A case report on indirect transmission of human rabies.

At present, virus infection is still a common threat to public health in developing countries. Human rabies remains a matter of great global concern with a case-fatality of almost 100%. The rabies virus belongs to the neurotropic type of virus of the Lyssavirus genus, and the disease presents as a deteriorating encephalomyelitis and is endemic throughout much of the world, particularly in Africa and Asia. Previous data have shown that, globally, approximately 59 000 human deaths are caused by rabies per year. Fortunately, human rabies can be treated through the timely administration of post-ex…

Rabies virus distribution in tissues and molecular characterization of strains from naturally infected non-hematophagous bats.

Bats are main reservoirs for Lyssavirus worldwide, which is an important public health issue because it constitutes one of the big challenges in rabies control. Yet, little is known about how the virus is maintained among bats, and the epidemiological relationships remain poorly understood. The aim of the present study was to investigate the distribution of the rabies virus (RABV) in bat tissues and organs and to genetically characterize virus isolates from naturally infected non-hematophagous bats. The heminested reverse transcriptase polymerase chain reaction (hnRT-PCR) and sequencing using …

Epidemiology of bat rabies in Germany.

Rabies in European bats was first reported in Germany in 1954. In concordance with Denmark and the Netherlands, Germany has reported one of the highest numbers (n = 187) of European bat lyssavirus (EBLV)-positive cases in bats in Europe so far (1954-2005). A combined descriptive epidemiological and phylogenetic analysis on bat rabies and prevailing EBLVs is presented, comprising the past 50 years. So far, only the two lineages of EBLV-1 (genotype 5), a and b, have been detected. Although only 50% of the rabies-positive bats have been identified by species, the Serotine bat (Eptesicus serotinus…

Histopathology and immunohistochemistry findings in kidney and urinary bladder of rabies virus-infected mice.

Rabies virus (RABV) is a lethal and neglected zoonosis responsible for over 60,000 deaths annually caused by the neurotropic virus Lyssavirus rabies. Although rabies is well-known for its severe nervous system impairment, little is known regarding the specific alterations caused in extraneural organs. Studies suggest an essential involvement of RABV in the kidneys. However, the extent of the pathological damage caused by RABV in this organ remains to be understood. This study describes the histopathological alterations and RABV antigen expression in the kidneys and urinary bladder. Viral immun…

Parasite diversity of European Myotis species with special emphasis on Myotis myotis (Microchiroptera, Vespertilionidae) from a typical nursery roost.

BACKGROUND: Bats belong to one of the most species-rich orders within the Mammalia. They show a worldwide distribution, a high degree of eco … site infections and pathogens. Morphometric data were recorded and the individuals were checked for Lyssavirus-specific antigen using a direct immunofluorescence test. For unambiguous species identification, the bats were analysed by cyt-b sequence comparison. RESULTS: Myotis myotis were parasitized by the six insect and arachnid ectoparasite species, i.e. …

The application of genetic markers for EBLV surveillance in European bat species.

The United Kingdom has performed passive surveillance for European bat lyssaviruses (EBLVs) since 1987, and species-targeted surveillance since 2003. One critical component of these studies is the accurate identification of bats either submitted for testing or sampled in the field. Identification is dependent on numerous morphological characteristics. Whilst this is an effective means of bat identification, a number of problems remain with this approach. It relies on the experience of bat specialists and can lead to problems in differentiating members of the Myotis genus, particularly between …

Active surveillance for antibodies confirms circulation of lyssaviruses in Palearctic bats.

BACKGROUND: Palearctic bats host a diversity of lyssaviruses, though not the classical rabies virus (RABV). As surveillance for bat rabies over the Palearctic area covering Central and Eastern Europe and Siberian regions of Russia has been irregular, we lack data on geographic and seasonal patterns of the infection. RESULTS: To address this, we undertook serological testing, using non-lethally sampled blood, on 1027 bats of 25 species in Bulgaria, the Czech Republic, Poland, Russia and Slovenia between 2014 and 2018. The indirect enzyme-linked immunosorbent assay (ELISA) detected rabies virus…

However, it hardly detected the Bokeloh Bat Lyssavirus (BBLV) and the European Bat Lyssaviruses types 1b and 2 (EBLV-1b and EBLV-2) in most laboratories resulting in a moderate inter-laboratory concordance (58.4%-82.7%) for these lyssaviruses. The two LFAs provided reliable and reproducible results on a …

Analysis of Mouse Brain Transcriptome After Experimental Duvenhage Virus Infection Shows Activation of Innate Immune Response and Pyroptotic Cell Death Pathway.

