Efficacy of a novel topical fipronil, (S)-methoprene, eprinomectin and praziquantel combination against naturally acquired intestinal nematode and cestode infections in cats.

The efficacy of a novel topical combination formulation of fipronil, (S)-methoprene, eprinomectin and praziquantel against naturally acquired intestinal nematode and cestode infections in cats was evaluated in seven negative control, blinded studies. Cats were selected based on a pre-treatment faecal examination indicating a patent infection with at least hookworms (two studies), Toxocara ascarids (one study), taeniid cestodes (two studies) or Dipylidium cestodes (two studies). In each study, cats were assigned randomly to blocks of two animals each, based on decreasing pre-treatment body weig

Efficacy of Metarhizium spp. against gastrointestinal nematodes in goats and evidence of conidia infection through the integument.

The search for sustainable alternatives to manage gastrointestinal nematodes in small ruminants has intensified due to rising anthelmintic resistance. This study evaluated in vitro the efficacy of several Brazilian isolates of the genus Metarhizium spp. on gastrointestinal nematodes (GINs) of goats which were naturally infected with mixed populations predominantly composed of Haemonchus spp. (63%), Trichostrongylus spp. (35%), and Oesophagostomum spp. (2%). Biological assays were conducted using quantitative coproculture after 10 days of incubation, and SEM was performed after 72 h of fungus-l

Albendazole resistance in naturally infected sheep with gastrointestinal nematodes in two northwest municipalities of Sinaloa, Mexico.

The main aim of this study was to evaluate the anthelmintic resistance to albendazole (ABZ) (from the benzimidazole [BZ] family) in sheep farms located in the northeast of Sinaloa, Mexico. The study was conducted on four sheep farms in the northwest of Sinaloa, Mexico. The faecal egg count reduction test (FECRT), along with endpoint and allele-specific polymerase chain reaction techniques, were used to determine the percentage and proportion of BZ resistance in gastrointestinal nematode (GIN) species before and after ABZ treatment. Genomic DNA (gDNA) from infective larvae (L3) was processed to

Response of the Symbiotic Microbial Community of Dioscorea opposita Cultivar Tiegun to Root-Knot Nematode Infection.

Yam is an important medicinal and edible dual-purpose plant with high economic value. However, nematode damage severely affects its yield and quality. One of the major effects of nematode infestations is the secondary infection of pathogenic bacteria or fungi through entry wounds made by the nematodes. Understanding the response of the symbiotic microbial community of yam plants to nematodes is crucial for controlling such a disease. In this study, we investigated the rhizosphere and how endophytic microbiomes shift after nematode infection during the tuber expansion stage in the Dioscorea opp

Purpureocillium lilacinum for plant growth promotion and biocontrol against root-knot nematodes infecting eggplant.

Purpureocillium lilacinum is a biocontrol Ascomycota fungus against various plant pathogens. In the present study, the efficacy of P. lilacinum was evaluated against a root-knot nematode, Meloidogyne incognita that infects eggplants. We performed an in vitro experiment in which the direct effects of P. lilacinum on the second-stage juvenile survival and egg hatching of M. incognita were tested at different exposure times. The results showed that P. lilacinum significantly reduced the rates of egg hatching and juvenile survival in a dose-dependent manner. Microscopic observation demonstrated th

First report of filarial nematodes in the genus Onchocerca infecting black flies (Diptera: Simuliidae) in Iran.

Black flies are blood-sucking insects of public health importance, and they are effective vectors of pathogens and parasites, such as filarial nematodes of the genus Onchocerca. Our previous surveys have shown that individuals of Simulium turgaicum are annoying pests of humans and livestock in the Aras River Basin of Iran. In the present study, adult black flies of S. turgaicum were trapped from different ecotopes of five villages in Khoda-Afarin County, Iran. By using a sensitive nested PCR assay and targeting the nuclear 18S rDNA-ITS1 marker, filarial infections were found in 38 (1.89%) of 2

Genome-Wide Identification of CircRNAs of Infective Larvae and Adult Worms of Parasitic Nematode, Haemonchus contortus.

CircRNAs, a novel class of ncRNA family, are endogenous transcriptional products involved in various biological and physiological processes in plants and animals. However, almost no information is available for circRNAs of parasitic helminths. In the present study, the circRNAs repertoire was comprehensively explored in Haemonchus contortus, a blood-sucking parasitic nematode of ruminants. In total, 20073 circRNAs were identified and annotated from three key developmental stages/genders of H. contortus including the free-living infective third-stage larvae (L3, 18883), parasitic adult female (

Protective immunity elicited by the nematode-conserved As37 recombinant protein against Ascaris suum infection.

