Online Monitoring and Control of Methanol Concentration in Pichia Pastoris Fermentations.

Culture broth methanol concentration is the key parameter in the production of recombinant proteins by using Pichia pastoris (Komagataella phaffii) as a cell factory under the regulation of the methanol-inducible Alcohol Oxidase 1 (AOX1) promoter (PAOX1) or other alternative promoters induced directly or indirectly by methanol. Accordingly, the implementation of an efficient methanol monitoring and control system is critical to guarantee the proper methanol concentration toward achieving an efficient and robust production while assuring reproducible bioprocessing conditions.

A dual-defective electrocatalyst for methanol oxidation reaction coupled seawater splitting.

Seawater electrolysis is considered a sustainable green hydrogen production route, but it is limited by high energy consumption, corrosion, and the competing chlorine evolution reaction (ClER). This study replaces oxygen evolution reaction (OER) with a low-potential methanol oxidation reaction (MOR) to reduce energy consumption and suppress ClER. We constructed a boron (B)-doped Cu/Mn bimetallic metal-organic framwork (MOF)-derived electrocatalyst (B-CuMn-TPA) on nickel foam, in which dual-defect regulation was achieved through Mn incorporation and B-defect introduction via NaBH4 reduction. Th…

Hydrogen Aggregation Enhances CO2 Hydrogenation to Methanol Over In2O3-Based Catalysts.

The hydrogen (H) spillover on the catalyst surface is crucial in the CO2 hydrogenation reaction, but its effects on product selectivity have been rarely investigated. Herein, we reveal the H-spillover mediated regulatory role of oxide supports, which changed the CO2 hydrogenation selectivity on In2O3-based catalysts. By replacing the supports from TiO2 to ZrO2, the primary product of CO2 hydrogenation experiences a significant shift from carbon monoxide (95.6%) to methanol (84.2%). In situ characterization and theoretical modeling evidence that the degree of H-spillover influences the distribu…

Interpretable machine learning for optimized dimethyl ether production from bio-methanol.

Hybrid models often referred to as gray-box models offer a promising approach by combining the flexibility of data-driven techniques with the accuracy and physical interpretability of first-principles models. This study evaluates a range of mathematical modeling techniques in the context of chemical reaction engineering, with a focus on the production of dimethyl ether (DME) from bio-methanol in a fixed-bed reactor. A comprehensive case study was conducted, beginning with the development of a first-principles model to solve a system of governing equations and generate 7,000 synthetic data poin…

Capture, Sampling and Analysis of Biogenic CO2 Streams for Methanol Synthesis.

The shipping sector is responsible for a considerable share of global CO2 emissions and is under pressure to reduce emissions and adopt carbon-neutral fuels. Among the proposed alternatives, methanol produced from green hydrogen and biogenic CO2 represents a promising option. However, the feasibility of its production is significantly influenced by the composition and variability of the bio-CO2 feedstock, which can negatively impact the complete value chain. To address these challenges, sampling campaigns were carried out at actual bio-CO2-emitting sites, namely biogas and biomass combustion f…

Why Is Methanol Formation Suppressed in CO2 Reduction Over Copper Electrocatalysts?

Electrocatalytic CO2 reduction (CO2RR) to produce methanol (CH3OH) provides a sustainable alternative to its energy-intensive industrial synthesis. However, C2+ species and CH4 are typically the dominant products on prototypical Cu catalysts, with no CH3OH formation. Herein, employing constant-potential explicit solvent methods, we systematically compared the thermodynamics and kinetics of C2+ products (covering 21 possible C-C coupling paths), CH4, and CH3OH formation to uncover the origin of intrinsic suppression of CH3OH. Nine C-C coupling pathways exhibit significantly lower barriers than …

Systematic metabolic engineering of Komagataella phaffii for methanol-to-lactate bioconversion.

Methanol is an attractive C1 feedstock for sustainable biomanufacturing. However, efficient conversion of methanol into value-added products remains challenging due to metabolic compartmentalization, accumulation of toxic intermediates, and carbon loss in methylotrophic yeasts. In this study, we established a methanol-to-lactate bioconversion platform in Komagataella phaffii through a systematic metabolic engineering strategy that integrates methanol assimilation, detoxification, and downstream carbon flux redirection. Initial expression of a heterologous lactate dehydrogenase enabled efficien…

Phase reconstruction of In-Mo oxide boosts CO2 hydrogenation to methanol.

An In0.5Mo0.5 oxide catalyst alleviates the activity-selectivity trade-off in CO2 hydrogenation to methanol. Reduction-driven reconstruction of In2(MoO4)3 generates intimate In-Mo oxide interfaces, while In-to-Mo electron transfer creates oxygen vacancies and promotes H2 activation, delivering the highest methanol space-time yield among the tested catalysts.

Methanol Metabolism in the Cytochrome and Menaquinone-Containing Acetogen Moorella thermoacetica.

Methanol derived from pectin degradation is common in oxic and anoxic sediments and is used by various aerobic and anaerobic microorganisms such as acetogenic bacteria or methanogenic archaea as a carbon and energy source. Acetogens use the Wood-Ljungdahl pathway to disproportionate methanol to CO2 and acetate. Methanol metabolism has been studied primarily in mesophilic acetogens such as Acetobacterium woodii and Eubacterium callanderi, that do not have cytochromes or quinones. Moorella thermoacetica, the model organism for elucidation of acetogenesis, is a thermophilic, cytochrome- and quino…

Electrosynthesis of Ethylene Glycol from Methanol via Oxidative C-C Coupling.

