Recent advances on the immunobiology of Bithynia spp. hosts of Opisthorchis viverrini.

This article reviews the past and present scientific reports regarding Bithynia spp. focusing on the biology, ecology and life cycle of Bithynia snails and their responses to Opisthorchis viverrini infection. Moreover, new data regarding comparative molecular genomics and proteomic approaches have recently revealed novel molecular components involved in the immune defence responses from Bithynia spp., providing additional perspectives for future studies. Studies on the specific interaction between Bithynia snails and their trematodes will contribute to further understanding the snail-parasite

Mosquito age and avian malaria infection.

BACKGROUND: The immune system of many insects wanes dramatically with age, leading to the general prediction that older insects should be more susceptible to infection than their younger counterparts. This prediction is however challenged by numerous studies showing that older insects are more resistant to a range of pathogens. The effect of age on susceptibility to infections is particularly relevant for mosquitoes given their role as vectors of malaria and other diseases. Despite this, the effect of mosquito age on Plasmodium susceptibility has been rarely explored, either experimentally or

The development of extra-intestinal cycle of Trypanosoma cruzi in Triatoma infestans and Panstrongylus megistus.

By applying various histological techniques the results obtained were similar to those in Lacombe's previous publications referring to the development of Trypanosoma cruzi in haemolymph, the later penetration in the Malpighian tubes and their return to the digestive tract. Trypomastigotes and sphaeromastigotes are found in the promesenteron after two hours of feeding the insect with mice blood containing numerous Trypanosoma cruzi. Some parasites move to the promesenteron from postmesenteron and others from the haemolymph of the insect. The amastigotes form inside the haemocytes, multiply, occ

Isolation and transmission of an unidentified Babesia sp. infective for cattle.

Engorged adult female ticks submitted from farms in South Africa were routinely screened for protozoan parasites by examination of haemolymph smears. An unidentified Babesia sp. was found in Hyalomma marginatum rufipes and its transmission to susceptible cattle was achieved both biologically (tick feeding) and mechanically (injection of infected blood). Attempts to transmit this species to susceptible rabbits and a horse using similar methods did not produce evidence of infection. This Babesia sp. was of low pathogenicity, even in splenectomized cattle. Morphologically, intra-erythrocytic piro

Transovarial transmission of murine typhus rickettsiae in Xenopsylla cheopis fleas.

It has been generally accepted that infected fleas do not pass on Rickettsia mooseri, or indeed any other known pathogen, to their progeny. It is reported here that such transovarial transmission does occur in laboratory-infected Xenopsylla cheopis fleas. By means of the direct fluorescent antibody test, Rickettsia mooseri was observed in cells of the hemolymph of infected fleas. As many as 11 percent of the adults and 2.9 percent of the larvae of the generation reared therefrom, had demonstrable rickettsiae. Moreover, batches of the F1 fleas were capable of transmitting the infection to more …

Host Compatibility of the Temnocephalid Diceratocephala boschmai: Experimental Transmission Between Invasive Cherax quadricarinatus and Native Macrobrachium rosenbergii in Malaysia.

BACKGROUND: The introduction of host-associated symbionts by invasive species threatens native aquatic biodiversity through symbiont spillover and host switching. This study investigated the experimental host-specificity and transmission potential of Diceratocephala boschmai from invasive Cherax quadricarinatus to the native giant freshwater prawn Macrobrachium rosenbergii. METHODS: Briefly, temnocephalid-infested C. quadricarinatus were exposed to symbiont-free M. rosenbergii and C. quadricarinatus for 30 days. Thereafter, symbiont transmission was assessed, and host hemolymph parameters, in…

Endocrine Control of Exaggerated Trait Growth in Rhinoceros Beetles.

Juvenile hormone (JH) is a key insect growth regulator frequently involved in modulating phenotypically plastic traits such as caste determination in eusocial species, wing polymorphisms in aphids, and mandible size in stag beetles. The jaw morphology of stag beetles is sexually-dimorphic and condition-dependent; males have larger jaws than females and those developing under optimum conditions are larger in overall body size and have disproportionately larger jaws than males raised under poor conditions. We have previously shown that large males have higher JH titers than small males during de

Participation of cell-free haemolymph of Biomphalaria tenagophila in the defence mechanism against Schistosoma mansoni sporocysts.

