Substrate and electron donor limitation induce phenotypic heterogeneity in different metabolic activities in a green sulphur bacterium.

Populations of genetically identical cells can display marked variation in phenotypic traits; such variation is termed phenotypic heterogeneity. Here, we investigate the effect of substrate and electron donor limitation on phenotypic heterogeneity in N2 and CO2 fixation in the green sulphur bacterium Chlorobium phaeobacteroides. We grew populations in chemostats and batch cultures and used stable isotope labelling combined with nanometer-scale secondary ion mass spectrometry (NanoSIMS) to quantify phenotypic heterogeneity. Experiments in H2 S (i.e. electron donor) limited chemostats show that

Modulation of carbonic anhydrase activity in two nitrogen fixing cyanobacteria, Nostoc calcicola and Anabaena sp.

The activity of enzyme carbonic anhydrase (CA) was investigated in two diazotrophic cyanobacteria, Anabaena sp. (ARM 629) and Nostoc calcicola, in the presence of CO2/NaHCO3 and different inhibitors. The CA activity increased when the cells were pretreated with a high concentration of CO2/NaHCO3 and then transferred to ambient level CO2. Maximum activity of CA was observed after 8 h of incubation in light on transfer of cells from high Ci to ambient level CO2, and was low when incubated in dark. Addition of the photosynthetic inhibitor DCMU brought about a differential reduction in CA activity

Toxicity of chromium and tin to Anabaena doliolum. Interaction with sulphur-containing amino acids and thiols.

Toxicity of chromium and tin on growth, heterocyst differentiation, nitrogenase activity and 14CO2 uptake of Anabaena doliolum and its amelioration by sulphur-containing amino acids and thiols has been studied. The final growth yield was found to be approximately 51% and 58% of control at sublethal concentration of chromium and tin respectively. Among various amino acids tested, cysteine (0.05 mM) significantly restored growth, heterocyst differentiation, nitrogenase and 14CO2 uptake of test alga. Dithiothreitol (1 mM) restored all the parameters and processes better than monothiol, mercaptoet

Mesopelagic N2 Fixation Related to Organic Matter Composition in the Solomon and Bismarck Seas (Southwest Pacific).

Dinitrogen (N2) fixation was investigated together with organic matter composition in the mesopelagic zone of the Bismarck (Transect 1) and Solomon (Transect 2) Seas (Southwest Pacific). Transparent exopolymer particles (TEP) and the presence of compounds sharing molecular formulae with saturated fatty acids and sugars, as well as dissolved organic matter (DOM) compounds containing nitrogen (N) and phosphorus (P) were higher on Transect 1 than on Transect 2, while oxygen concentrations showed an opposite pattern. N2 fixation rates (up to ~1 nmol N L-1 d-1) were higher in Transect 1 than in Tra

Metabolically-versatile Ca. Thiodiazotropha symbionts of the deep-sea lucinid clam Lucinoma kazani have the genetic potential to fix nitrogen.

Lucinid clams are one of the most diverse and widespread symbiont-bearing animal groups in both shallow and deep-sea chemosynthetic habitats … iazotropha. However, we did not detect the expression of these genes and thus the potential role of nitrogen fixation in this symbiosis remains to be determined. We also found the high expression of carbon fixation and sulfur oxidation genes, which indicate chemolithoautotrophy as the key physiology of Ca. Thiodiazotropha. However, we also detected the expression o …

Exsolution-Engineered Perovskite Catalysts for Durable Plasma-Assisted Ammonia Synthesis.

Ammonia is a cornerstone chemical for global food security and an emerging carbon-free energy carrier, yet its industrial synthesis via the Haber-Bosch process remains energy-intensive and carbon-emitting. Non-thermal plasma catalysis offers a promising decentralized alternative for nitrogen fixation under milder conditions, though the long-term structural stability of supported metal catalysts under harsh plasma environments remains a key open challenge. Plasma-assisted NH3 synthesis was investigated using exsolution-derived Co-La2O3/Al2O3 and Ni-La2O3/Al2O3 catalysts in a pulsed (1 kHz at 50…

Niche specialization and prevalence of comammox Nitrospira in the alkaline sediments of thermokarst lakes on the Tibetan Plateau.

Complete ammonia-oxidizing (comammox) Nitrospira are recognized as key players in the nitrogen cycle, yet their ecological roles in extreme environments remain poorly understood. Here, we show that comammox Nitrospira occupy distinct ecological niches in Tibetan Plateau thermokarst lakes (pH 7.5-10.2) using stable isotope probing, metagenomics, and metatranscriptomics. While gross nitrification rates are significantly higher in subalkaline lakes due to the active presence of ammonia-oxidizing archaea (AOA) and bacteria (AOB), these canonical nitrifiers are inhibited under elevated pH. In contr

Basal Plane Passivation of Ti3C2Tx MXenes Through Prussian Blue Analogue Nanoparticles Anchoring for Efficient and Selective Electrochemical Ammonia Synthesis.

