Adverse Effects of Pesticides on the Ovary: Evidence from Epidemiological and Toxicological Studies.

Pesticides are widely used in agriculture, public health, and residences to control pests and insects. The safety of exposure to pesticides has raised concerns due to their presence in the environment and their potential effects on human health. The ovary is the major female reproductive organ and is considered a potential target organ for pesticide toxicity. This comprehensive Review examines the adverse effects of pesticide exposure on the ovary based on evidence from human biomonitoring, epidemiological studies, and toxicological studies. Epidemiological studies have shown that pesticide ex

Juvenile hormone as a key regulator for asymmetric caste differentiation in ants.

Caste differentiation involves many functional traits that diverge during larval growth and metamorphosis to produce adults irreversibly adapted to reproductive division of labor. Investigating developmental differentiation is important for general biological understanding and has increasingly been explored for social phenotypes that diverge in parallel from similar genotypes. Here, we use Monomorium pharaonis ants to investigate the extent to which canalized worker development can be shifted toward gyne (virgin-queen) phenotypes by juvenile hormone (JH) treatment. We show that excess JH can a

Hormonal regulation and functional role of the "renal" tubules in the disease vector, Aedes aegypti.

The Aedes aegypti mosquito is a vector responsible for transmitting various arboviruses including dengue and yellow fever. Their ability to regulate the ionic and water composition of their hemolymph is a major physiological phenomenon, allowing the mosquito to adapt to a range of ecological niches. Hematophagus insects, including the female A. aegypti, face the challenge of excess salt and water intake after a blood meal. Post-prandial diuresis is under rigorous control by neuroendocrine factors, acting on the Malpighian "renal" tubules (MTs), to regulate primary urine production. The MTs are

Stage- and cell-specific expression of ecdysone receptors and ecdysone-induced transcription factors during midgut remodeling in the yellow fever mosquito, Aedes aegypti.

In insects, especially in mosquitoes that are adult blood feeders, midgut remodeling is an important event during metamorphosis. It involves two processes viz., programmed cell death (PCD) of larval cells, and proliferation and differentiation of imaginal cells to form pupal/adult midgut. These processes are regulated by 20-hydroxyecdysone (20E) and juvenile hormone (JH), but the signaling mechanisms, which trigger specific changes remain poorly understood. Here, we report stage- and cell-specific expression of ecydone receptor (EcR), ultraspiracle (USP), broad (Br), E75B and hormone receptor

The effect of pyriproxyfen on the concentration of circulating metabolic fuel molecules and chemical elements in the hemolymph of Acraea horta L. (Lepidoptera: Nymphalidae): A quantitative analysis.

Many pollinating insects expand their niche to adjacent agricultural areas and are, therefore, exposed to chemical insecticides. Acraea horta L. (Lepidoptera: Nymphalidae) is a pollinator butterfly widely distributed in the Southern African region. The objectives of this work were to evaluate carbohydrate, lipid and chemical elements in the hemolymph of A. horta exposed to pyriproxyfen, a juvenile hormone analog (JHA). Last instar larvae (L6: day 1 or day 2) were topically exposed to an aqueous solution of pyriproxyfen (100 μg of the active ingredient per insect) or to diluent (control group).

Environmental safety review of methoprene and bacterially-derived pesticides commonly used for sustained mosquito control.

Some pesticides are applied directly to aquatic systems to reduce numbers of mosquito larvae (larvicides) and thereby reduce transmission of pathogens that mosquitoes vector to humans and wildlife. Sustained, environmentally-safe control of larval mosquitoes is particularly needed for highly productive waters (e.g., catchment basins, water treatment facilities, septic systems), but also for other habitats to maintain control and reduce inspection costs. Common biorational pesticides include the insect juvenile hormone mimic methoprene and pesticides derived from the bacteria Bacillus thuringie

Endocrine Control of Exaggerated Trait Growth in Rhinoceros Beetles.

