Influence of gonadal factors on skin test reactivity of CFW mice to Candida albicans.

Sex differences in the delayed skin test response to Candida albicans were studied in gonadectomized and control CFW mice sensitized either actively with sonically treated antigen or passively with lymphoid cells. In mice actively sensitized with antigen, the responses to candida skin testing were larger in the female. In both sexes, the skin test responses were larger in mice gonadectomized prior to sensitization than in controls of the same sex. In mice passively sensitized with male lymphoid cells, the responses to candida skin testing were essentially equal in the sexes. In both sexes, gon

Ontogeny of the immune system.

Cellular and humoral aspects of the immune response develop sequentially in the fetus. Although there is evidence of immunologic activity to some antigens, many of the nonimmune effector systems are neither developed nor fully functional in the fetus. The lack of fully functional systems make the fetus a highly susceptible candidate for invading bacteria and viruses. Often, infection prior to immune competence may lead to abortion, malformation, and in some instances, viral persistence and immune tolerance. On the other hand, immune responses tend to elevate immunoglobulin values and cause spe

Immune expulsion of Trichuris muris from mice during a primary infection: analysis of the components involved.

Immune serum accelerated the expulsion of Trichuris muris when transferred into normal mice on days 0 and 3 after infection, but had no effect when the recipient mice had been immunosuppressed by sublethal irradiation or by cortisone treatment. Delaying serum transfer until days 7 and 8 in normal mice failed to accelerate expulsion, although immune mesenteric lymph node cells (MLNC) accelerated expulsion whether transferred early or late in infection. Expulsion from NIH mice, normally complete by 12 days, was prevented by sublethal irradiation given as late as 9 days after infection, but could

Passive transfer of delayed hypersensitivity to Blastomyces dermatitidis between mice.

This study further characterized the delayed hypersensitivity state induced in animals by Blastomyces dermatitidis exposure. Passive transfer of delayed hypersensitivity by transfer of cells and inhibition of migration of peritoneal exudate cells were studied, using sensitized mice of two inbred strains. Donor mice were subcutaneously inoculated with viable B. dermatitidis yeast cells. After 15 days, spleen cells or serum from these animals were injected intravenously into normal recipients of the same strain. After 24 h these mice were footpad tested with killed B. dermatitidis yeast cell ant

The immune response of the lactating rat to Nippostrongylus brasiliensis.

It has previously been shown that rats do not expel a primary infection of Nippostrongylus brasiliensis during lactation. In the present paper, three aspects of the immune response of the lactating rat to the parasite were examined and compared with the response of non-lactating rats. Protective and reaginic antibodies were present in the circulation of rats first infected during lactation, in titres as high or higher than those in non-lactating controls. The intestinal mast cell response was similar both in timing and magnitude and lactating rats became susceptible to shock with the i.v. inje

High-Starch Diet Prepartum Enhances IgG Concentration in Goat Colostrum Without Affecting the Transfer of Passive Immunity.

This study hypothesises that a high-starch (HS) diet during the last month of gestation enhances colostrum yield and composition as well as the dam and goat kid metabolism, immune status and performance. Thirty multiparous and pregnant Majorera dairy goats were randomly assigned to a prepartum dietary treatment (CON vs. HS) on wk -4 relative to the expected parturition. Goats were fed either a control (n = 15; 25% DM of starch) or HS (n = 15; 32% DM of starch) diet during the last month of gestation. Blood samples were collected on wk -4, -3, -2 and -1 relative to expected parturition, immedia

Transmission of maternal muscle-specific tyrosine kinase (MuSK) to offspring: report of two cases.

Familial occurrence of myasthenia gravis is uncommon and reports of maternal transmission of muscle-specific tyrosine kinase (MuSK) antibody myasthenia are rarer still. We report two families with maternal transmission of MuSK antibody myasthenia gravis to the offspring by different mechanisms. The first family demonstrates transmission genetic susceptibility of inheriting myasthenia gravis from MuSK antibodies, whereas the second one demonstrates transplacental transmission of MuSK antibodies at birth.

Transplacental IgG subclass concentrations in pregnancies at risk of haemolytic disease of the newborn.

The relationship of haemolytic disease of the newborn (HDN) to the transplacental passage of the four IgG subclasses was assessed at various gestational ages by comparing the maternal and fetal IgG subclass concentrations in 34 pregnancies at risk of HDN with those in 30 pregnancies not at risk. Higher maternal and fetal IgG1 levels were attained in pregnancies at risk of HDN than in pregnancies not at risk. In contrast, a slight decrease in maternal IgG2 and IgG4 levels occurred in pregnancies at risk of HDN, as compared with a slight rise in maternal IgG2 and IgG4 levels in pregnancies not a

Rat model of congenital toxoplasmosis: rate of transmission of three Toxoplasma gondii strains to fetuses and protective effect of a chronic infection.

