Nitric oxide synthase in aging rat skeletal muscle.

The neuronal isoform of nitric oxide synthase (NOS) is expressed at high concentrations in skeletal muscle, and NO influences muscle contractility, glucose utilization, and free radical damage or protection. NOS activity and expression was evaluated in extensor digitorum longus (EDL), soleus, and diaphragm of 8 and 24 month old Fisher 344 rats. In 8-month-old animals, NOS activity was highest in EDL, which contained the highest percentage of NOS containing fibers, and was lowest in soleus. NOS activity and percentage of NOS containing fibers was significantly reduced in all muscle groups with

Subclinical skeletal muscle involvement in long-QT syndrome.

Deafness is said to be the only extracardiac manifestation of long-QT syndrome. Whether long-QT syndrome manifests in the skeletal muscle as well, has not been investigated so far. Six affected members of two families with long-QT syndrome without deafness (Romano-Ward syndrome) underwent a clinical neurological examination, nerve conduction studies and needle electromyography. The clinical neurological examination and nerve conduction studies were normal but abundant spontaneous activity (fibrillations and bursts of fibrillations) could be recorded from the right biceps brachii muscle (one pa

The skeletal muscle sodium and chloride channel diseases.

The cause of several familial muscular diseases have recently been linked to mutations within skeletal muscle sodium and chloride channel genes. Thomsen's and Becker's diseases are autosomal dominant and recessive, respectively, and are caused by at least seven different mutations in the CLCN1 (ClC-1) skeletal muscle chloride channel gene on chromosome 7q35. Hyperkalaemic periodic paralysis, paramyotonia congenita and a small heterogeneous group of related 'pure' myotonias are autosomal dominant disorders and are due to at least 16 different mutations in the SCN4A (SkM1) adult skeletal muscle

Skeletal muscle characteristics: relationships to cardiovascular risk factors.

Recent studies have reported that skeletal muscle characteristics are altered in hypertension, insulin resistance, and android obesity. These conditions represent cardiovascular risk factors that are often "clustered" together, and have begun to be recognized as part of a metabolic/cardiovascular syndrome. This paper reviews the evidence correlating skeletal muscle characteristics to cardiovascular risk factors, and outlines the proposed mechanisms for the relationships. Muscle characteristics (e.g., fiber type, capillary density, oxidative capacity, insulin binding, GLUT 4 levels, and glucose

Skeletal muscle weakness in old age: underlying mechanisms.

Maintenance of muscle mass and strength contributes to mobility which impacts on quality of life. Although muscle atrophy, declining strength, and physical frailty are generally accepted as inevitable concomitants of aging, the causes are unknown. Clarification of the mechanisms responsible for these changes would enhance our understanding of the degree to which they are preventable or treatable. The decline in muscle function between maturity and old age is similar for muscles of many different animals including human beings, and is typified by the decreases of approximately 35% in maximum fo

Effects of skeletal change on muscle pattern formation.

The skeletal influence on muscle development was investigated taking an experimental and comparative approach. Chick hindlimbs of HH stage 23 were operated in order to induce the development of an enlarged, elongated fibula - similar to the condition in bird ancestors. The skeletal change resulted in a series of secondary alterations in the later-forming zeugopod muscles. By comparing the results to the zeugopod musculature in other birds and in reptiles it could be shown that changes did not occur at random but resembled the patterns of interspecific variation and ancestral arrangement. It is

[Ultrastructural changes in skeletal muscle in polymyalgia rheumatica].

Skeletal muscle from 21 patients with clinical confirmed polymyalgia rheumatica (PMR) were examined electronmicroscopically. The changes were classified according to 15 ultrastructure criteria. The focal changes have a regressive character and, while they are nonspecific, the systematic assessment of all criteria produced a characteristic pattern in which the ultrastructural picture of skeletal muscles provided a certain profile in cases of PMR. The functionally most important ultrastructural change concerns the mitochondria. Deposition and deformation of crystals result in mitochondrial chang

Polymyositis: reduction of acetylcholine receptors in skeletal muscle.

In polymyositis (PM), neither the precise target nor the mechanism of the attack against skeletal muscle have yet been defined. In this study, we report evidence of a pathologic process involving a membrane component of muscle, acetylcholine receptors (AChRs). Our results show that PM patients have significantly reduced AChRs at neuromuscular junctions, averaging 55% below control values (P less than 0.01). Incubation of cultured mammalian muscle cells with sera from PM patients significantly reduced (P less than 0.005) the number of surface AChRs and increased their rate of degradation in 7/8

Recovery of skeletal muscle after laceration and repair.

