MST4, a new Ste20-related kinase that mediates cell growth and transformation via modulating ERK pathway.

In this study, we report the cloning and characterization of a novel human Ste20-related kinase that we designated MST4. The 416 amino acid full-length MST4 contains an amino-terminal kinase domain, which is highly homologous to MST3 and SOK, and a unique carboxy-terminal domain. Northern blot analysis indicated that MST4 is highly expressed in placenta, thymus, and peripheral blood leukocytes. Wild-type but not kinase-dead MST4 can phosphorylate myelin basic protein in an in vitro kinase assay. MST4 specifically activates ERK but not JNK or p38 MAPK in transient transfected cells or in stable

Sanguinarine Induces Pyroptosis via the Hippo Signaling Pathway and Remodels the Tumor Immune Microenvironment in Lung Adenocarcinoma.

Sanguinarine (SAN), a natural alkaloid derived from Papaveraceae plants, is an active component in traditional Chinese medicines. SAN exerts inhibitory effects on tumor cells, but research on its effects on lung adenocarcinoma (LUAD) and the underlying mechanisms remains limited. The Hippo signaling pathway is commonly downregulated in LUAD, making it a key target for therapeutic intervention. Our study reveals that SAN significantly inhibits LUAD. Mechanistically, SAN bound to Large Tumor Suppressor 1 (LATS1) and promoted its phosphorylation, thereby activating the Hippo pathway and pyroptosi

Lats1/2 Are Essential for Developmental Vascular Remodeling and Biomechanical Adaptation to Shear Stress.

BACKGROUND: Mechanical cues exerted by shear stress from blood flow remodel an initial vascular plexus into a ramifying array of large and small vessels. Hemodynamic forces trigger changes in endothelial cell (EC) gene expression and dynamic alterations in cell shape and adhesion. The objective of this study is to elucidate the role of the Lats (large tumor suppressor) 1 and Lats2 (Lats1/2) Hippo pathway kinases in EC transducing of hemodynamic signals as vessels form. METHODS: Lats1/2 were genetically deleted in murine ECs (LatsiECDKO) and developing vessels were evaluated using immunofluore

Spontaneous recovery from sunitinib-induced disruption of sarcomere in human iPSC-cardiomyocytes and possible involvement of the Hippo pathway.

BACKGROUND: Sunitinib is known to cause cardiotoxicity in clinical settings. However, among sunitinib-treated patients experiencing adverse cardiac events, decreased cardiac function was reportedly reversible in > 50% of the patients. We previously showed that anti-cancer drugs such as sunitinib cause marked sarcomere disruption in human induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs), and the extent of sarcomere disruption can be used to predict drug-induced cardiotoxicity in humans. The aim of this study is to investigate whether the reversibility of sunitinib-induced cardiac

Collagen synthesis disruption and downregulation of core elements of TGF-β, Hippo, and Wnt pathways in keratoconus corneas.

To understand better the factors contributing to keratoconus (KTCN), we performed comprehensive transcriptome profiling of human KTCN corneas for the first time using an RNA-Seq approach. Twenty-five KTCN and 25 non-KTCN corneas were enrolled in this study. After RNA extraction, total RNA libraries were prepared and sequenced. The discovery RNA-Seq analysis (in eight KTCN and eight non-KTCN corneas) was conducted first, after which the replication RNA-Seq experiment was performed on a second set of samples (17 KTCN and 17 non-KTCN corneas). Over 82% of the genes and almost 75% of the transcrip

Mst1-mediated phosphorylation of Nur77 improves the endometrial receptivity in human and mice.

BACKGROUND: Successful embryo implantation requires the attachment of a blastocyst to the receptive endometrial epithelium, which was disturbed in the women with recurrent implantation failure (RIF). Endometrial β3-integrin was the most important adhesion molecule contributing to endometrial receptivity in both humans and mice. Nur77 has been proven indispensable for fertility in mice, here we explore the role of Nur77 on embryo-epithelial adhesion and potential treatment to embryo implantation failure. METHODS: The expression and location of Mst1 and Nur77 in endometrium from fertile women a

Defective Leukocyte Adhesion and Chemotaxis Contributes to Combined Immunodeficiency in Humans with Autosomal Recessive MST1 Deficiency.

PURPOSE: To investigate the clinical and functional aspects of MST1 (STK4) deficiency in a profoundly CD4-lymphopenic kindred with a novel homozygous nonsense mutation in STK4. Although recent studies have described the cellular effects of murine Mst1 deficiency, the phenotype of MST1-deficient human lymphocytes has yet to be fully explored. Patient lymphocytes were therefore investigated in the context of current knowledge of murine Mst1 deficiency. METHODS: Genetic etiology was identified by whole exome sequencing of genomic DNA from two siblings, combined with linkage analysis in the wider

Interactome network analysis identifies multiple caspase-6 interactors involved in the pathogenesis of HD.

Caspase-6 (CASP6) has emerged as an important player in Huntington disease (HD), Alzheimer disease (AD) and cerebral ischemia, where it is activated early in the disease process. CASP6 also plays a key role in axonal degeneration, further underscoring the importance of this protease in neurodegenerative pathways. As a protein's function is modulated by its protein-protein interactions, we performed a high-throughput yeast-2-hybrid (Y2H) screen against ∼17,000 human proteins to gain further insight into the function of CASP6. We identified a high-confidence list of 87 potential CASP6 interactor

An Integrated Analysis of circRNA and lncRNA Expression of Bovine Granulosa Cells Induced by Melatonin Reveals the Pathways Potentially Involved in Follicular Development.

