Gender differences in prevalence of substance use disorders among individuals with lifetime exposure to substances: results from a large representative sample.

BACKGROUND AND OBJECTIVES: Research regarding substance use and substance use disorders (SUDs) shows significant gender differences in prevalence of substance use and dependence. Though lifetime exposure to substances is higher among males, previous reports have not regarded gender differences in prevalence of SUDs among individuals formerly exposed to substances. In addition, though substance abuse is particularly important when exploring gender differences, previous reports have largely focused on rates of transition to substance dependence alone. In this study, we explored gender difference

Test-re-test reliability of DSM-IV adopted criteria for 3,4-methylenedioxymethamphetamine (MDMA) abuse and dependence: a cross-national study.

AIMS: This study evaluated the prevalence and reliability of DSM-IV adopted criteria for 3,4-methylenedioxymethamphetamine (MDMA) abuse and dependence with a purpose to determine whether it is best conceptualized within the category of hallucinogens, amphetamines or its own category. DESIGN: Test-re-test study. PARTICIPANTS: MDMA users (life-time use >5 times) were recruited in St Louis, Miami and Sydney (n=593). The median life-time MDMA consumption was 50 pills at the baseline. MEASUREMENTS: The computerized Substance Abuse Module for Club Drug (CD-SAM) was used to assess MDMA abuse and d

Opaque models: Using drugs and dreams to explore the neurobiological basis of mental phenomena.

On the basis of four historical and ethnographic case studies of modeling in neuroscience laboratories, this chapter introduces a distinction between transparent and opaque models. A transparent model is a simplified representation of a real world phenomenon. If it is not patently clear, it is at least much better comprehended than its objects of representation. An opaque model, by contrast, looks at one only partially understood phenomenon to stand in for another partially understood phenomenon. Here, the model is often just as complex as its target. Examples of such opaque models discussed i

Synthesis and Evaluation of Novel Aza-Aromatics as Dual 5‑HT2A and 5‑HT2C Receptor Agonists.

The 5-HT2A and 5-HT2C receptors are key therapeutic targets for CNS disorders. We investigated whether a nonhallucinogenic dual 5-HT2A/5-HT2C agonist could offer novel treatment potential. Large screening of in-house structurally diverse compounds revealed centhaquin, an FDA-approved hypovolemic shock drug, as a selective 5-HT2C agonist (EC50: 35 nM). We then synthesized 22 aza-aryl analogs with modified piperazine groups, and identified two dual agonists, 3ci and 3dh (EC50 < 1 μM), with no 5-HT2B activity up to 10 μM. Molecular docking highlighted critical interactions with Ser159 (5-HT2A) an

[Acute intoxication with "sobi-lobi" (Datura). Four cases in Niger].

The datura (Datura stramonium, Datura metel) belongs to a group of plants from the solanacies variety and can be the cause of voluntary intoxication. In Nigher, its flowers and seeds are used for their hallucinogenic properties. Its alcaloïds have an atropine-like effect. We are reporting here the first four cases of voluntary intoxication among teenagers in Niamey (Republic of Niger). The symptoms, which happened to be identical for the four patients, lead us quickly towards the possibility of a collective intoxication. During the questioning, the patients admitted they had smoked the plant's

Estimating Substance Use Homophily in the Sexual Network of a Large Cohort of Young Sexual and Gender Minorities Assigned Male at Birth.

Young sexual and gender minority (SGM) people assigned male at birth who use substances are at elevated risk of HIV acquisition. This brief report leverages a large cohort of SGM people assigned male at birth to estimate substance use homophily (i.e., same-same substance use status) in sexual partnerships. We found substance use homophily in this sexual network for marijuana, poppers, methamphetamine, and hallucinogens but not heavy episodic drinking, cocaine, or ecstasy. These results suggest substance use is associated with sexual network structure and may increase HIV disparities between in

How to spot illicit drug abuse in your patients.

Illicit drug abuse continues to become more widespread, especially in teenagers. Therefore, it is important for physicians to recognize the signs and symptoms of abuse in their patients. Drug abuse should be considered in differential diagnosis of many physical and nearly all psychiatric complaints. An understanding of the pharmacologic mechanisms and adverse effects of illicit drugs can enhance overall care of patients who abuse drugs. The primary classes of drugs of abuse--cannabinoids, opiates, stimulants, hallucinogens, and inhalants--produce clinically diverse presentations. By recognizin

Validity, reliability and clinical utility of ASSIST-Y in assessing risk of substance-related harm and dependence in Spanish male adolescents.

BACKGROUND: Substance use among adolescents is strongly associated with adverse physical, mental health, and social outcomes. Prevention and early intervention can reduce the likelihood of future problems, but requires valid and reliable screening tools capable of assessing risk across a range of substances. This study assessed the validity, reliability, and clinical utility of the Alcohol, Smoking and Substance Involvement Screening Test (ASSIST-Y) for adolescents aged 15-17 years. METHODS: A sample of adolescent males (N = 101), aged 15-17 years, held in a juvenile detention facility on sub

Psilocin and ketamine microdosing: effects of subchronic intermittent microdoses in the elevated plus-maze in male Wistar rats.

Short-term moderate doses of serotonergic and dissociative hallucinogens can be useful in the treatment of anxiety. Recently, a trend has developed for long-term intermittent 'microdosing' (usually one-tenth of a 'full' active dose), with reports of long-lasting relief from anxiety and related disorders; however, there is no scientific evidence for the efficacy of therapeutic microdosing nor to show its lasting effects. The objective of this study was to test for lasting effects on anxiety in rats after microdosing with ketamine or psilocin. Over 6 days, Wistar rats (N=40) were administered ke

Differentially classified methamphetamine-using men who have sex with men: A latent class analysis.

