Is Lipid Metabolism of Value in Cancer Research and Treatment? Part II: Role of Specialized Pro-Resolving Mediators in Inflammation, Infections, and Cancer.

Acute inflammation is the body's first defense in response to pathogens or injury that is partially governed by a novel genus of endogenous lipid mediators that orchestrate the resolution of inflammation, coined specialized pro-resolving mediators (SPMs). SPMs, derived from omega-3-polyunstaturated fatty acids (PUFAs), include the eicosapentaenoic acid-derived and docosahexaenoic acid-derived Resolvins, Protectins, and Maresins. Herein, we review their biosynthesis, structural characteristics, and therapeutic effectiveness in various diseases such as ischemia, viral infections, periodontitis,

Accelerated brain aging with opioid misuse and HIV: New insights on the role of glially derived pro-inflammation mediators and neuronal chloride homeostasis.

Opioid use disorder (OUD) has become a national crisis and contributes to the spread of human immunodeficiency virus (HIV) infection. Emerging evidence and advances in experimental models, methodology, and our understanding of disease processes at the molecular and cellular levels reveal that opioids per se can directly exacerbate the pathophysiology of neuroHIV. Despite substantial inroads, the impact of OUD on the severity, development, and prognosis of neuroHIV and HIV-associated neurocognitive disorders is not fully understood. In this review, we explore current evidence that OUD and neuro

Purification and partial characterization of an anticoagulant PLA2 from the venom of Indian Daboia russelii that induces inflammation through upregulation of proinflammatory mediators.

The present study describes the purification and partial characterization of a basic anticoagulant PLA2 enzyme named as Rv(i) PLA2 from the venom of Indian Daboia russelii. The molecular mass of the protein was found to be 13,659.65 Da, and peptide mass fingerprinting revealed that it belongs to group II PLA2 family. The peptide sequence showed similarity to uncharacterized basic PLA2 enzyme having an accession no. of P86368 reported from Sri Lankan D. russelii. Rv(i) PLA2 exhibited strong phospholipase A2 and anticoagulant activity. It also induced expression of COX-2 and TNF-α mRNA in a dose

Evidence of different mediators of central inflammation in dysfunctional and inflammatory pain--interleukin-8 in fibromyalgia and interleukin-1 β in rheumatoid arthritis.

The purpose of this study was to relate central inflammation to autonomic activity (heart rate variability (HRV)) in patients with rheumatoid arthritis (RA) and fibromyalgia (FM). RA patients had reduced parasympathetic activity and FM patients had increased sympathetic activity compared to healthy controls. Comparisons between RA and FM showed higher cerebrospinal fluid (CSF) interleukin (IL)-1β inversely correlated to parasympathetic activity in RA. The FM patients had higher concentrations of CSF IL-8, IL-1Ra, IL-4 and IL-10, but none of these cytokines correlated with HRV. In conclusion, w

Comparative analysis of two low-level laser doses on the expression of inflammatory mediators and on neutrophils and macrophages in acute joint inflammation.

Synovial membrane inflammation plays an important role in osteoarthritis (OA) pathophysiology. The synovial tissue of patients with initial OA is characterized by mononuclear cell infiltration and the production of pro-inflammatory cytokines and other mediators of joint injury. The study aims to evaluate the effect of low-level laser therapy (LLLT) at doses of 2 and 4 J on joint inflammation in rats induced by papain through histopathological analysis, differential counts of inflammatory cells; gene expression of IL-1β, IL-6, and IL-10; and TNF-α protein expression. Male Wistar rats (20) were

Indirect evidence of nasal inflammation assessed by titration of inflammatory mediators and enumeration of cells in nasal secretions of patients with chronic rhinitis.

Pathophysiologic mechanisms of perennial rhinitis are poorly understood. The characterization of inflammation was studied in nasal lavage of patients with perennial rhinitis by the enumeration of cells involved in the allergic inflammation and the measurement of six mediators released in nasal secretions to determine whether some mediators were relevant for the etiologic diagnosis and the occurrence of symptoms. Ten healthy subjects and 57 patients with perennial rhinitis were placed into four groups according to the symptoms they presented at the time of the study and the origin of the allerg

From biomarkers to therapeutic targets: Biomaterial-mediated cfDNA scavenging in inflammation.

Cell-free DNA (cfDNA) displays disease-dependent changes in abundance, fragmentation, composition, and tissue origin. Beyond serving as a noninvasive biomarker, persistent cfDNA can act as a damage-associated molecular pattern, activating TLR9, cGAS-STING, inflammasome signaling, and thromboinflammation. This review synthesizes advances in cfDNA biology, clinical applications, and therapeutic scavenging across inflammatory diseases. We critically evaluate the extent to which current clinical studies support diagnostic, prognostic, and causal conclusions while considering methodological heterog

NINJ1-mediated membrane rupture bridges cell death to tissue inflammation.

Plasma membrane rupture (PMR) is a terminal event in many forms of lytic cell death, yet was long regarded as a passive consequence of osmotic swelling and mechanical membrane failure. The identification of ninjurin-1 (NINJ1) as a key mediator of PMR fundamentally changed this view by establishing that membrane permeabilisation and terminal membrane rupture are mechanistically distinct processes. Across different cell death modalities, NINJ1 promotes the transition from membrane damage to catastrophic cellular disintegration. Structural studies have revealed how NINJ1 is restrained by autoinhi

Metformin triggers ATF4-mediated UPRmt and COX-2 inflammation in adipocytes.