Objective: Accumulating evidence demonstrates that melatonin is involved in modulating granulosa cell function and follicular development. lncRNAs (long non-coding RNAs) and circRNAs (circular RNAs) have been reported to participate in multiple biological processes. This study aimed to explore the candidate circRNAs and lncRNAs related to molecular mechanisms when exploring the role of melatonin in regulating ovarian function. Methods: Bovine ovary granulosa cells were collected 48 h after treatment with melatonin at 10-7 M. The lncRNA and circRNA profiles of bovine granulosa cells were furthe

Amphiregulin reflects brain metastasis progression and leads to PD-L1 expression in non-small cell lung cancer cells.

BACKGROUND: The Hippo kinase Nuclear Dbf2-related kinase 2 (NDR2) promotes brain metastasis (BM) in non-small cell lung cancer (NSCLC) by the disrupting Yes-associated protein 1 (YAP-1), suggesting a role in circulating tumor cells and/or brain colonization. The underlying mechanism remains to be clarified. METHODS: Human bronchial epithelial tumor cells (A549, H1975, H2030 and the brain-tropic H2030-BrM3 line) with or without NDR2 depletion (via siRNA or shRNA), were exposed to shear stress (up to 40 dyn/cm² for 3 h) using an Ibidi® pump system, in the presence or absence of exogenous Amphir

Expression of a hepatitis B virus pre-S2 deletion mutant in the liver results in hepatomegaly and hepatocellular carcinoma in mice.

Hepatocellular carcinoma (HCC) is the most common form of liver cancer and has a poor prognosis and a low survival rate; its incidence is on the rise. Hepatitis B virus (HBV) infection is one of the main causes of HCC. A high prevalence of pre-S deletions of HBV surface antigen, which encompass T-cell and/or B-cell epitopes, is found in HBV carriers; antiviral therapy and viral immune escape may cause and select for these HBV mutants. In particular, the presence of pre-S2 deletion mutants is an important risk factor associated with cirrhosis and HCC. We generated Alb-preΔS2 transgenic mice tha

Inhibition of MST1/2 attenuates the anti-hepatocarcinogenesis effect of Salvianolic acid B via the ERK/MAPK signaling pathway.

Salvianolic acid B (Sal B), extracted from Salvia miltiorrhiza, has a commendable effect on liver-related disease. The Hippo/YAP and MAPK pathways were reported to exert synergistic effect on hepatocarcinogenesis. Our group has demonstrated that Sal B could restrain the MAPK pathway and postpone the hepatic fibrosis process. Also, Sal B exhibits anti-tumor activity by regulating the Hippo/YAP signaling. Simultaneously, the inhibition of the MAPK pathway could diminish the YAP expression in HepG2 cells; thus, we investigated the impact of MST1/2 gene double knockout (DKO) on the MAPK signaling

Cloning and characterization of a new intestinal inflammation-associated colonic epithelial Ste20-related protein kinase isoform.

Intestinal epithelial cells respond to inflammatory extracellular stimuli by activating mitogen activated protein kinase (MAPK) signaling, which mediates numerous pathophysiological effects, including intestinal inflammation. Here, we show that a novel isoform of SPS1-related proline alanine-rich kinase (SPAK/STE20) is involved in this inflammatory signaling cascade. We cloned and characterized a SPAK isoform from inflamed colon tissue, and found that this SPAK isoform lacked the characteristic PAPA box and alphaF loop found in SPAK. Based on genomic sequence analysis the lack of PAPA box and

Hippo-YAP/TAZ signalling at the crossroads of brain ageing and neurological disorders: mechanisms, crosstalk, and therapeutic prospects.

As life expectancy increases globally, ageing has emerged as the main risk factor driving the increasing burden of neurological disorders. Although advances have been made in understanding their pathogenesis, current therapies remain largely symptomatic and do not prevent disease progression. There is a growing body of evidence that these disorders share common molecular mechanisms, including oxidative stress, mitochondrial dysfunction, neuroinflammation, impaired autophagy, ferroptosis, and blood-brain barrier disruption. Recently, the Hippo signalling pathway has been identified as a key reg

The Noncanonical Role of Hippo Signaling in Cancer.

Originally discovered in Drosophila, the Hippo pathway has emerged as an evolutionarily conserved signaling pathway that controls tissue growth and organ size in species ranging from insects to human. Unsurprisingly, dysregulation of Hippo pathway activity has been implicated in a broad spectrum of human cancers. While the prevailing view holds that the Hippo pathway suppresses tumor growth by inhibiting the oncogenic activity of the pathway effector Yes-associated protein (YAP)/transcriptional coactivator with PDZ-binding motif (TAZ), recent studies have uncovered context-dependent tumor supp

MMRN1 suppresses thyroid cancer progression via activation of the Hippo signaling pathway.

INTRODUCTION: Thyroid cancer (TC) represents the most frequently diagnosed malignancy of the endocrine system, with its incidence continuing to increase worldwide. Multimerin 1 (MMRN1), a glycoprotein implicated in various cancers, exhibits context-dependent roles, but its function in TC remains unexplored. The Hippo pathway, a key regulator of cell proliferation and apoptosis, is frequently dysregulated in TC. We hypothesized that MMRN1 inhibits TC progression by activating the Hippo signaling pathway. METHODS: Bioinformatics analysis of GSE33630 identified differentially expressed genes (DE
