[Neuropathology of non conventional infectious agents or prions].

The neuropathological diagnosis of infections by non conventional agents relies on four lesions: astrocytic gliosis (cell hypertrophy and proliferation) usually contrasting with absent mononuclear cell infiltrates (lymphocytes, monocytes-macrophages, and/or microglia) revealed by conventional techniques, and neuronal loss in the most affected areas are little specific findings. Amyloid plaques that are inconstantly found, and spongiosis of gray matter, a characteristic and very frequent finding, are most specific. PrP immunohistochemistry brings additional data. The main diagnostic difficultie

Developmentally moderated expressions of the neuropathology underlying schizophrenia.

A gradually accumulating body of literature suggests that behavioral dysfunction precedes the onset of the schizophrenic syndrome by many ye … aturational events in the central nervous system moderate the behavioral expression of a congenital neuropathology that affects subcortical regions of the brain that are part of multiple neural circuits. Specifically, it is suggested that the diathesis for schizophrenia involves a functional excess of dopamine activity in the basal ganglia that serves to disrupt …

Dementia of frontal lobe type: neuropathology and immunohistochemistry.

Brains from 12 patients dying with a clinical diagnosis of frontal lobe dementia have been examined at post mortem. In pathological terms four groups were encountered. Groups A and B showed severe frontal and temporal lobe atrophy characterised histologically in group A by severe neuronal loss, spongiform change of the superficial laminae, and mild astrocytosis; in group B severe neuronal loss was accompanied by intense gliosis but with little or no spongiform change. Two patients in this latter group also showed inclusions in frontal cortex and hippocampus typical of "Pick bodies"; such patie

Behavioural retardation in the neuropathology of mental retardation.

A series of experiments, involving diverse perinatal treatments of either rats or mice, have been performed in order to investigate the effects of these treatments upon certain selected spontaneous and learned behaviours in the laboratory in order to derive animal models of behavioural retardation. Rat dams were administered either methylazoxymethanol, metallic mercury, organic tin or neuroleptic compounds, and the offspring of these dams were studied on the behavioural tests at adult ages, prenatal studies. Newborn rat pups were administered either 6-OHDA (at various doses), or metallic mercu

[Neuropathology of human type 1 immunodeficiency virus infection].

The nervous system may be affected at any stage in the course of HIV-1 infection. Acute or subacute inflammatory demyelinating polyradiculoneuropathies occur often early and improve spontaneously. Distal symmetrical axonal, predominantly sensory, painful polyneuropathies occur in the late stages of the disease. Microvasculitis is frequent in the early neuropathies. Spastic and ataxic paraparesis associated with vacuolar myelopathy are rare and probably not related only to HIV-1. Aseptic lymphocytic meningitis may occur as the presenting or sole manifestation of HIV-1 infection. The more freque

Aspects on pathology and neuropathology in head injury.

Some principal differences of the head injury materal available to clinicians, hospital pathologists and forensic pathologists are discussed with reference to the primary and secondary findings at focal and diffuse brain injury and intracranial injuries after trauma. The importance of cooperative understanding between the different disciplines is stressed in order to cover the whole head injury pattern. Examples are given. The importance of intracranial arterial hemorrhages is stressed and may also be the cause of a particular group of acute subdural hematomas; it may be of special prognostic

Neuropathology of spiroplasma infection in the rat brain.

This study was designed to demonstrate the neuropathology of persistent spiroplasma infection in the rat brain. GT-48 spiroplasmas were inoculated intracranially into a series of suckling Sprague-Dawley rats. Their brains were evaluated at specific time intervals by microbiologic assay and by morphologic studies including histology, electron microscopy, and immunocytochemistry. The spiroplasmas were observed in the tissues by electron microscopy at peak infection 14 days after intracranial inoculation. At that time they were seen in vacuoles and neuronal processes within the neuropil as filame…

Spontaneous oral-facial dyskinesia: neuropathology of a case.