Biomphalaria tenagophila of Taim strain is able to completely destroy Schistosoma mansoni sporocyst few hours after parasite penetration, although the mechanism is still not well known. In this experimental work we show that passive transference of cell-free haemolymph, especially from B. tenagophila Taim, resulted in higher resistance of B. tenagophila Cabo Frio to S. mansoni infection. This effect was demonstrated in vivo, by the reduction in the infection rate, and the significantly lower production of sporocysts and cercariae of the parasite in snails treated with Taim cell-free haemolymph

Crustacean hyperglycemic hormone in the lobster nervous system: localization and release from cells in the subesophageal ganglion and thoracic second roots.

Crustacean hyperglycemic hormones (CHHs) are neuropeptides involved in the regulation of hemolymph glucose. The primary source of CHHs has been identified as the neurosecretory neurons of the eyestalk X-organ and its associated neurohemal organ, the sinus gland. We have identified another source of CHH-like peptides in the nervous system. With the use of immunocytochemistry, cells in the second roots of the thoracic ganglia have been observed to stain positively for CHH-reactive material. We also identified a pair of cells in the subesophageal ganglion that contain large amounts of CHH-reactiv…

Monoaminergic Systems in Flight-Induced Potentiation of Phonotactic Behavior in Female Crickets Gryllus bimaculatus.

We have recently shown that experience of flight remarkably enhanced subsequent terrestrial phonotaxis in females in response to the male calling song. Here, we elucidated the possible roles of octopamine and serotonin in the enhancing effect of flying on phonotactic behavior. Octopamine is known to be released into the hemolymph during flight in insects; however, the octopamine receptor antagonist epinastine did not abolish the effects of flight in our study. On the contrary, the drug significantly potentiated the influence of flying on phonotactic behavior. The octopamine receptor agonist ch…

Comparison between two methods of scorpion venom milking in Morocco.

BACKGROUND: The present study compared two methods used successfully in a large-scale program for the collection of scorpion venoms, namely the milking of adult scorpions via manual and electrical stimulation. RESULTS: Our immunobiochemical characterizations clearly demonstrate that regularly applied electrical stimulation obtains scorpion venom more easily and, most importantly, in greater quantity. Qualitatively, the electrically collected venom showed lack of hemolymph contaminants such as hemocyanin. In contrast, manual obtainment of venom subjects scorpions to maximal trauma, leading to …

Ablation of insulin-producing neurons in flies: growth and diabetic phenotypes.

In the fruit fly Drosophila, four insulin genes are coexpressed in small clusters of cells [insulin-producing cells (IPCs)] in the brain. Here, we show that ablation of these IPCs causes developmental delay, growth retardation, and elevated carbohydrate levels in larval hemolymph. All of the defects were reversed by ectopic expression of a Drosophila insulin transgene. On the basis of these functional data and the observation that IPCs release insulin into the circulatory system, we conclude that brain IPCs are the main systemic supply of insulin during larval growth. We propose that IPCs and …

Cyanoglycoside gynocardin fromAcraea horta (L.) (Lepidoptera: Acraeinae) : Possible implications for evolution of acraeine host choice.

All stages in the life cycle ofAcraea horta (L.) (Lepidoptera: Acraeinae) were found to release hydrogen cyanide (HCN) from their crushed tissues, and the source of cyanogenesis was present in the hemolymph of adults and larvae. Comparison with standards on thin-layer chromatograms (TLC) revealed the presence in adults of gynocardin, a cyclopentenyl cyanoglycoside also produced by the larval food plant,Kiggelaria africana L. (Flacourtiaceae). Analysis of adults reared on plant species (Passifloraceae) containing gynocardin and/or other cyanoglycosides suggested selective uptake of gynocardin b…

A serpin from the parasitoid wasp Leptopilina boulardi targets the Drosophila phenoloxidase cascade.

The insect phenoloxidase (PO) cascade is known to be tightly regulated by serine proteases and serine protease inhibitors of the serpin fami … y) type), and we show that either the venom or the recombinant LbSPNy inhibit the PO cascade in the hemolymph of Drosophila yakuba host larva. Altogether, our results identify the first serpin used as a virulence factor by a parasitoid wasp and show that it disrupts the activation pathway of the PO in the Drosophila host.

Heat stress is associated with disruption of ion balance in the migratory locust, Locusta migratoria.

Thermal tolerance is important in determining the spatial and temporal distributions of insects but the mechanisms which determine upper thermal limits remain poorly understood. In terrestrial insects heat tolerance is unlikely to be limited by oxygen supply but in some arthropods, heat stress has been shown to cause haemolymph hyperkalaemia which is known to have detrimental effects on neuromuscular excitability. It is however unresolved if heat-induced hyperkalemia is the cause or the result of cellular heat injury. To address the putative role of heat-induced hyperkalemia we quantified chan
