Mitochonic Acid 5 Binds Mitochondria and Ameliorates Renal Tubular and Cardiac Myocyte Damage.

Mitochondrial dysfunction causes increased oxidative stress and depletion of ATP, which are involved in the etiology of a variety of renal diseases, such as CKD, AKI, and steroid-resistant nephrotic syndrome. Antioxidant therapies are being investigated, but clinical outcomes have yet to be determined. Recently, we reported that a newly synthesized indole derivative, mitochonic acid 5 (MA-5), increases cellular ATP level and survival of fibroblasts from patients with mitochondrial disease. MA-5 modulates mitochondrial ATP synthesis independently of oxidative phosphorylation and the electron tr

Selection for biocontrol bacteria antagonistic toward Rosellinia necatrix by enrichment of competitive avocado root tip colonizers.

Biological control of soil-borne pathogens is frequently based on the application of antagonistic microorganisms selected solely for their ability to produce in vitro antifungal factors. The aim of this work was to select bacteria that efficiently colonize the roots of avocado plants and display antagonism towards Rosellinia necatrix, the causal agent of avocado white root rot. A high frequency of antagonistic strains (ten isolates, 24.4%) was obtained using a novel procedure based on the selection of competitive avocado root tip colonizers. Amplification and sequencing of the 16S rRNA gene, i

Effects of an endophytic bacterial consortium derived from Zingiberaceae on growth, yield, and curcuminoid biosynthesis in Curcuma longa L.

Endophytic bacteria, integral to the plant microbiome, possess considerable potential for improving the cultivation and sustainability of medicinal plants. These bacteria are essential for various plant functions, including growth enhancement, nutrient uptake, and resistance to biotic and abiotic stressors. This study recovered 29 endophytic bacterial isolates from the rhizomes of five Zingiberaceae species: Zingiber officinale, Curcuma longa, Kaempferia galanga, Alpinia purpurata, and Hedychium coronarium. These isolates were evaluated for plant growth-promoting (PGP) traits, including minera

Chitosan/indole-3-acetic acid/silver nanoparticle composite films: Preparation, characterization and efficacy in inhibiting postharvest potato dry rot.

This study aimed to prepare and characterize a chitosan (CS)-based composite film incorporating indole-3-acetic acid (IAA) and silver nanoparticles (AgNPs) via a solution casting method, and to evaluate its efficacy in controlling Fusarium oxysporum-induced dry rot in postharvest potatoes. Microstructural analysis revealed uniformly dispersed AgNPs and a compact network, with FTIR and XRD confirming that hydrogen bonding dominates CS and IAA interactions while coordination interactions govern CS and AgNP associations. This compact network significantly enhanced tensile strength (40.09% increas

Screening and optimization of salt-alkali tolerant phosphate-solubilizing Enterobacter sp. AD8: applications in soybean growth promotion and saline-alkali soil remediation.

Soil salinization severely limits crop growth, nutrient availability and phosphorus use efficiency. Combined saline-alkali stress and phosphorus deficiency greatly restrict soybean productivity. Applying salt-alkali tolerant phosphate-solubilizing bacteria is an effective biological strategy to improve crop performance and soil fertility. In this study, 15 strains were isolated from local saline-alkali soil. After functional screening, Enterobacter sp. AD8 showed excellent phosphate-solubilizing (264.21 mg/L soluble P) and IAA-producing (58.09 µg/mL) capacities. Its optimal culture conditions

Bioorthogonal catalytic centres engineered for gastrointestinal stabilization provide oral delivery for the treatment of gastric cancer.

BACKGROUND: Gastric cancer, the fifth most prevalent cancer globally, poses significant treatment challenges due to factors such as late diagnosis, early metastasis, limited surgical options, and the systemic toxicity of chemotherapy. Because luminal barriers are often compromised in gastric cancers , orally administered therapies that enable localized absorption and drug release represent a promising new direction for site-specific treatment with limited side effects. RESULTS: We introduced disulfide-linked thermostable exoshell system that orally delivered protein-based bioorthogonal cataly

Synergistic Effect of Indole-3-Acetic Acid and Homogeneously Dispersed Ulva prolifera-Derived Carbon Quantum Dots in Enhancing CO2 Fixation, Biomass Production, and Macromolecules in Chlorella vulgaris.