Rabies is an important neglected disease, characterized by invariably fatal encephalitis. Several studies focus on understanding the pathogenic mechanisms of the prototype lyssavirus rabies virus (RABV) infection, and little is known about the pathogenesis of rabies caused by other lyssaviruses. We sought to characterize the host response to Duvenhage virus infection and compare it with responses observed during RABV infection by gene expression profiling of brains of mice with the respective infections. We found in both infections differentially expressed genes leading to increased expression…

Compendium of animal rabies prevention and control, 2008: National Association of State Public Health Veterinarians, Inc. (NASPHV).

Rabies is a fatal viral zoonosis and a serious public health problem. The disease is an acute, progressive encephalitis caused by a lyssavirus. Although the United States has been declared free of canine rabies virus variant transmission, multiple viral variants are maintained in wild mammal populations, and there is always a risk of reintroduction of canine rabies. All mammals are believed to be susceptible to the disease, and for purposes of this document, use of the term "animal" refers to mammals. The recommendations in this compendium serve as a basis for animal rabies-prevention and -con…

Magnetic resonance imaging findings in a rare case of bovine to human rabies encephalitis.

Rabies encephalitis is a fatal zoonotic disease that poses diagnostic challenges due to its variable clinical presentation. This report describes a pediatric case involving an 8-year-old boy who presented with rapidly progressive neurological decline following unrecognized exposure to a rabid cow in a quarantined village, a rare but possible route of transmission. Although initial cerebrospinal fluid and saliva polymerase chain reaction assays for Lyssavirus were inconclusive, early magnetic resonance imaging (MRI) revealed characteristic involvement of the basal ganglia and limbic system. The…

Generation of recombinant rabies viruses encoding NanoLuc luciferase for antiviral activity assays.

Rabies is an invariably fatal disease caused by Rabies virus (RABV), a member of the family Rhabdoviridae, genus Lyssavirus. Once central nervous infection occurs and symptoms develop, the case fatality rate approaches 100% despite availability of post-exposure prophylaxis. Therefore, new antiviral therapies for rabies are urgently required. Antivirals which can inhibit virus replication can be identified through screening of small compounds, however, as RABV infection does not generate easily discernible cytopathic effects in vitro, cell viability assays may not be feasible to observe antivir…

Metabolomics of cerebrospinal fluid from humans treated for rabies.

Rabies is a rapidly progressive lyssavirus encephalitis that is statistically 100% fatal. There are no clinically effective antiviral drugs for rabies. An immunologically naïve teenager survived rabies in 2004 through improvised supportive care; since then, 5 additional survivors have been associated with use of the so-called Milwaukee Protocol (MP). The MP applies critical care focused on the altered metabolic and physiologic states associated with rabies. The aim of this study was to examine the metabolic profile of cerebrospinal fluid (CSF) from rabies patients during clinical progression o…

Transmission dynamics of rabies in China over the last 40 years: 1969-2009.

BACKGROUND: Rabies is a serious reemerging zoonosis in China. The molecular evolution and transmission patterns of rabies virus inferred fro … logenetic analysis revealed that Chinese rabies viruses are subdivided into 6 lineages (A-F) within Lyssavirus genotype 1. Lineage A represents the widely dispersed cosmopolitan lineage while lineage B is closely related to Arctic-like rabies viruses. The remaining lineages (C-F) are typical of those circulating across much of Southeast Asia. The evolutionar …

A molecular epidemiological study targeting the glycoprotein gene of rabies virus isolates from China.

A group of 31 rabies viruses (RABVs), recovered primarily from dogs, one deer and one human case, were collected from various areas in China between 1989 and 2006. Complete G gene sequences determined for these isolates indicated identities of nucleotide and amino acid sequences of >or=87% and 93.8%, respectively. Phylogenetic analysis of these and some additional Chinese isolates clearly supported the placement of all Chinese viruses in Lyssavirus genotype 1 and divided all Chinese isolates between four distinct groups (I-IV). Several variants identified within the most commonly encountered g…