Metformin is the most common drug for type 2 diabetes due to its action to improve insulin sensitivity and enhance glucose uptake in tissues, including adipose tissue. As a mitochondrial complex I inhibitor, treatment with metformin may cause deleterious effects. Here, we demonstrated that treatment of adipocytes 3T3-L1 cells with a high concentration of metformin (10 mM) led to increased reactive oxygen species (ROS) accumulation and triggered the mitochondrial unfolded protein response (UPRmt), as revealed by increased mRNA expression of UPRmt markers (mtHSP70, Lonp1, and FGF21). High-concen

The mediating role of inflammation and nutrition metabolism in the methylmalonic acid-stroke association.

BACKGROUND: This study explored the association between Methylmalonic Acid (MMA, the primary exposure factor) and stroke, and the potential mediating roles of systemic inflammatory marker (Neutrophil-to-Lymphocyte Ratio [NLR]) and nutritional-metabolic parameters (Triglycerides [TG], Fasting Plasma Glucose [FPG]). Based on biological plausibility and evidence-based rationale, these three indicators were selected a priori and incorporated as core mediating variables in the analytical model. METHODS: We included 9430 participants from NHANES 2011-2014, comprising 712 stroke cases and 8718 non-s

Contribution of endometrial microbiome to inflammation-mediated infertility in women undergoing ART.

STUDY QUESTION: Is the endometrial microbiome altered in women who fail to get pregnant after ART and do microbial-derived metabolites influence endometrial cellular mechanisms important for embryo implantation? SUMMARY ANSWER: The endometrial microbiome in women who fail to get pregnant after ART is more diverse and has fewer lactobacilli species than the endometrial microbiomes of women who become pregnant; the short-chain fatty acid butyrate, a common metabolite found in the presence of increased microbial diversity, diminishes endometrial epithelial barrier function and increases the expr

Specialized proresolving mediator-loaded extracellular vesicles mitigate pulmonary inflammation.

Extracellular vesicles (EVs) have emerged as versatile carriers of therapeutic cargo, including nucleic acids, proteins, and small molecules. However, their potential to deliver bioactive lipid mediators remains largely unexplored. Here, we present a novel synthetic biology-based strategy to selectively load EVs with proresolving lipid mediators of the resolvin D- and E-series by coexpressing the resolvin biosynthetic enzymes cyclooxygenase 2, 5-lipoxygenase, and 15-lipoxygenase using a custom-designed multigene expression vector. Human embryonic kidney 293 T cells transfected with the multige

IL10 is a potent modulator of IL1-mediated inflammation in bovine endometrial explants.

CONTEXT: Bovine uterine inflammation impairs fertility, animal welfare and economics. Balanced regulation of interleukin-1 family proteins is crucial to resolve endometrial inflammation. AIM: mRNA expression of interleukin 1B (IL1B) and its receptor antagonist (IL1RA) was examined in bovine endometrial explants. METHODS: Endometrial explants from 26 cows were incubated with heat-inactivated pathogenic Escherichia coli, opportunistic Bacillus pumilus or commensal and potentially protective Lactobacillus buchneri (3, 14 and 24 h) and with cytokines (24 h; IL1B, IL17A, IL10). mRNA expression of

AXL mediates influenza A virus entry and drives virus-induced inflammation.

Influenza A virus (IAV) infection remains a persistent global health challenge with limited antiviral drugs. Viral entry represents a potential multifaceted target for antiviral intervention; however, there is a limited number of approved inhibitors that effectively prevent IAV entry. Here, we identify the receptor tyrosine kinase AXL as an IAV functional entry factor with dual roles in viral invasion and host immune modulation. Although AXL has been previously reported as a negative regulator of inflammation, we reveal that its inhibition simultaneously reduces viral entry and virus-induced i

Quantification of Kidney Inflammation Using Nanobubble-mediated Contrast Enhanced Ultrasound.

Kidney inflammation is a central driver of acute kidney injury (AKI) and its progression to chronic kidney disease (CKD). While several imaging and biomarker-based approaches are under development, clinically validated non-invasive methods to directly quantify renal inflammation remain limited. This study introduces a novel approach using contrast-enhanced ultrasound (CEUS) with Cy5-labeled nanobubbles (NBs) to address this critical knowledge gap. Using a murine ischemia-reperfusion injury (IRI) model, CEUS imaging enabled real-time visualization of inflammation-induced changes in kidney perfu

Lipoprotein (a) integrates monocyte-mediated thrombosis and inflammation in atherosclerotic cardiovascular disease.

Elevated levels of lipoprotein (a) [Lp(a)], an apolipoprotein B particle, are causally linked to atherosclerotic cardiovascular disease (ASCVD). Lp(a) is thought to promote ASCVD through multiple mechanisms, including its effects on cholesterol transport, inflammation, and thrombosis. This study defines the mechanisms that integrate Lp(a)-mediated cholesterol accumulation, inflammation, and thrombosis. In this study, we employed systems biology approaches, including proteomics, transcriptomics, and mass cytometry, to define the immune cellular and molecular phenotypes in patients with ASCVD ha

Insulin resistance as potential mediator linking ApoB/ApoA1 to MAFLD, but not inflammation.

AIMS: Metabolic-associated fatty liver disease (MAFLD) is closely associated with insulin resistance (IR) and systemic inflammation. Apolipoprotein A1 (ApoA1) and Apolipoprotein B (ApoB), as notable non-traditional lipid markers, have demonstrated distinct advantages in identifying risks related to metabolic syndrome and coronary atherosclerosis, yet its association with MAFLD and the mediating roles of IR/inflammation remain unclear. METHODS: This retrospective investigation involved 1061 participants, categorized into a non-MAFLD group (n = 529) and an MAFLD group (n = 532). Univariate and
