treated with the composite mutagenesis of plasma and ultraviolet radiation. We found

A sensor histidine kinase from a plant-endosymbiont bacterium restores the virulence of a mammalian intracellular pathogen.

Alphaproteobacteria include organisms living in close association with plants or animals. This interaction relies partly on orthologous two-component regulatory systems (TCS), with sensor and regulator proteins modulating the expression of conserved genes related to symbiosis/virulence. We assessed the ability of the exoS+Sm gene, encoding a sensor protein from the plant endosymbiont Sinorhizobium meliloti to substitute its orthologous bvrS in the related animal/human pathogen Brucella abortus. ExoS phosphorylated the B. abortus regulator BvrR in vitro and in cultured bacteria, showing conserv

The Transcriptional Regulator Crt1 is Involved in The Pathogenic Lifestyle of Xanthomonas campestris pv. campestris B100.

The γ-proteobacterium Xanthomonas campestris pv. campestris B100 is the causal agent of black rot disease in a wide range of economically important crops. In addition, X. campestris pv. campestris is commercially relevant due to the synthesis of the exopolysaccharide xanthan. In this work, we first introduce a novel transcriptional regulator, termed Crt1, and present the effect that the deletion of the crt1 gene exerts on growth, xanthan production, virulence, and transcriptome and proteome profiles. Differential transcriptome analysis of the deletion mutant X. campestris pv. campestris B100 Δ

High levels of antisense transcription from numerous genes in the obligate intracellular bacterium Orientia tsutsugamushi.

Orientia tsutsugamushi (Ot) is a cytoplasmic, gram-negative, obligate intracellular bacterium that causes the human disease scrub typhus. It has a highly repetitive genome, whereby approximately 50% is composed of multiple copies of a proliferated integrative and conjugative element, the Rickettsial amplified Genetic Element (RAGE). Previous RNA sequencing analysis revealed a high level of antisense transcription in Ot, particularly from RAGE-encoded genes, with 18% of all genes having a sense:antisense ratio <1 and 5% with a ratio <0.1. In our current study, we have confirmed the earlier RNA

A feedback regulatory model for RifQ-mediated repression of rifamycin export in Amycolatopsis mediterranei.

BACKGROUND: Due to the important role of rifamycin in curing tuberculosis infection, the study on rifamycin has never been stopped. Although RifZ, which locates within the rifamycin biosynthetic cluster, has recently been characterized as a pathway-specific regulator for rifamycin biosynthesis, little is known about the regulation of rifamycin export. RESULTS: In this work, we proved that the expression of the rifamycin efflux pump (RifP) was regulated by RifQ, a TetR-family transcriptional regulator. Deletion of rifQ had little impact on bacterial growth, but resulted in improved rifamycin p

Xer1-independent mechanisms of Vpma phase variation in Mycoplasma agalactiae are triggered by Vpma-specific antibodies.

Despite their small genomes mycoplasmas maintain large multigene families devoted to surface antigenic variation. Although implicated as important factors for mycoplasma pathogenicity and persistence, the role of these antigenic switches in host immune evasion has never been unequivocally proven in these minimalist microbes. Mycoplasma agalactiae exhibits antigenic variation due to Xer1-mediated site-specific DNA inversions of vpma genes encoding abundant multiple surface lipoproteins. To evaluate the biological significance of Vpma oscillations the xer1 recombinase gene has been disrupted in

Regulation of toxin production in the pathogenic clostridia.

The genus Clostridium comprises a large, heterogeneous group of obligate anaerobic, Gram-positive spore forming bacilli. Members of this genus are ubiquitous in the environment and although most species are considered saprophytic, several are pathogenic to both humans and animals. These bacteria cause a variety of diseases including neuroparalysis, gas gangrene, necrotic enteritis, food poisoning, toxic shock syndrome and pseudomembraneous colitis, which in most cases arise as a consequence of the production of potent exotoxins. Treatment options are often limited, underscoring the need for ne

Facile construction of unmarked deletion mutants in Burkholderia pseudomallei using sacB counter-selection in sucrose-resistant and sucrose-sensitive isolates.

Burkholderia pseudomallei is the causative agent of melioidosis, a potentially fatal disease endemic or emerging world-wide. Here we report unmarked allele-replacement mutagenesis using efficient sacB counter-selection. Despite being genotypically sacB(+), most commonly used B. pseudomallei strains are sucrose-resistant and efficient sacB counter-selection is demonstrated in both resistant and sensitive strains.

AroC as a key virulence factor and live-attenuated vaccine target in Pseudomonas plecoglossicida.

Pseudomonas plecoglossicida is a major etiological agent of visceral white-spot disease in diverse aquaculture species. Chorismate synthase (AroC), a key enzyme in the aromatic amino acid biosynthetic pathway, is essential for central metabolism and has been implicated in bacterial virulence. In this study, an aroC deletion mutant (ΔaroC) and a complemented strain (CΔaroC) were constructed to investigate the role of AroC in virulence regulation and to evaluate its potential as a live-attenuated vaccine candidate. Compared with the wild-type (WT) strain, ΔaroC exhibited significantly reduced gr

Transcriptomic and network analysis reveal methionine sulfoxide reductases dependent regulation of flagellar motility in Salmonella Typhimurium.

Salmonella enterica serovar Typhimurium (S. Typhimurium), a food-borne pathogen is the major cause of gastroenteritis and systemic infections in humans. Its survival and pathogenicity depend upon coordinated regulation of motility, oxidative stress response, and virulence-associated pathways. By repairing oxidized methionine (Met-SO) residues, the methionine sulfoxide reductases (Msrs) system protects bacterial proteins from oxidative damage and aid in bacterial survival in the host. Flagella enable S. Typhimurium to swim and swarm, facilitating movement through the intestinal mucus thus aid i

A lineage-specific heat-induced feedback loop controls HrcA to promote chlamydial fitness under stress.

