[Comparison of integration processing technology of origin and traditional cutting processing technology of Moslae Herba for lung-Yang deficiency rats].

To compare the effect of integration processing technology of origin (IPTO) and traditional cutting processing technology (TCPT) of Moslae Herba for lung-Yang deficiency rats caused by complex factors, analyze the mechanism, and provide the modern pharmacology basis for the implementation of IPTO of Moslae Herba. The rat models of lung-Yang deficiency were established by smoking + swimming in ice water + drinking ice water. The model rats were randomly divided into different groups, and were treated with intragastric administration for 30 d. Then the general signs, anal temperature and autonom

[FREQUENCY OF IMMUNOGLOBULIN E DEFICIENCY AMONG PATIENTS WITH IMMUNODEPENDENT DISORDERS].

Isolated IgE deficiency is one of the most common primary immunodeficiency, which is still underestimated cause of health disorders of modern man. Recent genetic studies report that the cause of the IgE deficiency is immune dysregulation caused by polymorphisms of the gene, which is responsible for the synthesis of activation-induced cytidinedeaminase (AICDA). The Institute of Immunology and Allergology at Bogomolets NMU during the years 2012-2014 were examined 5298 patients with a range of different diseases, which could be suspected violations of immunity. All patients were conducted compreh

Deficiency of T helper cells in transient hypogammaglobulinemia of infancy.

We studied 17 patients with transient hypogammaglobulinemia of infancy to define the immunologic defect responsible for this disorder. The number of circulating B cells in these patients was normal, as was the ability of the B cells to synthesize immunoglobulins when stimulated with Epstein-Barr virus, a direct B-cell activator. However, the capacity of the B cells to synthesize IgG in response to pokeweed mitogen, a T-cell-dependent B-cell activator, was depressed. Experiments with cultured lymphocytes indicated that excess suppressor-cell activity was not present in these patients, but that

Clearance of Pneumocystis carinii in mice is dependent on B cells but not on P carinii-specific antibody.

Both CD4(+) T cells and B cells are critical for defense against Pneumocystis carinii infection; however, the mechanism by which B cells mediate protection is unknown. We show that P. carinii-specific IgM is not sufficient to mediate clearance of P. carinii from the lungs since CD40-deficient mice produced normal levels of specific IgM, but were unable to clear the organisms. Using chimeric mice in which the B cells were deficient in CD40 (CD40KO chimeras) we found that clearance of P. carinii infection is delayed compared with wild-type controls. These CD40KO chimeric mice produced normal lev

Overview of antibody-mediated immunity to S. pneumoniae: pneumococcal infections, pneumococcal immunity assessment, and recommendations for IG product evaluation.

Streptococcus pneumoniae (S. pneumoniae) strains colonize the nasopharynx and can cause mucosal infections in the upper airway and middle ear, pneumonias, and invasive infections like bacteremia, sepsis, and meningitis. Over 90 serotypes, defined by the structure of their capsular polysaccharides, are known. Twenty-three of these serotypes cause most infections and several of these serotypes can develop antibiotic resistance. Susceptibility factors that increase the susceptibility to S. pneumoniae mucosal and invasive infections include all forms of primary and secondary antibody deficiencies.

Malnutrition Decreases Antibody Secreting Cell Numbers Induced by an Oral Attenuated Human Rotavirus Vaccine in a Human Infant Fecal Microbiota Transplanted Gnotobiotic Pig Model.

Human rotavirus (HRV) is a leading cause of morbidity and mortality in children, especially in developing countries. Malnutrition is prevalent in these countries, which may contribute to the decreased oral vaccine efficacy, posing a concern for global health. Neonatal gnotobiotic (Gn) pigs closely resemble human infants in their anatomy, physiology, and outbred status and are a unique model to investigate malnutrition, oral live attenuated HRV (AttHRV) vaccination, and subsequent virulent HRV (VirHRV) challenge. We evaluated the impact of malnutrition on AttHRV vaccine efficacy and B cell immu

Expression of human IgG subclasses.

Human immunoglobulin G (IgG) can be divided into four subclasses that are selectively expressed. For instance, carbohydrate antigens preferentially elicit IgG2 antibodies, whereas protein antigens usually elicit IgG1 and IgG3. Elucidating the biological basis of the selective expression of these IgG subclasses is important to our understanding immunodeficiencies and B lymphocyte development. To investigate clinical importance of IgG subclass deficiencies, a sensitive and specific assay has been developed for IgG subclasses using particle concentration fluorescence immunoassay. Preliminary clin

Peripheral immunoglobulin secreting cells in immunodeficiencies; effects of intravenous immune globulin.