Two-dimensional (2D) MXene-based materials have emerged as promising electrocatalysts for aqueous nitrogen fixation. However, their performance is limited by inactive basal planes, weak N2 adsorption, and hydrophilic surface terminations that hinder N2 binding and promote the competing hydrogen evolution reaction (HER). Herein, we report basal-plane activation of Ti3C2T+ MXenes via decoration with Prussian blue analogue (PBA) nanoparticles. The PBA nanoparticles mask hydrophilic surface groups, suppress HER, and enhance N2 adsorption. Strong synergistic interactions between PBA nanoparticles a…

Amorphous Fe2CoOx cocatalysts enable efficient photocatalytic nitrogen reduction reaction: Synergistic nitrogen activation, charge separation and reaction pathway modulation.

Developing high-performance photocatalytic systems for nitrogen fixation is a crucial yet challenging for sustainable ammonia synthesis. Herein, an amorphous Fe2CoOx cocatalyst is successfully loaded onto Bi2MoO6 (BMO) to construct a highly efficient photocatalytic nitrogen reduction reaction (NRR) system. Visible-light-driven ammonia production over the optimized 1.0Fe2CoOx/BMO composite reaches 860.7 μmol g-1 h-1, about 15.4 times that of pristine BMO, outperforming most reported photocatalysts. The results of comprehensive characterizations and density functional theory calculations demonst…

Ammonium-accelerated recovery of rare earth elements from mine wastewater by an indigenous Pseudomonas oryzihabitans isolate.

The mining process of ion-adsorption-type rare earth ores generates a large amount of wastewater containing rare earth elements (REEs) and high concentrations of ammonium nitrogen (NH+4-N). It is urgent to develop single-stage, low-energy technologies to achieve REEs recovery and NH+4-N removal simultaneously. Here, we present a local Pseudomonas oryzihabitan, which is capable of achieving this goal. Interestingly, the typical pollutant ammonium does accelerate the recovery of REEs. Comprehensive characterizations show that the fixation of REEs mainly occurs extracellularly through surface com

Energy filtering transmission electron microscopy immunocytochemistry and antigen retrieval of surface layer proteins from Tannerella forsythensis using microwave or autoclave heating with citraconic anhydride.

Tannerella forsythensis (Bacteroides forsythus), an anaerobic Gram-negative species of bacteria that plays a role in the progression of periodontal disease, has a unique bacterial protein profile. It is characterized by two unique protein bands with molecular weights of more than 200 kDa. It also is known to have a typical surface layer (S-layer) consisting of regularly arrayed subunits outside the outer membrane. We examined the relationship between high molecular weight proteins and the S-layer using electron microscopic immunolabeling with chemical fixation and an antigen retrieval procedur

Hydrogen sulfide and rhizobia synergistically regulate nitrogen (N) assimilation and remobilization during N deficiency-induced senescence in soybean.

Hydrogen sulfide (H2 S) is emerging as an important signalling molecule that regulates plant growth and abiotic stress responses. However, the roles of H2 S in symbiotic nitrogen (N) assimilation and remobilization have not been characterized. Therefore, we examined how H2 S influences the soybean (Glycine max)/rhizobia interaction in terms of symbiotic N fixation and mobilization during N deficiency-induced senescence. H2 S enhanced biomass accumulation and delayed leaf senescence through effects on nodule numbers, leaf chlorophyll contents, leaf N resorption efficiency, and the N contents in

Bioprospecting osmotolerant phyllosphere bacteria: functional characterization and biotechnological application for enhanced nutrient use efficiency in solanum crops.

This study investigated a targeted foliar inoculation strategy using phyllosphere-competent epiphytic bacteria to enhance the agronomic performance of solanaceous crops, specifically tomato and brinjal, while circumventing barriers to rhizosphere establishment. Bacterial isolates from the phyllosphere of tomato and brinjal leaves, collected from two distinct agro-climatic regions, were screened for osmotolerance, nutrient mobilization, nitrogen fixation, and biocontrol traits. This was followed by molecular identification and the assessment of selected strains as foliar inoculants under condit…

A dynamic homodimer structure of the response regulator FixJ from Bradyrhizobium japonicum in its phosphorylated state in aqueous solution.

The symbiotic nitrogen-fixing bacterium Bradyrhizobium japonicum possess a two component regulatory system comprising FixL, a histidine kinase with O2-sensor, and FixJ, a response regulator controlling the expression of nitrogen fixation-related genes. The phosphotransfer from FixL to the N-terminal receiver domain (REC) of FixJ promote the association of the C-terminal DNA-binding domain (DBD) to DNA. To understand the structural basis of the activation, solution NMR approaches were employed to FixJ in the acetyl phosphate-mediated phosphorylated and the BeF3--bound states. The backbone reson…
