Interaction of the antiarrhythmic drug amiodarone with the muscarinic receptor in rat heart and brain.

The possible interaction between the muscarinic receptor and the antiarrythmic drug amiodarone was studied physiologically in the guinea pig ileum, as well as by competition binding experiments in rat brain and cardiac tissues, using the highly specific tritiated muscarinic antagonist N-methyl-4-piperidyl benzilate. In these studies, amiodarone was found to affect both antagonist and agonist binding to the muscarinic receptor. The drug's inhibitory effect on the binding of antagonist to cerebral cortex muscarinic receptors was consistent with mutually exclusive binding of the compounds [KI = (

Activation of tyrosine hydroxylase in the superior cervical ganglion by nicotinic and muscarinic agonists.

Both dimethylphenylpiperazinium (DMPP), a nicotinic agonist, and bethanechol, a muscarinic agonist, increase 3,4-dihydroxyphenylalanine (DOPA) synthesis in the superior cervical ganglion of the rat. DMPP causes approximately a fivefold increase in DOPA accumulation in intact ganglia whereas bethanechol causes about a two-fold increase in DOPA accumulation. These effects are additive with each other and with the increase in DOPA accumulation produced by 8-bromo cyclic AMP. The action of DMPP is dependent on extracellular Ca2+ while the actions of bethanechol and 8-bromo cyclic AMP are not depen

Ozone-induced changes in muscarinic bronchial reactivity by different testing methods.

We examined the effect of ozone (O3) on muscarinic bronchial reactivity in the guinea pig and compared reactivity determined by two different routes of agonist delivery. Reactivity before and from 4 h to 2 days after O3 exposure (3.0 ppm, 2 h) was determined by measuring specific airway resistance upon administration of intravenous acetylcholine and/or aerosolized methacholine challenge in 34 unanesthetized, spontaneously breathing animals. Before exposure, we observed more gradual and reproducible results to intravenous agonist. After exposure, hyperreactivity to parenteral agonist occurred c

Effects of clozapine in a selective muscarinic bioassay and on single cells of the rat hippocampus.

Acetylcholine was applied to isolated crayfish hindguts and caused dose-dependent increases in its contractile force. These effects were blocked by muscarinic but not by nicotinic antagonists. Dopamine, noradrenaline, enkephalin or GABA agonists had either no effect or one distinct from the action of acetylcholine. Of a number of antischizophrenic drugs, clozapine had the strongest antimuscarinic action with a pA 2 value of 7.5, followed by thioridazine with a pA 2 value of 6.7. All other drugs of the phenothiazine, butyrophenone or benzamide type were ineffective. From these results, it was c

Effects of trypsin-treatment on agonist binding affinity to the muscarinic acetylcholine receptor.

Trypsin-treatment of the microsome fraction of the ileum and the synaptic membrane fraction of the cerebral cortex of guinea-pig caused selective reduction in the apparent affinity of an agonist (carbachol), but not an antagonist (atropine), to muscarinic acetylcholine receptors (mAChR), measured as inhibition of binding of 3H-quinuclidinyl benzilate (3H-QNB). This effect was similar to that of Gpp(NH)p. The effects of trypsin and Gpp(NH)p were not additive. On the other hand, treatment of these fractions with 5,5'-dithiobis (2-nitrobenzoic acid) (DTNB) increased the apparent affinity of agoni

Molecular regulation of receptors: interaction of beta-estradiol and progesterone with the muscarinic system.

The effects of various substrates on the binding of agonists to muscarinic receptors were studied in the rat hypothalamus and adenohypophysis by competition experiments using the highly specific tritiated muscarinic antagonist N-methyl-4-piperidyl benzilate. It was found that agonist binding properties were affected only by the steroid sex hormones (beta-estradiol and progesterone), both of which resulted in a decrease in the proportion of high-affinity binding sites and a decrease in the dissociation constant. This suggests a link between the muscarinic system and the mechanism by which these

The muscarinic effects of acetylcholine on the action potential of bullfrog sympathetic ganglion cells.

The direct effects of acetylcholine (ACh) on Na+- or Ca2+-dependent action potentials of curarized sympathetic ganglion cells in bullfrogs were investigated under a condition where membrane depolarization caused by the muscarinic action of ACh was nullified by means of a hyperpolarizing current. ACh decreased the after-hyperpolarization of Na+-action potentials in Ringer's solution, and increased the after-depolarization of Ca2+-action potentials in the isotonic Ca2+ solution. In both solutions, the maximum rates of rise of the spikes were decreased and the slope membrane resistance at the ori

Further evidence for nicotinic and muscarinic receptors and their interaction in dog adrenal medulla.

Isolated adrenal glands of dogs were perfused through the adrenolumbar vein with Krebs-Ringer phosphate solution. Nicotine or acetylcholine (Ach) significantly increased the proportion of norepinephrine in the effluent whereas muscarine did not alter the relative proportions of epinephrine and norepinephrine. d-Tubocurarine and hexamethonium (C6) inhibited the response to nicotine completely but scarcely affected the response to Ach and significantly potentiated the response to muscarine. Atropine inhibited the response to muscarine completely, that to Ach partially and that to nicotine slight…

Lessons from the history of antipsychotic discovery: part 3 - muscarinic mechanisms.

After more than seven decades in which antipsychotic drug development has been dominated by dopaminergic mechanisms, the recent emergence of a muscarinic receptor-based treatment represents a conceptual shift in the pharmacotherapy of psychosis. This article reviews the history and development of the first clinically effective non-dopaminergic antipsychotic, tracing its origins to unexpected behavioural effects observed during trials of muscarinic agonists developed for Alzheimer's disease. Early studies of the muscarinic agonist xanomeline demonstrated antipsychotic efficacy, but were limited

Functional insights into autoantibodies targeting muscarinic acetylcholine receptors in Sjögren's disease.