Few data are available regarding structural and functional recovery of lacerated skeletal muscle after repair. This study used the extensor digitorum longus muscle of rabbits to document recovery after laceration and repair and a healing period of 12 weeks. Completely lacerated muscles recovered approximately 50% of their ability to produce tension but were able to shorten to 80% of normal. Partially lacerated muscle bellies recovered approximately 60% of their ability to produce tension and normal ability to shorten. Histologic specimens revealed that the lacerations did not cause necrosis of

Eccentric exercise-induced injury to rat skeletal muscle.

These experiments were designed to study skeletal muscle pathology resulting from eccentric-biased exercise in rats. The effects on the muscles of running on a treadmill on a 0 degrees incline (similar amounts of concentric and eccentric contractions), down a 16 degrees incline (primarily eccentric contractions), and up a 16 degrees incline (primarily concentric contractions) at 16 m . min-1 for 90 min were assessed by following postexercise changes in 1) plasma creatine kinase and lactate dehydrogenase activities, 2) glucose-6-phosphate dehydrogenase (G-6-PDase) activity (bio- and histochemic

High skeletal muscle adenylate cyclase in malignant hyperthermia.

Malignant hyperthermia occurs in humans with several congenital myopathies, usually in response to general anesthesia. Commonly, individuals who develop this syndrome lack symptoms of muscle disease, and their muscle lacks specific pathological changes. A biochemical marker for this myopathy has not previously been available; we found activity of adenylate cyclase and content of cyclic AMP to be abnormally high in skeletal muscle. Secondary modification of protein phosphorylation could explain observed abnormalities of phosphorylase activation and sarcoplasmic reticulum function.

Impact of Mitochondrial A3243G Mutation on Skeletal Muscle Energy Metabolism: Evidence from Human Induced Pluripotent Stem Cell-Derived Skeletal Muscle Cells.

The study of skeletal muscle disorders in patients with mitochondrial diseases is crucial for gaining insights into disease physiology; however, their molecular mechanisms have not been fully elucidated. We previously established human-induced pluripotent stem (iPS) cells in two patients with the mitochondrial DNA (mtDNA) A3243G mutation and isolated iPS cell clones with either undetectable or high levels of mutations. In the present study, we established skeletal muscle cells from iPS cells with mutation-high and mutation-undetectable clones and comparatively analyzed their mitochondrial func

Association of low skeletal muscle mass with the presence of advanced colorectal neoplasm: integrative analysis using three skeletal muscle mass indices.

AIM: This study aimed to evaluate an association between colorectal neoplasm (CRN) and skeletal muscle mass using three widely accepted skeletal muscle mass indices (SMIs) in a large population at average risk. METHOD: We performed a cross-sectional study using a screening colonoscopy database of 33 958 asymptomatic subjects aged 40-75 years. Appendicular skeletal muscle mass (ASM) was measured using a bioelectrical impedance analyser. ASM adjusted for height squared (ASM/ht2 ), weight (ASM/wt) and body mass index (ASM/BMI) were used as indices for muscle mass. Logistic regression models were

Relationship among muscle strength, muscle endurance, and skeletal muscle oxygenation dynamics during ramp incremental cycle exercise.

Peak oxygen uptake (VO2), evaluated as exercise tolerance, is a strong predictor of life prognosis regardless of health condition. Several previous studies have reported that peak VO2 is higher in those with a greater decrease in muscle oxygen saturation (SmO2) in the active muscles during incremental exercise. However, the skeletal muscle characteristics of individuals exhibiting a greater decrease in SmO2 during active muscle engagement in incremental exercise remain unclear. This study aimed to clarify the relationship among muscle strength, muscle endurance, and skeletal muscle oxygenation

Overexpression of HSP70 in mouse skeletal muscle protects against muscle damage and age-related muscle dysfunction.

Ageing is associated with skeletal muscle atrophy, a deficit in force generation, an increased susceptibility to contraction-induced injury, and a permanent force deficit following severe injury. Muscles of young mice adapt rapidly following exercise by an increase in the production of heat shock proteins (HSPs), whereas muscles of old mice show a severely diminished response. We hypothesized that overexpression of HSP70 in muscle throughout life would reduce age-related changes in function. The maximum tetanic force of extensor digitorum longus (EDL) muscles of adult and old wild-type (WT) an

STX4 Is Indispensable for Mitochondrial Homeostasis in Skeletal Muscle.

BACKGROUND: Mitochondrial homeostasis is vital for optimal skeletal muscle integrity. Mitochondrial quality control (MQC) mechanisms that are essential for maintaining proper functions of mitochondria include mitochondrial biogenesis, dynamics and mitophagy. Previously, Syntaxin 4 (STX4), traditionally considered a cell surface protein known for glucose uptake in skeletal muscle, was also identified at the outer mitochondrial membrane. STX4 enrichment was sufficient to reverse Type 2 diabetes-associated mitochondrial damage in skeletal muscle by inactivation of mitochondrial fission. However,
