Laboratory maintenance of the bacterial endosymbiont, Neorickettsia sp., through the life cycle of a digenean, Plagiorchis elegans.

The Digenea (Platyhelminthes: Trematoda) are a diverse and complex group of internal metazoan parasites. These parasites can serve as hosts to obligate intracellular bacteria belonging to the genus Neorickettsia (Family: Anaplasmataceae). Neorickettsiae persist within all stages of the fluke life cycle and thus are maintained through vertical transmission. However, the low prevalence of Neorickettsia in nature limits study of their transmission biology at different steps of digenean life cycles. To resolve this dilemma, we have developed for the first time a laboratory model allowing to mainta

Phenotypic Screens in Antimalarial Drug Discovery.

Phenotypic high-throughput screens are a valuable tool for identifying new chemical compounds with antimalarial activity. Traditionally, these screens have focused solely on the symptomatic asexual blood stage of the parasite life cycle; however, to discover new medicines for malaria treatment and prevention, robust screening technologies against other parasite life-cycle stages are required. This review highlights recent advances and progress toward phenotypic screening methodologies over the past several years, with a focus on exoerythrocytic stage screens.

Molecular and Morphological Elucidation of the Life Cycle of the Frog Trematode Langeronia macrocirra (Digenea: Pleurogenidae) in Los Tuxtlas, Veracruz, Mexico.

The genus LangeroniaCaballero and Bravo-Hollis, 1949, currently contains 6 species of amphibian trematodes distributed in North and Middle America. The type species of the genus, Langeronia macrocirraCaballero and Bravo-Hollis, 1949, occurs in Mexico and is relatively commonly found as a parasite of leopard frogs. However, information regarding its life cycle is lacking. In this paper, we study the life cycle of L. macrocirra in Laguna Escondida, Los Tuxtlas, Veracruz. Definitive hosts (Rana spp.) as well as potential intermediate hosts (gastropods, bivalves, crustaceans, tadpoles, hemipterans

Proteomic Analysis of the Schistosoma mansoni Miracidium.

Despite extensive control efforts, schistosomiasis continues to be a major public health problem in developing nations in the tropics and sub-tropics. The miracidium, along with the cercaria, both of which are water-borne and free-living, are the only two stages in the life-cycle of Schistosoma mansoni which are involved in host invasion. Miracidia penetrate intermediate host snails and develop into sporocysts, which lead to cercariae that can infect humans. Infection of the snail host by the miracidium represents an ideal point at which to interrupt the parasite's life-cycle. This research fo

Stage-specific and species-specific differences in the production of the mRNA and protein for the filarial nematode secreted product, ES-62.

Previous studies have shown that the secreted phosphorylcholine-containing glycoprotein of filarial nematodes, ES-62, is only present in the post-infective life-cycle stages, but that the mRNA is transcribed throughout the worm's life-cycle. The aim of this current study was to investigate whether the presence or absence of protein expression simply reflects differences in mRNA abundance. To this end, we investigated the relative abundance of ES-62 using TaqMan real time RT-PCR, in different life-cycle stages of 2 model filarial nematode parasites, Acanthocheilonema viteae and Brugia pahangi.

The adaptation of chlamydiae to facultative host multicellularity.

The phylum Chlamydiota consists of obligate intracellular bacteria comprising the human pathogen Chlamydia trachomatis and a large variety of species infecting animals and protists. Despite their diversity, a feature shared by all known chlamydiae is their biphasic developmental cycle, consisting of intra- and extracellular stages with substantial differences in morphology and physiology. Here, we report the isolation of a social amoeba, Dictyostelium giganteum, naturally infected with a chlamydial symbiont. The social life cycle of dictyostelids is characterized by multicellular stages throug

A cyclin protein governs the infectious and sexual life cycles of Cryptococcus neoformans.

Cell cycle is a fundamental process underlying growth and development in evolutionarily diverse organisms, including fungi. In human fungal pathogens, cell cycle control generally determines their life cycles, either in the environment or during infections. Thus, cell cycle components can potentially serve as important targets for the development of antifungal strategy against fungal infections. Here, in Cryptococcus neoformans, the most common cause of fatal fungal meningitis, we show that a previously uncharacterized B-type cyclin named Cbc1 is essential for both its infectious and sexual cy

There and back again: malaria parasite single-cell transcriptomics comes full circle.

The Malaria Cell Atlas (MCA) is an ambitious, ongoing project to profile the intensity and heterogeneity of gene expression throughout the entire malaria parasite life cycle with single-cell resolution. Real et al. now complete the cycle by adding the transmission stages of the most virulent malaria parasite, Plasmodium falciparum, to this easy-to-use resource.

Cellular remodeling during reversible life-stage transition in sessile ciliates: Genomic and transcriptomic insights.

Unraveling mechanisms driving dramatic life-stage transitions in response to environmental stress is central to understanding cellular adaptation. We studied a distinctive group of sessile ciliates that undergo profound morphological and organellar remodeling when transitioning from sessile trophonts to motile telotrochs. We generated chromosome-level genomes for two colonial species (Campanella sinica, Epistylis hentscheli) and one solitary species (Vorticella campanula), revealing genome sizes of 41.30 to 60.37 Mb. Notably, minichromosomes (10 to 100 kb) represent a shared, distinct chromoso

Apicortin defines the Plasmodium apical conoid body but is dispensable for the parasite life cycle.

