Novel α-Oxoamide Advanced-Glycation Endproducts within the N6-Carboxymethyl Lysine and N6-Carboxyethyl Lysine Reaction Cascades.

The highly reactive α-dicarbonyl compounds glyoxal and methylglyoxal are major precursors of posttranslational protein modifications in vivo. Model incubations of N2-t-Boc-lysine and either glyoxal or methylglyoxal were used to further elucidate the underlying mechanisms of the N6-carboxymethyl lysine and N6-carboxyethyl lysine reaction cascades. After independent synthesis of the authentic reference standards, we were able to detect N6-glyoxylyl lysine and N6-pyruvoyl lysine for the first time by HPLC-MS2 analyses. These two novel amide advanced-glycation endproducts were exclusively formed u

Advanced glycoxidation and lipoxidation end products (AGEs and ALEs): an overview of their mechanisms of formation.

Advanced lipoxidation end products (ALEs) and advanced glycation end products (AGEs) have a pathogenetic role in the development and progression of different oxidative-based diseases including diabetes, atherosclerosis, and neurological disorders. AGEs and ALEs represent a quite complex class of compounds that are formed by different mechanisms, by heterogeneous precursors and that can be formed either exogenously or endogenously. There is a wide interest in AGEs and ALEs involving different aspects of research which are essentially focused on set-up and application of analytical strategies (1

Argpyrimidine, a methylglyoxal-derived advanced glycation end-product in familial amyloidotic polyneuropathy.

FAP (familial amyloidotic polyneuropathy) is a systemic amyloid disease characterized by the formation of extracellular deposits of transthyretin. More than 80 single point mutations are associated with amyloidogenic behaviour and the onset of this fatal disease. It is believed that mutant forms of transthyretin lead to a decreased stability of the tetramer, which dissociates into monomers that are prone to unfolding and aggregation, later forming beta-fibrils in amyloid deposits. This theory does not explain the formation of beta-fibrils nor why they are toxic to nearby cells. Age at disease

Enhanced humic-like acid formation from waste activated sludge during alkaline thermal hydrolysis - Dual role of reducing sugars.

Recovering humic-like acids (HLAs) from waste activated sludge (WAS) using alkaline thermal hydrolysis (ATH) offers a promising route for simultaneous sludge reduction and resource recovery. However, the limited content of readily reactive organic compounds in WAS constrains HLA yield. In this study, fruit waste (FW) as a carbohydrate-rich waste was introduced as a co-substrate to enhance HLA formation. Results showed that HLA yield increased by 130.3%-188.8%, reaching a maximum of 0.53 ± 0.02 g/g volatile solid (VS) of initial WAS at a WAS-to-FW VS ratio of 1:3 under moderate ATH conditions (

Formation, Toxicity, and Analytical Techniques for Small-Molecule α-Dicarbonyl Compounds in Foods.

Small-molecule α-dicarbonyl compounds (α-DCs), including glyoxal, methylglyoxal and diacetyl, are electrophilic compounds characterized by two adjacent carbonyls. These compounds ubiquitously occur in various foods and food-medicine homologous herbs, generated via the Maillard reaction, caramelization, lipid peroxidation, and enzymatic reactions during thermal treatment and storage. Upon oral intake, small-molecule α-DCs are rapidly absorbed into systemic circulation, triggering protein and DNA damage, as well as inflammation. They also serve as important precursors to derive other hazards, su…

Optimization and Development of Albumin-Biopolymer Bioconjugates with Solubility-Improving Properties.

Bioconjugation is an emerging field in the food and pharmaceutical industry. Due to its biocompatibility and high ligand binding capacity, albumin is widely used in modern drug delivery systems. However, the protein is sensitive to environmental stresses; albumin conjugates, on the other hand, have improved functional properties. Biopolymers are gaining interest due to their biodegradability and safety, compared to synthetic polymers. In this study, albumin-biopolymer bioconjugates were prepared by nonenzymatic Maillard reaction at 60 °C and 80% relative humidity. This nonenzymatic conjugation…

Decoding the thermocyclic solar-driven fermentation: Multi-omics insights into microbial and metabolic dynamics of traditional Chishui river basin soy sauce.

Traditional Chishui River Basin soy sauce is produced through prolonged solar-cycle fermentation under diurnal temperature fluctuations and moisture absorption. Here, we employed integrated metagenomic and metabolomic analyses to investigate microbial and metabolic dynamics throughout fermentation. Results revealed a three-phase microbial succession: initial fungal-hydrolytic phase dominated by Aspergillus oryzae (82.28%), marked by proteolysis and amino acid accumulation; transitional phase enriched with Weissella (23.45%) and Zygosaccharomyces rouxii (5.85%), producing organic acids, esters,

Chitosan Film as a Replacement for Conventional Sulphur Dioxide Treatment of White Wines: A 1H NMR Metabolomic Study.

Chitosan-genipin (Ch-Ge) films have been proposed for the replacement of sulfur dioxide (SO2) in white wines preservation to circumvent the adverse health consequences caused by SO2 intake. To assess the effects of different-sized Ch-Ge films (25 and 100 cm2) on wine composition compared to SO2-treated and untreated wines, nuclear magnetic resonance metabolomics was applied. Relative to SO2, 100 cm2 films induced significant changes in the levels of organic acids, sugars, amino acids, 5-hydroxymethylfurfural, among other compounds, while 25 cm2 films appeared to induce only small variations. Recent advances in the biotechnological production of erythritol and mannitol.

Dietary habits that include an excess of added sugars have been strongly associated with an increased risk of obesity, heart disease, diabetes, and tooth decay. With this association in view, modern food systems aim to replace added sugars with low calorie sweeteners, such as polyols. Polyols are generally not carcinogenic and do not trigger a glycemic response. Furthermore, owing to the absence of the carbonyl group, they are more stable compared to monosaccharides and do not participate in Maillard reactions. As such, since polyols are stable at high temperatures, and they do not brown or ca…

Structural transitions and functional outcomes of soy protein isolate modified with carbohydrate conjugation via spray drying and ultrasonication.

Soy protein isolate (SPI) exhibits promising nutritional and techno-functional properties; however, its practical application is often restricted by limited solubility, structural instability, and poor interfacial performance under processing conditions. To address these limitations, this study investigated the structural and functional modification of SPI through conjugation with glucose, sucrose, and pectin using spray drying and ultrasound-assisted Maillard reaction pathways. The study comparatively evaluated how carbohydrate class and processing route influence grafting behavior, conformat…

Cooking Chemistry Transforms Proteins into High-Strength Adhesives.

In prior generations, proteins were taken from horses and other animals to make glues. Petroleum-derived polymers including epoxies and cyanoacrylates have since replaced proteins owing to improved performance. These modern materials come at a cost of toxicity as well as being derived from limited resources. Ideally, replacement adhesives will be made from benign, cheap, and renewable feedstocks. Such a transition to biobased materials, however, will not occur until similar or improved performance can be achieved. We have discovered that coupling of proteins and sugars gives rise to strong adh

UV-activated diacetyl for bacterial inactivation and browning inhibition in apple juice.

Microbial contamination and enzymatic browning remain key challenges in apple juice processing. This study introduces ultraviolet-activated diacetyl (UV/BD) as a non-thermal intervention that simultaneously ensures microbial safety and quality retention. Treatment with 0.5 mM BD and 1.5 kJ/m2 UV reduced native bacterial counts below detection limits and suppressed browning by 32.9% through two synergistic mechanisms: photolytically generated peroxyacetyl radicals selectively oxidize polyphenol oxidase, while oxygen-deprivation resulting from BD photooxidation halts quinone formation and starve