Peripheral blood B cells secreting IgG, IgA, IgM, and IgE were quantitated in normal adults (n = 12), newborns (n = 8), patients with antibody deficiency (n = 5), and patients with elevated IgE (four patients) using a reverse enzyme-linked immunospot (RELISPOT) assay. This technique measures immunoglobulin secreted by B cells by capture on an antibody-coated plate, and identified as a plaque on a nitrocellulose-membrane plate. Hypogammaglobulinemic patients and newborns (cord blood) showed no detectable IgG, IgA, or IgE secreting cells. Several cord blood and hypogammaglobulinemic patients, ho

Reciprocal effects of Schistosoma mansoni infection on spontaneous autoimmune arthritis in IL-1 receptor antagonist-deficient mice.

Schistosome infections have been shown to prevent inflammation in induced-type arthritis models. However, its effects on spontaneous arthritis remain unknown. We here investigated the effects of Schistosoma mansoni (Sm) infection on spontaneous autoimmune arthritis in IL-1 receptor antagonist (IL-1Ra)-deficient mice. Sm infection partially reduced the severity of arthritis in male IL-1Ra-deficient mice. The splenic responses of IL-17 and TNF-α were reduced, while those of IL-4 and IL-10 were enhanced by the infection. However, Sm infection increased IgG rheumatoid factor and anti-dsDNA IgG ser

Selective IgA deficiency and coeliac disease.

Twenty-five children and adults with concomitant coeliac disease and selective IgA deficiency are described. IgG-class reticulin antibodies were positive in 94%. The clinical course of coeliac disease did not differ from that of patients with normal serum IgA level. Patients with IgA deficiency also had other concomitant diseases, especially autoimmune diseases. Patients with selective IgA deficiency have at least a tenfold risk of coeliac disease compared with the population in general.

Kinetics of immunoglobulin and specific antibody responses of CBA mice infected with Trypanosoma rhodesiense.

Groups of CBA/CaJ and B-cell deficient CBA/N mice were infected with Trypanosoma rhodesiense EATRO 1886 strain. Survival, parasitaemia, serum Ig levels plus specific trypanosomal IgM and IgG antibodies were assayed and compared during infection. Whereas both strains of mice had similar parasitaemias during the first week of infection, CBA/N parasitaemias were lower than those observed in CBA/CaJ mice during the subsequent study period. Antibody responses, specific for T. rhodesience antigens, peaked on day 10 after infection in CBA/CaJ mice, then rapidly declined. However, antibody responses i

The use of intravenous immunoglobulin for the treatment of infection: an overview.

Immunoglobulin replacement therapy in the form of intravenous immunoglobulin (IVIgG) is clearly of benefit in primary hypogammaglobulinaemia and related disorders involving antibody deficiency. However, its use in the prevention of infection in other conditions is controversial and needs to be clarified. IVIgG therapy may be of benefit in selected patients with IgG subclass deficiency, and with a proven history of recurrent upper respiratory tract infections. IVIgG therapy may also benefit infants with AIDS and recurrent bacterial infections, but is only rarely of value in cases of adult AIDS.

Clinical uses of intravenous immune globulin: immunoglobulin replacement therapy and treatment of autoimmune cytopenias.

Intravenous immune globulin (IVIG) is indicated for IgG replacement in antibody deficiency syndromes and as immunoregulatory therapy in some autoimmune diseases. Two case histories illustrate both aspects of IVIG therapy. 1. Patient 1 is a 24-year-old male with agammaglobulinemia. He was successively treated with monthly IMIG, paternal plasma, and then over the last 5 years, IVIG. On IgG doses of 250 mg/kg q/4 weeks, IgG trough levels remained below 200 mg IgG/dl. IgG half-life was reduced. Although asymptomatic for prolonged periods of time, he eventually developed clinically evident inflamma

Periodontal disease mechanisms in immunocompromised patients.

Deficiency in the number and function of phagocytes is associated with gingival inflammation and periodontitis. A hereditary deficiency in membrane glycoproteins involved in granulocyte adherence causes impaired chemotaxis, reduced phagocytosis and periodontal problems. Virus infections of antigen-presenting cells interfere with immune responses and lead to seriously increased susceptibility to infections with bacteria which cause no problems in normal patients. Increased levels of IgG antibodies may limit penetration of antigens in the tissues, but at the cost of local inflammation and tissue

[Common variable immunodeficiency in children].

UNLABELLED: Common variable immunodeficiency (CVID) is one of the more frequent primary immunodeficiencies (PID), after IgA deficiency, and affects a heterogeneous group of patients of various ages and with autosomal recessive inheritance. Our objective is to present the group of children diagnosed with CVID treated in our Hospital Infantil Vall d'Hebron and comment on the diagnostic problems that can arise. Sixteen boys and girls were diagnosed between the ages of 7 months and 15 years. The diagnosis is based on low immunoglobulins and a clinical picture of infection. Differential diagnosis i
