Loss-of-function mutations in the gene encoding the integrin co-activator kindlin-1 cause Kindler syndrome. We report a novel kindlin-1-deficient keratinocyte cell line derived from a Kindler syndrome patient. Despite the expression of kindlin-2, the patient's cells display several hallmarks related to reduced function of β1 integrins, including abnormal cell morphology, cell adhesion, cell spreading, focal adhesion assembly, and cell migration. OBJECTIVE: To investigate the role of integrin α4β7 in the development of ulcerative colitis (UC) in rats.
METHODS: Sixty Sprague-Dawley rats were randomly divided into the control group (acetone enema), the model group (2,4-dinitrochlorobenzene, DNCB enema), and the α4 intervention group. Colonic mucosa of different groups was observed and compared in terms of pathology and cytokine changes(IL-2 and IL-6) using ELISA. Semi-quantitative RT-PCR was used to detect the colon α4β7 expression. Integrin α4β7(+) lymphocytes in the portal vein of rats were determined by flow cytometry.

The complete analysis of a cDNA clone encoding the mouse integrin beta 7 subunit that was isolated from a lambda gt10 cDNA library prepared from interleukin-4-activated mouse spleen B cells is reported. The 805-amino acid sequence deduced from the cDNA revealed a signal peptide, a 704-amino acid extracellular domain with eight N-glycosylation sites, a transmembrane domain, and a 61-amino acid intracellular domain. Several structural features were conserved with other integrin beta chains including the four cysteine-rich epidermal growth factor-like repeat sequences in the extracellular domain.

Immunohistological expression of integrins has been analyzed on 45 neuroblastoma specimens representative of the different clinical and histological forms of the tumor.