Ethylene glycol (EG) is a commodity chemical, but its industrial production heavily relies on fossil-based feedstocks (that are, oil-based ethylene and coal-based syngas) and faces high carbon footprint. Herein, we report one-step electrosynthesis of EG from an important C1 platform molecule─methanol (MeOH), via oxidative C-C coupling at the anode. Over a boron-doped diamond (BDD) electrode, EG was produced with a Faradaic efficiency of 34.3% and a selectivity of 48.7% at a current density of 100 mA cm-2, delivering a productivity of 639 μmol cm-2 h-1. Mechanistic studies revealed that the rea…

Manganese-Catalyzed Formate Synthesis via Coupled Methanol Dehydrogenation and Bicarbonate Reduction.

The formate salt represents an indispensable chemical found in a plethora of applications. A base-metal-catalyzed formate synthesis employing the simultaneous dehydrogenation of methanol and reduction of bicarbonate protocol is demonstrated in this paper. The reaction is catalyzed by a pincer Mn complex (Mn-1) bearing a PNHP ligand (PNHP = bis(2-(diphenylphosphino)ethyl)amine). The reaction proceeds via consecutive catalytic steps: a) the dehydrogenation of methanol, b) the reduction of bicarbonate to formate with the H2 generated from methanol dehydrogenation, and c) the formation of formate …

Direct air capture enables sustainable methanol production in water-scarce regions.

Methanol is an essential chemical for the global economy, but its production typically requires substantial water inputs. Here we investigate the techno-economic potential of water-conscious methanol production across more than 20,000 regions worldwide by integrating direct air capture (DAC) with electrolysis, methanol synthesis and a renewable energy supply system. In 97% of these regions, water availability from ambient air is sufficient, and constraints in the remaining regions can be mitigated through adjusted system design and operation. Our results further highlight the importance of mod…

Evolution of alcohol oxidase for improved methanol bioconversion and formaldehyde tolerance.

Methanol oxidation by alcohol oxidases (AOXs) is a key bottleneck in one-carbon (C1) bioconversion due to limited catalytic efficiency and poor formaldehyde tolerance. Here, we report the directed evolution of an alcohol oxidase from Gloeophyllum trabeum, yielding an optimized variant, GtAOXM3. The engineered enzyme exhibits a sixfold increase in catalytic efficiency (7.7 s-1 mM-1), together with enhanced formaldehyde tolerance, thermostability, and high methanol specificity. Molecular dynamics simulations suggest that increased global rigidity and cooperative residue dynamics contribute to th…

Methanol intoxication and acute kidney injury: pathophysiological mechanisms and therapeutic approaches.

Methanol intoxication is a potentially lethal condition that primarily affects the central nervous system, but it can also induce significant renal damage. Acute kidney injury (AKI) in this context is often underestimated, despite being an important prognostic marker associated with increased mortality and morbidity. This review aims to elucidate the pathophysiological mechanisms linking methanol intoxication to AKI, describe the cellular and metabolic pathways involved, and discuss current therapeutic approaches for renal protection and recovery. A descriptive and analytical review was conduc…

Mechanistic Understanding of Dissociated Hydrogen in Cu/CeO2-Catalyzed Methanol Synthesis.

The hydrogen dissociation and spillover mechanism on oxide-supported Cu catalysts play a pivotal role in the hydrogenation of carbon dioxide to methanol. This study investigates the hydrogen spillover mechanism on Cu/CeO2 catalysts using in situ spectral characterization under high-pressure reaction conditions and density functional theory (DFT) simulations. The research confirms that the Cu sites serve as the initial dissociation points for the hydrogen molecules. The chemically adsorbed hydrogen (H*) then spills over onto the CeO2 support and interacts with the lattice oxygen to form special…

Sun-simulated-driven production of high-purity methanol from carbon dioxide.

CO2 conversion to CH3OH under mild conditions is of particular interest yet rather challenging. Both electro- and thermo-catalytic CO2 reduction to CH3OH can only produce CH3OH in low concentration (typically mixed with water), requiring energy-intensive purification processes. Here we design a sun-simulated-driven tandem catalytic system comprising CO2 electroreduction to syngas, and further photothermal conversion into high-purity CH3OH (volume fraction > 97%). We construct a self-supporting electrocatalyst featuring dual active sites of Ni single atoms and encapsulated Co nanoparticles, whi

Engineering Komagataella phaffii to produce lycopene sustainably from glucose or methanol.

Lycopene, a potent carotenoid with high antioxidant capacity and extensive applications, holds significant potential for sustainable production via microbial engineering, particularly with the rising interest in methanol as an ideal non-grain feedstock for a carbon-negative economy. In this study, Komagataella phaffii was systematically engineered to enhance lycopene production using glucose and renewable methanol as alternative carbon sources. Firstly, we demonstrated that the cytoplasmic FPP could penetrate into the peroxisome, and thus achieved the dual-localized lycopene synthesis. Subsequ…
