Overexpression of YAP and TAZ is an independent predictor of prognosis in colorectal cancer and related to the proliferation and metastasis of colon cancer cells.

Источник пометил публикацию как отозванную

BACKGROUND AND OBJECTIVE: Yes-associated protein (YAP) and transcriptional co-activator with PDZ-binding motif (TAZ) are nuclear effectors of the Hippo pathway. Although they are abundantly expressed in the cytoplasm and nuclei of human colorectal cancer (CRC), and related to tumor proliferation status, there have been few studies on the predictive role of YAP and TAZ expression on the overall survival of patients with CRC. This study investigated YAP and TAZ expression in both CRC patients and colon cancer cell lines, and assessed their prognostic value. METHODS: Paraffin-embedded specimens

Armed oncolytic adenovirus elicits a "self-feeder" effect to supercharge NK cells for solid tumor control.

BACKGROUND: Natural killer (NK) cells are pivotal effectors in innate anti-tumor immunity, but their efficacy against solid tumors is constrained by inadequate tumor infiltration and functional suppression within the tumor microenvironment (TME). Although ex vivo expansion increases NK cell numbers, poor tumor homing and transient post-infusion activity persist as major limitations. This work aims to develop a combinatorial approach integrating ex vivo NK cell expansion with localized immunomodulation via engineered oncolytic adenoviruses (oAds) to address these challenges. METHODS: K562 feed

MT1H Inhibited Colon Cancer Development via Interacting With DUSP1 and MAPK Signaling Pathway Inactivation.

We focused on examining the impact of metallothionein 1H (MT1H) on colon cancer (CC) proliferation, migration, and invasion, as well as elucidating its underlying molecular mechanisms. Cell proliferation was assessed using the Cell Counting Kit-8 (CCK-8) assay. The invasive and migratory capacities of the cells were evaluated through transwell and wound healing assays. We found that both MT1H were lowly expressed in human CC tissues by immunohistochemistry (IHC). In HCT116 and SW480 cells and a xenograft mouse model, we found that overexpression of MT1H could inhibit CC cell proliferation, mig

Tailoring the biological activity of oxorhenium(V) complexes through Chrysin ligand functionalization.

Chrysin, structurally one of the simplest flavonoids, shows a broad spectrum of biological activities, particularly its anticancer potential. Additionally, it has the possibility of coordination to metal ions as a bidentate chelate ligand, forming highly stable metal complexes. This synergism of metal ions and Chrysin led to a significant improvement in its diverse biological activities. Chrysin (HL1) and its derivatives (methoxy (HL2), benzyloxy (HL3), acetylated (HL4), and silylated (HL5)) were coordinated as OO-bidentate ligands to the rhenium(V) ion, and all five complexes crystallized in

SPINK4 affected M2 macrophage polarization to promote colorectal cancer malignant phenotype by PI3K/AKT pathway.

This study aimed to elucidate the oncogenic role of SPINK4 in colorectal cancer (CRC) and its underlying mechanism regulating macrophage polarization in the tumor microenvironment. In vitro, we constructed SPINK4-overexpressing and SPINK4-knockdown CRC cell lines (HT29 and HCT116) to evaluate their effects on cell proliferation, migration, and invasion. We then established a macrophage-CRC cell co-culture system to explore whether SPINK4 promotes macrophage recruitment and M2 polarization. Mechanistically, we not only regulated the PI3K/AKT pathway using inhibitor LY294002 and activator 740Y-P

Seco-iridoid and lignans from Gymnosporia acuminata.

From the ethanol extract of Gymnosporia acuminata (syn. Maytenus hookeri), a new seco-iridoid derivative (1) and four lignans (2-5) were obtained. Significantly, the absolute configurations of the methoxypinoresinol derivatives 2 and 3 were unambiguously established for the first time by analysis of 2D NMR data combined with ECD calculations, identifying them as the 9'R and 9'S enantiomers. Meanwhile, compounds 4 and 5 exhibited moderate cytotoxic effects on HCT116 cells, with IC50 values of 41.0 and 49.0 μM.
