Moschus chrysogaster viral hemorrhagic disease (McVHD), caused by the Moschus chrysogaster hemorrhagic disease virus (McHDV), is an acute and highly fatal infectious disease of musk deer. At present, there is no prevention or treatment for this disease. In this study, we constructed a recombinant bacmid containing the McHDV VP60 gene and obtained the recombinant baculovirus rBac-McHDV VP60 by transfection into Sf9 (Spodoptera frugiperda) insect cells. The McHDV VP60 protein was successfully expressed in the insect cell-baculovirus expression system; furthermore, it was released in the supernat

Recombinant baculovirus expressing insect-selective neurotoxins derived from venomous animals are considered as an attractive alternative to chemical insecticides for efficient insect control agents. Recently we identified and characterized a novel lepidopteran-selective toxin, Buthus tamulus insect-selective toxin (ButaIT), having 37 amino acids and eight half cysteine residues from the venom of the South Indian red scorpion, Mesobuthus tamulus. The synthetic toxin gene containing the ButaIT sequence in frame to the bombyxin signal sequence was engineered into a polyhedrin positive Autographa

Understanding the biochemical impact of nontarget clinical drugs on insect models provides critical insights into xenobiotic toxicity. In this study, we investigated the biochemical effects of dietary AZR on the hemolymph of seventh-instar Galleria mellonella (Lepidoptera: Pyralidae) larvae. First-instar larvae of the insect were reared under laboratory conditions until the seventh-instar larvae. The larvae were fed an artificial diet supplemented with azithromycin (AZR) at different concentrations: 0.0012, 0.0072, 0.0432, 0.2592, 0.7776, and 1.0368%. Our results demonstrated that dietary AZR

Reproduction in insects is a tightly regulated process that relies on the accumulation of yolk protein precursors (YPPs) to support embryonic development. Vitellogenin, the main YPP, is synthesized in the fat body-analogous to the liver and adipose tissue of vertebrates-and transported through the hemolymph to the ovarian follicles, where it is internalized by developing oocytes via the vitellogenin receptor (VgR). Despite its essential role in vitellogenesis, the functional characterization of insect VgR remains incomplete. Triatomines are hematophagous insects and vectors of Trypanosoma cruz

The chironomid midges are the only insects that harbour true haemoglobin in their haemolymph. Here we report the identification of haemoglobin genes in two other nematoceran species. Two paralogous haemoglobin genes (glob1 and glob2) from the malaria mosquito Anopheles gambiae were cloned and sequenced. Furthermore, we identified two orthologous haemoglobin genes in the yellow fever mosquito Aedes aegypti. All four haemoglobins were predicted to be intracellular proteins, with the amino acids required for heme- and oxygen-binding being conserved. In situ-hybridization studies showed that glob1

Aenasius bambawalei Hayat (Encyrtidae: Hymenoptera) has been synonymized with Aenasius arizonensis (Girault) is a small, newly discovered endoparasitoid of the cotton mealybug Phenacoccuss solenopsis Tinsley (Pseudococcidae: Hemiptera), which completes its life cycle inside the body of its host and it is a potential insect control tool. Despite the acquired knowledge regarding host-parasitoid interaction, little information is available on the factors of parasitoid origin able to modulate mealybug physiology. The components of A. arizonensis venom have not been well studied but venom from othe

Among cockroaches (CR) that live in people's homes, two species, i.e., German CR (Blattella germanica) and American CR (Periplaneta americana) predominate in temperate and tropical areas, respectively. CR is an important source of inhalant indoor allergens that sensitize atopic subjects to (localized) type I hypersensitivity or atopy including allergic rhinitis and atopic asthma. In Thailand the predominant CR species is P. americana. CR allergens are found throughout CR infested houses; the number found in kitchens correlates with the degree of CR infestation while sensitization and reactivat

BACKGROUND: Caveolae are invaginated plasma membrane domains of 50-100 nm in diameter involved in many important physiological functions in eukaryotic cells. They are composed of different proteins, including the membrane-embedded caveolins and the peripheric cavins. Caveolin-1 has already been expressed in various expression systems (E. coli, insect cells, Toxoplasma gondii, cell-free system), generating intracellular caveolin-enriched vesicles in E. coli, insect cells and T. gondii. These systems helped to understand the protein insertion within the membrane and its oligomerization. There is

Genetically modified plants expressing insecticidal proteins from Bacillus thuringiensis (Bt) offer valuable options for managing insect pests with considerable environmental and economic benefits. Despite the benefits provided by Bt crops, the continuous expression of these insecticidal proteins imposes strong selection for resistance in target pest populations. Bt maize (Zea mays) hybrids have been successful in controlling fall armyworm (Spodoptera frugiperda), the main maize pest in Brazil since 2008; however, field-evolved resistance to the protein Cry1F has recently been reported. Insects have emerged as promising sustainable alternative protein sources, among which Protaetia brevitarsis (Lewis) (Coleoptera: Scarabaeidae) exhibits considerable potential for valorizing lignocellulosic wastes (e.g. crop straw) into high-value proteins. The larvae of this species are traditionally used in medicine and as a health food, but the underdeveloped rearing technology for adults has impeded large‑scale commercialization and application. Thus, the development and optimization of artificial diets represent a critical breakthrough for overcoming this bottleneck. Herein, we integrated

Gene expression in animal cells allows large scale production of proteins used for either structure and function studies or therapeutic purposes. Maximizing recombinant protein production is necessary to optimize cell growth and protein expression. Some studies have demonstrated the presence of pharmacologically active substances in insect hemolymph. In this work, we have identified and purified a protein from Lonomia obliqua hemolymph able to increase the production of the rabies virus glycoprotein, expressed in Drosophila melanogaster S2 cells, by about 59%.

