Characterization of spatial lipidomic signatures in tick-bitten guinea pig skin as a model for host-vector-pathogen interaction profiling.

Here, we demonstrate the adaptability of spatial "omics" methods to identify interphylum processes regulated at the vector-host interface of ticks during a mammalian blood meal. This approach enables a better understanding of complex bipartite or tripartite molecular interactions between hosts, arthropod vectors and transmitted pathogens, and contributes toward the development of spatially aware therapeutic target discovery and description.

The establishment of COPD organoids to study host-pathogen interaction reveals enhanced viral fitness of SARS-CoV-2 in bronchi.

Chronic obstructive pulmonary disease (COPD) is characterised by airflow limitation and infective exacerbations, however, in-vitro model systems for the study of host-pathogen interaction at the individual level are lacking. Here, we describe the establishment of nasopharyngeal and bronchial organoids from healthy individuals and COPD that recapitulate disease at the individual level. In contrast to healthy organoids, goblet cell hyperplasia and reduced ciliary beat frequency were observed in COPD organoids, hallmark features of the disease. Single-cell transcriptomics uncovered evidence for a

Functional characterization of the collagen-binding protein DIP2093 and its influence on host-pathogen interaction and arthritogenic potential of Corynebacterium diphtheriae.

Corynebacterium diphtheriae is typically recognized as the a etiological agent of diphtheria, a toxaemic infection of the respiratory tract; however, both non-toxigenic and toxigenic strains are increasingly isolated from cases of invasive infections. The molecular mechanisms responsible for bacterial colonization and dissemination to host tissues remain only partially understood. In this report, we investigated the role of DIP2093, described as a putative adhesin of the serine-aspartate repeat (Sdr) protein family in host-pathogen interactions of C. diphtheriae wild-type strain NCTC13129. Com…

Novel insights into host-pathogen interactions of large yellow croakers ( Larimichthys crocea) and pathogenic bacterium Pseudomonas plecoglossicida using time-resolved dual RNA-seq of infected spleens.

Host-pathogen interactions are highly complex, involving large dynamic changes in gene expression during infection. These interactions are fundamental to understanding anti-infection immunity of hosts, as well as the pathogenesis of pathogens. For bacterial pathogens interacting with animal hosts, time-resolved dual RNA-seq of infected tissue is difficult to perform due to low pathogen load in infected tissue. In this study, an acute infection model of Larimichthys crocea infected by Pseudomonas plecoglossicida was established. The spleens of infected fish exhibited typical symptoms, with a ma…

Molecular docking and dynamics simulation studies uncover the host-pathogen protein-protein interactions in soybean (Glycine max (L.) Merr.) and Groundnut bud necrosis virus: first report.

BACKGROUND: Soybean is a widely consumed oilseed crop with economic importance. It is enriched with numerous bioactive compounds that have health-promoting properties. Groundnut bud necrosis virus (GBNV) affects many agriculturally important crops, such as soybean, resulting in crop losses. GBNV is a single-stranded RNA virus from the genus Orthotospovirus and family Bunyaviridae. It is transmitted by insect vectors (thrips) and is further aggravated by secondary transmission from infected plants to others in the same field. There are no commercially available drugs against GBNV; thus, there i

Host responses and viral traits interact to shape the impacts of climate warming on highly pathogenic avian influenza in migratory waterfowl.

Emerging infectious diseases pose threats to wildlife populations, as exemplified by recent outbreaks of avian influenza viruses in wild birds. Climate change can affect infection dynamics in wildlife through direct effects on pathogens (e.g., environmental decay rates) and changes to host ecology, including shifting migration patterns. Here, we adapt an existing mechanistic model that couples migration and infection to study how traits of highly pathogenic avian influenza (HPAI) viruses contribute to HPAI outcomes in migratory waterfowl, then apply this model to explore potential impacts of c

Chemosensory protein 16 has an immune function and participates in host-pathogen interaction in Galleria mellonella infected with Pseudomonas entomophila.