OBJECTIVES: G protein-coupled receptors (GPCRs) participate in various pathophysiological processes in Sjögren's disease (SjD); in particular, autoantibodies against muscarinic acetylcholine receptor 3 (M3R) inhibit the secretion of saliva and tears by exocrine glands. We aimed to identify autoantibodies targeting other muscarinic receptors in SjD and assess their potential functional relevance to signalling pathways. METHODS: Autoantibodies against all muscarinic acetylcholine receptor subtypes were investigated in the serum of patients with SjD (n = 347) using enzyme-linked immunosorbent as

The Relationship between Muscarinic and Cannabinoid Receptors in Neuronal Excitability and Epilepsy: A Review.

BACKGROUND: Of the seventy million people who suffer from epilepsy, 40 percent of them become resistant to more than one antiepileptic medication and have a higher chance of death. While the classical definition of epilepsy was due to the imbalance between excitatory glutamatergic and inhibitory γ-aminobutyric acid (GABA)-ergic signalling, substantial evidence implicates muscarinic receptors in the regulation of neural excitability. SUMMARY: Cannabinoids have shown to reduce seizure activity and neuronal excitability in several epileptic models through the activation of muscarinic receptors w

Adreno-Muscarinic Synergy of Contractile Responses From Human Hyperplastic Prostate.

PURPOSE: Adreno-muscarinic synergy, a supra-additional contractile response to simultaneous application of α-adrenoreceptor and muscarinic receptor agonists, is a feature of several lower urinary tract regions that have dual sympathetic and parasympathetic innervation. We tested the hypothesis that synergy is also a feature of prostate tissue obtained from men with benign prostatic enlargement. METHODS: Isolated tissue strips were dissected from prostate 'chips', collected after transurethral prostate resection procedures for in vitro experiments, to measure isometric tension at 36°C. RESULT

Biased Profile of Xanomeline at the Recombinant Human M4 Muscarinic Acetylcholine Receptor.

Many Food and Drug Administration (FDA)-approved drugs are structural analogues of the endogenous (natural) ligands of G protein-coupled receptors (GPCRs). However, it is becoming appreciated that chemically distinct ligands can bind to GPCRs in conformations that lead to different cellular signaling events, a phenomenon termed biased agonism. Despite this, the rigorous experimentation and analysis required to identify biased agonism are often not undertaken in most clinical candidates and go unrealized. Recently, xanomeline, a muscarinic acetylcholine receptor (mAChR) agonist, has entered pha

Long-acting muscarinic antagonists and small airways in asthma: Which link?

Involvement of small airways, those of <2 mm in internal diameter, is present in all stages of asthma and contributes substantially to its pathophysiologic expression. Therefore, small airways are a potential target to achieve optimal asthma control. Airway tone, which is increased in asthma, is mainly controlled by the vagus nerve that releases acetylcholine (ACh) and activates muscarinic ACh receptors (mAChRs) post-synaptically on airway smooth muscle (ASM). In small airways, M3 mAChRs are expressed, but there is no vagal innervation. Non-neuronal ACh released from the epithelial cells that

Autoantibodies against M5-muscarinic and beta1-adrenergic receptors in periodontitis patients.

Autoantibodies against muscarinic and beta1-adrenergic receptors are considered a potential cause and/or risk factor for chronic heart failure. Association of periodontitis with such autoantibodies and with impaired heart function has been observed in patients exposed to endemic Chagas' disease, which triggers by itself cardiomyopathy and receptor immunization. Here we studied the association between periodontitis, markers of cardiac injury and receptor autoimmunization in periodontitis patients (n = 147) not exposed to Chagas' disease. The autoantibodies were determined by IgG binding to nativ

Effect of Muscarinic Cholinergic Receptor Blockade on Heart Activity in Rat Fetuses.

Cardiac activity parameters were analyzed in rat fetuses with preserved placental circulation on gestation days 17-20 after administration of muscarinic receptor blocker atropine either directly to the fetuses, or to the females. The obtained results attested to the absence of chronotropic effect from the cholinergic system in rats during the fetal period. At the same time, changes in heart rate variability observed after atropine injection showed that cholinergic receptors are involved into heart rhythm regulation in rat fetuses and that their role increases from the 17th to 20th day of the e

Characterization of binding of antipsychotics to muscarinic receptors using mouse cerebral cortex.

Antipsychotics are often the first-line treatment for behavioral and psychological symptoms of dementia. However, the potential anticholinergic effects of antipsychotics could counteract the therapeutic effects of cholinesterase inhibitors used to treat dementia. We investigated the inhibitory effects of 26 antipsychotics on [N-Methyl-3H]scopolamine specific binding in mouse cerebral cortex. At 10-5 M, chlorpromazine, levomepromazine, prochlorperazine, timiperone, zotepine, pimozide, blonanserin, olanzapine, quetiapine, and clozapine inhibited [N-Methyl-3H]scopolamine binding by > 45%. Structure-guided development of selective M3 muscarinic acetylcholine receptor antagonists.

Drugs that treat chronic obstructive pulmonary disease by antagonizing the M3 muscarinic acetylcholine receptor (M3R) have had a significant effect on health, but can suffer from their lack of selectivity against the M2R subtype, which modulates heart rate. Beginning with the crystal structures of M2R and M3R, we exploited a single amino acid difference in their orthosteric binding pockets using molecular docking and structure-based design. The resulting M3R antagonists had up to 100-fold selectivity over M2R in affinity and over 1,000-fold selectivity in vivo. The crystal structure of the M3R