The neuropathologic findings in a previously reported patient with spontaneous oral-facial dyskinesia are presented. The main microscopic abnormalities were limited to the dorsal halves of the caudate and putamen, which showed a unique pattern of neuronal cell loss and severe gliosis, giving a mosaic appearance. These findings differ from those of other known hereditary or acquired involuntary movement disorders.

Chronic traumatic encephalopathy-neuropathologic change in a routine neuropathology service: 7-year follow-up.

To follow our 2016 study of chronic traumatic encephalopathy neuropathologic change (CTE-NC) in our forensic autopsy service, we prospectively screened all cases with clinical histories of multiple concussions, persistent post-head injury symptoms, or ≥3 hospital investigations for head injuries from 2016 to 2022 inclusive using hyperphosphorylated tau (p-tau) immunostaining. The cases had routine brain sampling plus 4-6 additional lateral hemisphere samples. When "pathognomonic" CTE-NC lesions were identified, additional p-tau immunostaining was done for CTE-NC staging. Of ∼1100 adult brains

Angiotensin II-Stimulating Antihypertensive Medications and Dementia-Related Neuropathology.

IMPORTANCE: Antihypertensive medications that stimulate angiotensin II type 2 or 4 receptors (angiotensin II-stimulating medications) may be associated with lower risk of dementia. OBJECTIVE: To examine associations between cumulative exposure to angiotensin II-stimulating vs angiotensin II-inhibiting antihypertensive medications and neuropathology, accounting for blood pressure. DESIGN, SETTING, AND PARTICIPANTS: This community-based autopsy cohort study from the Adult Changes in Thought cohort was conducted at Kaiser Permanente Washington between February 24, 1994, and November 25, 2022, a…

Human brain matters: Navigating the neuropathology of COVID-19.

Infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused millions of deaths worldwide. Although the incidence of severe acute cases has declined, the prevalence of long COVID, also known as post-acute sequelae of COVID-19 (PASC), is rising. The pathological mechanisms underlying severe COVID-19, along with the relationship to neurological disorders and potential risk for neurodegeneration, remain poorly understood. The aim of this narrative review is to summarize neuropathological features described in postmortem human COVID-19 brains (n = 352). Furthermore, analy

Dopaminergic neuropathology in calves naturally infected with Neospora caninum.

Neospora caninum is a protozoan parasite that causes abortion and congenital neurological disorders in cattle. Although its reproductive effects are well established, the neurochemical alterations associated with congenital infections remain unclear. This study aimed to evaluate the spatial relationship between N. caninum antigen distribution and the expression of dopaminergic markers in the brains of naturally infected neonatal calves. Brain tissues from infected and healthy calves were examined histopathologically and immunohistochemically. Immunohistochemistry was performed using antibodies

Role of GBA variants in Lewy body disease neuropathology.

Rare and common GBA variants are risk factors for both Parkinson's disease (PD) and dementia with Lewy bodies (DLB). However, the degree to which GBA variants are associated with neuropathological features in Lewy body disease (LBD) is unknown. Herein, we assessed 943 LBD cases and examined associations of 15 different neuropathological outcomes with common and rare GBA variants. Neuropathological outcomes included LBD subtype, presence of a high likelihood of clinical DLB (per consensus guidelines), LB counts in five cortical regions, tyrosine hydroxylase immunoreactivity in the dorsolateral

Glucocerebrosidase deficiency leads to neuropathology via cellular immune activation.

Mutations in GBA (glucosylceramidase beta), which encodes the lysosomal enzyme glucocerebrosidase (GCase), are the strongest genetic risk factor for the neurodegenerative disorders Parkinson's disease (PD) and Lewy body dementia. Recent work has suggested that neuroinflammation may be an important factor in the risk conferred by GBA mutations. We therefore systematically tested the contributions of immune-related genes to neuropathology in a Drosophila model of GCase deficiency. We identified target immune factors via RNA-Seq and proteomics on heads from GCase-deficient flies, which revealed b…