Bacterial stress responses rely on transcriptional regulation by sigma factors and repressors. In Chlamydia, which lacks a dedicated heat shock sigma factor, repressor HrcA limits chaperone gene expression under non-stressed conditions. While HrcA function may be enhanced by the chaperone GroESL in a positive feedback loop, here we identify a heat-induced negative feedback loop mediated by HagF, a protein unique to Chlamydiota and conserved across Chlamydia species. We show that HrcA represses hagF, but heat shock induces hagF expression, leading to HagF accumulation and binding to HrcA. Struc

Early gene expression in Pseudomonas fluorescens exposed to a polymetallic solution.

The molecular response of Pseudomonas fluorescens cells exposed to a mixture of heavy metals remains largely unknown. Here, we studied the temporal changes in the early gene expression of P. fluorescens cells exposed to three doses of a polymetallic solution over two exposure times, through the application of a customized cDNA microarray. At the lowest metal dose (MD/4), we observed a repression of the Hsp70 chaperone system, MATE and MFS transporters, TonB membrane transporter and histidine kinases, together with an overexpression of metal transport (ChaC, CopC), chemotaxis and glutamine synt

Probable Interspecies Transfer of the bla(VIM-4) Gene between Enterobacter cloacae and Klebsiella pneumoniae in a Single Infant Patient.

We report the interspecies transfer of the bla(VLM-4) gene in MBL-producing Enterobacter cloacae and Klebsiella pneumoniae isolates from a newborn patient who had received meropenem therapy. We show evidence that gene bla(VIM-4) was transmitted as a part of the class-1 integron on a ca. -90 kb conjugative plasmid. High homology of nucleotide sequence was observed between the integron found in VIM-4 producing E. cloacae and K. pneumoniae strains tested and class-1 integrons previously reporteded in Pseudomonas aeruginosa from Hungary and Poland. This finding may suggest P. aeruginosa as a poten

Biofilm state transitions: quorum sensing inhibition and network destabilization.

Biofilms are traditionally viewed as surface-associated, extracellular polymeric substance (EPS)-encased aggregates. Here, we present a theoretical perspective positioning biofilm formation as a coordinated regulatory state transition, drawing on a synthesis of published transcriptomic, proteomic, and metabolic studies that document systemic reprogramming of gene regulatory networks, second-messenger signaling (e.g., cyclic di-GMP), quorum sensing circuits, and metabolic flux distribution. Spatial stratification within biofilms generates metabolically distinct subpopulations-including persiste

Differential expression of the human gonadotropin alpha gene in ectopic and eutopic cells.

We have analyzed the regulation of the alpha gonadotropin gene in eutopic placental cells and ectopic tumor cells by constructing a series of plasmid vectors containing alpha genomic 5' flanking DNA placed upstream of the gene encoding the bacterial enzyme chloramphenicol acetyltransferase (CAT). These plasmid DNAs were transfected into a eutopic (JAr) and an ectopic (HeLa) cell line. Both cell types expressed the CAT gene from plasmid constructs containing as much as 1,500 base pairs (bp) and as little as 140 bp of alpha 5' flanking DNA; JAr cells were considerably more efficient than HeLa ce

Growth phase-dependent expression of drug exporters in Escherichia coli and its contribution to drug tolerance.

Drug exporters contribute to the intrinsic drug resistance in many organisms. Although there are at least 20 exporter genes in Escherichia coli, most of them apparently do not confer drug resistance in complex laboratory media except for the AcrAB, EmrE, and MdfA efflux systems. In this study, we comprehensively investigated the growth phase-dependent expression of drug exporter genes. The expression of acrAB, emrAB, emrD, emrE, emrKY, mdfA, and ydgFE is stable at moderate levels during any growth phase, whereas mdtEF promoter activity greatly increased with cell growth and reached the maximum

Co-evolution and exploitation of host cell signaling pathways by bacterial pathogens.

Bacterial pathogens have evolved by combinations of gene acquisition, deletion, and modification, which increases their fitness. Additionally, bacteria are able to evolve in "quantum leaps" via the ability to promiscuously acquire new genes. Many bacterial pathogens - especially Gram-negative enteric pathogens - have evolved mechanisms by which to subvert signal transduction pathways of eukaryotic cells by expressing genes that mimic or regulate host protein factors involved in a variety of signaling cascades. This results in the ability to cause diseases ranging from tumor formation in plants

Increased expression of the type IV secretion system in piliated Neisseria gonorrhoeae variants.

Neisseria gonorrhoeae produces a type IV secretion system that secretes chromosomal DNA. The secreted DNA is active in the transformation of other gonococci in the population and may act to transfer antibiotic resistance genes and variant alleles for surface antigens, as well as other genes. We observed that gonococcal variants that produced type IV pili secreted more DNA than variants that were nonpiliated, suggesting that the process may be regulated. Using microarray analysis, we found that a piliated strain showed increased expression of the gene for the putative type IV secretion coupling

Evidence for moonlighting functions of the θ subunit of Escherichia coli DNA polymerase III.

The holE gene is an enterobacterial ORFan gene (open reading frame [ORF] with no detectable homology to other ORFs in a database). It encodes the θ subunit of the DNA polymerase III core complex. The precise function of the θ subunit within this complex is not well established, and loss of holE does not result in a noticeable phenotype. Paralogs of holE are also present on many conjugative plasmids and on phage P1 (hot gene). In this study, we provide evidence indicating that θ (HolE) exhibits structural and functional similarities to a family of nucleoid-associated regulatory proteins, the Hh
