Protective role of V-set and immunoglobulin domain-containing 4 expressed on kupffer cells during immune-mediated liver injury by inducing tolerance of liver T- and natural killer T-cells.

UNLABELLED: V-set and Ig domain-containing 4 (VSIG4, CRIg, or Z39Ig), a newly identified B7-related cosignaling molecule, is a complement receptor and a coinhibitory ligand that negatively regulates T-cell immunity. Despite its exclusive expression on liver Kupffer cells (KCs) that play key roles in liver tolerance, the physiological role of VSIG4 in liver tolerance remains undefined. Mice lacking VSIG4 had poor survival rates and severe liver pathology in a concanavalin A (ConA)-induced hepatitis (CIH) model, which could be prevented by adoptive transfer of VSIG4(+) KCs. The absence of VSIG4 …

Toll-like receptor 4-mediated inflammatory stimulation in Kupffer cell enhances arsenite-induced liver fibrosis by triggering hepatic stellate cell activation.

As a metalloid element with toxic properties, arsenic (As) has been shown to induce liver fibrosis (LF). Activation of hepatic stellate cell (HSC) is a pivotal factors in the progression of LF, yet the key sentinel cells of the liver tissue, Kupffer cell (KC), also exert a central regulatory role in driving LF. However, the specific functions of these cells in As-induced LF remain to be elucidated. Toll-like receptor 4 (TLR4) triggering the release of inflammatory cytokines via activating downstream nuclear transcription factor-κB (NF-κB) signaling pathways. This study employed primary rat KC

Carbon monoxide-loaded red blood cells ameliorate metabolic dysfunction-associated steatohepatitis progression via enhancing AMP-activated protein kinase activity and inhibiting Kupffer cell activation.

Metabolic dysfunction-associated steatohepatitis (MASH) is a progressive form of nonalcoholic fatty liver disease characterised by fat accumulation, inflammation, oxidative stress, fibrosis, and impaired liver regeneration. In this study, we found that heme oxygenase-1 (HO-1) is induced in both MASH patients and in a MASH mouse model. Further, hepatic carbon monoxide (CO) levels in MASH model mice were >2-fold higher than in healthy mice, suggesting that liver HO-1 is activated as MASH progresses. Based on these findings, we used CO-loaded red blood cells (CO-RBCs) as a CO donor in the liver,

Kupffer Cells Regulate Natural Killer Cells Via the NK group 2, Member D (NKG2D)/Retinoic Acid Early Inducible-1 (RAE-1) Interaction and Cytokines in a Primary Biliary Cholangitis Mouse Model.

BACKGROUND Kupffer cells and natural killer (NK) cells has been identified as contributing factors in the pathogenesis of hepatitis, but the detailed mechanism of these cell types in the pathogenesis of primary biliary cholangitis (PBC) is poorly understood. MATERIAL AND METHODS In this study, polyinosinic: polycytidylic acid (poly I: C), 2-octynoic acid-bovine serum albumin (2OA-BSA) and Freund's adjuvant (FA) were injected to establish a murine PBC model, from which NK cells and Kupffer cells were extracted and isolated. The cells were then co-cultivated in a designed culture system, and the…

Kupffer Cell Capture-Evading Modifiable Sub-20 nm Lipid Nanodisc-Based 19F Magnetic Resonance Imaging Probes.

19F magnetic resonance imaging (MRI) is an innovative imaging method that enables sensitive visualization of 19F-containing probes and has been applied to biomedical imaging. A key strategy in probe design is incorporation of a large number of highly mobile 19F nuclei that can enhance the MRI signal intensity. However, conventional emulsion-based probes are difficult to reduce to below 80 nm in diameter, making them prone to uptake by Kupffer cells and resulting in poor delivery efficiency to target tissues. Moreover, smaller probes based on nanocrystals or polymers suffer from low 19F mobilit…

Targeting Kupffer Cell CD44-Mediated Ammonia Death via the ELAVL1-GLS Metabolic Circuit in Radiation-Induced Liver Disease.

Radiation-induced liver disease (RILD) remains a major dose-limiting toxicity in liver cancer radiotherapy. However, the underlying mechanisms, particularly the immunometabolic reprogramming that sustains inflammatory amplification following irradiation, are still poorly understood. Here, we report that Kupffer cell (KC) depletion significantly attenuated radiation-induced liver inflammation and injury. Irradiation markedly upregulated CD44 expression in KCs, and single-cell RNA sequencing revealed a CD44⁺ macrophage subset associated with injury, inflammatory activation, and metabolic stress

Production system-associated hepatic histomorphometric adaptations in swine: Comparative analysis of glycogen deposition, Kupffer cell abundance, and liver microarchitecture.

BACKGROUND AND AIM: Swine production systems differ considerably in housing, nutrition, biosecurity, and environmental exposure, all of which may influence hepatic metabolism, immune function, and tissue architecture. Histological indicators such as hepatic glycogen deposition, Kupffer cell abundance, and sinusoid-hepatic cord organization provide valuable insight into physiological adaptations associated with different management practices. However, comparative information on these hepatic characteristics across commercial swine production systems remains limited. This study aimed to compare

Kupffer cell depletion attenuates IL-6/STAT3 mediates hepatocyte apoptosis in immunological liver injury of trichloroethylene sensitized mice.

Trichloroethylene (TCE) induced TCE hypersensitivity syndrome which makes immune injuries in multi-system. The multiple organ damage included skin, liver, kidney and so on. The main manifestations of liver injuries were apoptosis and edema of hepatocytes. In our previous research, we found the activation of Kupffer cells (KCs) which increased IL-6 can aggravate liver cell apoptosis in TCE sensitized mice. However, the mechanism of IL-6 in liver damages induced by TCE was not clear. This study explored the function of IL-6/STAT3 signal pathway on the TCE induced apoptosis of liver cell. We esta…

Kupffer cell depletion protects against the steatosis, but not the liver damage, induced by marginal-copper, high-fructose diet in male rats.

High-fructose feeding impairs copper status and leads to low copper availability, which is a novel mechanism in obesity-related fatty liver. Copper deficiency-associated hepatic iron overload likely plays an important role in fructose-induced liver injury. Excess iron in the liver is distributed throughout hepatocytes and Kupffer cells (KCs). The aim of this study was to examine the role of KCs in the pathogenesis of nonalcoholic fatty liver disease induced by a marginal-copper high-fructose diet (CuMF). Male weanling Sprague-Dawley rats were fed either a copper-adequate or a marginally copper…

Hemorrhage induces enhanced Kupffer cell cytotoxicity while decreasing peritoneal or splenic macrophage capacity. Involvement of cell-associated tumor necrosis factor and reactive nitrogen.

Studies indicate that simple hemorrhage produces a profound depression of cell-mediated immunity, thereby contributing to an enhanced susceptibility to septic challenge in the host. However, it remains unknown whether or not the macrophages' cytotoxic capacity is altered after hemorrhage. To study this, C3H/HeN mice were bled to and maintained at a blood pressure of 35 mm Hg for 60 min, and adequately resuscitated. Mice were then killed at 2 or 24 h after hemorrhage to obtain peritoneal macrophage, splenic macrophage, and Kupffer cells. Cytotoxicity was assessed by determining the capacity of …