Apicomplexan parasites such as Plasmodium spp. and Toxoplasma gondii possess unique tubulin-based structures, including subpellicular microtubules and apical polar rings, which are essential for parasite motility, host cell invasion, and replication. Apicortin, a microtubule-associated protein, contains a doublecortin (DC) domain and a partial tubulin polymerization-promoting protein (TPPP) domain, both implicated in microtubule binding and stabilization. How tubulin-based structures are maintained is poorly understood, but it may involve Apicortin, so far found only in apicomplexans and the p

An Overview of Trypanosoma cruzi Biology Through the Lens of Proteomics: A Review.

The protozoan parasite Trypanosoma cruzi, causative agent of Chagas disease, affects millions of people in endemic Latin American countries and beyond. In Latin America, Chagas disease is an important cause of death and disability, for which vaccines are lacking and improved treatment options are required. Additionally, the factors governing the development of a variety of clinical manifestations during Chagas disease, ranging from complete lack of symptoms to severe irreversible chronic organ damage (mainly cardiac or digestive), remain largely unknown. Much remains to be learned regarding th

Successful completion of the life cycle of Amblyomma variegatum using tick artificial membrane feeding system.

The three-host tick Amblyomma variegatum, commonly known as the tropical bont tick, poses a major threat to livestock health and productivity in tropical and subtropical regions worldwide. The inability to control the tick and prevent the diseases it transmits is partly due to the necessity of rearing the tick on animals. Thus, the goal of this study was to adapt a previously developed artificial membrane feeding system to complete the life cycle of A. variegatum. All life stages of A. variegatum were fed using fresh goat blood at 38 °C and blood replacement occurred every 12 h. Key parameters

Automated detection and quantification of two-spotted spider mite life stages using computer vision for high-throughput in vitro assays.

The two-spotted spider mite (Tetranychus urticae Koch) is a globally significant agricultural pest with high reproductive capacity, rapid development, and frequent evolution of miticide resistance. Breeding and selection of resistant host cultivars represent a promising complement to chemical control, but widespread adoption is limited primarily due to the labor-intensive nature of conventional in vitro phenotyping methods. Here, we present a high-throughput, semi-automated image analysis pipeline integrating the Blackbird CNC Microscopy Imaging Robot with computer vision models for mite life

Gas-propelled navigation: CO2 prompts parasitic worm behavior.

Skin-penetrating nematodes such as Strongyloides stercoralis depend on a suite of chemosensory cues to find their host. Banerjee et al. uncovered life-stage-specific behavioral preferences for CO2 in S. stercoralis and revealed its distinct influence on navigation behaviors during the complex life cycle in this species.

Selenium Differentially Influences Methylmercury Retention across Mayfly Life Stages.

Though high mercury and selenium concentrations are individually toxic to organisms, there is a hypothesized antagonistic relationship. This potential mercury-selenium interaction is under-studied in aquatic macroinvertebrates, particularly in relation to complex life histories. We examined the proposed effect of selenium on methylmercury accumulation between four life stages for a parthenogenetic mayfly (Neocloeon triangulifer). We exposed diatoms to elevated methylmercury concentrations and fed them to mayflies exposed to elevated aqueous selenomethionine. We found some support for the mercu

Insights into the biology of the rat lungworm, Angiostrongylus cantonensis.

Angiostrongylus cantonensis is a parasitic nematode with a complex life cycle involving rats as definitive hosts and gastropods as intermediate hosts. The parasite can infect other organisms, including humans, in which it causes neuroangiostrongyliasis, a globally emerging but neglected disease. This primer reviews the biology of A. cantonensis including its life cycle and development in its natural, accidental, and paratenic hosts, as well as its expanding geographic distribution. It then considers recent advances in A. cantonensis research followed by exploring areas that are ripe for furthe

Freshwater trematodes in Iceland and the surrounding north - current advances and questions.

Iceland is an isolated, sub-Arctic, oceanic island of volcanic origin in the northern North Atlantic. With a limited faunal diversity and being the most northern point in the distributional range for some species, it is an intriguing model region to study parasite biodiversity and biogeography. Since 2006, there has been a history of intense biodiversity discoveries of freshwater trematodes (Trematoda, Digenea), thanks to the use of integrative taxonomic methods. The majority of digeneans (28 out of 41 known) were characterised with molecular genetic methods and morphological analyses, with so

Disruption of mitochondrial function in blood and sexual stages of Plasmodium falciparum by ferulenol.

BACKGROUND: Malaria continues to be a significant global health challenge, necessitating the development of novel antimalarial compounds. This study explores the effects of ferulenol on multiple lifecycle stages of Plasmodium falciparum. Ferulenol has been identified as a promising antimalarial candidate, demonstrating high efficacy in inhibiting asexual blood-stage parasites at low micromolar concentrations. However, its effects on other parasite stages remain unexplored, despite its mitochondrial target being critical for sexual stage development. This study aims to investigate ferulenol's p
