Design, Synthesis, and Bioactivities of Novel 4-(Trifluoromethyl)nicotinamide Derivatives for Use as Insecticides.

Hemiptera pests damage crops and transmit viruses through piercing-sucking mouthparts, causing enormous economic losses and inducing resistance to insecticides. Novel insecticides are critical to combat rising resistance. Nicotinamidase, a newly validated target present in the insect chordotonal organ, has attracted attention. A series of novel sulfonyl, carbonyl, and amide group-based 4-(trifluoromethyl)nicotinamide derivatives was designed using the prodrug strategy and an intermediate derivatization method. Bioactivity assays indicated that most compounds possessed enhanced insecticidal act…

Acute toxicity and sublethal effects of neonicotinoid insecticides on the predatory ladybird Propylea japonica.

BACKGROUND: Neonicotinoid insecticides are widely used for pest control, but their impacts on non-target natural enemies remain insufficiently characterized, particularly at sublethal concentrations. The predatory ladybird Propylea japonica is an important biological control agent in agroecosystems. Here, we evaluated the acute toxicity, ecological risk, sublethal and transgenerational effects, and physiological and transcriptomic responses of P. japonica to multiple neonicotinoid insecticides. RESULTS: Seven neonicotinoid insecticides showed substantial variation in acute toxicity to second-…

Residual activity of insecticides against adult Systena frontalis (Coleoptera: Chrysomelidae) under semi-field conditions.

Systena frontalis (Fabricius) (Coleoptera: Chrysomelidae) is a serious insect pest in nursery production. Insecticides are an important control tactic for S. frontalis adults to reduce economic losses associated with its injury. Because densities and activity of S. frontalis adults are influenced by many biotic (e.g., adult emergence timing) and abiotic (e.g., fluctuating temperatures) factors in the nurseries, it is challenging to reliably determine the efficacy and residual activity of insecticides under field conditions. Thus, the objective was to determine the residual activity of common a…

Screening of Toxic Effects of Neonicotinoid Insecticides with a Focus on Acetamiprid: A Review.

Recently, neonicotinoids have become the fastest-growing class of insecticides in conventional crop protection, with extensive usage against a wide range of sucking and chewing pests. Neonicotinoids are widely used due to their high toxicity to invertebrates, simplicity, flexibility with which they may be applied, and lengthy persistence, and their systemic nature ensures that they spread to all sections of the target crop. However, these properties raise the risk of environmental contaminations and potential toxicity to non-target organisms. Acetamiprid is a new generation insecticide, which …

Zebrafish Larvae Rapidly Recover from Locomotor Effects and Neuromuscular Alterations Induced by Cholinergic Insecticides.

Owing to the importance of acetylcholine as a neurotransmitter, many insecticides target the cholinergic system. Across phyla, cholinergic signaling is essential for many neuro-developmental processes including axonal pathfinding and synaptogenesis. Consequently, early-life exposure to such insecticides can disturb these processes, resulting in an impaired nervous system. One test frequently used to assess developmental neurotoxicity is the zebrafish light-dark transition test, which measures larval locomotion as a response to light changes. However, it is only poorly understood which structur…

Nicotinic acetylcholine receptor modulator insecticides act on diverse receptor subtypes with distinct subunit compositions.

Insect nicotinic acetylcholine receptors (nAChRs) are pentameric ligand-gated ion channels mainly expressed in the central nervous system of insects. They are the directed targets of many insecticides, including neonicotinoids, which are the most widely used insecticides in the world. However, the development of resistance in pests and the negative impacts on bee pollinators affect the application of insecticides and have created a demand for alternatives. Thus, it is very important to understand the mode of action of these insecticides, which is not fully understood at the molecular level. In…

Non-covalent acetylcholinesterase inhibitors: In vitro screening and molecular modeling for novel selective insecticides.

Insecticides represent the most crucial element in the integrated management approach to malaria and other vector-borne diseases. The evolution of insect resistance to long-used substances and the toxicity of organophosphates (OPs) and carbamates are the main factors contributing to the development of new, environmentally safe pesticides. In our work, fourteen compounds of 7-methoxytacrine-tacrine heterodimers were tested for their insecticidal effect. Compounds were evaluated in vitro on insect acetylcholinesterase from Anopheles gambiae (AgAChE) and Musca domestica (MdAChE). The evaluation w…

Chloroorganic (DDT) and organophosphate (malathion) insecticides impair the motor function of the bovine cervix.