CONTEXT: Substance use interventions for methamphetamine-using men who have sex with men (MSM) are limited by the assumption that they are a uniform group. We hypothesized that an LCA would identify distinct patterns of substance use and demographic and psychosocial variables associated with different substance-using groups would aid in understanding distinctions. Using cross-sectional data from 343 methamphetamine-using MSM, we conducted an LCA to model the patterns of polysubstance use then examined how the classes varied on psychosocial variables defined by the Information-Motivation-Behavi

Shifting Narratives in Media Coverage Across a Decade of Drug Discourse in the Philadelphia Inquirer: Qualitative Sentiment Analysis.

BACKGROUND: The media has immense power in shaping public narratives surrounding sensitive topics such as substance use. Its portrayals can unintentionally fuel harmful stereotypes and stigma, negatively impacting individuals struggling with addiction, influencing policy decisions, and hindering broader public health efforts. OBJECTIVE: This study aimed to examine how the regional newspaper, The Philadelphia Inquirer, covered events related to illicit drug use between 2013 and 2022, focusing on linguistic patterns and themes associated with specific types of substances. METHODS: We collected

The Role of Environment, Early Use, Genetic Influences, and Neurobiology in Substance Use Disorder.

Substance use disorders (SUDs) have dire consequences for the individual, family, and community. Although SUDs were once viewed as a moral issue, they are now understood to be complex, chronic brain illnesses associated with both genetic and non-genetic influences. The American Psychiatric Association recognizes SUDs associated with the use of 10 different substances, for example, alcohol, nicotine, opioids, cannabis, caffeine, hallucinogens, inhalants, sedatives, hypnotics, and stimulants. Environmental exposure to substances, early use, genetic influences, and neurobiology have all been corr

Phencyclidine and chemical "stroking".

The current wave of drug abuse is but the latest manifestation of the real human need for "stroking." Drugs which affect the pleasure center of the brain are prime objects of abuse. Phencyclidine ("PCP" or "angel dust") is a cyclohexylamine which has properties of the amphetamine-stimulants, the narcotic-depressants and the hallucinogens. Previously unrecognized population groups include children less than six years of age and pregnant women and their intoxicated offspring. Successful treatment of present and future patients must be multidisciplinary and will ultimately depend upon a clear und

Classifying Psychedelic-Related Complications.

Classic psychedelics such as psilocybin, lysergic acid diethylamide (LSD), and 5-methoxy-dimethyltryptamine (5-MeO-DMT) have shown promising effects in the treatment of certain mental health conditions. Enthusiastic claims about their therapeutic potential have led to overly optimistic reactions in the media and the public, subsequently resulting in increased use outside of clinical contexts and a heightened rate of psychedelic-related complications. As psychedelics exhibit low toxicity and hardly any habit-forming potential, the typical risks and harms of these substances are often overlooked

Anxiety sensitivity and substance use disorders: Associations across multiple drug classes and tests of a transdiagnostic mechanism.

Anxiety sensitivity (AS), defined as the fear of anxiety-related symptoms, is a well-established transdiagnostic vulnerability linked to internalizing psychopathology (INT). AS is theorized to amplify distress and promote substance use through negative reinforcement processes. Although prior research has demonstrated associations between AS and substance use, most studies have focused on alcohol and cannabis use in undergraduate samples. The present study examined associations between AS and lifetime substance use disorder (SUD) diagnoses across multiple drug classes, including underexamined s

Psychoactive substances 25H-NBOMe and 25H-NBOH induce antidepressant-like behavior in male rats.

Ring-substituted phenethylamines are believed to induce psychedelic effects primarily by interacting with 5-hydroxytryptamine 2 (5-HT2A) receptors in the brain. We assessed the effect of the psychedelic substances 25H-NBOMe and 25H-NBOH on the depressive-like behavior of male adult rats. Naive Wistar rats were divided into groups to assess the effects of different doses (0.1 mg/kg, 1 mg/kg, and 3 mg/kg) of 25H-NBOMe and 25H-NBOH. The substances were administered intraperitoneally and the hallucinogenic properties were evaluated using the head twitch response test (HTR). Additionally, we assess

A cluster of 25B-NBOH poisonings following exposure to powder sold as lysergic acid diethylamide (LSD).

INTRODUCTION: 25B-NBOH is a synthetic hallucinogen closely related to the "NBOMe" family of N-substituted 2C phenethylamine derivatives. There have been no published reports documenting the clinical toxicity of NBOH derivatives. CASE SERIES: Five patients presented to the Emergency Department (ED) with altered conscious state following exposure to powder sold as "powdered LSD" at a party. A 24-year-old male who ingested the powder developed mydriasis, tachycardia, hypertension, and severe agitation requiring parenteral sedation. A 22-year-old male who insufflated the powder developed status e

LSD-stimulated behaviors in mice require β-arrestin 2 but not β-arrestin 1.

Recent evidence suggests that psychedelic drugs can exert beneficial effects on anxiety, depression, and ethanol and nicotine abuse in humans. However, their hallucinogenic side-effects often preclude their clinical use. Lysergic acid diethylamide (LSD) is a prototypical hallucinogen and its psychedelic actions are exerted through the 5-HT2A serotonin receptor (5-HT2AR). 5-HT2AR activation stimulates Gq- and β-arrestin- (βArr) mediated signaling. To separate these signaling modalities, we have used βArr1 and βArr2 mice. We find that LSD stimulates motor activities to similar extents in WT and