BACKGROUND: Ascaris lumbricoides is one of the three major soil-transmitted gastrointestinal helminths (STHs) that infect more than 440 million people in the world, ranking this neglected tropical disease among the most common afflictions of people living in poverty. Children infected with this roundworm suffer from malnutrition, growth stunting as well as cognitive and intellectual deficits. An effective vaccine is urgently needed to complement anthelmintic deworming as a better approach to control helminth infections. As37 is an immunodominant antigen of Ascaris suum, a pig roundworm closely

Sequential Changes in the Host Gut Microbiota During Infection With the Intestinal Parasitic Nematode Strongyloides venezuelensis.

Soil-transmitted helminths (STHs) are medically important parasites that infect 1. 5 billion humans globally, causing a substantial disease burden. These parasites infect the gastrointestinal tract (GIT) of their host where they co-exist and interact with the host gut bacterial flora, leading to the coevolution of the parasites, microbiota, and host organisms. However, little is known about how these interactions change through time with the progression of infection. Strongyloidiasis is a human parasitic disease caused by the nematode Strongyloides stercoralis infecting 30-100 million people.

A Novel Method for Predicting Anisakid Nematode Infection of Atlantic Cod Using Rough Set Theory.

Atlantic cod ( Gadus morhua L.) is one of the most important fish species in the fisheries industries of many countries; however, these fish are often infected with parasites. The detection of pathogenic larval nematodes is usually performed in fish processing facilities by visual examination using candling or by digesting muscles in artificial digestive juices, but these methods are both time and labor intensive. This article presents an innovative approach to the analysis of cod parasites from both the Atlantic and Baltic Sea areas through the application of rough set theory, one of the meth

Infection levels and diversity of anisakid nematodes in blackspot seabream, Pagellus bogaraveo, from Portuguese waters.

The blackspot seabream, Pagellus bogaraveo, is a sparid fish of great economic importance in the northeast Atlantic. The main aim of this work was to assess the infection levels and diversity of anisakid nematodes parasitizing P. bogaraveo from Portuguese waters. The anisakid larvae were identified by polymerase chain reaction-restriction fragment length polymorphism analysis and ten different patterns were observed, four of which were not previously reported in the literature. Moreover, several species were detected for the first time in this host: Anisakis simplex × Anisakis pegreffii hybrid

Antibody isotype analysis of malaria-nematode co-infection: problems and solutions associated with cross-reactivity.

BACKGROUND: Antibody isotype responses can be useful as indicators of immune bias during infection. In studies of parasite co-infection however, interpretation of immune bias is complicated by the occurrence of cross-reactive antibodies. To confidently attribute shifts in immune bias to the presence of a co-infecting parasite, we suggest practical approaches to account for antibody cross-reactivity. The potential for cross-reactive antibodies to influence disease outcome is also discussed. RESULTS: Utilising two murine models of malaria-helminth co-infection we analysed antibody responses of

Histoplasma capsulatum and Caenorhabditis elegans: a simple nematode model for an innate immune response to fungal infection.

Histoplasma capsulatum is a primary fungal pathogen of mammals responsible for histoplasmosis. During pathogenesis H. capsulatum yeast proliferate in phagosomes of macrophages. This extensive host/pathogen interaction involves a complex cascade of responses in both organisms. In the mammalian host, infection results in complex branched immunity that is initiated with an innate response and later induces an adaptive response but each response is difficult to resolve during fungal infection. Therefore, in an effort to identify less complex systems and to gain understanding of the host innate res

Osteopenia and reduced serum alkaline phosphatase activity in grazing lambs naturally infected with gastrointestinal nematodes.

The effect of gastrointestinal nematode infections on bone development was investigated in growing sheep on pasture. Forty-five weaned lambs from six groups in a two-factorial design incorporating stocking rate (SR; low, medium and high) and presence or absence of infection on pasture were sampled in the late grazing season. Worm counts were performed at slaughter, and the left metacarpal bones were excised for bone assessment. Faecal egg counts and worm burdens, primarily of Ostertagia circumcincta and Trichostrongylus vitrinus, were considerably higher in the high SR infected group ("I-High"…

Intestinal inflammation and activation of sensory nerve pathways: a functional and morphological study in the nematode infected rat.

BACKGROUND: In the rat, gastric distension elicits an intensity dependent pseudoaffective bradycardia mediated via capsaicin sensitive afferent and cholinergic efferent vagal pathways. Inflammation alters visceral perception although the mediators responsible have not been identified. In the nematode infected rat, there is a substantial increase in neuronal substance P (SP) content of the gut. AIMS: To examine the effects of inflammation on perception of a noxious visceral stimulus and on SP and neurokinin 1 (NK-1) receptor immunoreactivity (IR) in visceral afferent pathways. METHODS: Immuno