Dichlorodiphenyltrichloroethane (DDT) is a representative organochlorine insecticide and a known endocrine disruptor. Malathion is an organophosphate insecticide and a next-generation pesticide. Previously, it was shown that oxytocin (OT) and prostaglandins (PGs) are involved in the mechanism of the adverse effect of DDT on bovine myometrial contractions. However, disruption of myometrial contractions without disruption of cervical activity may not be sufficient to cause preterm delivery. Hence, the aim of this study was to determine the effects of insecticides on the function of the bovine ce…

The stability of four organophosphorus insecticides in stored cucumber samples is affected by additives.

The influence of some additives, including metal ions, antioxidants, enzyme inhibitors and organic solvents, on the storage stability of four organophosphorus pesticides in cucumber samples were investigated. It was found that metal ions, including Al3+, Fe3+, and Co2+, increased the stability of dichlorvos, malathion, and chlorpyrifos. Conversely, Al3+, Fe3+, Fe2+, and Co2+ caused catalytic degradation of diazinon. With the addition of organic solvents (CH2Cl2, CHCl3, CCl4, CH3OH and CH3COCH3), remaining of diazinon residues was higher (16-54%) after storage for seven days. CCl4 was associate

Assessing the acute toxicity of insecticides to the buff-tailed bumblebee (Bombus terrestris audax).

The buff-tailed bumblebee, Bombus terrestris audax is an important pollinator within both landscape ecosystems and agricultural crops. During their lifetime bumblebees are regularly challenged by various environmental stressors including insecticides. Historically the honey bee (Apis mellifera spp.) has been used as an 'indicator' species for 'standard' ecotoxicological testing, but it has been suggested that it is not always a good proxy for other eusocial or solitary bees. To investigate this, the susceptibility of B. terrestris to selected pesticides within the neonicotinoid, pyrethroid and…

Molecular basis of resistance to organophosphate insecticides in the New World screw-worm fly.

BACKGROUND: The emergence of insecticide resistance is a fast-paced example of the evolutionary process of natural selection. In this study, we investigated the molecular basis of resistance in the myiasis-causing fly Cochliomyia hominivorax (Diptera: Calliphoridae) to dimethyl-organophosphate (OP) insecticides. METHODS: By sequencing the RNA from surviving larvae treated with dimethyl-OP (resistant condition) and non-treated larvae (control condition), we identified genes displaying condition-specific polymorphisms, as well as those differentially expressed. RESULTS: Both analyses revealed …

High-throughput analysis of insecticides on malaria vectors using liquid chromatography tandem mass spectrometry.

BACKGROUND: Different setups and protocols have been developed for investigating insecticide effects on Anopheles (An.) mosquitoes, vectors of malaria. However, chemical uptake resulting from their tarsal contact with insecticide-treated material has seldom been investigated. To address the challenges encountered in the interpretation of bioassay data, a high throughput method for chemical analysis on malaria vectors was developed and validated for five selected insecticides including alpha-cypermethrin (aCYP), deltamethrin (DM), etofenprox (EPX), permethrin (PM), pirimiphos-methyl (PPM). Preferences of Pet Owners in Regard to the Use of Insecticides for Flea Control.

A survey conducted from 2002 to 2012 of 759 customers from 84 veterinary practices allows us to examine the habits of dog and cat owners regarding the use of insecticides for flea control. The results indicate that the percentage of animals treated during the 12 mo prior to the survey was not very high (71% in dogs and 50% in cats), considering that 100% of animals included in the study were flea-infested. Statistical analysis shows that animals older than 4 mo are treated more frequently and that dogs are more likely to be treated than cats. Among cats, those living in houses with gardens or …

Lethal and sublethal toxicity of neonicotinoid and butenolide insecticides to the mayfly, Hexagenia spp.

Neonicotinoid insecticides are environmentally persistent and highly water-soluble, and thus are prone to leaching into surface waters where they may negatively affect non-target aquatic insects. Most of the research to date has focused on imidacloprid, and few data are available regarding the effects of other neonicotinoids or their proposed replacements (butenolide insecticides). The objective of this study was to assess the toxicity of six neonicotinoids (imidacloprid, thiamethoxam, acetamiprid, clothianidin, thiacloprid, and dinotefuran) and one butenolide (flupyradifurone) to Hexagenia sp…

Effects of Insecticides on Oviposition and Host Discrimination Behavior in Trichogramma chilonis (Hymenoptera: Trichogrammatidae).

Trichogramma chilonis Ishii is an important natural enemy of many lepidopterous pests on various crops. Effects of two conventional insecticides on oviposition and host discrimination behavior in T. chilonis were evaluated in the laboratory through video tracking. During the oviposition period, when the number of host eggs was limited (only one host egg), females exposed to beta-cypermethrin LC1 or LC20 exhibited significantly higher feeding and re-drilling rate and significantly longer post-oviposition duration than control females. Spinosad LC20 treatment not only decreased the wasp oviposit…