Juvenile hormone (JH) is a key insect growth regulator frequently involved in modulating phenotypically plastic traits such as caste determination in eusocial species, wing polymorphisms in aphids, and mandible size in stag beetles. The jaw morphology of stag beetles is sexually-dimorphic and condition-dependent; males have larger jaws than females and those developing under optimum conditions are larger in overall body size and have disproportionately larger jaws than males raised under poor conditions. We have previously shown that large males have higher JH titers than small males during de

Physiological responses and transcriptome analysis of Hemerocallis citrina Baroni exposed to Thrips palmi feeding stress.

Thrips are serious pests of Hemerocallis citrina Baroni (daylily), affecting crop yield and quality. To defend against pests, daylily has evolved a set of sophisticated defense mechanisms. In the present study, induction of systemic resistance in Hemerocallis citrina 'Datong Huanghua' by Thrips palmi feeding was investigated at both biochemical and molecular levels. The soluble sugar content of daylily leaves was significantly lower than that in control check (CK) at all time points of feeding by T. palmi, whereas the amino acid and free fatty acid contents started to be significantly lower th

Juvenile hormone enhances viral activity in bumble bees.

Bees are susceptible to viral infections that can compromise individual and colony health. As viruses depend on host cellular resources for replication, we investigated the influence of a key insect regulator, Juvenile hormone (JH), which enhances reproduction and metabolic rate on viral activity. We hypothesized that elevated JH levels would promote viral replication in a tissue-specific manner. To test this hypothesis, we used the bumble bee, Bombus terrestris, and the Israeli acute paralysis virus (IAPV). In our first experiment, we compared IAPV replication in pre- and post-diapause queens

Western diet consumption by host vertebrate promotes altered gene expression on Aedes aegypti reducing its lifespan and increasing fertility following blood feeding.

BACKGROUND: The high prevalence of metabolic syndrome in low- and middle-income countries is linked to an increase in Western diet consumption, characterized by a high intake of processed foods, which impacts the levels of blood sugar and lipids, hormones, and cytokines. Hematophagous insect vectors, such as the yellow fever mosquito Aedes aegypti, rely on blood meals for reproduction and development and are therefore exposed to the components of blood plasma. However, the impact of the alteration of blood composition due to malnutrition and metabolic conditions on mosquito biology remains und

Intraluminal Farnesol and Farnesal in the Mealworm's Alimentary Canal: An Unusual Storage Site Uncovering Hidden Eukaryote Ca2+-Homeostasis-Dependent "Golgicrine" Activities.

Farnesol, the sesquiterpenoid precursor of the six presently known insect juvenile hormones (JHs) was for the first time chemically identified in 1961, not in JH synthesizing glands or whole body extracts, but in excrements of the mealworm Tenebrio molitor. This finding was thought to be irrelevant and remained unexplored. In 1970, it was reported that the fall to zero of the JH titer in both prediapausing adults and in last instar larvae of the Colorado potato beetle causes severe malfunctioning of the Golgi system in the fat body, among various other effects. This endomembrane system in the

RNAi-mediated knockdown of juvenile hormone acid methyltransferase depresses reproductive performance in female Aethina tumida.

Small hive beetles, Aethina tumide, are free-flying parasites of social bee colonies where they feed and reproduce. In case of mass infestation, A. tumida can cause significant economic losses. There is an urgent need to explore novel green molecular approaches for sustainable control of A. tumida. It has been confirmed that juvenile hormone acid methyl transferase (JHAMT) plays a crucial role in regulating the synthesis of juvenile hormone (JH). However, its impact on female reproduction of A. tumida remains unclear. In the present study, a novel JHAMT gene was identified from A. tumida with

CYP321F3 mediates metabolic resistance to methoxyfenozide in rice stem borer, Chilo suppressalis.

The development of insecticide resistance in insect populations is a major challenge to sustainable agriculture and food security worldwide. Methoxyfenozide, an insect growth regulator that acts as an agonist of 20-hydroxyecdysone (20E), has severely declined in its efficacy against the rice stem borer (Chilo suppressalis), a notorious pest of rice crops in East and Southeast Asia. To date, however, the genes involved in methoxyfenozide resistance in target pests remain unclear. We conducted a long-term (seven years from 2017 to 2023) and large geographical scale (8 provinces and 45 cities in

A South African Scenario: Structure and Function of Peptides of the Adipokinetic Hormone Family of the Brown Locust, Locustana pardalina, and the Putative Role of These Peptides in Plague Management.