The incidence of congenital toxoplasmosis in Fischer rats infected between the 8th and 12th days of pregnancy with three different strains of Toxoplasma gondii (RH, 76K, and Prugniaud) were 58.2, 35.2, and 62.8%, respectively. No infected fetuses were collected from rats previously infected with RH or Prugniaud strain parasites, even if the rats were reinfected during pregnancy. Since pups from chronically infected mothers are protected from congenital toxoplasmosis, rat infection could thus constitute a relevant model for immunological studies and vaccine design.

Malaria in pregnancy.

In its booklet 'Tropical Diseases 1990' the World Health Organization (WHO) Division of Control of Tropical Diseases (CTD) said, "Malaria remains the most important of the tropical diseases--widespread throughout the tropics, but also occurring in many temperate regions". The size of the malarial scourge was estimated as: number of people infected 267 million; clinical cases 107 million/year; mortality 1-2 million/year; number of people at risk 2100 million. Immunity to malaria is reduced in pregnancy. This makes the disease particularly dangerous for pregnant women, especially for nulliparae.

Natural history of perinatal cytomegaloviral infection.

Epidemiological data presented here indicate that cytomegaloviral (CMV) infection is one of the most common perinatal infections found in human beings. Transmission to the offspring occurs in utero at birth and postnatally. Intrauterine infection results from primary or recurrent maternal involvement, the latter being more common in populations where infection is initially acquired during childhood or adolescence, such as in low socioeconomic settings. Congenital infection is usually subclinical with either type of maternal involvement but primary infection has a greater tendency to produce di

Current prospects for immunization against cytomegaloviral disease.

Cytomegaloviral (CMV) seronegative women who acquire a primary CMV infection during pregnancy are at the greatest risk for delivering infants who may be deaf or intellectually handicapped because of congenital CMV infection. Maternal immunization before pregnancy may protect newborns from congenital disease because mothers who are naturally seropositive are protected against secondary infection and because newborns who acquire CMV either via transfusion or transplacentally are protected if their mothers had antibodies to CMV prior to pregnancy. Further evidence for the feasibility of immunizat

Vaccination against Hendra virus of previously unvaccinated mares and their foals demonstrates the effectiveness of the current prime and boost vaccination schedule for foals.

Hendra virus (HeV) is a high consequence pathogen that causes severe respiratory and/or neurological disease in both horses and humans with high mortality. The equine vaccine, Equivac® HeV, is a recombinant soluble version of the G glycoprotein of HeV (HeV-sG) that elicits a potent HeV-neutralising antibody response in adult horses and also provides indirect protection for humans. Maternally derived HeV-G specific antibodies could interfere with subsequent foal vaccination, although other factors such as foal age and the suitability of vaccination protocols that have been optimised for adult h

Prophylactic use of canine parvovirus monoclonal antibody induces blockade of vaccinal canine parvovirus immunization similar to maternally derived passive immunity.

OBJECTIVE: To determine initial antibody titer, degradation profile, and duration of passive antibody blockade of active immunization by modified-live viral (MLV) canine parvovirus (CPV-2) vaccine following CPV monoclonal antibody (CPMA) prophylactic subcutaneous (SC) administration to CPV-2-naïve puppies. METHODS: Purpose-bred, CPV-2-seronegative male and female Beagle dogs, 7 to 8 weeks of age, were randomized to CPMA treatment (n = 13) and control (7) groups. Dogs were commingled and group housed in a shower-in/shower-out facility. At study day 0, dogs were administered either 0.1 mL/kg bo

Maternal immunization. Clinical experiences, challenges, and opportunities in vaccine acceptance.

Maternal immunization holds tremendous promise to improve maternal and neonatal health for a number of infectious conditions. The unique susceptibilities of pregnant women to infectious conditions, as well as the ability of maternally-derived antibody to offer vital neonatal protection (via placental transfer), together have produced the recent increased attention on maternal immunization. The Advisory Committee on Immunization Practices (ACIP) currently recommends 2 immunizations for all pregnant women lacking contraindication, inactivated Influenza and tetanus toxoid, reduced diphtheria toxo

Adoptive transfer of immunity to Histoplasma capsulatum in athymic nude mice.

Studies of adoptive transfer of resistance to Histoplasma capsulatum were performed with athymic nude (nu/nu) recipients and heterozygous (nu/+) donor mice. This model has the benefit of employing a recipient mouse (nu/nu) which cannot be immunized actively against H. capsulatum. Nu/nu mice were protected against H. capsulatum by whole spleen cells, fractioned T lymphocytes, and macrophages from immunized nu/+ donor mice. Similar cell populations from nonimmune nu/+ mice were not protective. Treatment with gold sodium thiomalate, an inhibitor of macrophage function, increased susceptibility to