Chemosensory protein 16 was identified in the hemolymph of Galleria mellonella as a protein with an amount increasing after oral infection with 10^3 CFU of Pseudomonas entomophila, and decreasing after infection with a higher dose (10^5 CFU) of bacteria. The expression of the CSP16 gene occurred in the fat body and in the gut and correlated with changes in the protein level in the hemolymph. The CSP16 protein inhibited P. entomophila growth in the concentration range from 0.15 to 6 nM. Additionally, the CSP16 protein showed bactericidal activity against P. entomophila, Bacillus thuringiensis,

Increased Pilus Production Conferred by a Naturally Occurring Mutation Alters Host-Pathogen Interaction in Favor of Carriage in Streptococcus pyogenes.

Studies of the human pathogen group A Streptococcus (GAS) define the carrier phenotype to be an increased ability to adhere to and persist on epithelial surfaces and a decreased ability to cause disease. We tested the hypothesis that a single amino acid change (Arg135Gly) in a highly conserved sensor kinase (LiaS) of a poorly defined GAS regulatory system contributes to a carrier phenotype through increased pilus production. When introduced into an emm serotype-matched invasive strain, the carrier allele (the gene encoding the LiaS protein with an arginine-to-glycine change at position 135 [li

Might Interspecific Interactions between Pathogens Drive Host Evolution? The Case of Plasmodium Species and Duffy-Negativity in Human Populations.

Malarial infections have long been recognized as a driver of human evolution, as demonstrated by the influence of Plasmodium falciparum on sickle-cell anemia persistence. Duffy-negativity is another blood disorder thought to have been selected because it confers nearly complete resistance against Plasmodium vivax infection. Recent evidence suggests that the benefits of being Duffy-negative cannot be expected to play a strong selective pressure on humans, whereas its costs cannot be considered as negligible. Here, we suggest that the cross-talk between P. falciparum and P. vivax in coinfected c

Maintenance of brucellosis in Yellowstone bison: linking seasonal food resources, host-pathogen interaction, and life-history trade-offs.

The seasonal availability of food resources is an important factor shaping the life-history strategies of organisms. During times of nutritional restriction, physiological trade-offs can induce periods of immune suppression, thereby increasing susceptibility to infectious disease. Our goal was to provide a conceptual framework describing how the endemic level bovine brucellosis (Brucella abortus) may be maintained in Yellowstone bison based on the seasonality of food resources and the life-history strategies of the host and pathogen. Our analysis was based on active B. abortus infection (measu

Virus and host-associated variations in the interaction of low-pathogenic avian influenza viruses with the epithelial target tissue of the chicken reproductive tract.

Low-pathogenic avian influenza viruses (LPAIVs) show pathogenesis variations, suggested of being impacted by virus and host characteristics. This study aimed to understand the interaction of virus and host factors in LPAIV reproductive tract infection. Oviduct organ cultures (OOCs) were prepared from two chicken lines, brown layers (BL) and white layers (WL), to compare virus replication, microscopical lesions, and host-immune responses between two atypical virulent LPAIV strains H3N1 (A/chicken/Belgium/460/2019) and H6N1 (A/chicken/Netherlands/917/2010), and an avirulent H9N2 strain (A/chicke

Development of a high-brightness, genetically stable fluorescent reporter system for Mycoplasma pneumoniae and its application in host-pathogen interaction studies.

This study aimed to develop a genetically stable, high-brightness fluorescent reporter system for Mycoplasma pneumoniae (MP) and to demonstrate its utility in investigating host-pathogen interactions. The system is based on the mNeonGreen fluorescent protein. Its expression level was enhanced by utilizing the promoter of the MP tuf gene, while transposition efficiency was improved by codon optimization of the transposase gene. To facilitate the selection of recombinant mycoplasmas containing the transposon, strategies involving antibiotic resistance gene and promoter optimization were employed…

Salivarian Trypanosomes Have Adopted Intricate Host-Pathogen Interaction Mechanisms That Ensure Survival in Plain Sight of the Adaptive Immune System.