In response to high temperatures, insect metabolic rates increase, favoring the release of higher amounts of reactive oxygen species (ROS). These ROS need to be counteracted by antioxidants to avoid oxidative stress, which can lead to cell damage and death. In this manuscript, we review evidence in insects showing the effects of high temperatures on ROS production, the antioxidant defenses reported in insects in response to high and extremely high temperatures and the extent to which they contribute to preventing oxidative damage. Endogenously produced antioxidants can be enzymatic or non-enzy

Flufenoxuron, a widely used chitin synthesis inhibitor, is effective against several insect and mite pests. This study evaluated the reproductive toxicity in male sixth-instar larvae of Spodoptera mauritia, a sporadic lepidopteran pest of rice crops. Larvae were topically treated with sublethal concentrations of flufenoxuron (ED25-0.00068 μg and ED50-0.00015 μg), and biochemical responses were assessed at 48, 96, and 144 h post-treatment. Significant reductions in lipid, protein, and carbohydrate levels were observed in the haemolymph and testes of treated larvae, indicating a potential impair

Juvenile hormone (JH) acts as a gonadotrophic hormone stimulating insect vitellogenesis and oogenesis. Paracellular transport of yolk proteins through intercellular channels (patency) in the follicular epithelium is a developmentally regulated and evolutionarily conserved process during vitellogenesis. However, the mechanisms underlying patency opening are poorly understood. Using the migratory locust Locusta migratoria as a model system, we report here that JH-regulated remodeling of zonula adherens (ZA), the belt-like adherens junction maintaining physical linking between follicle cells cont

Latrodectus species are among the most venomous of spiders, with abundant toxic proteinaceous components in their venomous glands and other tissues, as well as their eggs. To date, several proteinaceous toxins with insecticidal potential, including α-insectotoxin and δ-insectotoxin, two of the most potent known insecticidal toxins, have been purified and characterized by comprehensively utilizing conventional biochemical techniques. This has greatly enhanced our knowledge of the molecular basis and mechanism of action of their toxicity. Application of proteomic and transcriptomic techniques fu

West Nile and dengue viruses are (re)emerging mosquito-borne flaviviruses that cause significant morbidity and mortality in man. The identification of mosquito proteins that associate with flaviviruses may provide novel targets to inhibit infection of the vector or block transmission to humans. Here, a tandem affinity purification (TAP) assay was used to identify 18 mosquito proteins that interact with dengue and West Nile capsid, envelope, NS2A or NS2B proteins. We further analyzed the interaction of mosquito cadherin with dengue and West Nile virus envelope protein using co-immunoprecipitati

BACKGROUND: The fall armyworm, Spodoptera frugiperda, is a destructive pest threatening global food security. The sterile insect technique (SIT) is a promising eco-friendly control strategy, which relies on exploring genes regulating male fertility. Small heat shock proteins (sHSPs) act as molecular chaperones and participate in insect reproduction, yet their roles in lepidopteran dimorphic spermatogenesis remain poorly understood.
RESULTS: Here, we functionally characterized a testis-enriched sHSP gene, SfHsp20.2, in S. frugiperda. Using CRISPR/Cas9-mediated knockout, we found that loss of S

Plants perceive diverse extracellular molecules derived from microbes and insects and activate immune responses. While significant progress has been made in understanding plant recognition of microbial proteins and carbohydrates such as chitin, the mechanisms underlying lipid recognition have remained largely elusive. Lipids are essential components of cells, and various microbe- and insect-derived lipid molecules have been reported to induce or modulate immune responses in plants. Recent findings have revealed that lipid perception involves not only canonical receptor-mediated recognition but

Mucins are highly glycosylated proteins that are characterized by a higher proportion of threonine, serine, and proline residues in their sequences. Although mucins in humans and vertebrates have been implicated in many biological processes, their roles in growth and development in invertebrates such as in insects remain largely unknown. Based on bioinformatic analyses, we identified eight mucin or mucin-like genes in the migratory locust, Locusta migratoria. RNA interference against these genes demonstrated that three Lmmucin genes were essential for the survival of L. migratoria nymphs, and

Endoparasitoid wasps deliver a variety of maternal factors, such as venom proteins, viruses, and virus-like particles, from their venom and calyx fluid into hosts and thereby regulate the hosts' immune response, metabolism and development. The endoparasitoid, Microplitis mediator, is used as an important biological agent for controlling the devastating pest Helicoverpa armigera. In this study, using an integrated transcriptomic and proteomic analysis approach, we identified 75 putative venom proteins in M. mediator. The identified venom components were consistent with other known parasitoid wa