To address light attenuation caused by self-shading in dense microalgal cultures within photobioreactors, this study proposes a synergistic strategy employing Ulva prolifera-derived carbon quantum dots (U-CQDs) combined with indole-3-acetic acid (IAA). Hydrothermally synthesized U‑CQDs exhibiting fluorescence emission at 580 nm were homogeneously suspended in the culture, converting UV‑blue light into orange‑red wavelengths readily absorbed by microalgae, thereby improving light distribution uniformity. The combination of 1 mg·L-1 U-CQDs and 5 mg·L-1 IAA increased biomass production to 2.17 g·

Minute-scale control of ubiquitin-mediated degradation reveals dynamics of bacterial secreted effector-functions.

Precise temporal control of protein abundance is essential for dissecting dynamic cellular processes. While degron-based systems enable rapid protein depletion in eukaryotic cells, comparable tools are lacking for bacterial effectors delivered into host cells during infection. Here, we establish AIDE (Auxin-Inducible Degradation of Effectors), a host-directed degradation platform that harnesses the ubiquitin-proteasome system to selectively eliminate secreted bacterial proteins, including membrane-integrated effectors. By integrating a minimal auxin-inducible degron (AID) tag into effector gen

Unraveling the molecular mechanisms underlying flat stem formation in Atractylodes lancea in response to phytoplasmas.

For centuries, the herb Atractylodes lancea (Thunb.) has been revered for its pharmacological properties, particularly its efficacy in combating spleen and stomach ailments. However, prolonged artificial cultivation of A. lancea has led to an increased presence of serious plant diseases, including flat branch disease caused by phytoplasmas. To elucidate the mechanisms of flat stem (FS) formation, physiological and biochemical indicators, as well as related metabolic pathways were assessed in A. lancea stems following phytoplasma infection. The results demonstrated significant changes in the le

Alteration in Indole Metabolites After Cardiopulmonary Bypass Surgery in Neonates and Infants.

IMPORTANCE: Cardiopulmonary bypass (CPB) surgery is associated with changes in the intestinal microbiome. Metabolism of tryptophan into the indole pathway is entirely facilitated by the intestinal microbiome, and indole metabolites play a critical role in intestinal epithelial integrity, intestinal and systemic vascular tone, and intestinal and systemic immune response. OBJECTIVES: To evaluate the impact of CPB on microbial-derived indole metabolites and their association with clinical outcomes. DESIGN, SETTING, AND PARTICIPANTS: Prospective cohort study of neonates and infants younger than

H2S-Mediated GH3.1 Persulfidation Regulates IAA Homeostasis to Enhance Nodulation Formation and Nitrogen Fixation in Robinia pseudoacacia.

Hydrogen sulphide (H2S), a gaseous signalling molecule, plays a multifaceted role in plant physiology by enhancing adaptability to environmental stresses. However, the regulatory mechanism of symbiotic nitrogen (N) fixation by H2S in indeterminate nodules of woody legumes remains unclear. In this study, we investigated the mechanism by which H2S promotes nodulation and N fixation in the woody legume Robinia pseudoacacia. Exogenous H2S significantly enhanced rhizobium infection, nodule formation and nitrogenase activity, demonstrating its positive role in the symbiotic process. Transcriptomic a

SHADE AVOIDANCE 4 Regulates Magnesium Deficiency-Induced Root Hair Development by Regulating Auxin Transport in Arabidopsis.

Root hairs enlarge root surface and facilitate plant exploration for edaphic resources, particularly under nutrient-limiting conditions. While magnesium (Mg) deficiency is known to markedly stimulate root hair development, the underlying molecular mechanisms remain poorly characterised. Here, we elucidate the critical role of SHADE AVOIDANCE 4 (SAV4) in regulating Mg deficiency-induced root hair elongation through modulating epidermal auxin level in Arabidopsis. Root hair elongation under Mg deficiency was significantly suppressed in sav4 mutant, and SAV4 epidermal-specific expression partiall

Ethylene response factor 113 regulates the growth and development of poplar through the tryptophan-dependent auxin metabolism.