Salivarian trypanosomes are extracellular parasites affecting humans, livestock and game animals. Trypanosoma brucei rhodesiense and Trypanosoma brucei gambiense are human infective sub-species of T. brucei causing human African trypanosomiasis (HAT-sleeping sickness). The related T. b. brucei parasite lacks the resistance to survive in human serum, and only inflicts animal infections. Animal trypanosomiasis (AT) is not restricted to Africa, but is present on all continents. T. congolense and T. vivax are the most widespread pathogenic trypanosomes in sub-Saharan Africa. Through mechanical tra

Differential Viral-Host Immune Interactions Associated with Oseltamivir-Resistant H275Y and Wild-Type H1N1 A(pdm09) Influenza Virus Pathogenicity.

Oseltamivir is a common therapy against influenza A virus (IAV) infections. The acquisition of oseltamivir resistance (OR) mutations, such as H275Y, hampers viral fitness. However, OR H1N1 viruses have demonstrated the ability to spread throughout different populations. The objective of this work was to compare the fitness of two strains of OR (R6 and R7) containing the H275Y mutation, and a wild-type (F) pandemic influenza A (H1N1) 2009 (pdm09) virus both in vitro and in vivo in mice and to select one OR strain for a comparison with F in ferrets. R6 showed faster replication and pathogenicity

Tracing amino acid exchange during host-pathogen interaction by combined stable-isotope time-resolved Raman spectral imaging.

This study investigates the temporal and spatial interchange of the aromatic amino acid phenylalanine (Phe) between human retinal pigment epithelial cell line (ARPE-19) and tachyzoites of the apicomplexan protozoan parasite Toxoplasma gondii (T. gondii). Stable isotope labelling by amino acids in cell culture (SILAC) is combined with Raman micro-spectroscopy to selectively monitor the incorporation of deuterium-labelled Phe into proteins in individual live tachyzoites. Our results show a very rapid uptake of l-Phe(D8) by the intracellular growing parasite. T. gondii tachyzoites are capable of

DNAM-1 mediates NK-cell activation and host-pathogen interaction via direct binding to fungal cell wall proteases.

Natural killer (NK) cells play a key role in combating invasive mycoses, with surface proteins crucial for immune cell-target interactions. Here, we investigate the role of DNAX accessory molecule 1 (DNAM-1; CD226) in the recognition of Aspergillus fumigatus and Candida albicans hyphae. Super-resolution microscopy reveals a uniform distribution of DNAM-1 on naïve NK cells that is maintained upon contact with fungal hyphae. However, soluble DNAM-1 binds to the fungal cell wall and blocking DNAM-1 reduces antifungal activity in colony-forming assays. Furthermore, in silico domain-domain interact

Draft genome of the Crimson rosella (Platycercus elegans elegans), an Australian parrot and key resource for the study of host-pathogen interaction in Psittaciformes.

OBJECTIVES: The Crimson rosella (Platycercus elegans) is a common parrot of Australia, and most individuals are infected with Beak and Feather Disease Virus (BFDV). In other parrot species, the virus can cause feather loss, beak deformities, immune suppression and mortality. All parrots globally are considered susceptible to the virus and thus there are sizable conservation benefits from better understanding of its host-pathogen interactions and evolution, particularly because parrots are already among the most threatened bird taxa globally. The draft genome reported here is both a valuable sc…

Integrated clinical and metabolomic analysis of dengue infection shows molecular signatures associated with host-pathogen interaction in different phases of the disease.

PURPOSE: Dengue is a mosquito vector-borne disease caused by the dengue virus, which affects 125 million people globally. The disease causes considerable morbidity. The disease, based on symptoms, is classified into three characteristic phases, which can further lead to complications in the second phase. Molecular signatures that are associated with the three phases have not been well characterized. We performed an integrated clinical and metabolomic analysis of our patient cohort and compared it with omics data from the literature to identify signatures unique to the different phases. METHOD
