Fatty Acid Modification of the Anticancer Peptide LVTX-9 to Enhance Its Cytotoxicity against Malignant Melanoma Cells.

Spider venom is a valuable resource for the development of novel anticancer drugs. In this study, we focused on novel linear amphipathic α-h … TX-9, fatty acid modification of this peptide was performed, and five different carbon chain length lipopeptides named LVTX-9-C12-C20 were produced. Among them, the lipopeptide LVTX-9-C18 showed the highest cytotoxic activity in relation to B16-F10 cells, whether in a serum or serum-free medium. Most importantly, the cytotoxic activity of LVTX-9-C18 was improv …

Diabetic pregnancy activates the innate immune response through TLR5 or TLR1/2 on neonatal monocyte.

Diabetes mellitus (DM) during pregnancy causes congenital malformation, macrosomia, respiratory distress syndrome, and other abnormalities in neonates, but whether maternal DM affects the neonatal innate immune system is unknown. Therefore we aimed to reveal the influence of DM in pregnancy on the toll-like receptor (TLR)-mediated innate immune response in neonates. Cord blood was collected after full-term vaginal or cesarean delivery and classified into a DM group (n=8) and non-DM (control) group (n=7). Mononuclear cells were harvested from cord blood by using density gradient centrifugation,

Occidiofungin's chemical stability and in vitro potency against Candida species.

Occidiofungin is a cyclic glyco-lipopeptide produced by Burkholderia contaminans. MICs against Candida species were between 0.5 and 2.0 μg/ml. Occidiofungin retains its in vitro potency in the presence of 5% and 50% human serum with a minimal lethal concentration (MLC) of 2 and 4 μg/ml, respectively. Time-kill and postantifungal effect (PAFE) experiments of occidiofungin against Candida albicans were performed. The results demonstrate that occidiofungin is fungicidal. Occidiofungin was also found to be a very stable molecule. It is resistant to extreme temperatures and pH and maintains its act

Inhibition of daptomycin by pulmonary surfactant: in vitro modeling and clinical impact.

The lipopeptide daptomycin has been approved for use in skin and skin-structure infections but has failed to meet statistical noninferiority criteria in a clinical trial for severe community-acquired pneumonia. Daptomycin exhibited an unusual pattern of activity in pulmonary animal models: efficacy in Staphylococcus aureus hematogenous pneumonia and inhalation anthrax but no activity against Streptococcus pneumoniae in simple bronchial-alveolar pneumonia. Daptomycin was shown to interact in vitro with pulmonary surfactant, resulting in inhibition of antibacterial activity. This effect was spec

Bactericidal activity of daptomycin against Streptococcus pneumoniae compared with eight other antimicrobials.

A spectrum of pneumococci with varying susceptibilities to beta-lactams, macrolides and quinolones was tested for susceptibility to nine antibiotics, including the novel lipopeptide daptomycin. Daptomycin was active against all strains (MIC range </=0.5 mg/L; MIC(50) 0.125 mg/L; MIC(90) 0.25 mg/L). All pneumococci were susceptible to vancomycin, teicoplanin, linezolid and quinupristin/dalfopristin, with MICs < 4.0 mg/L. Time-kill assays with eight strains demonstrated that daptomycin (2 x MIC) was bactericidal in seven of eight strains tested at 24 h, with >90% killing at 1 h.

Characterization of the antifungal activity of the bacterial isolate Bacillus velezensis TCG15.

Bacillus velezensis TCG15 is a bacterial isolate from soil that shows antifungal activity against Aspergillus spp. and Candida spp. This study aimed to characterize its antifungal activity. TCG15 produces cyclic lipopeptides (cLPs), including surfactin A, plipastatin, and bacillomycin L. We obtained the complete sequence of the operon encoding nonribosomal peptide synthetase for bacillomycin L biosynthesis, bmyL operon (bmyLDABC), using genome analysis of TCG15. The bmyLD gene-disrupted mutant did not produce bacillomycin L, plipastatin, or surfactin A. The mutant showed no antifungal activity…

Role of C17 fengycin B from Bacillus subtilis DB9011 in enterotoxigenic Escherichia coli inhibition.

Enterotoxigenic Escherichia coli (ETEC) is a leading cause of post-weaning diarrhea in piglets, and the rise of antimicrobial resistance underscores the urgent need for antibiotic alternatives in swine production. Bacillus species are promising candidates owing to their ability to produce antimicrobial secondary metabolites, particularly cyclic lipopeptides such as surfactins, iturins, and fengycins. Although Bacillus lipopeptides are known to exhibit antibacterial activity against E. coli, the specific compounds responsible for the inhibition of ETEC in probiotic-relevant Bacillus strains rem…

Daptomycin for eradication of a systemic infection with a methicillin-resistant-Staphylococcus aureus in a biventricular assist device recipient.

The rate of infection in patients who require ventricular assist devices is estimated at more than 35%. Infections with multi-resistant organisms such as methicillin-resistant Staphylococcus aureus in ventricular assist device recipients are often difficult to treat and present a high mortality rate. Daptomycin is a new cyclic lipopeptide antibiotic, useful in gram-positive organisms resistant to standard treatment. We report a case of a 65-year-old man suffering from a dilatative cardiomyopathy and concomitant MRSA infection who received a biventricular assist device. The patient had MRSA sep

Targeted Killing of Staphylococcus aureus Using Specific Peptides Displayed on Yeast Vacuoles.

Staphylococcus aureus is a common pathogen that causes health care-related and community-associated infections. In this study, we provide a novel system that can recognize and kill S. aureus bacteria. The system is specifically based on a combination of the phage display library technique and yeast vacuoles. A phage clone displaying a peptide capable of specific binding to a whole S. aureus cell was selected from a 12-mer phage peptide library. The peptide sequence was SVPLNSWSIFPR. The selected phage's ability to bind specifically with S. aureus was confirmed using an enzyme-linked immunosorb

Lipid lysination by MprF contributes to hemolytic pigment retention in group B Streptococcus.

Group B Streptococcus (GBS) is the leading cause of neonatal sepsis and meningitis. A major virulence factor is a pigmented beta-haemolytic/cyto-lysin (β-h/c) toxin with an ornithine rhamnolipid structure. We initially observed that absence of MprF enzyme altered pigmentation and haemolytic activity in GBS. Next, we showed that MprF-dependent lipid lysination contributes to the retention of the ornithine rhamnolipid within GBS membrane. Furthermore, cationic lipidation by MprF altered membrane properties contributing to resistance to the cyclic lipopeptide daptomycin and to acidic pH. This stu

Therapeutic immunization with a mixture of herpes simplex virus 1 glycoprotein D-derived “asymptomatic” human CD8+ T-cell epitopes decreases spontaneous ocular shedding in latently infected HLA transgenic rabbits: association with low frequency of local PD-1+ TIM-3+ CD8+ exhausted T cells.

UNLABELLED: Most blinding ocular herpetic disease is due to reactivation of herpes simplex virus 1 (HSV-1) from latency rather than to primary acute infection. No herpes simplex vaccine is currently available for use in humans. In this study, we used the HLA-A*02:01 transgenic (HLA Tg) rabbit model of ocular herpes to assess the efficacy of a therapeutic vaccine based on HSV-1 gD epitopes that are recognized mainly by CD8(+) T cells from "naturally" protected HLA-A*02:01-positive, HSV-1-seropositive healthy asymptomatic (ASYMP) individuals (who have never had clinical herpes disease).