The brown locust, Locustana pardalina, is a major agricultural pest in southern Africa during swarm formation. Like other locust species, L. pardalina has much higher carbohydrate concentrations in circulation than lipid; carbohydrates are predominantly used in the first minutes of flight and with sustained flight, the metabolic fuel switches to lipids mobilised from fat body stores. We isolated three peptides from the corpora cardiaca of the brown locust; through sequence elucidation by Edman degradation, mass spectrometry and chromatographic confirmation, we show that the brown locust has th

Skin lesions caused by Tabanus bovinus bites.

We report seven Caucasian adult patients who were bitten by horseflies (Tabanus bovinus Linnaeus, 1758) during a trip to Bolivia. Clinical picture was superimposable in all patients: it was characterized by multiple, erythematous, roundish, flattened, large plaques, often with a central point corresponding to horsefly bite, surrounded by satellite, similar, although smaller, lesions. All lesions were located on the neck and/or the upper chest and/or the shoulders; one patient had lesions also on the face. All patients complained of pain at the site of the bite. No systemic symptoms and/or sign

Hormonal regulation of microRNA expression dynamics in the gut of the yellow fever mosquito Aedes aegypti.

The yellow fever mosquito Aedes aegypti is an obligatory blood feeder and a major arboviral disease vector, evoking severe public health concerns worldwide. In adult female mosquitoes, the gut is critical for blood digestion and pathogen entry. We aimed for a systematic exploration of microRNA expression dynamics in the gut during the gonadotrophic cycle. Small RNA libraries were constructed from female mosquito gut tissues at five time points. Unsupervised hierarchical clustering revealed three expression clusters (early, mid and late) peaking at sequential time points - 24, 48 and 72 h poste

Role of two insect growth regulators in integrated pest management of citrus scales.

Portions of two commercial citrus orchards were treated for two consecutive years with buprofezin or three consecutive years with pyriproxyfen in a replicated plot design to determine the long-term impact of these insect growth regulators (IGRs) on the San Joaquin Valley California integrated pest management program. Pyriproxyfen reduced the target pest, California red scale, Aonidiella aurantii Maskell, to nondetectable levels on leaf samples approximately 4 mo after treatment. Pyriproxyfen treatments reduced the California red scale parasitoid Aphytis melinus DeBach to a greater extent than

Knockout of DOPA decarboxylase induces complete albinism in Spodoptera frugiperda.

Dopa decarboxylase (DDC) is a crucial enzyme that regulates melanin synthesis and plays an essential role in cuticular pigmentation and insect development. However, the function of DDC in the fall armyworm, Spodoptera frugiperda, is not yet well understood. In this study, we knocked out the DDC gene and found that mosaic mutants displayed an albino phenotype across all life stages, including larval, pupal, and adult stages. Notably, homozygous DDC mutants showed complete larval albinism, exhibited developmental delays, and frequently failed to pupate, ultimately leading to death. Histological

IAG Regulates the Expression of Cytoskeletal Protein-Encoding Genes in Shrimp Testis.

Insulin-like androgenic gland hormone (IAG) is the master regulator of sexual differentiation and testis development in male crustaceans. However, the molecular mechanism on how IAG functions during testis development is still largely unknown. Here, the transcriptional changes were analyzed in the testes of shrimp after LvIAG knockdown in Litopenaeus vannamei. Differential expression analysis identified 111 differentially expressed genes (DEGs), including 48 upregulated DEGs and 63 downregulated DEGs, in testes of shrimp after LvIAG knockdown. Gene ontology (GO) analysis showed that these DEGs

Mutations of a Drosophila NPC1 gene confer sterol and ecdysone metabolic defects.

The molecular mechanisms by which dietary cholesterol is trafficked within cells are poorly understood. Previous work indicates that the NPC1 family of proteins plays an important role in this process, although the precise functions performed by this protein family remain elusive. We have taken a genetic approach to further explore the NPC1 family in the fruit fly Drosophila melanogaster. The Drosophila genome encodes two NPC1 homologs, designated NPC1a and NPC1b, that exhibit 42% and 35% identity to the human NPC1 protein, respectively. Here we describe the results of mutational analysis of t