ERF transcription factors play a pivotal role in orchestrating plant growth and development processes. In this study, PagERF113 was isolated from the 84 K poplar (Populus alba × P. glandulosa) and its function was detected using molecular and genetic methods. Subcellular localization and trans-activation assays indicated that PagERF113 lacks transcriptional activation capability, residing in both the cell membrane and nucleus. Phenotypic analyses revealed that transgenic poplar lines overexpressing PagERF113 (OE) exhibited weedy growth compared to wild-type (WT), characterized by small leaves,

RLI2 regulates Arabidopsis female gametophyte and embryo development by facilitating the assembly of the translational machinery.

Eukaryotic protein translation is a complex process that requires the participation of different proteins. Defects in the translational machinery often result in embryonic lethality or severe growth defects. Here, we report that RNase L inhibitor 2/ATP-BINDING CASSETTE E2 (RLI2/ABCE2) regulates translation in Arabidopsis thaliana. Null mutation of rli2 is gametophytic and embryonic lethal, whereas knockdown of RLI2 causes pleiotropic developmental defects. RLI2 interacts with several translation-related factors. Knockdown of RLI2 affects the translational efficiency of a subset of proteins inv

Auxin-independent effects of apical dominance induce changes in phytohormones correlated with bud outgrowth.

The inhibition of shoot branching by the growing shoot tip of plants, termed apical dominance, was originally thought to be mediated by auxin. Recently, the importance of the shoot tip sink strength during apical dominance has re-emerged with recent studies highlighting roles for sugars in promoting branching. This raises many unanswered questions on the relative roles of auxin and sugars in apical dominance. Here we show that auxin depletion after decapitation is not always the initial trigger of rapid cytokinin (CK) increases in buds that are instead correlated with enhanced sugars. Auxin ma

The N-Terminal Acetyltransferase Naa50 Regulates Arabidopsis Growth and Osmotic Stress Response.

N-terminal acetylation (Nt-acetylation) is one of the most common protein modifications in eukaryotes. The function of Naa50, the catalytic subunit of the evolutionarily conserved N-terminal acetyltransferase (Nat) E complex, has not been reported in Arabidopsis. In this study, we found that a loss of Naa50 resulted in a pleiotropic phenotype that included dwarfism and sterility, premature leaf senescence and a shortened primary root. Further analysis revealed that root cell patterning and various root cell properties were severely impaired in naa50 mutant plants. Moreover, defects in auxin di

Rice dwarf and low tillering 10 (OsDLT10) regulates tiller number by monitoring auxin homeostasis.

Tiller number is a crucial agronomic trait that directly affects the number of effective panicles and yield formation in rice. Here, we report a semi-dwarf and low tillering mutant Osdlt10 (dwarf and low tillering 10) that exhibited reduced tiller number, semi-dwarfism, increased grain width, low seed-setting rate, curled leaf tip and a series of abnormalities of agronomic traits. Phenotypic observations showed that Osdlt10 mutants had defects in tiller bud formation and grew slowly at the tillering stage. Map-based cloning revealed that LOC_Os10g41310 was the responsible gene for OsDLT10, whi

Auxin production in diploid microsporocytes is necessary and sufficient for early stages of pollen development.

Gametophytic development in Arabidopsis depends on nutrients and cell wall materials from sporophytic cells. However, it is not clear whether hormones and signaling molecules from sporophytic tissues are also required for gametophytic development. Herein, we show that auxin produced by the flavin monooxygenases YUC2 and YUC6 in the sporophytic microsporocytes is essential for early stages of pollen development. The first asymmetric mitotic division (PMI) of haploid microspores is the earliest event in male gametophyte development. Microspore development in yuc2yuc6 double mutants arrests befor

Development of potent fluorescent polyamine toxins and application in labeling of ionotropic glutamate receptors in hippocampal neurons.

The natural product argiotoxin-636 (ArgTX-636) found in the venom of the Argiope lobata spider is a potent open-channel blocker of ionotropic glutamate (iGlu) receptors, and recently, two analogues, ArgTX-75 and ArgTX-48, were identified with increased potency and selectivity for iGlu receptor subtypes. Here, we have exploited these analogues as templates in the development of fluorescent iGlu receptor ligands to be employed as unique tools for dynamic studies. Eighteen fluorescent analogues were designed and synthesized, and subsequently pharmacologically evaluated at three iGlu receptor subt
